Antioxidant activity of folic acid: From mechanism of action to clinical application

Abstract:The presumed protective effect of folic acid (FA) on the pathogenesis of cardiovascular, hematological and neurological diseases and cancer has been associated with the antioxidant activity of FA. The actual antioxidant activity of FA depends on its consumption and reduction into its reduced forms. The more the FA is supplemented and/or reduced, the higher its antioxidant activity becomes. In rats subjected to acute cardiac volume overload, there was progressive left ventricular (LV) hypertrophy, and reduced capillary density which was prevented by FA supplementation (30 mg/kg). This effect was associated with prevention of apoptosis of endothelial cells as demonstrated by downregulation of active caspase‐3 expression. Moreover, FA prevented collagen loss and decreased both mast cell degranulation and MT1‐MMP expression. The antioxidant effect of FA was reflected in elevation of GSH/GSSG ratio and a reduction of protein nitration. Furthermore, in dyslipidemic patients with stable angina pectoris, FA supplementation (10 mg/day) for three months improved endothelial dysfunction and decreased carotid intima‐media thickness. Our results indicate that the beneficial effects of FA may in part be due to its potent antioxidant activity.
