Abstract:AIM: To observe the effects of hypoxia on the levels of nitric oxide (NO), endothelin (ET-1) and the expression of inducible nitric oxide synthase (iNOS) mRNA in human umbilical vein endothelial cells (HUVECs), and further investigate the mechanism of hypoxic pulmonary hypertension. METHODS: On the basis of the HUVECs culture model, the methods of nitrate reductase and radioimmunoassay were used to determine the changes of NO and ET-1 in the medium secreted by HUVECs, and the expression of iNOS mRNA was analyzed by semi quantitative RT-PCR after exposure to hypoxia (3% O_2) for 6, 12 or 24 h. RESULTS: The contents of NO_2-/NO_3-and ET-1 in hypoxia group in the medium was significantly higher than that in control group at different time points (P0.05). Also, iNOS mRNA expression increased significantly (P0.05). CONCLUSION: Hypoxia stimulates the release of NO and ET-1 from HUVECs, also induces iNOS-mRNA expression. The change of NO may be the result of iNOS mRNA upregulation induced by hypoxia.
[Relationship Between Expression of Endothelial Nitric Oxide Synthase and NADPH Oxidase in Lungs of Mice Exposed to Chronic Hypoxia].

Abstract:OBJECTIVE:To explore the relationship between the expression of endothelial nitric oxide synthase (eNOS) and NADPH oxidase (NOX) in the lungs of mice treated by chronic hypoxic exposure. METHODS:Thirty male wild-type (WT) C57Bl/6 mice and thirty male eNOS-knockout (KO) C57BL/6 mice were randomly divided into normoxic groups (exposed to normoxia for 7 days or 21 days), hypoxic groups (exposed to 10% oxygen for 7 days or 21 days), and treatment groups (exposed to 10% oxygen and orally administrated 10 mmol/L 4-hydroxy TEMPO in drinking water for 7 days or 21 days) (n=6 in each group). The remodeling of the small pulmonary arteries was evaluated by the percentage of media wall thickness (MT%). The weight ratio of right ventricle to left ventricle plus septum (RV/[LV+S]) was calculated to evaluate the hypertrophy of right ventricle. Real-time PCR was used to measure the mRNA expression of NOX2, NOX4, and eNOS in mouse lungs. ELISA was used to determine the concentration of reactive oxygen species (ROS) in mouse lungs. RESULTS:In WT mice and KO mice, the hypoxic groups had significantly increased pulmonary vascular remodeling and RV/[LV+S] compared with the normoxic and treatment groups (P<0.05), but there were no significant differences between the normoxic and treatment groups (P>0.05). In WT mice, the hypoxic and treatment groups had significantly lower ROS concentrations than the normoxic group (P<0.05), but there were no significant differences between the hypoxic and treatment groups (P>0.05). In WT mice, the mRNA expression of eNOS, NOX2, and NOX4 was significantly higher in the hypoxic group than in the normoxic group (P<0.05), and 4-hydroxy TEMPO reversed their over-expression. In the normoxic group, the KO mice had significantly higher NOX2 and NOX4 mRNA expression than the WT mice (P<0.05); in KO mice, the hypoxic group showed no significant changes in NOX4 mRNA expression (P>0.05), but had significantly reduced NOX2 mRNA expression (P<0.05), as compared with the normoxic group; the treatment group had reduced expression of NOX2 mRNA expression and increased NOX4 mRNA expression (P<0.05), as compared with the hypoxic group. CONCLUSIONS:eNOS plays a key role in the regulation of expression of NOX2 and NOX4 in the lungs exposed to hypoxia. It suggests that NOX and eNOS may physically interact with one another in pulmonary vascular remodeling induced by chronic hypoxia.
Effect of inhaled nitric oxide on aquaporin expression in newborn rat lung in acute hyperoxic lung injury

