Abstract:<p>The developmental neurotoxicity of calcium cyclamate was evaluated in Sprague Dawley [Crl:CD(SD)] rats, administered in drinking water, in comparison to a concurrent control group (water) and a positive control group given propylthiouracil (PTU). Calcium cyclamate was administered to F0 females for 4 weeks prior to pairing, throughout mating, gestation and lactation and to F1 offspring from weaning to 12 weeks of age, PTU was administered by gavage to F0 females from Day 6 of gestation up to Day 20 of lactation. Target calcium cyclamate doses were 0, 250, 500 and 1,000 mg/kg bw/day, while the PTU dose was 0.5 mg/kg bw/day. No treatment-related effects of cyclamate were observed in either the F0 or F1 generations on reproductive performance or neurobehavioral development. In comparison, PTU exposure resulted in developmental delays, memory impairment and a number of neuropathological and morphometric outcomes. The results from the unique developmental neurotoxicity study design, corroborate the absence of hyperactivity and any other neurotoxic effects following cyclamate administration at levels up to 878 mg/kg bw/day in F0 females and 784 mg/kg bw/day in F1 animals. This demonstrates the suitability of PTU as a positive control and confirms the safe use of cyclamate as a no-calorie sweetener.</p>

Abstract:CALCIUM cyclamate is widely used as a sweetening agent in foods and beverages. We now find that oral administration of calcium cyclamate to hamsters induces myocardial lesions accompanied by coronary sclerosis as well as soft-tissue calcification in other organs. This adverse effect of calcium cyclamate overdosage remained unnoticed in earlier toxicological tests1–3 and it seems from our present studies that cyclamate may enhance the potentially toxic actions of the cation with which it forms a stable, water-soluble salt.

Abstract:Calcium cyclamate, an artificial sweetener, was studied for its effectiveness in inducing transmissible chromosomal aberrations in germ cells of male mice. Both the dominant-lethal and the heritable translocation tests were carried out following daily treatment (on weekdays) of males by oral intubation with the maximum tolerated dose for 6 weeks. Calcium cyclamate is negative in both tests; therefore, there is no evidence of induced chromosome breakage and exchange.

Abstract:The growth and development of animals depend strongly on the balanced nutrition in the diet. This research aims is to characterize the weight variations of nano-sized calcium carbonate (CaCO3) in purified diet that to be fed to animal model of rat. The nano-sized CaCO3 was prepared by milling the calcium carbonate particles for 20 hours at a rotation speed of 1000 rpm and resulting particle size in a range of 2-50 nm. Nano-sized CaCO3 added to purified diet to the four formulas that were identified as normal diet (N), deficiency calcium (DC), rich in calcium (RC), and poor calcium (PC) with containing in nano-sized CaCO3 much as 0.50 %, 0.00 %, 0.75 % and 0.25 % respectively. The nutritional content of the purified diet was proximate analyzed, it resulted as followed moisture, ash, fat, protein, crude fiber. The quantities of chemical element were analyzed by atomic absorption spectrometry (AAS), it resulted iron, magnesium, potassium and calcium. The results showed that N diet (Ca: 16,914.29 ppm) were suggested for healthy rats and RC diet (Ca: 33,696.13 ppm) for conditioned osteoporosis rats. The crystalline phases of the samples that were examined by X-ray diffraction showed that crystalline phase increased with the increasing concentration of CaCO3.

Abstract:In this paper, the effect of commonly used food sweetener (sodium cyclamate) on the proliferation and differentiation of osteoblasts has been researched. The morophology change of osteoblasts was investigated by confocal laser scanning microscopy. Cell viability was studied by MTT analysis. BMP2 expression was analyzed by western blot and immunofluorescence. Mineralization ability of osteoblasts was researched by using alizarin red staining method. The results indicate that a very low concentration (0.06 μM) of sodium cyclamate can curle and fold microfilament and microtubule of osteoblasts. The increase addition of sodium cyclamate resulted significantly decrease of cells viability. The expression of bone morphogenetic protein-2 (BMP2) was seriously suppressed by sodium cyclamate. Alizarin Red staining experiment revealed that sodium cyclamate decreased the mineralization ability of osteoblasts. The present results suggest that sodium cyclamate can seriously inhibit the proliferation and differentiation of osteoblasts.

