Abstract:The determination of particle characteristics from light scattering patterns is a challenging inversion problem and, not least, a demanding instrumentation problem. Despite the importance of the knowledge of size distribution in several technological dairy processes, often laser diffractometers and other instrumentation for particle size analysis are not available in dairy laboratories and, therefore, such information is not easily available, except for research purposes. Near infrared (NIR) instrumentation, instead, is largely available in dairy labs. Laser granulometers are based on the principle that particles scatter light from one or two laser beams with an angular pattern directly related to their size. Consequently, a suspension of particles forms an angular pattern of scattered light that is characteristic of its size distribution. In a similar manner, a NIR spectrometer in transmission mode can be considered as a tool for studying the behaviour of forward scattering at different wavelengths. In this work, a model based on an approximation of Mie scattering was developed for the calculation of scattering due to fat globules in the NIR transmission spectrum of milk. The inversion of the model was applied to raw milk spectra in the spectral regions from 1000 nm to 1360 nm and from 1580 nm to 1800 nm, free from strong absorption bands, in order to estimate the fat particle size distribution. More than 700 samples, collected monthly for two years from 50 Friesian–Holstein, 7 Jersey and 5 Brown cows, were analysed. Four hundred of these samples were also analysed using a laser granulometer. The correlation ( r 2 ) between NIR and laser granulometric data was equal to 0.95 for the mean volume surface diameter ( d3,2) with a root mean square error ( RMSE) of 0.11 microns. A sub-class of Weibull distribution with only one freedom parameter proved to be sufficient in order to describe milk fat globule distribution and fit spectral data. The method developed in this work can be useful both for genetic selection and technological purposes and easily extended to the analysis of other dietary fat emulsions.

Abstract:A compact laser scatterometric device for determining the fat percentage of milk filling a cylindrical tube has been designed. The device operates by detecting the angular distribution of the scattered radiation of a semiconductor laser using an axial array of photodiodes. We have experimentally found that the light-scattering indicatrix in cow milk has a monotonous dependence on milk fat content. The intensity at side- or forward-scattering angles normalized to the backscattering intensity proves to be a reliable, informative parameter. A polynomial approximation for the calibration curve of fat percentage versus normalized scattering intensity is constructed to enable fat content measurements in the fairly wide range of ~0.01–10%. Furthermore, the intensity at forward scattering angles responds to the presence of large-scale particles in milk. The device was tested in a laminar flow regime at milk flow rates up to 100 mL/s.

Abstract:Mie scattering theory was shown in this paper to be suitable for analyzing the forward scattered light intensity distribution of micro-sized air bubble defects in glass, shining by a monochrome laser with a wavelength of 532um. The scattered light was measured by a high definition CCD camera. The scattering process can be classified as uncorrelated single scattering according to the properties of optical media. After calculating and smoothing the gray value of split rings of picture, Chahine algorithm was applied to reverse the size of defects. This technique was accurate to within 5% for defects with radii of <50um.

Abstract:Mie scattering theory is used in this paper to analyze the forward scattered light intensity distribution of an air bubble in the subsurface of optical glass, shining by a monochrome laser with a wavelength of 632.8um. The scattering process can be classified as uncorrelated single scattering .according to the properties of optical media. The finite difference time domain (FDTD) software is used to establish a 3-d simulation model to calculate for forward scattered intensity distribution of different sized air bubbles. Moreover, according to the relationship between Mie scattering intensity pattern and the size of bubbles, the size of bubbles are figured out with the help of neural network algorithm. The errors are lower than 10%. The simulation results can improve the precision of defects detection in optical glass.
Determination of Fat and Protein Content of Milk Using He-Ne laser light Scattering

Abstract:The main aim of of this thesis is to build up simple, an accurate and fast method for the determination of fat and protein content in milk. Based on the laser scattering theory, the ratio of the scattered light intensity to the transmitted light intensity, which is called scattered-transmitted method,is taken on as the optical parameter characterizing the fat and protein content. In this way, the influence of the fluctuation of the power of the light source is reduced and the accuracy of determination is enhanced consequently. The method we used can be implemented in diary industries due to its simple methodology and its economic importance because all milk products are sold on basis of fat and protein content.Finally, the data analysis is carried out using curve and surface fitting method. The fat contents of mama and shola homogenized milk types are determined to be 2.9% and 2.7%, respectively. The protein content of mama milk is 3.6% and for shola milk is 3.4%.

