Abstract:Objective To investigate the effects of miRNAs-107 (miR-107) on pancreatic cancer proliferation,senescence and invasion.Methods MiR-107 expression levels in 3 pancreatic cancer cell lines PANC-1,ASPC-1,BXPC-3 and normal pancreatic HTERT-HPNE cells were studied by quantitative reverse transcription-polymerase chain reaction (qRT-PCR).PANC-1 cells were transfected with 50 nmol/L anti-miR-107 or negative control using Lipofectamine 2000.After transfection,the miR-107 expression was measured by qRT-PCR.Cell proliferation was tested by methylthiazol tetrazolium (MTT) assay.Cell senescence was detected by β-galactosidase staining.The expression levels of PCNA,P16INK4A and MMP2 were measured by qRT-PCR.Results Compared with the HTERT-HPNE cells,the expression level of miR-107 in 3 pancreatic cancer cell lines was significantly increased (P ＜ 0.01).After transfected with 50 nmol/L anti-miR-107,cell proliferation was inhibited,and cell senescence were increased in PANC-1 cells (P ＜ 0.05),and there was no obvious change in cell invasion.Compared with the HTERT-HPNE cells,after transfected with anti-miR-107,the PCNA expression was significantly decreased and P16INK4A was significantly increased,but expression of M MP2 didn't change significantly.Conclusions These results demonstrate that miR-107 promotes the proliferation and escapes cell senescence in PANC-1 cells by targeting PCNA and P16INK4A.But it has no obvious effects on cell invasion.Therefore,it may be a new target for the biologic therapy for pancreatic cancer.

Abstract:Increasing evidence demonstrates that microRNAs (miRNAs/miRs), a type of non-coding small RNA, can regulate tumor cell migration, invasion and metastasis, and may therefore serve a major function in the occurrence and development of tumors. The present study investigated the effect of miR-383 on the proliferation, migration and invasion of colon cancer HT-29 and LoVo cell lines. The expression of miR-383 in colon cancer and adjacent non-tumor tissues was examined by reverse transcription-quantitative polymerase chain reaction. MiR-383 upregulation was stimulated by transfection with a miR-383 mimic. Cell proliferation was measured with MTT and colony formation assays, and cell migration and invasion potential were examined by Transwell chamber assays. A proliferating-inducing ligand (APRIL), myeloid cell leukemia-1 and cyclooxygenase-2 protein expression was analyzed by western blotting. The expression of miR-383 was decreased in colon cancer tissues compared with adjacent non-tumor tissues (P<0.05). Transfection with a miR-383 mimic suppressed proliferation and inhibited cell migration and invasion in HT-29 and LoVo colon cancer cell lines. Overexpression of miR-383 in HT-29 and LoVo cells resulted in the suppression of APRIL protein expression. In conclusion, miR-383 was downregulated in colon cancer. The upregulation of miR-383 inhibited proliferation, migration and invasion of colon cancer cells, potentially through the regulation of target gene APRIL.

Abstract:Objective To investigate the role of miR-223 on the proliferation,cell cycle and apoptosis of colon cancer cell line SW480.Methods PLL3.7-miR-223 and PLL3.7-miR-EV plasmid were transiently transfected into SW480 cells.The expression of miR-223 was examined using Real-time PCR.The proliferation of cells were detected by using Cell Counting Kit-8 at respective time of 12,24,48 hours after tansfection.The cell cycle and apoptosis of transfected cells were detected by flow cytometry assay.The levels of IGF-1R and AKT protein expression were determined by Western blot.Results 24 hours after transfection,the expression of miR-223 in PLL3.7-miR-223 transfected group(6.55±2.04)was higher than that of PLL3.7-miR-EV group,which was defined as 1(P<0.01).Compared with the control group,the miR-223 group showed a significant inhibition of proliferation,G1 arrest[(61.61±4.02)% vs.(35.99±1.62)%]and an increased rate of apoptosis[(67.43±4.75)% vs.(44.23±3.11)%],all P<0.01.The expression of IGF-1R and AKT of miR-223 group were lower than that of EV group.Conclusions In this study,we found that miR-223 could inhibit the proliferation,promote G1 arrest and apoptosis of colorectal cell line SW480 through down-regulate IGF-1R and AKT.
Mir-139-5p Inhibits the Epithelial-Mesenchymal Transition and Enhances the Chemotherapeutic Sensitivity of Colorectal Cancer Cells by Downregulating BCL2

