Abstract:The aqueous two-phase extraction process (APTE) for polysaccharides from the fruits of Ilex cornuta (Lindl. et Paxt.) (PFIC) was studied, and its monosaccharide composition and antioxidant activities were analyzed. After screening and selection of isopropanol/ammonium sulfate as the aqueous two-phase system, various parameters of crude extract amount, ultrasonic time, extraction temperature and extraction time on the extraction process was investigated in detail and optimized by response surface methodology (RSM). The obtained PFIC was characterized by FT-IR and its monosaccharide composition was analyzed by GC-MS. Furthermore, the antioxidant activities of PFIC were evaluated by measuring the scavenging rates of DPPH radical, hydroxyl radical and superoxide anion radical. Under the optimal conditions in which crude extract amount was 2.23 g, ultrasonic time was 17.5 min, extraction temperature was 69 â„ƒ and the extraction time was 1.55 h, the yield of polysaccharides reached 50.87% which was close to the predicted value of 50.78%. FT-IR analysis confirmed that the extract was polysaccharide, and GC-MS analysis indicated that the monosaccharide components of the polysaccharides were rhamnose, arabinose, xylose, mannose, glucose and galactose. Antioxidant results showed that PFIC showed strong scavenging ability to DPPH radical, fine scavenging ability to hydroxyl radical and certain scavenging ability to superoxide anion radical. The ultrasonic-assisted APTE process for PFIC optimized by RSM is reasonable and reliable, and the obtained PFIC has the potential to be utilized as natural antioxidants.
Process Optimization of Enzyme-Assisted Extraction of Polysaccharides from Artificially-Cultivated Cordyceps cicadae and Its Kinetic, Thermodynamic and Antioxidant Activities Analysis

Abstract:Objective: To optimize the enzyme-assisted extraction process of polysaccharides from artificially-cultivated Cordyceps cicadae was conducted in this study. Four different parameters were considered to evaluate the yield of polysaccharides including liquid to solid ratio, enzyme dosage, enzymatic temperature and extraction time. Methods: A four-factor-three-level experimental design was employed to establish a mathematical model by Box-Behnken method, and the scavenging capacity of polysaccharides against three radicals was examined. Afterward, Fick's second law was used to build the kinetic model for the extraction of polysaccharide from artificially-cultivated Cordyceps cicadae. The parameters including rate constants, relative extraction rate, and activation energy were employed to analyze the kinetic and thermodynamic features. Results: Based on the response surface analysis, the optimal extraction process was presented to be as following: Liquid to solid ratio 1:30 g/mL, enzyme dosage 1.6%, enzymatic digestion temperature 67 ℃ and extraction time 90 min. The polysaccharide yield under the above condition was 7.91%, which was close to the predicted value. Moreover, the results of antioxidant capacities indicated that the obtained crude polysaccharides under optimal conditions showed strong DPPH radical scavenging and hydroxyl radical scavenging with IC50 values for 0.60 and 0.54 mg/mL, respectively, and its ORAC value was 45.62 Trolox μmol/g, suggesting potent antioxidant activity in vitro. Conclusion：The study of enzyme-assisted extraction of artificially-cultivated Cordyceps cicadae flower polysaccharides provide theoretical support for the production of polysaccharide fractions from this kind of commercialized Cordyceps cicadae resources.

Abstract:Dendrobium chrysotoxum has long been used as a health-promoting and therapeutic tonic in China. The polysaccharides from D. chrysotoxum (DCP) are considered to play an important role in its medical effects. The present study aimed at optimizing the technology for enzyme-assisted extraction (EAE) of DCP and at investigating the physicochemical characteristics and functional properties of DCP-E obtained by EAE and DCP-H obtained by hot water extraction (HVVE). Based on single factor tests and orthogonal experiments of extraction pH, extraction temperature, cellulase amount and extraction time, the optimum conditions for CP-E were extraction pH value of 5.5, extraction temperature of 40 degrees C, cellulase amount of 10 g/L, extraction time of 3.0 h and solid/liquid ratio of 1:25. Under these conditions, the yield of DCP-E was 8.41 g/100 g dry weight, which was 1.25 times of that of DCP-H. As compared to DCP-H, DCP-E displayed an increased purity, a decreased molecular weight and relative viscosity as well as a changed monosaccharide compositions. Foam stabilization and splenic cell proliferation experiments showed that DCP-E had higher cell proliferation rate while DCP-H had better foam stabilization activity. These results indicate that different extraction methods would influence the physicochemical characteristics and subsequently functional properties of polysaccharides, suggesting that the suitable extraction method should be selected according to the application intention of polysaccharides. Abstract:Ultra-high pressure extraction technique was used to extract polysaccharides from Dendrobium candidum. Extraction pressure, pressure holding time and liquid-solid ratio were found to have a significant influence on the polysaccharides extraction and were optimized by the response surface method (RSM). The obtained optimum conditions were: extraction pressure 445.28MPa, pressure holding time 6.7 minutes and liquid-solid ratio 237.91ml/g. The maximum polysaccharide yield could reach 596.75mg/g under these conditions. The ultra-high pressure extraction process was a suitable method for its higher yield and shorter extraction time compared to the water-heating extraction.

