Possible identity of lysolecithin acyl-hydrolase with lysolecithin-lysolecithin acyl-transferase in rat-lung soluble fraction

Abstract:Previous studies have shown that rat-lung soluble fraction contains a lysolecithin-lysolecithin acyl-transferase activity in addition to a lysolecithin acylhydrolase (lysophospholipase, EC 3.1.1.4) activity. No separation of these two activities was achieved when the soluble fraction was subjected to hydroxylapatite column chromatography followed by gel filtration on Sephadex G-100 and G-200 columns. Instead, the two activities were copurified about 100-fold with a yield of 30%. The ratio of specific activity of the acyl-hydrolase to that of the acyl-transferase (30:1) remained unchanged throughout the purification procedure. Both activities of the purified preparation were colsely similar to or almost identical with each other with respects to Km values for lysolecithin, pH dependence, stability, etc. It is suggested that the same enzyme is responsible for both the acyl-hydrolase and acyl-transferase activities.