Abstract:AIM: To investigate the effect of inhaled nitric oxide on aquaporin expression and alveolar epithelial fluid transport in newborn rats with acute hyperoxic lung injury. METHODS: 32 newborn SD rats were randomized to breathe for 48 h room air (C), >95%O2 (O), >95%O2+10-5 NO (NO only in the first 24 h, ONO), room air + NO (CN). Then, the rats were killed, the lung wet-to-dry weight ratio (QW/QD), the histology, and AQP1, AQP5, α1-NKA, α-ENaC mRNA expressions in the lungs were measured. RESULTS: Compared with C group, the QW/QD in O group significantly increased (P<0.01), and AQP1 mRNA expression decreased significantly (P<0.01). Compared with O group, ONO group had a lower level of QW/QD (P<0.05), and AQP1 mRNA expression increased (P<0.05). AQP5 mRNA expression in all groups remained unchanged. CONCLUSION: In newborn rats with acute hyperoxic lung injury, inhaled 10-5 nitric oxide for 24 h may attenuate lung edema and increase AQP1 mRNA expression, suggesting that inhaled 10-5 nitric oxide for 24 h may promote the AQP1 expression in lung in this model of acute lung injury.
Expressions of NOX4 and eNOS in pulmonary vasculature in mouse model of chronic hypoxia

Abstract:Objective To investigate the pulmonary vascular endothelial nitric oxide synthase(eNOS)and NOX4 expressions in mice following chronic hypoxia.Methods C57BL/6J male mice were exposed to either normoxia or hypoxia(10±0.5% O2) for 1,3,7 days or 2 weeks.HE staining was performed to observe pulmonary vascular remodeling of chronic hypoxia in mice.Immunohistochemisty applied for detecting eNOS and α-SMA protein.The real-time PCR was used to investigate eNOS and NOX4 mRNA.Results Mice exposed to hypoxia exhibited significant pulmonary vascular remodeling characterized by thickening of the vascular wall and muscularization of previously nonmuscularized more distal small vessels.The expressions of mRNA of eNOS or NOX4 were significantly increased in the lungs in chronically hypoxic mice compared with normoxia controls.The immunohistochemical staining using an isoform-specific eNOS antibody demonstrated significant increase in eNOS expression in the lung vascular endothelium.Conclusion Both NOX4 and eNOS may play important roles in the pathogenesis of chronic hypoxia pulmonary hypertension.

Abstract:Objective: To investigate the impact of nitric oxide (NO) on platelet-derived growth factor (PDGF) expression in intrapulmonary arteries of rats with hypoxic pulmonary vascular structural remodeling and to examine the possible mechanism responsible for hypoxic pulmonary hypertension. Methods: Twenty-five male Wistar rats were randomly divided into four groups: nomoxic group (n=6), hypoxic group (n=6), hypoxic + L-arginine group (hypoxic + L-Arg group) (n=7) and hypoxic + N (1) -L-nitro-arginine methyl ester group (hypoxic + L-NAME group) (n=6). Relative medial thickness (RMT) of small and median muscularized pulmonary arteries and the percentage of muscularized arteries in small pulmonary arteries were observed in lung sections of rats by using a light microscope. Immunohistochemistry technique was used to assess the abundance and localization of PDGF in intrapulmonary arteries of rats in four groups. Results: RMT of pulmonary arteries and the percentage of muscularized arteries in small pulmonary arteries were obviously increased in hypoxic rats as compared with normal controls (P<0.01). L-arginine significantly reduced RMT and the percentage of muscularized arteries of chronically hypoxic rats (P<0.01). L-NAME, however, markedly increased RMT and the percentage of muscularized arteries of chronically hypoxic rats (P<0.01). There were PDGF expressions in intrapulmonary artery endothelial cells and smooth muscle cells in four groups. PDGF expressions were augmented in intrapulmonary artery endothelial cells and smooth muscle cells in hypoxic rats as compared with normal controls (P<0.01). L-Arg significantly inhibited the PDGF expressions by intrapulmonary artery endothelial cells and smooth muscle cells in hypoxic rats. However, L-NAME markedly strengthened the PDGF expressions by intrapulmonary artery endothelial cells and smooth muscle cells. There was a positive correlation between RMT and PDGF expression of endothelial cells and smooth muscle cells of median pulmonary arteries (r=0.8636, 0.8264, P<0.01, respectively). There was also a positive correlation between RMT and PDGF expression of endothelial cells and smooth muscle cells of small pulmonary arteries (r=0.8898, 0.8688, P<0.01, respectively). Conclusion: Nitric oxide inhibited PDGF expression in intrapulmonary arteries of rats with hypoxic pulmonary vascular structural remodeling.
The Role of Inos-Derived NO in the Antihypertrophic Actions of B-type Natriuretic Peptide in Neonatal Rat Cardiomyocytes