Abstract:The conventional extraction methods for polysaccharides were time-consuming, laborious and energy-consuming. Microwave-assisted extraction (MAE) technique was employed for the extraction of Potentilla anserina polysaccharides (PAPMAE), which is a traditional Chinese functional food. The extracting parameters of PAPMAE were optimized by Box–Behnken design. The optimum conditions were ratio of water to raw material 14.5:1, microwave power 369W, extraction temperature 63.3°C and extraction time 76.8min with an enhanced yield of 13.33%. In microwave heating process, a sharp decrease in Mw was detected in SEC-LLS measurement. A df value of 3.03 indicated that PAPMAE exhibited a compact hard sphere conformation in an aqueous solution. Furthermore, it showed stronger antioxidant activities compared with hot water extraction by evaluating in superoxide radical (O2-), hydroxyl radical (OH), 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical, reducing power and metal chelating assay. The data obtained clearly showed that the molecular weights played a more important role in antioxidant activities.

Abstract:To obtain the optimal process for the enzyme−assisted aqueous extraction of polysaccharides from Acanthopanax senticosus, and study the physicochemical properties of polysaccharides of different molecular weights, the extraction of Acanthopanax polysaccharides was optimized using the BBD response surface test. The polysaccharides with different molecular weights were obtained by ethanol−graded precipitation at 40%, 60%, and 80%, which were presented as ASPS40, ASPS60, and ASPS80. The polysaccharides were analyzed by HPGPC, ion chromatography, FT−IR, UV, SEM, TGA, XRD, Congo red, and I2−KI tests. The antioxidant assay was used to evaluate their antioxidant properties in vitro. The findings demonstrated that the recovery rate of Acanthopanax polysaccharide was 10.53 ± 0.682%, which is about 2.5 times greater compared to the conventional method of hot water extraction. Based on FT−IR, TGA, polysaccharides with different molecular weights did not differ in their structure or thermal stability. The XRD suggests that the internal structure of ASPSs is amorphous. Congo red and I2−KI showed that all three polysaccharides had triple helix structures with longer branched chains and more side chains. Furthermore, the antioxidant results showed the antioxidant activity of polysaccharides is not only related to the molecular weight size but also can be related to its composition and structure. These studies developed a green, and scalable method to produce polysaccharides from Acanthopanax senticosus and evaluated the properties of Acanthopanax polysaccharides of different molecular weights.

Abstract:AbstractThe polysaccharide from Morinda citrifolia fermentation liquor was extracted by the hot water method. The acetylated polysaccharide, phosphorylated polysaccharide, carboxymethylated polysaccharide, and sulfated polysaccharide were identified by IR and NMR spectra. The results showed that Morinda citrifolia polysaccharide and its derivatives showed the good antioxidant activity, and them up to Vc level. These results provide a good basis for studying the antioxidant activity and structural‐activity relationship of Morinda citrifolia polysaccharide and its derivatives.

Abstract:Dendrobium huoshanense is a famous edible and medicinal herb, and polysaccharides are the main bioactive component in it. In this study, response surface methodology (RSM) combined with a Box–Behnken design (BBD) was used to optimize the enzyme-assisted extraction (EAE), ultrasound–microwave–assisted extraction (UMAE), and hot water extraction (HWE) conditions and obtain the polysaccharides named DHP-E, DHP-UM, and DHP-H. The effects of different extraction methods on the physicochemical properties, structure characteristics, and bioactivity of polysaccharides were compared. The differential thermogravimetric curves indicated that DHP-E showed a broader temperature range during thermal degradation compared with DHP-UM and DHP-H. The SEM results showed that DHP-E displayed an irregular granular structure, but DHP-UM and DHP-H were sponge-like. The results of absolute molecular weight indicated that polysaccharides with higher molecular weight detected in DHP-H and DHP-UM did not appear in DHP-E due to enzymatic degradation. The monosaccharide composition showed that DHPs were all composed of Man, Glc, and Gal but with different proportions. Finally, the glycosidic bond types, which have a significant effect on bioactivity, were decoded with methylation analysis. The results showed that DHPs contained four glycosidic bond types, including Glcp-(1→, →4)-Manp-(1→, →4)-Glcp-(1→, and →4,6)-Manp-(1→ with different ratios. Furthermore, DHP-E exhibited better DPPH and ABTS radical scavenging activities. These findings could provide scientific foundations for selecting appropriate extraction methods to obtain desired bioactivities for applications in the pharmaceutical and functional food industries.