Abstract:Rapid and safe detection of contactless liquid components is an important technological development priority for milk and food safety. However, there is few practical methods to detect fat or protein content by non-destructive methods. Herein, we present a simple and rapid method for the determination of fat and protein content in milk products based on the laser scattering light theory that was developed. The intensities of the transmitted light and 90¬į scattered light with respect to transmission direction are collected and processed. The fat contents of 40 milk samples determined by the transmitted light method were less than 10% error. The scattering light method also accurately represented the component of milk. In this paper, the theory and operating principle of the instrument are introduced. Through data analysis and graph illustration, concrete schemes for testing the milk component are given. The method we used is real-time and satisfies online requirements for milk products. The results of this study have great potential in employing one low-cost technology for fresh milk content analysis.

Abstract:The angular intensity distribution of 650 nm laser radiation multiply scattered by bovine milk samples of various fat content, containing large-scale particles with a size of ~20 μm, is studied in cylindrical geometry. It is shown that the forward scattering intensity measured for milk with a fat content in the range of 0–6% decreases monotonically with increasing concentration of somatic cells in the range of 5 × 10 4 –10 6 cm ‒3 . However, the angular dependence of the side- and backscattering intensity, used to determine the fat content in milk, practically does not change when somatic cells are added. A prototype of a milk composition sensor is assembled, based on measuring the scattering indicatrix in the angular range of 18°–162° using an axisymmetric array of photodiodes in combination with a photodetecting CMOS matrix that measures the small-angle forward light scattering intensity (near 0°).

Abstract:Laser light scattering (LLS) including the angular dependence of the absolute integrated scattered intensity (static LLS) and of the line-width distribution (dynamic LLS) has been used to characterize the molecular weight distributions of dextran samples with different branching densities. In the process of converting a translational diffusion coefficient distribution (G(D)) obtained from the precisely measured intensity-intensity time correlation function into a molecular weight distribution (MWD), we encountered the following two problems: the change of the dextran conformation as a function of molecular weight and the lack of a set of narrowly distributed dextran standards. A procedure to solve these two problems simultaneously has been presented, wherein the weight-average molecular weight (M(w)) obtained from static LLS is used to constrain the conversion of G(D) to MWD. By using this procedure, we were able to obtain a calibration of D (cm2/s) = 1.98 X 10(-4)M-[0.657-0.0201 log(M)] with a set of broadly distributed dextrans and to accomplish the calculation of MWD of dextran from the measured spectral distribution. The calculated molecular weight distributions are fairly comparable to the ones obtained from gel filtration experiments.
Laser Light‐scattering Characterization of Gelatin in Formamide

Abstract:Laser light scattering (LLS) including angular dependence of absolute integrated scattered intensity (static LLS) and of the spectral distribution (dynamic LLS) has been used successfully to characterize gelatin in formamide at room temperature. In static LLS, the use of formamide as a single solvent instead of an aqueous salt solution avoids the well-known problem of preferential sorption of salts in the domain of gelatin molecules. Therefore the true weight-average molecular weight M(w), the z-average radius of gyration, and the second virial coefficient have been determined. In dynamic LLS, precise measurements of the intensity-intensity time correlation function permit a Laplace inversion to obtain an estimate of the normalized characteristic linewidth distribution which could be reduced to a translational diffusion coefficient distribution, G(D). This report shows that the calibration between D and M can be established from M(w) and G(D) by using only two broadly distributed gelatins instead of a set of narrowly distributed gelatin standards. After establishing a calibration between D and M, we were able to estimate the molecular weight distribution of gelatin from G(D). (C) 1994 John Wiley & Sons, Inc.

Abstract:The luminescence spectra of milk containing rhodamine 6G are shown to exhibit typical signatures of random lasing when excited with 532 nm laser pulses. Experiments carried out on whole and skim forms of two commercial brands of UHT milk, with fat volume concentrations ranging from 0 to 4%, presented lasing threshold values dependent on the fat concentration, suggesting that a random laser technique can be developed to monitor such important parameter.

