Abstract:Good sense: A highly sensitive and selective ratiometric fluorescent probe for Fe2+ was developed by linking an Fe2+-insensitive fluorophore to an Fe2+-sensitive reporter–fluorophore fragment. The fluorescence intensity ratio increased linearly with the concentration of Fe2+ (see scheme). The probe was applied to the ratiometric imaging of intracellular and adscititious fluctuations of Fe2+ in live HL-7702 and HepG2 cells.

Abstract:Cancer cells require large amounts of iron to maintain their proliferation. Iron metabolism is considered a hallmark of cancer, making iron a valid target for anti-cancer approaches. The development of novel compounds and the identification of leads for further modification requires that proof of mechanism assays be carried out. There are many assays to evaluate the impact on proliferation; however, the ability to chelate iron is an important and sometimes overlooked end-point measure due to the high costs of equipment and the challenge to quickly and reproducibly quantify the strength of chelation. Here, we describe a quantifiable and inexpensive cell-free fluorescent method to confirm the ability of novel compounds to chelate iron. Our assay relies on the commercially available inexpensive fluorescent dye Calcein, whose fluorescence can be quantified on most fluorescent microtiter plate readers. Calcein is a weak iron chelator, and its fluorescence is quenched when it binds Fe2+/3+; fluorescence is restored when a novel chelator outcompetes Calcein for bound Fe2+/3+. The removal of fluorescent quenching and the resulting increase in fluorescence allows the chelation ability of a novel putative chelator to be determined. Therefore, we offer an inexpensive, high-throughput assay that allows the rapid screening of novel candidate chelator compounds.

Thirty Femtograms Detection of Iron in Mammalian Cells

Abstract:Inorganic nanomaterials and particles with enhanced optical, mechanical or magnetic attributes are currently being developed for a wide range of applications. Safety issues have been formulated however concerning their potential cyto- and genotoxicity. For in vivo and in vitro experimentations, recent developments have heightened the need of simple and facile methods to measure the amount of nanoparticles taken up by cells or tissues. In this work, we present a rapid and highly sensitive method for quantifying the uptake of iron oxide nanoparticles in mammalian cells. Our approach exploits the digestion of incubated cells with concentrated hydrochloric acid reactant and a colorimetric based UV-Visible absorption technique. The technique allows the detection of iron in cells over 4 decades in masses, from 0.03 to 300 picograms per cell. Applied on particles of different surface chemistry and sizes, the protocol demonstrates that the coating is the key parameter in the nanoparticle/cell interactions. The data are corroborated by scanning and transmission electron microscopy and stress the importance of resiliently adsorbed nanoparticles at the plasma membrane.

Abstract:Iron plays an important role in various physiological processes. However, the detailed biological functions of iron have not been sufficiently explored because of a lack of effective methods to monitoring iron, especially the labile ferrous ion (Fe2+). In the current study, a novel turn-on phosphorescent probe for Fe2+ quantification and visualization has been proposed based on the hybrid nanocomposite of manganese dioxide and gemini iridium complex (MnO2-GM-Ir). The surfactant-like GM-Ir with positive charges was beneficial to combine with the negatively charged manganese dioxide (MnO2) nanosheets, and thus endowing the MnO2-GM-Ir nanocomposite excellent dispersion ability in the water as well as efficiently avoiding the interference to the detection caused by the agglomeration of nanocomposite. Phosphorescence of GM-Ir was effectively quenched by MnO2 nanosheets through fluorescence resonance energy transfer (FRET) and the inner filter effect (IFE), while the phosphorescence could be significantly recovered in the presence of Fe2+ via a selective Fe2+-mediated reduction of MnO2 nanosheets, indicating a highly-specific selectivity towards Fe2+ with a low detection limit (80 nM). The drug test assay and in vitro imaging studies further proved that the MnO2-GM-Ir nanocomposite could be employed as a promising probe for the quantitative detection of exogenous Fe2+ in drug and in vitro imaging of living cells.

Abstract:Iron is one of the essential trace elements in the human body, which is involved in many important physiological processes of life. The abnormal amount of iron in the body will bring many diseases. Therefore, a novel near-infrared fluorimetric method was developed. The method is based on a fluorescent probe (E)-4-(2-(3-(dicyanomethylene)-5,5-dimethylcyclohex-1-en-1-yl)vinyl)-N, N-diethylaniline oxide (DDED) which uses N-oxide as a recognition group to real-time monitoring and imaging of Fe2+ in vivo and in vitro. The method exhibits excellent selectivity and high sensitivity (LOD = 27 nM) for Fe2+, fast reaction rate (< 4 min), extremely large Stokes shift (> 275 nm), low cytotoxicity. The strip test strongly illustrates the potential application of DDED in real environment. In particular, DDED has been successfully applied to real-time monitoring and imaging of Fe2+ in HepG2 cells and zebrafish. That is, the method has great potential for the detection of Fe2+ in living systems.