Abstract:In the infarcted rat heart, the increase of NO occurs in the hypertrophied myocardium of non-infarcted areas and its antihypertrophic efficacy has been well established. As another endogenous regulator and the reliable index of heart pathology, B-type natriuretic peptide also exhibits the antihypertrophic properties in many tissues by elevating intracellular cGMP. Several studies indicate that natriuretic peptides family may exert some actions in part via a nitric oxide pathway following receptor-mediated stimulation of iNOS. Therefore, it raises our great interest to ask what role NO plays in the antihypertrophic actions of B-type natriuretic peptide in cardiomyocytes. Incubation of cardiomyocytes under mild hypoxia for 12 h caused a significant increase in cellular protein content, protein synthesis and cell surface sizes. This growth stimulation was suppressed by exogenous B-type natriuretic peptide in a concentration dependent manner. Furthermore, the generation of intracellular cGMP, the upregulation of iNOS mRNA expression, the increase of iNOS activity and subsequent nitrite generation in hypertrophic cardiomyocytes was also increased by B-type natriuretic peptide. AG, a selective iNOS inhibitor, inhibited the upregulation of iNOS expression and the increase of iNOS activity by the combination of B-type natriuretic peptide/mild hypoxia or by the combination of 8-bromo-cGMP/mild hypoxia. Rp-8-br-cGMP, cGMP dependent protein kinase inhibitor, attenuated the actions of B-type natriuretic peptide and 8-bromo-cGMP which increases intracellular cGMP independent of B-type natriuretic peptide. In conclusion, our present data suggest that B-type natriuretic peptide exerted the antihypertrophic effects in cardiomyocytes, which was partially attributed to induction of iNOS-derived NO by cGMP pathway.
[Nitric Oxide Regulates C-Fos Expression in Osteoblastic Cells Induced by Wall-Shear].

Abstract:OBJECTIVE:To observe regulation of nitric oxide on c-fos expression in osteoblastic cells in response to changes in wall-shear stress in vitro.METHODS:Isolated and purified osteoblastic cells from the calvaria of newborn SD rats were cultured and passaged. The third generation cells, pre-treated with 10% FBS DMEM, 0.3 mmol/L L-NMMA DMEM and 0.1 mmol/L SNP DMEM separately, were subjected to wall-shear stress of 1.2 Pa. Gene expression of the c-fos and NOS activity were studied before (0 min) and 10 min, 15 min, 30 min, 60 min after treated with wall-shear stress.RESULT:The expression of c-fos mRNA was increased transiently after application of 1.2 Pa wall-shear stress in osteoblastic cells and peaked at 15 min. The expression of c-fos mRNA was decreased after pre-application with L-NMMA and increased after use of SNP.CONCLUSION:Changes in the osteoblastic cells mechanical environment may cause a dramatic induction of NO and c-fos expression.
Nitric Oxide Regulation of Myocardial O2 Consumption and HEP Metabolism.

Abstract:NO and O(2) compete at cytochrome-c oxidase, thus potentially allowing NO to modulate mitochondrial respiration. We previously observed a decrease of myocardial phosphocreatine (PCr)/ATP during very high cardiac work states, corresponding to an increase in cytosolic free ADP. This study tested the hypothesis that NO inhibition of respiration contributes to this increase of ADP. Infusion of dobutamine + dopamine (DbDp, each 20 microg.kg(-1).min(-1) iv) to more than double myocardial oxygen consumption (MVo(2)) in open-chest dogs caused a decrease of myocardial PCr/ATP measured with (31)P NMR from 2.04 +/- 0.09 to 1.85 +/- 0.08 (P < 0.05). Inhibition of NO synthesis with N(omega)-nitro-L-arginine (L-NNA), while catecholamine infusion continued, caused PCr/ATP to increase to the control value. In a second group of animals, L-NNA administered before catecholamine stimulation (reverse intervention of the first group) increased PCr/ATP during basal conditions. In these animals L-NNA did not prevent a decrease of PCr/ATP at the high cardiac work state but, relative to MVo(2), PCr/ATP was significantly higher after L-NNA. In a third group of animals, pharmacological coronary vasodilation with carbochromen was used to prevent changes in coronary flow that might alter endothelial NO production. In these animals L-NNA again restored depressed myocardial PCr/ATP during catecholamine infusion. The finding that inhibition of NO production increased PCr/ATP suggests that during very high work states NO inhibition of mitochondrial respiration requires ADP to increase to drive oxidative phosphorylation.
Influence of endothelin-1 on nitric oxide and hydrogen sulfide pathways in vascular smooth muscle cells in rats