Abstract:Cytogenetic studies in Mongolian gerbils, Meriones unguiculatus, have shown that calcium cyclamate when given intraperitoneally can induce hyperploidy and chromosomal breaks, gaps, and fragments in bone marrow cells in vivo. Increasing concentrations of the sweetener produced increasing incidence of chromosomal abnormalities. Analysis of means showed that a 'saturation effect' of calcium cyclamate to induce hyperploidy, breaks and fragments was reached at 150 mg/kg; whereas a dose-response relationship was found between increasing concentrations (except at 50 mg/kg) and increasing percentage of cells showing gaps. The ability of calcium cyclamate to produce hyperploid cells and fragments is revealed for the first time in this study.

Abstract:In the late 1960s the artificial sweetener cyclamate was implicated as a bladder carcinogen in rats. This finding and other concerns about its safety ultimately led to a ban on cyclamate in the U.S. and restrictions on its use in many other countries. Since that time, the carcinogenic potential of cyclamate and cyclohexylamine, its principal metabolite, has been reevaluated in a group of well-controlled, well-designed bioassays that have failed to substantiate the earlier findings. This review of the published and unpublished literature on cyclamate attempts to evaluate the carcinogenicity question and other important aspects of the toxicity of cyclamate and cyclohexylamine, including their effects on various organ systems, their genotoxic potential, and their effects on reproduction. In addition, the physiological disposition of cyclamate is reviewed, with particular attention directed toward the site and extent of its conversion to cyclohexylamine.

Abstract:To determine the effect of cyclamate calcium on the humoral immune response in rabbits, one group was fed a 3% and another a 5% water solution of cyclamate calcium. A third group served as controls. After 150 days, the animals were tested for their ability to respond immunologically to bovine serum albumin (BSA). Other aspects of immunity investigated were the presence or absence of neoplasia of the urinary bladder, spleen, thymus, and lymph nodes, the number of white and red blood cells, and cyclamate conversion and excretion in the urine. The results indicated that the 5% group had a delayed and suppressed response to BSA. No neoplasia was detected. This group had fewer red cells and excreted increased concentrations of cyclohexylamine in the urine.

Abstract:The aim of this study was to investigate the effect of either sheep or cow milk supplementation to a low calcium and phosphorus diet on growth and organ mineral distribution in weanling rats. Rats were fed diets consisting of either a control chow, a 50% reduced calcium and phosphorous chow (low Ca/P), low Ca/P and sheep milk, or low Ca/P and cow milk diet for 28 days. Food intake of the rats, the growth rate of the rats, and the concentrations of minerals in the soft organs and serum were determined. Rats fed the low Ca/P diet alone had lower weight gain than rats consuming either of the milk-supplemented diets (p < 0.05). Both sheep milk and cow milk supplementation overcame the effects of consuming a diet restricted in calcium and phosphorus but the sheep milk was effective at a significantly lower level of milk intake (p < 0.05). Significant differences (p < 0.05) in essential and trace mineral concentrations due to milk type were observed in the kidney, spleen, and liver. For non-essential minerals, significant differences (p < 0.05), related to diet, were observed in all organs for arsenic, cesium, rubidium, and strontium concentrations.

Abstract:[Objective] To compare the effects of different calcium agents on growth and development in rats, and to study the effect of different form calcium agents on the absorption and utilization of calcium in rats. [Methods] Weaning female SD rats were randomly divided into 9 groups of calcium nano chelat, Calcium Citrate Malate, calcium L-aspartic acid, Yak bone calcium, cattle bone calcium, coral calcium, fish bone calcium、 calcium carbonate and low calcium meal, and fed on different calcium agents and low calcium meal in semi-purified diets, respectively, for 4 weeks. Meanwhile 3-day calcium metabolism experiment was carried out in the fourth week. [Results] The body weight in groups of Yak bone calcium and cattle bone calcium were lower than that of basic low calcium meal group (P﹤0.05). The body weight in group of Yak bone was lower than that of calcium carbonate group (P﹤0.05). The body length in group of cattle bone carbonate was lower than that in groups of calcium carbonate and basic low calcium meal (P﹤0.05). The food consumption efficiency in group of cattle bone carbonate was 37.39%, which was lower than that in group of low calcium meal (P﹤0.05). The apparent absorptivity of calcium in groups of calcium nano chelat, Yak bone calcium, coral calcium, fish bone calcium and calcium carbonate was 91.04%, 91.96%, 92.55%, 93.24%, respectively, which were higher than that in low calcium meal group (P﹤0.05). The retention rate of calcium in groups of Yak bone coral, fish bone calcium and calcium carbonate was 89.26%, 89.78%, 90.97%, 91.66%, respectively, which were higher than that in low calcium meal group (P﹤0.05). [Conclusion] Yak bone calcium and cattle bone calcium are worse than the other test groups in aspects of effect on promoting growth and development of the rats. With the same content of calcium from eight kinds of calcium agents, the absorptivity is not significantly different.

