Abstract:Objective To determine the expression pattern of mcpr1 in murine palatogenesis between the normal murine palatal shelves and the cleft palatal ones.Methods The murine embryonic model with cleft palate was established by inducing with retinoic acid.Immunohistochemical staining was used to detect the expression of mcpr1 gene during the murine embryonic development.Results The embryonic mouse with cleft palate was established successfully.At embryonic day 12(E12),when palatal shelves have elongated and grown vertically,MCPR1 was expressed strongly in epithelial cells in normal palatal shelves,and in mesenchymal cells in cleft palatal shelves.At E16,MCPR1 was expressed in the mesenchymal cells near the epithelial components.At the corresponding time points,the expression of MCPR1 protein was stronger in normal palatal shelves than in cleft ones respectively.Conclusion The expression of MCPR1 protein was different between the normal and the cleft palate during the palatalogenesis.

Abstract:AIM:To investigate the expression of mouse cleft palate related gene in murine embryonic palatal ectomesenchymal (MEPM) cell,mandibular ectomesenchymal cell,dental follicle cell,dental pulp cell and periodontal ligament cell.METHODS:Primary embryonic palatal ectomesenchymal cell cultures were done.MEPM cells,ectomesenchymal cells of human mandibular process,dental follicle cells,dental pulp cells and periodontal ligament cells were detected by immunohistochemistry to investigate the expression of MCRP1.RESULTS:Primary MEPM cells were established and transferred successfully.The result of immunohistochemistry showed that MCPR1 expressed in cytoplasm,but not nuclei.It expressed strongly in MEPM cells and periodontal ligament cells,but weakly in mandibular ectomesenchymal cells,dental follicle cells and dental pulp cells.CONCLUSION: MCPR 1 expressed mainly in cytoplasm of different cells in different levels,which inferred that MCPR 1 might take part in the development of many organs,and expressed differently during different periods of embryonic development. Abstract:AIM:To investigate the expression of mouse cleft palate related gene in different patterns during murine embryonic development.METHODS:Gestation day(GD) 12,14,18 C57BL/6N pregnant female murine were sacrificed.The fetus were immediately removed from uterus and were detected in polyclonal antibodies of MCPR1 by immunohistochemistry to investigate the expression of MCRP1.RESULTS:The results of immunohistochemistry showed that the MCPR1 expressed extensively in different patterns during murine embryonic development,however there was no tissue specification or obvious tendency.CONCLUSION:MCPR1 expressed differently in different tissue with different patterns.According to which it could be inferred that the MCPR1 might take part in the development of many organs during different periods of embryonic development.
Identification and Characterization of a Novel Gene, Mcpr1, and Its Possible Function in the Proliferation of Embryonic Palatal Mesenchymal Cells

Abstract:We cloned a novel mouse cDNA, Mcpr1 (mouse cleft palate-related gene 1), between retinoic acid (RA)-treated murine embryonic palatal and control shelves by improved subtractive hybridization. Its transcript was identified by Northern blotting. The open reading frame encodes 132 amino acids and shows almost no identity to other genetic products. Mcpr1 expression could be detected extensively in adult mouse tissues and during murine embryonic development. It was identified to be significantly stimulated by RA in murine palatal shelves at embryonic day 12 and in palatal mesenchymal cells in vitro. We demonstrate that MCPR1 protein was localized primarily in the cytoplasm and could be synthesized and secreted by transfected COS-7 cells. Both the secretory and recombinant proteins of Mcpr1 inhibited proliferation of murine embryonic palatal mesenchymal cells and impeded the progression from the G1 to S phase in the cell cycle. The cells were prone to apoptosis after exposure to glutathione S-transferase-MCPR1. Furthermore, knockdown of MCPR1 protein levels by antisense oligodeoxynucleotides promoted progression of cells from the G1 to S phase and completely abolished the RA-induced block of the cell cycle from the G1 to S phase. These findings suggest that Mcpr1 might function as one of the RA-up-regulated genes involved in inhibiting cell proliferation during palatogenesis and RA-induced cleft palate by regulating proliferation and apoptosis of embryonic palatal mesenchymal cells and might even play a role in the development of many other organs.
Immunolocalization of epidermal growth factor (EGF), EGF receptor and transforming growth factor alpha (TGF alpha) during murine palatogenesis in vivo and in vitro.