Abstract:MicroRNAs (miRNAs) are important regulators involved in various cancers, including colorectal cancer (CRC). The functions and mechanisms of the miRNAs involved in CRC progress and metastasis are largely unknown. In this study, miRNA microarray analysis was performed to screen crucial miRNAs involved in CRC progress, and miR-139-5p was chosen for further study. The functional roles of miR-139-5p in colon cancer were demonstrated by CCK-8 proliferation assay, cell invasion and migration, cell apoptosis and in a KO mouse study. miR-139-5p expression was significantly decreased in cancer tissues compared to normal tissues. The miR-139-5p expression level was associated with tumour stage (P < 0.01). Function studies revealed that miR-139-5p was significantly correlated with the metastasis potential and drug resistance of colon cancer cells by affecting the epithelial-mesenchymal transition (EMT). Then, we identified BCL2 as a direct target of miR-139-5p cells in vitro. The patient samples and KO mice model showed that BCL2 expression was inversely correlated with the expression of miR-139-5p. In conclusion, we found that miR-139-5p targeted the BCL2 pathway to reduce tumour metastasis and drug sensitivity in CRC. This axis provided insight into the mechanism underlying miRNA regulation of CRC metastasis and a novel therapeutic target for CRC therapy.
MicroRNA-206 Attenuates Tumor Proliferation and Migration Involving the Downregulation of NOTCH3 in Colorectal Cancer.

Abstract:Colorectal cancer (CRC) is the most common cancer diagnosed worldwide, and the development of metastases is a major cause of mortality. Accumulating evidence suggests that microRNAs are important in carcinogenesis by affecting the expression of genes that regulate cancer progression. A number of studies have shown that miR-206 is frequently downregulated in many human malignancies, including CRC, and is associated with a more malignant phenotype. Previous studies involving HeLa and C2C12 cells have validated the inhibitory mechanism of miR-206 via NOTCH3 targeting. However, whether or not the interplay between miR-206 and NOTCH3 also occurs in CRC is unknown. Therefore, we investigated the tumor suppressive and metastatic effects of miR-206 and its target, NOTCH3, in CRC. Based on the inverse association between the expression of miR-206 and NOTCH3 in CRC tissues, miR-206 mimics were transiently transfected into the SW480 (and its metastatic strain) and SW620 colon cancer cell lines. Upregulation of miR-206 inhibited cancer cell prolife-ration and migration, blocked the cell cycle, and activated apoptosis. The tumor suppressive capacity of miR-206 had a similar effect on CRC cells, although with a different metastatic potential, and may be explained by direct NOTCH3 signaling inhibition and indirect cross-talk with other signaling pathways involving CDH2 and MMP-9. These results support miR-206 as a tumor suppressor in CRC and suggest a potential therapeutic target for clinical intervention.

Abstract:Background and study aims MicroRNA (miRNA) is involved in diverse biological and physiological processes of tumors. Dysregulation of miRNA will induce a series of human diseases. miR-129-2-3p has vital effects in the pathogenesis of various tumors. However, the regulatory function of miR-129-2-3p in colon cancer remains to be clarified. This study investigated the role of miR-129-2-3p targeting BZW1 in proliferation, apoptosis, migration, and invasion of colon cancer. Patients and methods Here, RT-qPCR was applied to measure the miR-129-2-3p levels in colon cancer tissues. The predicted targets of miR-129-2-3p were identified by bioinformatics and verified using luciferase reporter assay. The effects of miR-129-2-3p on colon cancer were detected by CCK-8, colony formation, transwell chamber test, wound healing, and flow cytometry assays. Finally, the influence of miR-129-2-3p on tumor growth was studied. Nude mice were xenografted with transfected Lovo cells by subcutaneous injection of 5 × 10 5 cells in 100 μl. HE staining and TUNEL were used to assess metastasis ability. Results miR-129-2-3p level in colon cancer tissue was significantly reduced. Furthermore, it was verified that BZW1 was a target of miR-129-2-3p, and its expression in colon cancer cells was inhibited by miR-129-2-3p. Additionally, miR-129-2-3p inhibited colon cancer cell proliferation, colony formation, mobility ability and tumor growth, and promoted cell apoptosis by targeting BZW1. miR-129-2-3p overexpression in tumor xenografts in vivo decreased BZW1 expression, and suppressed tumor growth. Conclusion Collectively, these findings indicated that miR-129-2-3p exerts a suppressive role in colon cancer cells by directly targeting BZW1, and may have significant therapeutic implications for patients with colon cancer.