Abstract:1. 14C-labelled cyclamate has been administered to guinea pigs, rabbits, rats and humans. When given orally to these species on a cyclamate-free diet, cyclamate is excreted unchanged. In guinea pigs some 65% of a single dose is excreted in the urine and 30% in the faeces, the corresponding values for rats being 40 and 50%, for man, 30–50% and 40–60%, and for rabbits, 90 and 5%, the excretion being over a period of 2–3 days. 2. Cyclamate appears to be readily absorbed by rabbits but less readily by guinea pigs, rats and humans. 3. If these animals, including man, are placed on a diet containing cyclamate they develop the ability to convert orally administered cyclamate into cyclohexylamine and consequently into the metabolites of the latter. The extent to which this ability develops is variable, the development occurring more readily in rats than in rabbits or guinea pigs. In three human subjects, one developed the ability quite markedly in 10 days whereas two others did not in 30 days. Removal of the cyclamate from the diet caused a diminution in the ability to convert cyclamate into the amine. 4. In rats that had developed the ability to metabolize orally administered cyclamate, intraperitoneally injected cyclamate was not metabolized and was excreted unchanged in the urine. The biliary excretion of injected cyclamate in rats was very small, i.e. about 0.3% of the dose. 5. The ability of animals to convert cyclamate into cyclohexylamine appears to depend upon a continuous intake of cyclamate and on some factor in the gastrointestinal tract, probably the gut flora.

Effects of various diets on the calcium and phosphorus composition of mealworms (Tenebrio molitor larvae) and superworms (Zophobas morio larvae)

Abstract:OBJECTIVE To evaluate whether the nutritive quality of Tenebrio molitor larvae and Zophobas morio larvae, which are commonly cultured as live food sources, is influenced by 4 commercially available diets used as nutritional substrates; identify which diet best improved calcium content of larvae; and identify the feeding time interval that assured the highest calcium intake by larvae. ANIMALS 2,000 Zophobas morio larvae (ie, superworms) and 7,500 Tenebrio molitor larvae (ie, mealworms). PROCEDURES Larvae were placed in control and diet treatment groups for 2-, 7-, and 10-day intervals. Treatment diets were as follows: wheat millings, avian hand feeding formula, organic avian mash diet, and a high-calcium cricket feed. Control groups received water only. After treatment, larvae were flash-frozen live with liquid nitrogen in preparation for complete proximate and mineral analyses. Analyses for the 2-day treatment group were performed in triplicate. RESULTS The nutrient composition of the high-calcium cricket feed groups had significant changes in calcium content, phosphorus content, and metabolizable energy at the 2-day interval, compared with other treatment groups, for both mealworms and superworms. Calcium content and calcium-to-phosphorus ratios for larvae in the high-calcium cricket feed group were the highest among the diet treatments for all treatment intervals and for both larval species. CONCLUSIONS AND CLINICAL RELEVANCE A 2-day interval with the high-calcium cricket feed achieved a larval nutrient composition sufficient to meet National Research Council dietary calcium recommendations for nonlactating rats. Mealworm calcium composition reached 2,420 g/1,000 kcal at 48 hours, and superworm calcium composition reached 2,070g/1,000 kcal at 48 hours. These findings may enable pet owners, veterinarians, insect breeders, and zoo curators to optimize nutritive content of larvae fed to insectivorous animals.

Abstract:Objective: To establish fluorosis model in the ameloblasts of rat offspring for further study on the pathogenesis of dental fluorosis. Methods: Three male and three female SD rats weighing around 250 g were matched and divided into three groups. Rats in each group were fed with water containing fluoride of 0 mg/L, 50 mg/L and 100 mg/L separately till parturition. The offsprings were killed at day 3 and 5 of birth, respectively, to make mandible sections and observe the morphological and ultrastructural changes of ameloblasts in the tooth germ under optical microscope and transmission electron microscope. Results: In the 100 mg/L group, vacuole changes were found in part of ameloblasts in mature phase; cavity-like lesions could be seen in ameloblasts and enamel matrix, which were not seen in the 50 mg/L group and 0 mg/L groups under optical microscope. Observations under transmission electron microscope revealed various degrees of vacuolization, expansion of the endoplasmic reticulum, mitochondrial swelling in ameloblasts, and expansion of the gap between the cells in ameloblasts of rats in the 50 mg/L and 100 mg/L groups. Conclusion: The toxic-like changes in ameloblasts of rat offsprings may be produced by feeding pregnant rats with drinking water containing various concentration of fluoride.
Establishment of A Rat Model of Low Calcium Diet Related Hyperoxaluria