Abstract:The distribution of epidermal growth factor, the epidermal growth factor receptor and transforming growth factor alpha during murine palatogenesis was investigated immunocytochemically. On embryonic day 12 staining for transforming growth factor alpha was present throughout the palatal mesenchyme, with little in the epithelia. On embryonic day 13 staining increased in the palatal epithelia and in the mesenchyme at the tip of the palate. As the palatal shelves fused together (embryonic day 14.5) intense staining for transforming growth factor alpha was seen in the midline epithelial seam and in the subjacent mesenchyme. On embryonic day 15 there was a generalised increase in palatal epithelial staining; this was most marked in the remnants of the degenerating epithelial seam. Mesenchymal staining was, however, uniform. Whilst palatal staining for epidermal growth factor was sparse, at all stages, staining for its receptor was present throughout the palatal epithelia and mesenchyme. This was most intense in the palatal medial edge epithelia at the time of midline epithelial seam degeneration. The regional and temporal differences in staining for the epidermal growth factor receptor and transforming growth factor alpha suggested that these molecules may play an important role in normal palate development in vivo, particularly in degeneration of the midline epithelial seam.

Characterization of Pancreatic Lesions from MT-tgf Alpha, Ela-myc and MT-tgf Alpha/ela-Myc Single and Double Transgenic Mice.

Abstract:In order to identify good animal models for investigating therapeutic and preventive strategies for pancreatic cancer, we analyzed pancreatic lesions from several transgenic models and made a series of novel findings. Female MT-tgf alpha mice of the MT100 line developed pancreatic proliferation, acinar-ductal metaplasia, multilocular cystic neoplasms, ductal adenocarcinomas and prominent fibrosis, while the lesions in males were less severe. MT-tgf alpha-ES transgenic lines of both sexes developed slowly progressing lesions that were similar to what was seen in MT100 males. In both MT100 and MT-tgf alpha-ES lines, TGF alpha transgene was expressed mainly in proliferating ductal cells. Ela-myc transgenic mice with a mixed C57BL/6, SJL and FVB genetic background developed pancreatic tumors at 2-7 months of age, and half of the tumors were ductal adenocarcinomas, similar to what was reported originally by Sandgren et al 1. However, in 20% of the mice, the tumors metastasized to the liver. MT100/Ela-myc and MT-tgf alpha-ES/Ela-myc double transgenic mice developed not only acinar carcinomas and mixed carcinomas as previously reported but also various ductal-originated lesions, including multilocular cystic neoplasms and ductal adenocarcinomas. The double transgenic tumors were more malignant and metastasized to the liver at a higher frequency (33%) compared with the Ela-myc tumors. Sequencing of the coding region of p16ink4, k-ras and Rb cDNA in small numbers of pancreatic tumors did not identify mutations. The short latency for tumor development, the variety of tumor morphology and the liver metastases seen in Ela-myc and MT-tgf alpha/Ela-myc mice make these animals good models for investigating new therapeutic and preventive strategies for pancreatic cancer.

Abstract:OBJECTIVE:To invesitgate the expression patterns of amelogenin and enamelin in the developing tooth germs.METHODS:Mandible sections of postnatal day 1, 3, 7 and 14 mouse were prepared, immunohistochemical analysis and reverse transcriptase polymerase chain reaction (RT-PCR) were performed to detect the expression patterns of amelogenin and enamelin in mandibular first molars.RESULTS:Amelogenin was observed in the cytoplasm of secretory ameloblasts and the whole enamel matrix layer. It was also transiently expressed in the odontoblasts of postnatal day 1 molars. Enamelin proteins were observed in the enamel layer deposited by secretory ameloblasts, especially intense beneath the ameloblast process and dentino-enamel junction. The mRNA levels of both amelogenin and enamelin were highest on postnatal day 7 (the ratio to glyceraldehyde phosphate dehydrogenase of amelogenin and enamelin: 0.813 ± 0.085 and 0.799 ± 0.064, respectively, P < 0.05).CONCLUSIONS:Amelogenin and enamelin were enamel matrix proteins predominately expressed by secretory ameloblasts. The temporal-spatial expression patterns of amelogenin and enamelin indicate the important roles they played in amelogenesis and biomineralization.
Spatiotemporal Evolution of Developing Palate in Mice