Effects of different extraction methods on the physico-chemical characteristics and biological activities of polysaccharides from Clitocybe squamulosa

Abstract:This study comparatively evaluated the effects of the commonly used six extraction methods (acidic, alkaline, enzymatic, ultrasonic, high-pressure, and microwave) on the physico-chemical properties, processing characteristics, and biological activities of polysaccharides from Clitocybe squamulosa (CSFPs). The results show that polysaccharides extracted using an enzyme-assisted extraction method has a relatively high extraction yield (4.46 ± 1.62 %) and carbohydrate content (70.79 ± 6.25 %) compared with others. Furthermore, CSFPs were all composed of glucose, galactose, mannose, xylose, and glucosamine hydrochloride. Only ultrasonic-assisted extraction of polysaccharides (CSFP-U) has a triple helix chain conformation. Scanning electron microscopy (SEM) revealed significant differences in the microstructure of polysaccharides prepared using different methods. Besides that, the polysaccharides prepared by alkali extraction (CSFP-B) and high-pressure assisted extraction (CSFP-H) have good water (2.86 ± 0.29 g/g and 3.15 ± 0.29 g/g) and oil (8.13 ± 0.32 g/g and 7.97 ± 0.04 g/g) holding properties. The rheological behavior demonstrated that CSFPs solutions were typical non-Newtonian fluid. Apart from this, the antioxidant capacity (clearing DPPH (IC50 = 0.29) and ABTS free radicals (IC50 = 0.19), total reduction ability (IC50 = 3.02)) of polysaccharides prepared by the microwave-assisted extraction (CSFP-M) method was significantly higher than that of other extraction methods. By contrast, the polysaccharide prepared by acid extraction (CSFP-A) has the optimum binding capacity (bile acid salt (71.30 ± 6.78 %) and cholesterol (57.07 ± 3.26 mg/g)). The antibacterial activity of CSFPs was positively correlated with their concentration. Thus, the research results can provide a theoretical basis for the development and utilization of polysaccharides from C. squamulosa.

Abstract:The polysaccharides from Stemona tuberosa Lour, a kind of plant used in Chinese herbal medicine, have various pharmacological activities, such as anti-inflammatory and antioxidant properties. However, the effects of the extraction methods and the activity of polysaccharides from different parts are still unknown. Therefore, this study aimed to evaluate the effects of different extraction methods on the yields, chemical compositions, and bioactivity of polysaccharides extracted from different parts of Stemona tuberosa Lour. Six polysaccharides were extracted from the leaves, roots, and stems of Stemona tuberosa Lour through the use of hot water (i.e., SPS-L1, SPS-R1, and SPS-S1) and an ultrasound-assisted method (i.e., SPS-L2, SPS-R2, and SPS-S2). The results showed that the physicochemical properties, structural properties, and biological activity of the polysaccharides varied with the extraction methods and parts. SPS-R1 and SPS-R2 had higher extraction yields and total sugar contents than those of the other SPSs (SPS-L1, SPS-L2, SPS-S1, and SPS-S2). SPS-L1 had favorable antioxidant activity and the ability to downregulate MUC5AC expression. An investigation of the anti-inflammatory properties showed that SPS-R1 and SPS-R2 had greater anti-inflammatory activities, while SPS-R2 demonstrated the strongest anti-inflammatory potential. The results of this study indicated that SPS-L1 and SPS-L2, which were extracted from non-medicinal parts, may serve as potent natural antioxidants, but further study is necessary to explore their potential applications in the treatment of diseases. The positive anti-inflammatory effects of SPS-R1 and SPS-R2 in the roots may be further exploited in drugs for the treatment of inflammation.