[Effect of Microrna143 Expression on Cell Proliferation in Colonic Carcinoma].

Abstract:OBJECTIVE:To investigate the effect of microRNA143 on cell proliferation and K-ras expression in colorectal carcinoma.METHODS:Northern blot was used to examine the expression of miR-143 in colorectal carcinoma and adjacent normal tissues. A miR-143 expression vector was constructed and transfected into a human colon adenocarcinoma cell line SW480. Cell proliferation was evaluated by MTT assay. RT-PCR and Western blot were used to examine the expression of K-ras oncogene in transfected cells.RESULTS:The level of mature miR-143 was lower in tumors compared with adjacent normal tissues in 81% of colorectal carcinoma specimens. In transfected cells, the increased accumulation of miR-143 inhibited the cell proliferation, and resulted in approximately 40.3% decrease of K-ras protein levels, but had no effect on level of K-ras mRNA.CONCLUSION:The increased accumulation of miR-143 inhibits the proliferation of transfected cells, and results in down-regulation of K-ras protein in colorectal carcinoma.

Abstract:To investigate the function of over-expression of miR-490 and miR-363 as well as the mechanism of effecting colon cancer,we collected colon cancer cell line SW620,HCT116,normal colon epithelial cells,and used the transient transfection to express miR-490 and miR-363.Then we studied the function of miR-490 and miR-363 in colon cancer cells through the analysis of cell apoptosis,cell cycle and the survival rate,etc.Western blotting was used to detect the protein expression of WEE 1 and Cdc2.We obtained the results that the transfection of miR-490 and miR-363 could inhibit the proliferation of colon cancer SW620 cell line,promote its apoptosis and down regulate the ratio ofG2/M;MiR-490 and miR-363 shared the work in coordination,which could inhibit WEE1 protein kinase and thus regulate Cdc2.The results showed that miR-490 and miR-363 could inhibit the function of colon cancer cell line SW620.

Abstract:Previous research has showed that miR-99a-5p was a tumor suppressor. The aim of our study was to explore the effect of miR-99a-5p on the vitality and proliferation, migration together with the invasion of oral tumor cells via inhibiting the expression of NOX4. QRT-PCR and Western blot were applied to examine the expression level of miR-99a-5p and NOX4 in human oral tumorous and adjacent tissues. Dual luciferase reporter gene assay was applied to confirm that miR-99a-5p negatively regulated directly on NOX4 in TSCC1 cells. Cell transfection and lentiviral vectors were used to up-regulate expression of miR-99a-5p and NOX4, respectively. Cell proliferation, cell cycle, apoptosis and invasion along with the migration in different groups were assessed using MTT assay, colony formation assay, the flow cytometry, transwell assay and the wound healing assay, respectively. MiR-99a-5p was under-expressed in human oral tumor, while NOX4 was over-expressed. There was a negative relationship between miR-99a-5p and NOX4. Up-regulating miR-99a-5p or down-regulating NOX4 suppressed the vitality, proliferation, migration together with invasion of TSCC1 cells. MiR-99a-5p affected the vitality and proliferation, migration together with the invasion of oral tumor cells through targeting NOX4.
Effects of miR-26 on Proliferation and Invasion of Colon Cancer Cells by BID Protein

Abstract:Objective To investigate the role of miR-26 in the rcgulation of BID protein and its effect on proliferation and invasion of colon cancer cells.Methods The expression of miR-26 in colon cancer tissues and colon cancer cells was detected by qPCR.The relationship between miR-26 and clinicopathological staging of colorectal cancer was examined.Immunofluorescence was used to detect the transfection efficiency of miR-26-inhibitor in colon cancer cells.MTT assay was used to detect the effect of miR-26 on the proliferation of colon cancer cells.Transwell invasion assay was used to detect the effect of miR-26 on the invasion of colon cancer cells.Results The expression of miR-26 in colon cancer tissues was higher than in normal tissues.The expression of miR-26 was significantly increased with the pathological staging of colon cancer.Silencing miR-26 could increase the expression of BID but inhibit the proliferation and invasive ability of colon cancer cells.Conclusion MiR-26 is up-regulated in colon cancer,andcan regulate the expression of BID and effect the proliferation and invasion of colon cancer cells.
In vitro study of miRNA-369-3p targeting TCF4 regulating the malignant biological behavior of colon cancer cells