Abstract:Journal of Dental Research, Ahead of Print. The intricate formation of the palate involves a series of complex events, yet its mechanistic basis remains uncertain. To explore major cell populations in the palate and their roles during development, we constructed a spatiotemporal transcription landscape of palatal cells. Palate samples from C57BL/6 J mice at embryonic days 12.5 (E12.5), 14.5 (E14.5), and 16.5 (E16.5) underwent single-cell RNA sequencing (scRNA-seq) to identify distinct cell subsets. In addition, spatial enhanced resolution omics-sequencing (stereo-seq) was used to characterize the spatial distribution of these subsets. Integrating scRNA-seq and stereo-seq with CellTrek annotated mesenchymal and epithelial cellular components of the palate during development. Furthermore, cellular communication networks between these cell subpopulations were analyzed to discover intercellular signaling during palate development. From the analysis of the middle palate, both mesenchymal and epithelial populations were spatially segregated into 3 domains. The middle palate mesenchymal subpopulations were associated with tooth formation, ossification, and tissue remodeling, with initial state cell populations located proximal to the dental lamina. The nasal epithelium of the palatal shelf exhibited richer humoral immune responses than the oral side. Specific enrichment of Tgfβ3 and Pthlh signals in the midline epithelial seam at E14.5 suggested a role in epithelial–mesenchymal transition. In summary, this study provides high-resolution transcriptomic information, contributing to a deeper mechanistic understanding of palate biology and pathophysiology.

Abstract:BACKGROUND: The submandibular gland in mice is a typical study model for salivary development. Transforming growth factor beta is an essential growth factor during development and diseases. Till now the receptor of transforming growth factor beta and its function during salivary development has not been known well. OBJECTIVE: To observe the expression of transforming growth factor beta typeⅠreceptor (Tgfbr1), transforming growth factor beta type Ⅱ receptor (Tgfbr2) and phospholated extracellular signal-regulated kinase 1/2 in mouse submandibular glands in order to uncover the mechanism of transforming growth factor beta signaling in gland development. METHODS: Male and female C57BL/6J mice were mated to get embryos at embryonic day 14.5 (E14.5) and newborn stage. The samples were fixed in 4% paraformaldehyde and processed into paraffin embedded serial sections. The sections were stained with hematoxylin and eosin and immunohistochemistry by the antibodies of Tgfbr1, Tgfbr2 and phospholated extracellular signal-regulated kinase 1/2 using standard procedures. RESULTS AND CONCLUSION: (1) At newborn stage, the submandibular gland was located under the mandible in mice. The submandibular gland alveolus, gland duct including intercalary duct developed well. (2) At E14.5, Tgfbr1 and Tgfbr2 were expressed strongly in the submandibular glands epithelium, but not in the mesenchyme. (3) Expression of phospholated extracellular signal-regulated kinase 1/2, similar to Tgfbr1 and Tgfbr2, was only detected in the epithelium. Therefore, transforming growth factor beta might regulate extracellular signal-regulated kinase signaling in epithelial cells of the submandibular glands in order to control the submandibular gland development.
α‐Amylase expressed in human small intestinal epithelial cells is essential for cell proliferation and differentiation