Abstract:Cellulase-assisted extraction of polysaccharides from Polygonatum odoratum (CPP) was optimized by response surface methodology (RSM) and the extracted CPP's preliminary chemical characteristics, as well as antioxidant and immunomodulatory activities were also investigated. The optimal extraction parameters comprised an extraction temperature of 58.21 °C, an extraction time of 3.18 h, pH value of 5.8, and cellulase amount of 6.0%. Under these conditions, the relative yield was 15.76%, higher than the yield achieved with hot water extracted polysaccharide (HPP). Chemical composition analysis demonstrated that CPP and HPP consisted of mannose, glucosamine, rhamnose, glucose, galactose, and arabinose with a molecular ratio of 7.80:1.08:1.63:65.93:3.58:1.00 and 11.22:0.23:0.23:17.59:2.73:9.10, respectively. The molecular weight distribution of CPP was lower and more homogeneous compared with HPP. CPP exhibited stronger antioxidant activities than HPP, including DPPH radical scavenging activity and reducing power. Both CPP and HPP could significantly promote the proliferation and neutral red phagocytosis of RAW 264.7 macrophage cells in vitro. These results indicate that the cellulase-assisted extraction method influenced the physicochemical characteristics, and consequently, the functional activities of polysaccharides, suggesting the cellulose-assisted method may be a viable option for extraction polysaccharides from P. odoratum.

Abstract:The purpose of the study was to improve the extraction of polysaccharide from the leaves of Cercis chinensis Bunge using ultrasound, compare the difference between boiling and ultrasound extraction in terms of polysaccharide content, monosaccharide compounds, and evaluate how the factors affected the bioactivity. The best conditions, according to the single factor experiments and the Box-Bohnken design (BBD), were an intensity of ultrasound of 180 W, duration of extraction of 40 min, proportion of water to material of 15:1 (g/g), and a higher polysaccharide yield of 20.02±0.55 (mg/g) than in boiling extraction (16.09±0.82 mg/g). The antioxidative experiment suggested the polysaccharide by ultrasound exhibited higher DPPH, hydroxyl radical scavenging capacities, and reducing power at 1.2–1.4 mg/mL, which was superior to the boiling polysaccharide. Further analysis shows that the ultrasonic purified polysaccharides like gla, N-glu, and gluA contained more total sugar and uronic acids than the boiling method did. This may indicate that the ultrasonic isolation of the polysaccharides increase the antioxidant activity of the polysaccharides.

Abstract:In this study, five polysaccharides were extracted from processed Cistanche deserticola . The processing included crude product, enzymatic hydrolysis, hot air drying, stir-baking with wine and high-pressure steaming, and these polysaccharides were named as CP-CDPs, EH-CDPs, HAD-CDPs, SBW-CDPs and HPS-CDPs, respectively. The structural characteristics and biological activities were explored. The results showed that processing changed properties of C. deserticola polysaccharides. CP-CDPs had the highest brightness value L *(93.84) and carbohydrate content (61.27 %). EH-CDPs had minimum M w (1531.50 kDa), while SBW-CDPs had maximum M w (2526.0 kDa). Glucose was major predominant monosaccharide in CP-CDPs (89.82 %), HAD-CDPs (79.3 %), SBW-CDPs (59.41 %) and HPS-CDPs (63.86 %), while galactose was major monosaccharide in EH-CDPs (29.44 %). According to SEM, SBW-CDPs showed compact structures, while HPS-CDPs and HAD-CDPs had similar looser structure than SBW-CDPs; meanwhile, CP-CDPs showed irregular agglomeration shape and EH-CDPs was dense blocky shape. The AFM showed SBW-CDPs had the largest molecular chain than other polysaccharides. When scavenging activity reaching 50 %, the concentrations of CP-CDPs, EH-CDPs, HAD-CDPs, SBW-CDPs, HPS-CDPs are 2.25, 0.25, 0.75, 1.8 and 1.5 mg/mL, respectively. This study sheds light on the effects of traditional Chinese medicine processing on characteristics, bioactivities of C. deserticola polysaccharides, and provides the basis for applications in food and pharmaceutical industries.

Abstract:Ultrasound-assisted extraction of Cissus repens polysaccharides (CRPs) was optimized through response surface methodology (RSM) based on Box-Behnken design (BBD). The maximum CRPs yield (16.18 %) was achieved under the optimum extraction conditions: extraction time 72 min, extraction temperature 74 °C, extraction power 240 W. Then three-phase partitioning (TPP) method combined with gradient alcohol precipitation was used to obtained CRP20, CRP40, CRP60 and CRP80 from CRPs, and CRP80 has a higher purity than others. The primary chemical and structural characteristics of CRP80 were investigated by UV, FT-IR, high-performance liquid chromatography (HPLC) and high-performance gel-permeation chromatography (HPGPC). CRP80 is mainly composed of glucose, galactose, arabinose and mannose, with a molecular weights of approximately 2.95 kDa. Furthermore, the antioxidant activity and hypoglyceamic activity of CRP80 in vitro were evaluated. The results showed that CRP80 had strong scavenging activities on ABTS, hydroxyl and DPPH radicals, as well as high scavenging activities on α-glucosidase and α-amylase. Our research provided an efficient method for the extraction of polysaccharides from C. repens and CRP80 has potential as a promising source of natural antioxidants and hypoglycemic agent for the functional food and medicinal industries.