Abstract:Background: Colorectal carcinoma (CRC) is a common malignant tumor of the digestive tract. It is characterized by a high degree of malignancy, early metastasis and poor prognosis. Studies have shown the effect of miR-369-3p on the biological function of a variety of tumors. However, the mechanism by which miR-369-3p and its potential target genes participate in the pathogenesis of CRC has not been elucidated. This study aims to study the relationship between miR-369-3p and transcription factor 4 (TCF4), to reveal the mechanism of the occurrence and development of CRC, and to provide a promising target for the treatment of CRC. Methods: Real-time quantitative polymerase chain reaction (RT-qPCR) was used to detect the miR-369-3p levels in CRC tissues and cells. miR-369-3p mimics and/or TCF4 overexpression vectors were transfected into SW480 cells. The expression of miR-369-3p and TCF4 mRNA was detected using RT-qPCR. Bioinformatics analysis predicted the binding site of miR-369-3p to the TCF4 3'UTR, and the targeting relationship was verified by a dual luciferase reporter gene assay. Cell proliferation and invasion were investigated by labeled immunofluorescence assay using BrdU antibody and Transwell assay. The oxidative stress ability of cells was determined by commercial kits. The levels of proteins related to cell proliferation and invasion were measured by western blotting. Results: The level of miR-369-3p was significantly down-regulated in CRC tissues and cell lines, especially in SW480 cells (P<0.05). The expression of TCF4 was negatively correlated with that of miR-369-3p. High levels of miR-369-3p targeting TCF4 suppressed cell proliferation and downregulated the protein expression of Ki67 and PCNA (P<0.05). Overexpressed miR-369-3p binding TCF4 inhibited cell invasion and decreased the protein levels of vascular endothelial growth factor (VEGF) and E-cadherin (P<0.05). Furthermore, upregulation of miR-369-3p increased the activity of superoxide dismutase (SOD) while decreasing the content of malondialdehyde (MDA) and activity of lactate dehydrogenase (LDH) by blocking the expression of TCF4 (P<0.05). Conclusions: MiR-369-3p inhibits the proliferation, invasion and oxidative stress of CRC cells by targeting TCF4, thus defining miR-369-3p as a potential target for the diagnosis and treatment of CRC.
MicroRNA-204 Inhibits Expression of B Cell Lymphoma-2 in Non-Small Ceil Lung Cancer and Regulate Cell Biological Behaviors

Abstract:Objective To investigate the expression of microRNA-204 (miR-204) in non-small cell lung cancer (NSCLC) tissues and the molecular mechanism of targeting B cell lymphoma-2 (bcl-2) in the regulation of cell proliferation,apoptosis and invasion in A549 cell line.Methods The expression of miR-204 was detected by real-time polymerase chain reaction in 45 NSCLC tissues.Cell counting kit-8 (CCK-8) assay,Transwell assay and cysteinyl aspartate-specific protease (Caspase)-3 assay were performed to access cell line proliferation,invasion and apoptosis ability of A.549 cell line.The target gene of miR-204 was analyzed by bioinformatics combined with Dual-luciferase assay and function recovery assays.Results miR-204 expression level was down-regulated in NSCLC tissues compared with adjacent normal tissues (1.28 ± 0.31 and 3.75 ± 0.27,P =0.000) while the expression of bcl-2 was up-regulated in NSCLC tissues (3.46 ±0.40 and 0.78 ±0.48,P =0.000).Furthermore,the expression levels were correlated with the pathologic grades.MiR-204 can significantly decrease the proliferation and invasion ability,as well as increase apoptosis (P =0.000).Furthermore,a significant reduction of luciferase activity was detected in A549 cell after co-transfected with miR-204 and Dual-luciferase expression vector containing bcl-2 wide-type 3'-untranslated region when compared with muted-type region group (0.12 ±0.02 and 0.29 ±0.03,P =0.000).The biological behavior effect on A549 cell can be recovered through over express the level of bcl-2.Conclusion miR-204 is down-regulated in NSCLC,and regulate the proliferation,invasion and apoptosis of A549 cells via targeting bcl-2.
Expression and Effect of Mir-769-5p on Proliferation, Migration and Invasion in Non-Small Cell Lung Cancer