Abstract:Abstract A simple, sensitive, automated and environmentally friendly FI/MZ technique was suggested for the indirect determination of iron (III) as ferric chloride in bulk and biological. These method involve the reduction of Fe (III) to Fe (II) using ascorbic acid as reducing agent, the reduced ion was reacted with 1,10- phenanthroline as a selective organic reagent to form red-orange complex, measured at λ=510 nm. The complexation ratio between matel ion with ligand was 1:3 ratio (M: L). The colored product obeyed Beer’s Law with linear ranges 1-30 and 8-75 μg/mL with detection limits 1.02, 0.28 μg/mL for batch and FIA/MZ methods, respectively. The rate of sampling was 54 sample/h, the %RSD less than 2% and the recovery was about 99%. The developed technique was simply and low cost with high throughput, it provides the use of an aqueous medium as carrier of chemicals in flow system that is nontoxic and causes no pollution, thereby belonging to green chemistry. The proposed procedure can be successfully applied to estimate the content of Iron (III) in biological samples. The results of estimation are satisfactory as compared with those given by a reference method, showed the new method to be accurate and precise at the 95% confidence level.

Abstract:Two imidazole‐based color‐changing fluorescent probes 4‐(4,5‐bis(4‐chlorophenyl)‐1H‐imidazol‐2‐yl)‐N, N‐diphenylaniline (CIN) and 4‐(4,5‐di(furan‐2‐yl)‐1H‐imidazol‐2‐yl)‐N, N‐diphenylaniline (FIN) were synthesized and characterized. The new probes CIN and FIN exhibit excellent selectivity and sensitivity towards Fe3+ with relatively low detection limits and strong anti‐interference properties. Both the probes CIN and FIN complexed 1 : 1 with Fe3+ ion. Two imidazole‐based color‐changing fluorescent probes 4‐(4,5‐bis(4‐chlorophenyl)‐1H‐imidazol‐2‐yl)‐N, N‐diphenylaniline (CIN) and 4‐(4,5‐di(furan‐2‐yl)‐1H‐imidazol‐2‐yl)‐N, N‐diphenylaniline (FIN) were synthesized and characterized. The new probes CIN and FIN exhibit excellent selectivity and sensitivity towards Fe3+ with relatively low detection limits (0.26 μM and 0.10 μM) and strong anti‐interference properties. Both the probes CIN and FIN complex 1 : 1 with Fe3+ ion, and the complexation constants were 1.165×105 M−1 (CIN) and 1.899×105 M−1 (FIN), respectively. Moreover, CIN and FIN could be recycled by EDTA and could effectively monitor Fe3+ ions at pH 4–10, which ensures the sensor's potential for detecting Fe3+ in actual aqueous solutions. The probes were further applied to the detection of Fe3+ in the actual reaction of Fe2+ oxidation, and the filter paper and the probe solution are combined to make an indicator to detect Fe3+ more conveniently and quickly.

Abstract:Iron is an essential element in the composition of living organisms and plays a crucial role in a wide range of biological activities. The human body primarily obtains essential iron through the consumption of food. Therefore, it is vital for the health of human body to maintain iron homeostasis. The reducing character of the cellular microenvironment enables Fe2+ to occupy a dominant position within the cell. Hence, there is an urgent need for a simple and sensitive tool that can detect a large amount of Fe2+ in organisms. In this work, a highly specific fluorescent chemodosimeter NPCO ("NP" represents the naphthalimide fluorophore, and "CO" represents the carbamoyl oxime structure) for the detection of Fe2+ with excellent sensitivity (LOD = 82 nM) was constructed by incorporating a novel carbamoyl oxime structure as the recognition group. NPCO can be effectively employed for the detection of Fe2+ in food samples, living cells, and zebrafish. Furthermore, by using soybean sprouts as a model plant, the application of NPCO was expanded to detect Fe2+ in plants. Therefore, NPCO could be used as an excellent assay tool for detecting Fe2+ in organisms and is expected to be an important aid in exploring the mechanism of iron regulation.