Abstract:We demonstrate a first simultaneous measurement of both the refractive index and the attenuation coefficient (defined as the sum of the scattering and absorption coefficients) of highly turbid milk and milk-cream mixtures. We achieve this by observing the real-time reflectance profile of a divergent laser beam made incident on the surface of the milk sample. The experiments were carried out on commercial milk samples with fat volume concentrations of 0.5 or less, 1.6, and 3.3%, and on milk-cream mixtures with fat volume concentrations of 10 and 33.3%, without any dilutions of these samples. We find that the reflectance data are well described, for the first time without any empirical fit-parameters, by Fresnel theory that correctly includes the effect of angle-dependent penetration into the turbid medium on the total internally reflected signal. Therefore, our method provides the most accurate determination to date of the refractive index and attenuation coefficient of milk and milk-cream mixtures. Our sensor is compact, portable, and inexpensive.

Abstract:The applicability of laser polarimetric scatterometry in the analysis of the dispersed composition of water-based multicomponent bioorganic systems has been investigated experimentally using cow milk samples with different fat contents before and after dilution with water. It is shown that the procedure of volume dilution with water makes it possible to determine simultaneously the fat and casein contents in milk on the basis of the angular dependences of light scattering matrix. The empirical dependence of scattering-matrix elements on the fat percentage in milk is revealed. The size distributions of fat and casein particles in the investigated milk samples has been obtained by solving the inverse problem for the measured scattering matrices.

Abstract:Monitoring the composition of milk products is an important factor in the management of dairy farms and industry. Information on the quantitative content of milk components is necessary to control milk quality, as well as to optimize dairy cow nutrition and diagnose their clinical condition. The content of fat and protein is considered the main criterion for determining the market value of milk. Increasing the efficiency of dairy production requires the use of inexpensive and compact devices that are capable of performing multicomponent analysis of milk both directly on the farm and in technological lines. We investigated the possibility of fast simultaneous determination of fat and protein content in milk by laser polarimetric scatterometry. The block-diagonal elements of the scattering matrix were measured for a series of commercially produced milk samples with the indicated fat percentage, which were diluted by volume with water. From the measured scattering matrices, the size distributions of fat droplets and casein aggregates were reconstructed. Using the size histograms, the content of fat and protein and protein-to-fat ratio in the studied milk samples are estimated.
Detecting concentrations of milk components by an iterative optical technique

Abstract:This paper introduces a theoretical and practical model for reconstructing the scattering properties of a participating media. Our theory is based on a robust generalization of the Gerchberg-Saxton (G-S) algorithm. At the end of this algorithm the reduced scattering coefficient μ's of a given substance, can be estimated from the standard deviation (STD) of the retrieved phase of the remitted light. We use the theory to compute the phase's STD that directly correlated to the optical properties for different types of milk components, and we derive a novel appearance model for milk parameterized by the lactose and protein contents. Our results show that we are able to detect the possibility of lactose and milk proteins' quantitative signature by the G-S optical tool, en route to the design of a novel milk-content-monitoring tool. Abstract:Dairy milk composition analysis is critical for quality control and pricing purposes. Particularly, fat content in dairy milk determines its market value, and free fatty acids can be used as a proxy for milk quality. Milk with high fat contents (4% and above) is progressively skimmed to obtain a chain of products (butter, cheese, milk, powder milk, etc.), increasing the profitability in a traditionally low-margin industry. Besides total fat percentage, the composition of fatty acids is also relevant, as C14, C16 and C18 free fatty acids can be used to monitor cattle food intake. Established chemical separation techniques to measure fat content, such as the Gerber method, are off-line, manual, labor intensive, cannot distinguish fats and have a high error margin. We present a monitoring system concept, free of labelling and sample processing, for fat milk content using Raman spectroscopy in flowing microfluidic channels. The technique has been tested using several microfluidic flow rates (20 μl/min to 1000 μl/min) with Raman measurements of 5 seconds of exposure time and with a single accumulation. We show that Raman spectra remain the same even after continuously refreshing the fluid during measurements at increasing flow rates. Proof of concept Raman measurements at the region of interest for undiluted whole (3.5% of fat), semi-skimmed (1.5%) and buttermilk (0.5%) are presented under different microfluidic flows, showing the potential of the technique. The technique can also be used to identify different fats, proteins, and minerals.