Abstract:α‐Amylase, which plays an essential role in starch degradation, is expressed mainly in the pancreas and salivary glands. Human α‐amylase is also detected in other tissues, but it is unclear whether the α‐amylase is endogenously expressed in each tissue or mixed exogenously with one expressed by the pancreas or salivary glands. Furthermore, the biological significance of these α‐amylases detected in tissues other than the pancreas and salivary glands has not been elucidated. We discovered that human α‐amylase is expressed in intestinal epithelial cells and analyzed the effects of suppressing α‐amylase expression. α‐Amylase was found to be expressed at the second‐highest messenger RNA level in the duodenum in human normal tissues after the pancreas. α‐Amylase was detected in the cell extract of Caco‐2 intestinal epithelial cells but not secreted into the culture medium. The amount of α‐amylase expressed increased depending on the length of the culture of Caco‐2 cells, suggesting that α‐amylase is expressed in small intestine epithelial cells rather than the colon because the cells differentiate spontaneously upon reaching confluence in culture to exhibit the characteristics of small intestinal epithelial cells rather than colon cells. The α‐amylase expressed in Caco‐2 cells had enzymatic activity and was identified as AMY2B, one of the two isoforms of pancreatic α‐amylase. The suppression of α‐amylase expression by small interfering RNA inhibited cell differentiation and proliferation. These results demonstrate for the first time that α‐amylase is expressed in human intestinal epithelial cells and affects cell proliferation and differentiation. This α‐amylase may induce the proliferation and differentiation of small intestine epithelial cells, supporting a rapid turnover of cells to maintain a healthy intestinal lumen.

Selachian tooth development: II. Immunolocalization of amelogenin polypeptides in epithelium during secretory amelogenesis in Squalus acanthias

Abstract:We have determined the distribution of amelogenin polypeptides in an order of elasmobranchs using indirect immunofluorescence with rabbit polyclonal antibodies prepared to purified murine amelogenins. We find that amelogenins are definitely present within the inner enamel epithelium prior to the production of the extracellular matrix component termed "enameloid" (row II developing tooth organs). During subsequent stages of selachian tooth development (row III tooth organs), immunofluorescence staining data indicated localization of amelogenin antigens within epithelium as well as the enameloid extracellular matrix. The results from these immunohistochemical studies suggest that the 16-20 kdalton amelogenins, which are characteristic of murine inner enamel epithelial cells undergoing terminal biochemical differentiation into secretory ameloblasts, may also be regarded as molecular markers for amelogenesis in developing teeth in the spiny dogfish, Squalus acanthias.

Abstract:Objective: To compare the different expression of amelogenin(AMELX), ameloblastin(AMBN) and enamelin(ENAM) in tooth germs, thus to clarify the functions of these proteins in the development of tooth. Methods: The mice whose tooth germs were in bell stage, late bell stage and enamel mature stage were selected. The expression of these three proteins in the sagital sections of these mice was detected by immunohistochemistry, and EnVision two steps method was applied. The antibodies were presented from Dr. Simmer. Results: Immunohistochemistry showed that the temporal and spatial expression patterns of AMELX, AMBN and ENAM in ameloblasts and enamel matrix were different. With the mineralization of enamel matrix, expression of AMELX decreased, but it weakly expressed in adjacent bone. At the same time, expression of AMBN and ENAM were more stable. Conclusion: AMELX, AMBN and ENAM are the major matrix proteins of enamel, and play different roles during different developing stages of tooth germs. AMELX may facilitate the growth of crystals in the early mineral stage, and act in the repairing of bone. AMBN may promote the mineralization of enamel matrix, and also keep the shape and size of crystals. ENAM may act in remineralization and promoting the growth of crystals.
Selective Fluorescence of Zymogen Granules of Pancreatic Acinar Cells Stained with Hematoxylin and Eosin

Abstract:Following staining with hematoxylin and eosin Y, paraffin sections of mouse pancreas were examined by transmitted light, epifluorescence and confocal laser scanning microscopy. Light microscopy revealed that the nuclei of pancreatic acinar cells were located basally, while the apices of the cells appeared eosinophilic, although the secretory granules were difficult to visualize. Under violet-blue light excitation, the zymogen granules at the apices of the acinar cells showed strong yellowish fluorescence; the other part of the cytoplasm was only faintly fluorescent and the nuclei and the supporting tissues were nonfluorescent. Confocal laser scanning microscopy resulted in clear pictures of the zymogen granules and their distribution within the cell. The fluorescent emission of zymogen granules was certainly the result of eosin Y staining, because hematoxylin is not a fluorochrome and the zymogen granules are not autofluorescent. Staining with eosin Y alone, however, did not result in clear fluorescent images of zymogen granules or any other cellular structures. Our observation shows that the fluorescence emission of eosin Y allows easy and precise recognition of zymogen granules of pancreatic cells.
Single-Cell RNA-Seq Analysis Identifies Angiotensinogen and Galanin as Unique Molecular Markers of Acinar Cells in Murine Salivary Glands