Abstract:Ferroptosis is a form of nonapoptotic cell death characterized by the unchecked accumulation of lipid peroxides. Ferrostatin-1 and its analogs (ferrostatins) specifically prevent ferroptosis in multiple contexts, but many aspects of their molecular mechanism of action remain poorly described. Here, we employed stimulated Raman scattering (SRS) microscopy coupled with small vibrational tags to image the distribution of ferrostatins in cells and found that they accumulate in lysosomes, mitochondria, and the endoplasmic reticulum. We then evaluated the functional relevance of lysosomes and mitochondria to ferroptosis suppression by ferrostatins and found that neither is required for effective ferroptosis suppression.

Abstract:Fe2+ plays a crucial role in biological systems as an essential trace element in the body. Iron is absorbed by the body through food and Fe2+ is also an important component of living cells; therefore, quantitative testing of Fe2+ in food and in living cells is of great importance. This paper presents the development of a novel π-bridge EDOT-based N-oxide turn-on fluorescent probe, designated as FeE, for the detection of Fe2+. The probe has been shown to exhibit excellent sensitivity, a favorable linear relationship between fluorescence signal intensity and Fe2+ concentration, and an effective immunity to interference. The probe has low cytotoxicity and has been successfully used to detect Fe2+ in cells using laser confocal fluorescence microscopy. It is also possible to determine the presence of Fe2+ in animal blood, spinach, apple juice, red wine, mineral water and metal cans.
Iron fortification of infant cereals: a proposal for the use of ferrous fumarate or ferrous succinate

Abstract:Hemoglobin-repletion tests in rats, organoleptic studies, and iron-absorption studies in humans were used to search for Fe sources with high bioavailability that could be added to infant cereals as alternatives to the Fe compounds currently used for fortification. From rat and organoleptic studies on 11 alternative Fe sources, ferrous fumarate, ferrous succinate, and ferric saccharate were selected as the most suitable for infant-cereal fortification and, by use of radioactive labels, absorption of those compounds from fortified cereal was measured in adult human volunteers. There was no difference in absorption between ferrous fumarate and ferrous sulfate whereas the values for ferrous succinate, ferrous saccharate (10% Fe), and ferric pyrophosphate were 92%, 74%, and 39% of the ferrous sulfate values, respectively. We conclude that ferrous fumarate and ferrous succinate are highly available Fe sources in man that can be used to fortify infant cereals without causing fat oxidation or discoloration.

Abstract:Iron, a crucial biologically active ion essential for metabolic processes in living organisms, plays a vital role in biological functions, and imbalances in iron levels can lead to various diseases. In this study, we have developed two simple "turn-on" fluorescent probes, NOPy and NOCN, for the quick and selective detection of Fe 2+ at nanomolar levels (LOD of 35 nM), accompanied by significant absorption and emission shifts, along with colorimetric demarcation. Both fluorophores exhibit an excellent "turn-on" emission response upon encountering Fe 2+ in the cells. Flow cytometry and confocal fluorescence imaging studies demonstrate enhanced fluorescence signals in response to labile iron, efficiently detecting heme during erastin-induced ferroptosis. Interestingly, we also observed that the product formed after Fe 2+ sensing localizes within the lipid droplets. These water-soluble and highly sensitive reactive probes, NOPy and NOCN, enable investigations of iron-dependent physiological and pathological conditions. The development of these probes represents an advancement in the field, offering a rapid and selective means for detecting Fe 2+ with minimal cytotoxicity.

Abstract:Dysregulation of iron homeostasis causes iron-mediated cell death, recently described as ferroptosis. Ferroptosis is reported in many chronic diseases, such as hepatic cancer, renal, and cardiovascular diseases (heart failure, atherosclerosis). However, there is a notable scarcity of research studies in the existing literature that explore treatments capable of preventing ferroptosis. Additionally, as far as the author is aware, there is currently no established model for studying ferroptosis within cardiovascular cells, which would be essential for assessing metal-chelating molecules with the potential ability to inhibit ferroptosis and their application in the treatment of cardiovascular diseases. In this study, a smooth muscle cell-based ferroptosis model is developed upon the inhibition of the system Xc− transporter by erastin associated or not with Fe(III) overload, and its rescue upon the introduction of well-known iron chelators, deferoxamine and deferiprone. We showed that erastin alone decreased the intracellular concentration of glutathione (GSH) without affecting peroxidized lipid concentrations. Erastin with ferric citrate was able to decrease intracellular GSH and induce lipid peroxidation after overnight incubation. Only deferiprone was able to rescue the cells from ferroptosis by decreasing lipid peroxidation via iron ion chelation in a 3:1 molar ratio.