Abstract:OBJECTIVE:To establish a rat model of low calcium diet related hyperoxaluria and explore its features.METHODS:By means of randomized blocks design, totally 24 SD male rats were divided into low calcium diet group, medium calcium diet group, and high calcium diet group. Each group was sequentially fed on different calcium diets for 3 days. The urinary volume within 24 hours was recorded, the consistency of urinary oxalate by high-efficiency liquid chromatography, and the consistency of urine creatinine by automatic biochemical analyzer. The consistency was corrected to the output of urinary oxalate of rats in 24 hours, and the results were evaluated by repeated measurement of variance analysis and multivariate analysis of variance.RESULTS:The output of urinary oxalate of rats in 24 hours varied with time (F=7.893, P0.05). The output of urinary oxalate of rats in 24 hours varied with group division (F=3.565, P<0.05). The output of urinary oxalate in 24 hours in three groups on the third day was significantly higher than that on the first day (P<0.05).CONCLUSION:By controlling the calcium intake, we successfully established the model of low calcium diet related hyperoxaluria in rat.

Abstract:High sugar content in beverage or food can affect the aging process, and thus natural/artificial sweeteners are widely used as substitutes. However, whether sweeteners have such adverse effects as sugar remains to be clarified. Therefore, in the current study, three sulfa sweeteners, namely, saccharin sodium salt hydrate (SAC(2)), sodium cyclamate (CYC3) and acesulfame potassium (AceK(4)) were evaluated for their effects on the lifespan, deposition of lipofuscin, exercise activity, food intake, and intestinal fat deposition (IFD5) of Caenorhabditis elegans (C. elegans(6)). It was shown that SAC at 0.3 and 10 mg/mL shortened the lifespan of C. elegans and impaired the exercise capacity, while at other concentrations no significant effects were observed. In contrast, CYC at 0.1, 1 and 10 mg/mL prolonged the lifespan of C. elegans. On the other hand, AceK at 1 mg/mL increased the lifespan of C. elegans, and could decrease both lipofuscin deposition and IFD in a dose-dependent manner. Taken together, these results indicated that although SAC, CYC, and AceK all belong to the sulfa sweeteners, each has distinct effects on different physiological activities associated with aging, at least in C. elegans.
Retention of Calcium from Various Xylitol-Calcium Combinations in Rats

Abstract:Previous studies have shown that xylitol, the five-carbon sugar alcohol, increases the intestinal absorption of Ca in the rat. In this study an optimum xylitol:Ca molar ratio for Ca absorption and retention was determined in 10-week-old Long-Evans male rats. Xylitol was intubated with 20 mg of 45Ca (as 45CaCl2 or 45CaCO3) into the stomach of fasted rats in 1 ml water using xylitol:Ca molar ratios of 0:1, 0.5:1, 1:1, 1.5:1 or 2:1. Radioactivity of urine, feces and humerus was determined from 48-hr samples. The highest retention rate of 45Ca in the whole body or in the humerus was found when 114 mg of xylitol was given with Ca (xylitol:Ca molar ratio 1.5). Xylitol affected humeral retention more significantly than whole body retention. For comparison, the retention of 45Ca was determined from four lactose:CaCl2 combinations and from Ca-lactate and Ca-citrate salts. None of these combinations increased Ca retention significantly, although lactose at 114 mg dose showed slight positive effect. The results favor the use of a xylitol:Ca molar ratio of 1.5 in Ca supplements.

Cariogenicity of High Fructose Corn Syrup: an Experimental Study on Rats

Abstract:AIM: To evaluate the cariogenicity of high fructose corn syrup(HFCS).METHODS:30 SPF Sprague-Dawley(SD)rats inoculated orally with Streptococcus mutans UA159 were randomly divided into 3 groups and were respectively provided with non-sugar,sucrose-containing and HFCS-containing diets.After 3 months,the Jaw bones were removed.A binocular microscope was used to examine the teeth for buccal and lingual caries.Grinding sections were then prepared,stained with Rhodamine B,and examined for proximal and fissure caries by fluorescence microscope.The caries lesions were assessed by Keyes method.RESULTS: The prevalence of dental caries was 100% in all 3 groups.Keyes caries scores in sucrose group were similar to those in HFCS group(P0.05) and significantly higher than in non-sugar group(P0.05).While no significant difference was observed in the levels of lesions extending into the inner 3/4 of dentine on smooth surfaces between HFCS group and non-sugar group(P0.05),the lesions on occlusal and smooth surfaces in HFCS group were significantly more severe than those in non-sugar group(P0.05).CONCLUSION: In SD rats HFCS containing diet may induce dental caries similar to sucrose.