Abstract:Development of cost-effective methods and instruments to accurately determine optical parameters of turbid materials can have wide applications in life science research and clinical applications such as blood test and assay of cell suspensions. We report a fast and photodiode based method without integrating spheres to measure three light scattering signals and determine absorption coefficient, scattering coefficient and anisotropy factor of the sample using only one optically thick sample. Results of optical parameters for microsphere suspension samples have been obtained from 460 to 1000nm and validated against the results predicted by Mie theory. Using a GPU executed Monte Carlo code and gradient decent based inverse algorithm, the inverse solutions for determination of optical parameters and their spectra can be achieved in real-time. The uniqueness of the inverse solutions has been proved.

Abstract:Colloidal suspensions made of smart core-shell structures are of current interest in many fields. Their properties come from the possibility of varying the core and shell materials for modifying the composite particles' chemical, biological, and optical properties. These particles are formed with a material with a constant refractive index core and a shell with a refractive index decaying until it matches the solvent refractive index. Poly(N-IsoPropyl AcrylaMide) (PNIPAM) is a typical example of materials forming shells. In this report, we present how to apply Mie scattering theory to predict and understand the static light scattering of large nonhomogeneous colloidal particles with spherical symmetry whose size is comparable with or larger than the light wavelength used for developing scattering experiments, where the Rayleigh-Gans-Debye approximation is not valid. Here, the refractive index decay was approximated by a Gaussian RI profile numerically evaluated through a multilayer sphere. We calculated the form factor functions of suspensions of PNIPAM microgels previously reported and core-shell suspensions made of polystyrene/PNIPAM at 20 and 40 °C synthesized by us. In all the cases, our method succeeded in providing the scattering intensity as a function of the angle. The software for using the numerical method is fairly straightforward and is accessible as an open-source code. The results can not only help predict and understand the photonic properties of microgels with large core-shell structures but also for any particle with a refractive index distribution with spherical symmetry, as in the case of microgels with super chaotropic agents, hollow microgels, or microparticles.
Developing an optical backscatter method for determining casein micelle particle size in heated milk

Abstract:A plethora of different factors, such as heat treatment, pH, soluble calcium and phosphate concentrations, colloidal calcium phosphate, ionic strength, redox potential, etc., affect functionally of critical milk components such as casein micelles, fat globules and whey proteins. These physicochemical changes induce fat- or protein-protein interactions that would be associated to changes in particle size that might be revealed using light backscatter measurements. We hypothesized that inline, simple, low-cost light backscatter measurements might have the potential to provide functionally related information, representing an interesting opportunity for process control. Casein micelle particle size and near infrared light backscatter spectra were measured in milks heat treated at 80 and 90 °C and pH 6.3, 6.7 and 7.1 in order to obtain prediction models for estimating changes in casein micelle particle size during milk heat treatment. Light intensity was measured over a spectral range of 200-1100 nm using a simple optical backscatter sensor and was implemented into models for particle size predictions as a function of heat treatment temperature and pH. Models which included an exponential factor containing a ratio of two specific wavebands were found to improve R2 when compared to single wavelength models. The best model exhibited an R2 of 0.993 and SEP of 2.36 nm. The developed prediction models show promise for in-line monitoring of whey protein denaturation and casein micelle particle size.

Abstract:We present new observational data on small‐angle light scattering properties of natural, random shaped particles, as contrasted with spherical particles. The interest in this “shape effect” on scattering arises from the need for a suitable kernel matrix for use in the laser diffraction method (LD) of particle sizing. LD is now used broadly for measuring size distribution of suspended marine particles. LD involves the measurement of small‐angle forward scattering at multiple angles. This data is inverted using the kernel matrix to produce size distribution. In the absence of a suitable matrix for random shaped particles, past practice has been to use a model based on Mie theory, applicable strictly only to homogeneous spheres. The present work replaces Mie theory with empirical data. The work was motivated in part by anomalous field observations of size distribution and settling velocity distributions reported in literature. We show that a kernel matrix for random shaped particles results in improved interpretation of field multiangle scattering observations. In particular, a rising edge at the fine particle end of the size spectrum is shown to be associated with shape effects.