Abstract:Objective To explore the expression of miRNA-769-5p (miR-769-5p) in non-small cell lung cancer(NSCLC) and its effect on proliferation,migration and invasion of NSCLC cells.Methods The miR-769-5p expression in NSCLC cell lines (A549,NCI-H1299,NCI-H157,ANIP-973 and GLC-82) was detected by QRT-PCR,and the cell lines of highest and lowest expression of miR-769-5p were applied for the further study.NSCLC cell lines were divided into transfection group and control group,and respectively transfected with miR-769-5p mimics/inhibitor and miRNA mimics control/inhibitor control.Furthermore,cell proliferation,colony formation migration and invasion were respectively analyzed by MST,colony formation,and transwell chamber experiments.Results The expression of miR-769-5p in A549,NCI-H1299,NCI-H157,ANIP-973 and GLC-82 were 0.06,0.40,0.09,1.04 and 2.31.miR-769-5p mimics and miR-769-5p inhibitor were respectively transfected into A549 and GLC-82 cell lines.MST test showed that transfection with miR-769-5p mimics in A549 cells for 48,72,96 h significantly inhibited the cell viability (P＜0.05).The colony formation (124.7±14.7 vs.399.4±46.0,P＜0.05),migration (94.4±18.1 vs.157.8±24.8,P＜0.05) and invasion (84.4±15.1 vs.135.6±16.7,P＜0.05) ability of A549 cell were also suppressed compared with control group after transfection.Down-regulation of miR-769-5p by transfected with miR-769-5p inhibitor could enhance the malignant phenotype of GLC-82 cells in vitro,including cell growth (P＜0.05),colony formation (555.7±29.7 vs.366.3±28.7,P＜0.05),as well as cell migration (226.7±40.3 vs.153.3± 38.7,P＜0.05) and invasion (196.7±39.1 vs.138.9±40.4,P＜0.05).Conclusion The expression of miR-769-5p can inhibit the proliferation,migration and invasion of NSCLC cells.

Abstract:Background: MicroRNAs have been discovered to have the possibility to play a significant role in cancer development. It has been found that miR-141-5p is upregulated in various cancers. However, the functions of miR-141-5p in cervical cancer have rarely been reported. Methods: The expression level of miR-141-5p was assessed in cervical cancer tissues and cell lines by RT-qPCR. The function of miR-141-5p in C33A and HeLa cells was detected by CCK-8, and colony formation, wound-healing, transwell chamber, and flow cytometry assays. Dual luciferase reporter was carried out to identify the interaction between miR-141-5p and BTG antiproliferation factor 1 (BTG1). Results: miR-141-5p was upregulated in cervical cancer and was negatively associated with the prognosis of patients with cervical cancer. Functional analyses demonstrated that silenced miR-141-5p expression inhibited the cell proliferation, migration, and invasion, and alleviated apoptosis of C33A and HeLa cells. In addition, miR-141-5p suppresses the activity of BTG1-3 '-UTR. Rescue assays demonstrated that the cervical cancer progression is suppressed by miR-141-5p inhibitor and retrieved by sh-BTG1. Conclusions: The authors' findings reveal that miR-141-5p exerts its role through targeting BTG1 in cervical cancer progression, indicating that miR-141-5p may represent a promising target for the treatment of cervical cancer patients. Mir-449B Inhibits the Proliferation of SW1116 Colon Cancer Stem Cells Through Downregulation of CCND1 and E2F3 Expression

Abstract:Colorectal cancer is one of the leading causes of cancer-related mortality worldwide. Cancer stem cells are cell populations with stem cell nature presenting in tumor tissues and are the root of tumor formation and metastasis. CCND1 and E2F3 play important roles in cell cycle regulation. The 3'UTRs of CCND1 and E2F3 contain miR-449 binding sites. By transfecting pre-miR-449b and inhibiting miR-449b, we found that cell cycle, cell proliferation ability and cell cycle regulatory protein expression levels of colon cancer stem cells were altered. The correlation between CCND1, E2F3 and miR-449b showed that miR-449b could downregulate CCND1 and E2F3 expression. This, in turn, reduced the proliferative ability of colon cancer stem cells. These data suggest that miR-449b plays a tumor-suppressive role in colon cancer stem cells.
miR‐140‐3p inhibits colorectal cancer progression and its liver metastasis by targeting BCL9 and BCL2