Abstract:Objective To explore the effect of different concentrations of endothelin-1 (ET-1)on the en-dogenous nitric oxide (NO)and hydrogen sulfide (H2S)pathways of vascular smooth muscle cells (A7r5 cell lines)in rats.Methods A7r5 cell lines were divided into the control group and the experimental group.ET-1 at a concentra-tion of 10 -8-10 -6 mol/L was added into the experimental group,and as for the control group,the same volume of sterile phosphate buffered saline (PBS)buffer solution was added.The content of NO and H2S in A7r5 cell lines was detected by fluorescent NO probe and H2S probe after ET-1 stimulation for 48 h,respectively.The content of NO in the supernatant was measured by NO assay kit at 48 h of the incubation.The content of H2S in the supernatant was measured by polarographic H2S sensor at 48 h of the incubation. The expressions of inducible nitric oxide synthase (NOS2),endothelial nitric oxide synthase (NOS3),cystathionine -γ -lyase (CSE),cystathionine -β -synthase (CBS)and proliferating cell nuclear antigen (PCNA)were detected by the Western blot method.Results The rela-tive fluorescence intensity of the content of NO in the A7r5 cell lines of ET-1 10 -8,10 -7 and 10 -6mol/L groups (0. 078 ± 0. 080,0.075 ± 0.002,0.056 ± 0.009)was markedly lower than that in the control group(0.094 ± 0. 061), and the differences were statistically significant(F＝15.248,P＜0.05);Compared with the control group[(2. 131 ± 0. 484)μmol/L],the content of NO in the supernatant of the experimental groups [(1.391 ± 0.134 )μmol/L, (1.219 ± 0. 280)μmol/L,(1.116 ± 0.181)μmol/L]was significantly decreased,and the differences were statistically significant(F＝20.833,P＜0.01);NOS2 protein expression(0.457 ± 0.097,0.462 ± 0.116,0.438 ± 0.180)was decreased markedly compared with that of the control group(0.721 ± 0.222),and the differences were statistically sig-nificant(F＝6.196,P＜0.01),but the expression of NOS3 showed no significant differences(F＝2.669,P＞0.05). The relative fluorescence intensity of the content of H2S in the A7r5 cell lines of ET-1 10 -8,10 -7 and 10 -6mol/L groups (0.063 ± 0.002,0.056 ± 0.008,0.042 ± 0.009)was markedly lower than that in the control group (0.082 ± 0. 006),and the differences were statistically significant(F ＝16.297,P＜0.01);Compared with the control group [(29.439 ±4.236)μmol/L],the content of H2S in the supernatant of the experimental groups [(17.516 ±5.144) μmol/L,(14.481 ± 4.885)μmol/L]was significantly decreased,and the differences were statistically significant (F＝12.518,P ＜0.01).CBS protein expression(0.359 ± 0.096,0.270 ± 0.038,0.174 ± 0.051)was decreased markedly compared with that of the control group(0.707 ± 0.107),and the differences were statistically significant (F＝20.833,P＜0.01),and the expression of CSE showed no significant differences(F＝0.708,P＞0.05).The data showed that PCNA protein expression in the 10 -7mol/L ET-1 group(0.686 ± 0.180)significantly increased com-pared with that of the control group(0.437 ± 0.191),and the difference was statistically significant (t＝ -2.840,P＜0.01).Conclusion ET-1 stimulation can lead to the proliferation of vascular smooth muscle cells and down-regu-late its endogenous NO and H2S pathways.
Effects of Pharmacological Inhibitors of NADPH Oxidase on Myogenic Contractility and Evoked Vasoactive Responses in Rat Resistance Arteries