Abstract:The submandibular gland (SMG) and sublingual gland (SLG) are two of three major salivary glands in mammals and comprise serous and mucous acinar cells. The two glands share some functional properties, which are largely dependent on the types of acinar cells. In recent years, while ScRNA-seq (single-cell sequencing) with a 10 × platform has been used to explore molecular markers in salivary glands, few studies have examined the acinar heterogeneity and unique molecular markers between SMG and SLG. This study aimed to identify the molecular markers of acinar cells in the SLG and SMG. We performed ScRNA-seq analyses in 4-week-old mice and verified the screened molecular markers using reverse transcription-quantitative real-time PCR, immunohistochemistry, and immunofluorescence. Our results showed prominently heterogeneous acinar cells, although there was great similarity in the cluster composition between the two glands at 4 weeks. Furthermore, we demonstrated that Agt is a specific marker of SMG serous acinar cells, whereas Gal is a specific marker of SLG mucous acinar cells. Trajectory inference revealed that Agt and Gal represent two types of differential acinar cell clusters during late development in adults. Thus, we reveal previously unknown specific markers for salivary acinar cell diversity, which has extensive implications for their further functional research.

Abstract:The nucleotide sequence of the coding and regulatory regions of the alpha-amylase gene (aml) of Streptomyces limosus was determined. High-resolution S1 mapping was used to locate the 5' end of the transcript and demonstrated that the gene is transcribed from a unique promoter. The predicted amino acid sequence has considerable identity to mammalian and invertebrate alpha-amylases, but not to those of plant, fungal, or eubacterial origin. Consistent with this is the susceptibility of the enzyme to an inhibitor of mammalian alpha-amylases. The amino-terminal sequence of the extracellular enzyme was determined, revealing the presence of a typical signal peptide preceding the mature form of the alpha-amylase.

Amylase secretion by cultured porcine parotid cells

Abstract:The similarity of porcine and human physiology and the availability of slaughterhouse tissues suggests the use of porcine parotid cells as a model for amylase secretion. A procedure is described for the isolation of porcine parotid cells by collagenase-P/dispase digestion of the tissue. The preparation consisted of individual cells and small aggregates that were maintained in primary culture, during which the cells formed aggregates that firmly attached to the plastic substrate. The amylase content of the cultured cells remained adequate for assay of secretory activity during culture for one week after isolation. Depending upon variations in experimental treatments, the cultured cells secreted approx. 35-65% of cellular amylase in response to a carbachol challenge. The cells were slightly responsive to long exposures to isoproterenol, and were unresponsive to nicotine, elevated extracellular K+ or substance P. Secretion induced by carbachol required extracellular Ca2+, was inhibited by atropine and occurred with a nearly linear response over a 30-min period. The Ca2+ ionophore A23187 was also a potent secretagogue for amylase secretion, producing levels of secretion equal to that induced by carbachol. The ease of preparation and the retention of amylase during primary culture suggests that the preparation will be useful in studies on muscarinic receptor-mediated control of amylase secretion.
Alpha‐fetoprotein is Dynamically Expressed in Rat Pancreas During Development

Abstract:To identify proteins involved in pancreatic development, we used a differential proteomics approach by comparing pancreatic extracts from four biologically significant stages of development: embryonic day (E) 15.5, E18.5, postnatal (P) days 0 and adult. By two-dimensional gel electrophoresis (2D-E) and MALDI-TOF MS (Matrix Assisted Laser Desorption/Ionization Time-Of-Flight Mass Spectrometry) following database searching and protein annotation, 15 proteins were identified as being differently expressed in the pancreas between the four phases. The expression pattern and the localization of alpha-fetoprotein (AFP), one of significant changed proteins observed, were further determined. Four isoforms of AFP (72 kDa, 60 kDa, 48 kDa and 37 kDa) were found by Western blotting in the pancreas tested, most of them showed a stronger signal in E18.5 followed by a steady decrease and only a 60-kDa isoform was detected in the adult pancreas. Immunolocalization for AFP revealed that a positive reactivity was detectable at E15.5 pancreas, became stronger in the cytoplasm of mesenchyme cells at E18.5, and declined after birth to a nearly undetectable level in adults. Prohormone convertases in mouse submandibular gland: co-localization of furin and nerve growth factor