Abstract:<p>Recent studies have identified microRNAs (miRNAs) as a compelling novel class of biomarker in colorectal cancer (CRC) development and metastasis. Here, we demonstrated that the level of plasma exosomal miR‐140‐3p in CRC patients was lower than that in healthy controls. The decreased miR‐140‐3p level was also observed in CRC patients with liver metastasis. The expression of miR‐140‐3p in CRC tissues were significantly lower than that in matched normal tissues. Functionally, miR‐140‐3p overexpression suppressed proliferation, migration, invasion, and β‐catenin nuclear translocation, as well as promoted apoptosis in LoVo cells, while inhibition of miR‐140‐3p reversed these cellular processes in HCT 116 cells. Notably, BCL9 and BCL2 were recognized as direct targets of miR‐140‐3p. BCL9 knockdown abrogated miR‐140‐3p inhibitor‐induced effects on HCT 116 cells with decreased proliferation, migration, and invasion. BCL2 knockdown increased apoptosis of miR‐140‐3p inhibitor‐transfected HCT 116 cells. In vivo experiments revealed that miR‐140‐3p overexpression inhibited tumor growth in LoVo xenograft model and diminished metastatic nodules in nude mice liver. Taken together, this work supports that miR‐140‐3p exerts as a tumor suppressor in CRC progression via targeting BCL9 and BCL2, and suggests miR‐140‐3p‐BCL9/BCL2 axis may be applied in miRNA‐based therapy and prognostication of CRC.</p>

Abstract:Abstract Background It has been shown that miR-144-3p regulates cell proliferation, apoptosis, migration and invasion in various cancers. However, the function and expression of miR-144-3p in colorectal cancer (CRC) remained obscure. Methods Immunohistochemical (IHC) staining was performed to investigate the protein expression of BCL6 in CRC tissues. The effect of BCL6 and miR-144-3p on CRC cells was explored through methylthiazolyl tetrazolium (MTT) assay, colony formation and cell cycle assays. Luciferase reporter assays, reverse transcription quantitative polymerase chain reaction (RT-qPCR) and Western blot assay were carried out to determine that BCL6 is directly regulated by miR-144-3p. Results Our results showed that miR-144-3p is down-regulated in CRC and correlates with the tumor progression of CRC patients. miR-144-3p inhibits cell proliferation and delays G1/S phase transition of CRC cells. Moreover, we found that BCL6 is a new target of miR-144-3p. Furthermore, BCL6 is a mediator of miR-144-3p repression of cell proliferation and cell cycle arrest in CRC cells. miR-144-3p repression of Wnt/β-catenin signaling is mediated by BCL6 in CRC cells. Conclusions Overall, the effect of the miR-144-3p/BCL6 axis on regulating CRC carcinogenesis was demonstrated, and miR-144-3p was identified as a potential prognostic and therapeutic target in colorectal cancer.

Abstract:MicroRNAs are small non-coding RNAs that may also function as oncogenes and tumor-suppressor genes, as the abnormal expression of microRNAs is associated with various human tumors. However, the effect of miR-335 on the lung cancer cells remains unclear. The aim of the paper was to study the expression of miR335 in non-small cell lung cancer (NSCLC) and miR335’s relation to the metastasis, invasion, and apoptosis in lung cancer cells A549 and H1299. qRT-PCR was used to identify the miR-335 expression. The effects of miR-335 on cell proliferation, apoptosis, and invasion were further analyzed. Luciferase reporter assay and Western blot were to verify Bcl-w and SP1 as potential major target genes of miR-335. Finally, the effect of Bcl-w on miR-335-induced cell survival was determined. Our results showed that miR-335 expression was significantly lower in NSCLC tissue, which was significantly associated with lymph node metastasis. In contrast to cells in blank and negative control groups, incidence of apoptosis was significantly higher (P < 0.05) and the number of cells migrating through matrigel was significantly lower (P < 0.05) in miR-335 mimics transfected group. Western blot and luciferase reporter assay demonstrated that miR-335 could bind to the putative binding sites in Bcl-w (or SP1) mRNA 3′-untranslated region to visibly lower the expression of Bcl-w (or SP1). The introduction of Bcl-w cDNA without 3′-untranslated region abrogated miR-335-induced cell survival. These results indicated that upregulation of miR-335 can simultaneously suppress the invasiveness and promote apoptosis of lung cancer cell A549 and H1299 by targeting Bcl-w and SP1. Therefore, miR-335 may be a potential therapeutic target in NSCLC treatment.
The Studies on Effect of Survivin siRNA for Apoptosis,Proliferation and Invasion of Colon Carcinoma Cell Line

