Identification of Novel Interferon Stimulated Genes in Differentiated Hepatocytes

Abstract:Pluripotent stem cells are being actively studied as a cell source for regenerating damaged liver. For long term survival of engrafting cells in the body, not only do the cells have to execute liver‐specific functions but also withstand metabolic stressors and invading pathogens. The cellular innate immune system orchestrated by the interferon pathway provides the first line of defense against infectious agents. The objective of this study is to assess the innate immune function as well as to systematically profile the IFN‐induced genes during hepatic differentiation of pluripotent stem cells. We derived endodermal cells, hepatoblast and immature hepatocytes from hESC using stage‐specific growth factor cocktails. The cells were stimulated with IFN‐α and subjected to type I IFN pathway analysis. Transcriptome analysis was carried out by RNA sequencing. The results showed that IFN‐α treatment activated JAK‐STAT pathway in differentiating cells. Unbiased RNA sequencing analysis indicated stage specific expression of classical and novel IFN‐stimulated genes demonstrating an intact innate immune response. Reverse transcription‐qPCR confirmed the IFN‐induced expression of novel ISGs including RASGRP3, CLMP, PPM1J, and TRANK1. Infection of these hESC‐derived hepatic cells with Hepatitis C virus also induced the expression of ISGs including the classical and novel genes – LAMP3, ETV7, and TRANK1 further confirming intact innate immunity. hESC‐derived hepatic cells contain an intact innate system.