Abstract:Reactive oxygen species (ROS), such as superoxide anions and hydrogen peroxide, are reported to contribute to the dynamic regulation of contractility in various arterial preparations, however, the situation in pressurized, myogenically active resistance arteries is much less clear. In the present study, we have utilized established pharmacological inhibitors of NADPH oxidase activity to examine the potential contribution of ROS to intrinsic myogenic contractility in adult Sprague–Dawley rat resistance arteries and responses to vasoactive agents acting via the endothelium (i.e., acetylcholine, SKA-31) or smooth muscle (i.e., sodium nitroprusside, phenylephrine). In cannulated and pressurized cremaster skeletal muscle and middle cerebral arteries, the NOX inhibitors 2-acetylphenothiazine (2-APT) and VAS2870, selective for NOX1 and NOX2, respectively, evoked concentration-dependent inhibition of basal myogenic tone in a reversible and irreversible manner, respectively, whereas the non-selective inhibitor apocynin augmented myogenic contractility. The vasodilatory actions of 2-APT and VAS2870 occurred primarily via the vascular endothelium and smooth muscle, respectively. Functional responses to established endothelium-dependent and –independent vasoactive agents were largely unaltered in the presence of either 2-APT or apocynin. In cremaster arteries from Type 2 Diabetic (T2D) Goto-Kakizaki rats with endothelial dysfunction, treatment with either 2-APT or apocynin did not modify stimulus-evoked vasoactive responses, but did affect basal myogenic tone. These same NOX inhibitors produced robust inhibition of total NADPH oxidase activity in aortic tissue homogenates from control and T2D rats, and NOX isozymes 1, 2 and 4, along with superoxide dismutase 1, were detected by qPCR in cremaster arteries and aorta from both species. Based on the diverse effects that we observed for established, chemically distinct NOX inhibitors, the functional contribution of vascular NADPH oxidase activity to stimulus-evoked vasoactive signaling in myogenically active, small resistance arteries remains unclear.

Abstract:Background: Under hypoxia, exaggerated compensatory responses may lead to acute mountain sickness. The excessive vasodilatory effect of nitric oxide (NO) can lower the hypoxic pulmonary vasoconstriction (HPV) and peripheral blood pressure. While NO is catalyzed by various nitric oxide synthase (NOS) isoforms, the regulatory roles of these types in the hemodynamics of pulmonary and systemic circulation in living hypoxic animals remain unclear. Therefore, this study aims to investigate the regulatory effects of different NOS isoforms on pulmonary and systemic circulation in hypoxic rats by employing selective NOS inhibitors and continuously monitoring hemodynamic parameters of both pulmonary and systemic circulation. Methods: Forty healthy male Sprague-Dawley (SD) rats were randomly divided into four groups: Control group (NG-nitro-D-arginine methyl ester, D-NAME), L-NAME group (non-selective NOS inhibitor, NG-nitro-L-arginine methyl ester), AG group (inducible NOS inhibitor group, aminoguanidine), and 7-NI group (neurological NOS inhibitor, 7-nitroindazole). Hemodynamic parameters of rats were monitored for 10 min after inhibitor administration and 5 min after induction of hypoxia [15% O2, 2200 m a. sl., 582 mmHg (76.5 kPa), Xining, China] using the real-time dynamic monitoring model for pulmonary and systemic circulation hemodynamics in vivo. Serum NO concentrations and blood gas analysis were measured. Results: Under normoxia, mean arterial pressure and total peripheral vascular resistance were increased, and ascending aortic blood flow and serum NO concentration were decreased in the L-NAME and AG groups. During hypoxia, pulmonary arterial pressure and pulmonary vascular resistance were significantly increased in the L-NAME and AG groups. Conclusions: This compensatory mechanism activated by inducible NOS and endothelial NOS effectively counteracts the pulmonary hemodynamic changes induced by hypoxic stress. It plays a crucial role in alleviating hypoxia-induced pulmonary arterial hypertension.

After 21 days, mean pulmonary artery pressure (mPAP) of each rat was evaluated and the ratio of right ventricular mass to left ventricular plus septum mass was detected. Meanwhile, the production of NO in pulmonary tissue, the plasma concentration of H 2S and the activity of CSE in pulmonary tissue were detected.Results Mean PAP and RV/LV+SP significantly increased in hypoxic rats as compared with those of normal controls ( P 0.01). Meanwhile, compared with controls, the production of NO in pulmonary tissue, the plasma concentration of H 2S and the activity of CSE in pulmonary tissue markedly decreased in hypoxic rats( P 0.01, respectively). However, mPAP and RV/LV+SP were significantly increased in hypoxic + L-NAME groups as compared with hypoxic groups( P 0.05), at the same time, the production of NO in pulmonary tissue was markedly decreased ( P 0.01 , respectively). However, the plasma concentration of H 2S and the activity of CSE in pulmonary tissue in hypoxic + L-NAME group increased significantly as compared with those of hypoxic group( P 0.05).Conclusion The endogenous nitric oxide might inhibit H 2S production in pulmonary artery of hypoxic rats.