Abstract:Ferric ion (Fe 3+ ) serves as an essential and indispensable inorganic element in various biological process. Excess Fe 3+ is harmful for biological systems and human health. Herein, highly sensitive carbon quantum dots (Fe-CQDs) for ferric ions were synthesized by a high-efficiency, one-step hydrothermal route utilizing L-arginine and citric acid. The prepared Fe-CQDs exhibited good water solubility, high photostability, and bright blue florescence with relatively high quantum yields (QYs) over 21.33% and a Stokes shift exceeding 100 nm. The functionalized Fe-CQDs offer rapid, sensitive, and selective recognition of Fe 3+ with a linear relationship from 0.6 to 80 μM and a low detection limit of 0.2 μM. Moreover, the synthesized Fe-CQDs illustrated strong anti-interference properties in samples and buffer solutions. The developed sensor demonstrated high biocompatibility and was successfully applied for intracellular Fe 3+ determination in SHSY5Y and HT22 cells, illustrating excellent imaging application. The present work provides a promising platform for the early clinical diagnosis of Fe 3+ -associated diseases.

A new spectrophotometric method for measuring ceruloplasmin ferroxidase activity: an innovative approach

Abstract:Ferroxidases are enzymes that participate in the iron metabolism of different organisms. They catalyze the oxidation of ferrous iron, Fe 2 +, into ferric iron, Fe 3 +, which is essential in iron homeostasis and physiological functioning. The present study describes a novel spectrophotometric method of serum ceruloplasmin ferroxidase activity. This method is easy to perform; it is also sensitive, specific, and rapid. In this method, ferrous ions are used as a substrate for the enzyme, with either salicylic acid or sulfosalicylic acid being taken as a chromogenic compound. These chromogens easily form a colored complex with ferric ions but are not formed with ferrous ions. In the enzymatic reaction, the ceruloplasmin ferroxidase enzyme catalyzes the oxidation of ferrous to ferric ions. The resulting increase in ferric ion concentration is then measured spectrophotometrically, following the formation of the colored complex. The complex formed has maximum absorbance at 540 nm in the case of salicylic acid and 490 nm in the case of sulfosalicylic acid. Comparatively, it was tested against the standard method to ascertain the new method's effectuality and reliability for assaying ferroxidase activity. The determined correlation coefficient amounted to 0.99, showing a strong correlation between the results obtained by the two methods. This new spectrophotometric technique offers a simplified, sensitive, specific, and fast means of estimating ferroxidase activity. It avoids using concentrated strong acids in the procedure and correlates excellently with the standard technique. This sets up a potential alternative for accurately determining ferroxidase activity in biological samples.

Abstract:A turn-on fluorescent probe for the detection of Fe2+ is facilely synthesized via a nucleophile substitution reaction. The fluorescent probe, N-butyl-4-phenyltellanyl-1,8-naphthalimide (Naph-Te), shows excellent selectivity to Fe2+ in a mixed solution of acetonitrile and phosphate buffer under aerobic conditions. The coexistence of biological abundant metal ions such as Na+, K+, Ca2+ and Mg2+ has little effect on the fluorescence signal. This turn-on response is achieved via a redox-involved reaction triggered by Fe2+ at neutral pH and room temperature, which removes the heavy-atom effect of the tellurium atom on the naphthalimide fluorophore to afford a fluorescent product (N-butyl-4-hydroxyl-1,8-naphthalimide). The probe has excellent cell membrane permeability and is further applied successfully to monitor supplementary Fe2+ in live HL-7702 cells using a laser confocal fluorescence microscope.

Abstract:Iron ions play an important role in our lives. Excessive or lack of iron ion intake leads to many diseases. At the same time, the water environment is easily polluted by these metal ions with the acceleration of industrialization. Therefore, the detection of iron ions in the water environment and the human body is particularly important. In this paper, we prepared a RhB-EDA fluorescent probe by condensing rhodamine B (RhB) with ethylenediamine (EDA) for high recognition of Fe3+. A RhB-EDA molecule itself is colorless and has no fluorescence emission in an alcohol solution. When Fe3+ was added, the lactam ring structure of the fluorescent probe opened, and the UV and fluorescence spectra changed. At the same time, the color of the mixed solution gradually deepened toward pink. Therefore, dual spectral detection and naked-eye observation of Fe3+ were realized. In addition, with the decrease of the pH value and the prolongation of chelating time, the ultraviolet absorbance and fluorescence emission intensity were enhanced and the color of the mixed solution deepened. The RhD-EDA fluorescent probe is simple and accurate and provides good technical support for the detection of Fe3+.This article has not yet been cited by other publications.