Abstract:Objective: To study the efficiency of silencing survivin gene expression level of survivin small RNA expression plasmid and its impact on the apoptosis, proliferation and invasive ability of colorectal cancer cell line SW480. Methods: The expression plasmid of survivin siRNA (pRNAT/sur-siRNA) was constructed and transduced into SW480 cell line, was measured by the Western blot analysis and semi-quantitative reverse transcription polymerase chain reaction (RT-PCR) were used to measure the change in expression level of the survivin mRNA and protein, after transduction of the expression plasmid into the SW480. The apoptosis, proliferation, and invasive ability of SW480 cell line was evaluated by flow cytometry, MTT assay and Boyden Chamber Assay, respectively. Results: The expression plasmid (pRNAT/sur-siRNA) against survivin siRNA downregulated the survivin expression level of protein and mRNA dramatically in SW480 cell line, with a down-regulation of 85% and 80%, respectively. After transfection of the pRNAT/sur-siRNA in SW480, the apoptosis rate of pRNAT/sur-siRNA/SW480 was 16.9%, proliferation of pRNAT/sur-siRNA/SW480 cell was 37.4% and there was a remarkable difference comparing with the controls (P < 0.01). Cell invasive experiment (Boyden Chamber Assay) showed that cell penetrating membrane number of the pRNAT/sur-siRNA/SW480, pRNATV SW480 and the SW480 cell was 153 ± 66, 505 ± 65 and 578 ± 98, respectively (P< 0.01). Conclusion: The siRNA against survivin can significantly inhibit the expression level of the survivin, thus promote apoptosis of SW480 cell and inhibit proliferation and invasive ability of the SW480 cell.
miR-22 suppresses cell proliferation, invasion and migration invasion in human colon cancer cell based on targeting NLRP3

Abstract:Abstract Background To investigate the effect of the cell proliferation, migration and invasion in human colon cancer cells which based on the mechanism of miR-22 targeting NLR family pyrin domain containing3(NLRP3) Methods Human colonic epithelial cells (HCoEpiC) and human colon cancer cell lines (HCT116, HCT8, HT29, LS174T, LOVO, SW480) were prepared for the present study. The HCT116 cells were divided into 2 parts with different processes, and each part was randomly divided into different groups (part1: BC, NC1, miR-22, NC2, si-miR and part2: BC, miR-22, NC3, si-NLRP3, miR + NC4, miR + NLRP3). Quantification the expression levels of miR-22, NLPR3 and miR-22 targeting NLRP3 related proteins by means of qRT-PCR and Western blot. The colon cancer cell proliferation, invasion and migration were assessed via CCK8, colony formation and transwell assays. Subsequently, nude xenografts mice model was constructed and mice were divided into 4 groups: Model, miR-22, si-NLRP3 and miR + NLRP3. Through the changes of tumor volume and weight, the expression of Ki67 and the protein levels of NLRP3, IL-1B, MMP-9, MMP-2, E-catherine, N-catherine and Vimentin to verify the results. Results The cell proliferation, migration and invasion in miR-22, si-NLRP3 and miR + NLRP3 groups were significantly weaker than BC group. In compared to normal tissues, the growth rate of tumor was evidently slowed down, the volume and quality of tumor were evidently reduced and the proportion of Ki67 positive cells were evidently decreased. Both in tumor tissues and in cancer cells, the expression of IL-1B, NLRP3, MMP-9, MMP-2, N-Catherine and Vimentin were evidently decrease and the levels of E-Catherine were evidently increased than in normal tissues or cells. In compared to si-miR and CYLD groups, treatment with si-miR + si-C could effectively reverse the effect of suppressed the tumor progression.