Abstract:Nitric oxide (NO),well known as the endothelium-derived relaxing factor (EDRF),has become increasingly recognized as an important determinant of cardiovascular diseases.A molecular pathway by which NO accomplishes various biochemical processes is the cGMP-independent post-translational modification of protein,S-nitrosylation.Here we review the biochemistry and regulating mechanisms of NO-related S-nitrosylation of proteins,and discuss the multiple roles of S-nitrosylation in vascular functions and pathology in particular.

Abstract:Nitric oxide (NO) inhibits vascular inflammation, but the molecular basis for its anti-inflammatory properties is unknown. We show that NO inhibits exocytosis of Weibel-Palade bodies, endothelial granules that mediate vascular inflammation and thrombosis, by regulating the activity of N-ethylmaleimide-sensitive factor (NSF). NO inhibits NSF disassembly of soluble NSF attachment protein receptor (SNARE) complexes by nitrosylating critical cysteine residues of NSF. NO may regulate exocytosis in a variety of physiological processes, including vascular inflammation, neuro-transmission, thrombosis, and cytotoxic T lymphocyte cell killing.
Selective and sensitive fluorescence imaging reveals microenvironment-dependent behavior of NO modulators in the endothelial system

Abstract:1)An imaging based method for evaluating the efficacy of NO modulators in live cells was developed.Some NO modulators were observed yielding unexpected effects on cellular NO levels in some endothelial cell lines.Further study unveiled that intracellular microenvironments have profound effects on the efficacy of NO modulators in these cell lines.This result highlights the pitfall of the specific cellular microenvironments on drug efficacy.Nitric oxide (NO) is a second messenger playing crucial roles in the signaling of a variety of cellular functions. Due to its pathophysiological significance, various NO modulators have been developed to interrogate NO pathways and some have been used as therapies. These modulators are often used directly to observe pharmacological effects in cell lines but their actual effect on intracellular NO level is seldom analyzed. Herein, facilitated by a selective and sensitive fluorescence probe, we observed that some NO modulators display unexpected behaviors with both NO scavenger carboxy-PTIO and endothelial nitric oxide synthase (eNOS) inhibitor N(ω)-nitro-l-arginine methyl ester (l-NAME) failing to decrease intracellular free NO level in EA. hy926 cells while NO donor diethylamine-NONOate (DEA·NONOate) and eNOS activator calcimycin (A23187) failing to increase free NO level in human umbilical vein endothelial cell line (HUV-EC-C), although the reagents were confirmed to work normally in the primary human umbilical vein endothelial cell (primary HUVEC) and RAW 264.7 macrophage cells. Further research suggests that these unusual behaviors may be attributed to the cellular microenvironments including both the NO synthase (NOS) level and the endogenous glutathione (GSH) level. Genetically manipulating eNOS level in both cells restores the expected response, while decreasing GSH level restores the ability of DEA·NONOate to increase NO level in HUV-EC-C. These results reveal that the cellular microenvironment has a profound impact on pharmacological effect. Abstract:Introduction: Hypoxic persistent pulmonary hypertension in the newborn (PPHN) is usually treated with oxygen and inhaled nitric oxide (NO), both pulmonary arterial relaxants. But treatment failure with NO occurs in 25% of cases. We previously demonstrated that 72 h exposure to hypoxia, modeling PPHN, sensitized pulmonary artery smooth muscle cells (PASMC) to the contractile agonist thromboxane and inhibited relaxant adenylyl cyclase (AC) activity. Methods: In this study, we examined the effects of sodium nitroprusside (SNP), as NO donor, on the thromboxane-mediated contraction and NO-independent relaxation pathways and on reactive oxygen species (ROS) accumulation in PASMC. In addition, we examined the effect of the peroxynitrite scavenger 5,10,15,20-Tetrakis (4-sulfonatophenyl)porphyrinato Iron (III) (FeTPPS) on these processes. Results: Exposure of PASMC to 72 h hypoxia increased total intracellular ROS compared to normoxic control cells and this was mitigated by treatment of cells with either SNP or FeTPPS. Total protein nitrosylation was increased in hypoxic PASMC compared to controls. Both normoxic and hypoxic cells treated with SNP exhibited increased total protein nitrosylation and intracellular nitrite; this was reduced by treatment with FeTPPS. While cell viability and mitochondrial number were unchanged by hypoxia, mitochondrial activity was decreased compared to controls; addition of FeTPPS did not alter this. Basal and maximal mitochondrial metabolism and ATP turnover were reduced in hypoxic PASMC compared to controls. Hypoxic PASMC had higher basal Ca2+, and a heightened peak Ca2+ response to thromboxane challenge compared to controls. Addition of SNP further elevated the peak Ca2+ response, while addition of FeTPPS brought peak Ca2+ response down to control levels. AC mediated relaxation was impaired in hypoxic PASMC compared to controls but was normalized following treatment with FeTPPS. Addition of SNP inhibited adenylyl cyclase activity in both normoxic and hypoxic PASMC. Moreover, addition of the Ca2+ chelator BAPTA improved AC activity, but the effect was minimal. Discussion: We conclude that NO independently augments contraction and inhibits relaxation pathways in hypoxic PASMC, in part by a mechanism involving nitrogen radical formation and protein nitrosylation. These observations may partially explain impaired effectiveness of NO when treating hypoxic pulmonary hypertension.