Abstract:Selected the extract conditions of ramulus mori polysaccharide by orthogonal design, and combination of microwave extraction,ultrasonic extraction and enzymatic extraction method the effect of extraction was observed. we compared the ethanol precipitation method and the macroreticular resin purification effect by purify the crude extract. The results were that the best extract conditions were the time being 100min, the solution-material ratio being more than 20 times and pH5 in general extraction method. The extraction rate absolute enhanced to 18.75% combined with enzyme extraction. With AB-8 macroporous resin-based purification , the ramulus mori polysaccharide of 84.57% purity was achieved.

Abstract:The ultrasonic-assisted deep eutectic solvent method was used to extract the polysaccharides of Pericarpium Citri Reticulatae (PCRP), and the ultrasound-assisted DES extraction process was optimized by Box-Behnken response surface test using the extraction rate of the PCRP as an index; the in vitro activities of purified the PCRP(PCRPs-1) were investigated by determining the scavenging rate of DPPH• and ABTS• + as well as by enzyme inhibition assay. The monosaccharide composition was analyzed by HPLC. The best process conditions for response surface optimization were a material-liquid ratio of 1:37 g/mL, water content of 44%, time of 89 min, and power of 320 W. The polysaccharide extraction rate was measured to be 5.41%, which was well optimized when compared with that of the ordinary aqueous extraction method of 3.92%. By α-glucosidase and α-amylase inhibition activity test, it showed that the PCRPs-1 had hypoglycemic activity. The DPPH radical scavenging activity test and ABTS + scavenging activity test indicated that the PCRPs-1 had good biological activity. Analysis of the monosaccharide fractions showed that the PCRPs-1 consisted of mannose, rhamnose, glucuronic acid, galacturonic acid, glucose, galactose, xylose, and arabinose, with molar ratios of 1:39.24:4.41:8.91:7.83:86.00:1.02:9.17. The activity studies showed that PCRPs-1 possessed certain hypoglycaemic and antioxidant activities.

Optimization of Multi-Enzymatic Extraction of Polysaccharides from Cardamine hupingshanensis and Their Antioxidant Activity

Abstract:The extraction of crude polysaccharides from cultivated Cardamine hupingshanensis was optimized by the combined use of one-factor-at-a-time method and response surface methodology.The results of one-factor-at-a-time experiments demonstrated hydrolysis with a mixture (2:1,m/m) of cellulase and pectinase at a dosage of 2％ (m/m) at 60 ℃ and an initial pH of 4.0 for 90 min to be optimum for the extraction ofpolysaccharides.Furthermore,the extraction conditions op tmized by response surface methodology were as follows:hydrolysis time,91.8 min;hydrolysis temperature,57.1 ℃;and intial pH,4.17.Under these conditions,the predicted maximum yield of polysaccharides of 4.14％ was obtained,and experiments gave an average value of 4.07％,indicating a relative error of about 1.62％ the predicted and experimental values.The results of antioxidant assays showed that Cardamine hupingshanensis.polysaccharides possessed antioxidant activity better than that of VC.Our experimental results provide an experimental basis for studying the extraction and properties of Cardamine hupingshanensis polysaccharides.

Abstract:Polysaccharides as the biopolymers are showing various structural and modulatory functions. Effective separation of carbohydrate structures is essential to understanding their function. In this study, we choose an efficient organic acid in combination with recyclable organic solvent three-phase partitioning technology for the simultaneous extraction of polysaccharides from Ampelopsis japonica (AJPs) to ensure the integrity of linear and branched polysaccharide. The monosaccharide composition, glycosidic linkage information, structural and physicochemical analyses and associations with antioxidant activities were extensively analyzed. Synergistic extraction was compared with the conventional hot water extraction method and the results showed that AJPs-HNP exhibited better elastic properties and excellent antioxidant activity. Correlation analysis confirmed that the antioxidant activity of AJPs was significantly correlated with relative molecular weight, uronic acid content and terminal glycoside linkage molar ratios. The collaborative processing has significantly improved the utilization potential of AJPs and provides a sound theoretical foundation for the effective extraction and separation of polysaccharides. Overall, this work provides systematic and comprehensive scientific information on the physicochemical, rheological and antioxidant properties of AJPs, revealing their potential as natural antioxidants in the functional food and pharmaceutical industries.