Abstract:Nerve growth factor (NGF) in mouse submandibular glands (SGs) is generated from a 35-kD precursor by proteolytic enzymes that have yet to be identified. Prohormone convertases (PCs) cleave the NGF precursor in vitro, and in this study we questioned whether PCs could process salivary NGF in vivo. mRNA coding for PC2 (but not PC1) was detected on Northern blots of SG mRNA and also by in situ hybridization within parasympathetic neurons of intralobular ganglia. Northern blot and in situ hybridization analyses also detect mRNA coding for furin. In SGs of male mice, furin mRNA levels are high at birth and remain high throughout development. In glands from female mice, levels decline during postnatal development and are lower in adults than in newborns. Immunocytochemistry detects furin immunoreactivity in pro-acinar and ductal cells of glands from newborn and pubescent mice. In glands of adults, furin immunoreactivity is detectable in acinar cells but highest levels are present in NGF-containing granular convoluted tubule cells. These data, taken together with those from previous studies, suggest that furin is a candidate processing enzyme for NGF in mouse submandibular glands.
Expression of Muc19/Smgc gene products during murine sublingual gland development: cytodifferentiation and maturation of salivary mucous cells

Abstract:Muc19/Smgc expresses two splice variants, Smgc (submandibular gland protein C) and Muc19 (mucin 19), the latter a major exocrine product of differentiated murine sublingual mucous cells. Transcripts for Smgc were detected recently in neonatal sublingual glands, suggesting that SMGC proteins are expressed during initial salivary mucous cell cytodifferentiation. We therefore compared developmental expression of transcripts and translation products of Smgc and Muc19 in sublingual glands. We find abundant expression of SMGC within the initial terminal bulbs, with a subsequent decrease as Muc19 expression increases. During postnatal gland expansion, SMGC is found in presumptive newly formed acinar cells and then persists in putative acinar stem cells. Mucin levels increase 7-fold during the first 3 weeks of life, with little change in transcript levels, whereas between postnatal days 21 and 28, there is a 3-fold increase in Muc19 mRNA and heteronuclear RNA. Our collective results demonstrate the direct transition from SMGC to Muc19 expression during early mucous cell cytodifferentiation and further indicate developmentally regulated changes in Muc19/Smgc transcription, alternative splicing, and translation. These changes in Muc19/Smgc gene expression delineate multiple stages of salivary mucous cell cytodifferentiation and subsequent maturation during embryonic gland development through the first 4 weeks of postnatal life.
Analysis of low abundance membrane-associated proteins from rat pancreatic zymogen granules

Abstract:Zymogen granules (ZG) are specialized storage organelles in the exocrine pancreas that allow the sorting, packaging, and regulated apical secretion of digestive enzymes. As there is a critical need for further understanding of the key processes in regulated secretion to develop new therapeutic options in medicine, we applied a suborganellar proteomics approach to identify peripheral membrane-associated ZG proteins. We focused on the analysis of a "basic" group (pH range 6.2-11) with about 46 spots among which 44 were identified by tandem mass spectrometry. These spots corresponded to 16 unique proteins, including rat mast cell chymase (RMCP-1) and peptidyl-prolyl cis-trans isomerase B (PpiB; cyclophilin B), an ER-resident protein. To confirm that these proteins were specific to zymogen granules and not contaminants of the preparation, we conducted a series of validation experiments. Immunoblotting of ZG subfractions revealed that chymase and PpiB behaved like bona fide peripheral membrane proteins. Their expression in rat pancreas was regulated by feeding behavior. Ultrastructural and immunofluorescence studies confirmed their ZG localization. Furthermore, a chymase-YFP fusion protein was properly targeted to ZG in pancreatic AR42J cells. Interestingly, for both proteins, proteoglycan-binding properties have been reported. The importance of our findings for sorting and packaging during ZG formation is discussed.