Abstract:By regulation the expression of nitric oxide synthase (NOS) mRNA, endothelin binded ET_(A)—R and ET_(B)—R on the vascular smooth muscle cells (VSMC) of rat affects the mitogenesis and proliferation of VSMC with reverse transcription-polymerase chain reaction(RT-PCR) and ~(3)H-thymidine incorporation. Interleulinl enhances the expression of NOS gene and affects the mitogenesis of VSMC partly through the pathway of endothelin and its receptors.

Abstract:The study aimed to investigate the regulation of heme oxygenase/carbon monoxide system of pulmonary arteries by endogenous nitric oxide in hypoxic rats so as to explore the regulatory mechanism responsible for hypoxic pulmonary vascular structural remodeling. Twenty-one rats were randomly divided into three groups: normoxia group, hypoxia group and hypoxia +L-NAME group. Immunohistochemistry technique was used to assess the abundance and localization of heme oxygenase-I in pulmonary arterial smooth muscle cells. At the same time the levels of carbon monoxide in plasma and lung tissue homogenates were detected by spectrophotometer. The expression of heme oxygenase-1 by pulmonary arteries smooth muscle cells of hypoxic rats increased significantly compared to that in normoxic rats (P < 0.01). However, when L-NAME was used, expression of heme oxygenase-1 by pulmonary artery smooth muscle cells of hypoxic rats decreased significantly compared to that in rats of hypoxia group (P < 0.01). The levels of carbon monoxide in plasma and lung tissue homogenates of hypoxia rats also increased significantly in hypoxic rats as compared to those of normoxic rats (P < 0.0l). But they were markedly reduced in rats of hypoxia +L-NAME group compared to hypoxia group (P < 0.01). The results suggest that endogenous nitric oxide might regulate pulmonary circulation via carbon monoxide pathway under hypoxia.

Abstract:Induction of the inducible form of nitric oxide synthase (iNOS) in the vascular and cardiac tissue by several inflammatory stimuli may result in the production of large amounts of nitric oxide (NO) for a sustained period. Recent data obtained in the rat aorta in which iNOS was induced by lipopolysaccharide (LPS) have demonstrated that adventitial cells represent the main site of NO production. Adventitial-derived NO can exert an immediate down-regulatory effect on smooth muscle contraction (via activation of the cyclic GMP pathway) but may also initiate longer lasting effects through the formation of NO stores within the medial layer. One candidate for such NO stores are dinitrosyl non-heme iron complexes. Low molecular weight thiols interact with preformed NO stores and promote vasorelaxation by a cyclic GMP-independent mechanism involving the activation of potassium channels. In the heart, the induction of iNOS is involved in delayed protection against ischemia-reperfusion-induced functional damages. Recent data obtained with monophosphoryl lipid A, a non-toxin derivative of LPS, strongly suggest that iNOS-derived NO in the rat heart does not act as an immediate mediator of the cardioprotection but rather as a trigger of long-term protective mechanisms. Thus, the present data reveal the important role of adventitial cells as a site of iNOS expression and activity in intact blood vessels. The induction of adaptive mechanisms in the heart and the formation of releasable NO stores in blood vessels are examples of long-term consequences of iNOS induction. These new information are relevant for a better understanding of the circumstances in which NO overproduction by iNOS may play either a beneficial or deleterious role in these tissues.
