Abstract:Weaning may be associated with negative energy balance and body weight loss when calves are still immunologically immature, predisposing them to infectious diseases. The aim of the present experiment was to investigate the effects of treatment of preweaning dairy calves with recombinant bovine somatotropin (rbST) on the somatotropic axis, selected immune parameters, and hematology of calves around weaning. Thirty-six Holstein female calves were randomly assigned to receive 1.5 to 1.8 mg of rbST (Posilac, Elanco Animal Health, Greenfield, IN) per kilogram of body weight or to receive injections of saline (saline solution 0.9%, Valley Vet Supply, Marysville, KS) every 7 d from 21 to 63 d of life. Calves were fed milk replacer ad libitum from birth to 38 d of age (d -11), when progressive weaning started, and calves were weaned at 49 d of age (d 0). Calves were weighed at birth and weekly from 21 to 63 d of age, when wither height also was measured. Calves were vaccinated with 0.5 mg of ovalbumin on study d -28 and -7. Blood samples were collected on d -28, -25, -21, -11, 0, 3, 7, and 14. Polymorphonuclear leukocytes were isolated and challenged ex vivo with Escherichia coli to determine phagocytosis and oxidative burst capacity. Additionally, expression of cluster of differentiation (CD)62L and CD18 by granulocyte, lymphocyte, and CD14+ monocyte were determined. Blood samples were also used to determine hematological parameters and concentrations of growth hormone, insulin-like growth factor-1, insulin, glucose, fatty acids, β-hydroxybutyrate, haptoglobin, and anti-ovalbumin IgG. Calves treated with rbST had greater concentrations of growth hormone and insulin-like growth factor-1 from d -25 to 14 than control calves, whereas insulin, fatty acid, and β-hydroxybutyrate concentrations did not differ. On d -11, glucose concentration was greater for rbST-treated calves. Treatment did not affect polymorphonuclear lymphocyte phagocytosis and oxidative burst, but intensity of expression of CD62L and CD18 by granulocytes tended to be increased by rbST treatment. Treatment did not affect the concentration of anti-ovalbumin IgG in serum. Haptoglobin concentration was reduced in rbST treated calves on d 3 and we noted a tendency for hematocrit to be lower in rbST-treated calves. Treatment did not affect body weight, wither height, and average daily gain, despite the fact that rbST-treated calves had lower daily milk replacer intake. The relatively minor improvements in immune responses resulting from rbST treatment of weaning calves may not be sufficient to reduce the incidence of infectious diseases.
Blood immune transcriptome analysis of artificially fed dairy calves and naturally suckled beef calves from birth to 7 days of age

Abstract:Neonatal calves possess a very immature and naïve immune system and are reliant on the intake of maternal colostrum for passive transfer of immunoglobulins. Variation in colostrum management of beef and dairy calves is thought to affect early immune development. Therefore, the objective of this study was to examine changes in gene expression and investigate molecular pathways involved in the immune-competence development of neonatal Holstein dairy calves and naturally suckled beef calves using next generation RNA-sequencing during the first week of life. Jugular whole blood samples were collected from Holstein (H) dairy calves (n = 8) artificially fed 5% B.W. colostrum, and from beef calves which were the progenies of Charolais-Limousin (CL; n = 7) and Limousin-Friesian beef suckler cows (LF; n = 7), for subsequent RNA isolation. In dairy calves, there was a surge in pro-inflammatory cytokine gene expression possibly due to the stress of separation from the dam. LF calves exhibited early signs of humoral immune development with observed increases in the expression genes coding for Ig receptors, which was not evident in the other breeds by 7 days of age. Immune and health related DEGs identified as upregulated in beef calves are prospective contender genes for the classification of biomarkers for immune-competence development, and will contribute towards a greater understanding of the development of an immune response in neonatal calves.
Acute pancreatitis: an under-recognized risk of percutaneous transhepatic distal biliary intervention.

Abstract:OBJECTIVE Percutaneous transhepatic biliary intervention (PTBI) plays an important role in the management of biliary obstruction, and this may be complicated by acute pancreatitis. The aim of this study was to assess the incidence of acute pancreatitis following PTBI. PATIENTS AND METHODS Patients who underwent PTBI between January 1992 and December 2003 in a tertiary referral centre were identified from the hospital database. Patients who did not have their amylase measured post-procedure were excluded, as acute pancreatitis might have been missed. Acute pancreatitis was defined as hyperamylasaemia of three times or more above normal in association with abdominal pain. RESULTS Over a 12-year period, 331 patients underwent 613 procedures. Serum amylase was measured after 134 procedures (21.9%) and was elevated in 26 of those (19.4%). There was no difference in the frequency of hyperamylasaemia between proximal and distal PTBI (14/73 [19.2%] vs 12/61 [19.7%] procedures, p=NS). However, acute pancreatitis developed after 4 of 61 (6.6%) distal PTBI (stent, n=3; internal-external catheter insertion, n=1) but not after proximal PTBI (cholangiography or external drainage) (p=0.041). The attacks were mild in three of the four patients. No pancreatitis-related deaths occurred. CONCLUSION The risk of acute pancreatitis after distal PTBI is under-recognized and should be considered as a consent issue in patients scheduled for distal PTBI and when post-procedure abdominal pain ensues.

Abstract:Abstract Two 89-d experiments (Exp 1 and Exp 2) evaluated the effects of feeding an immunomodulatory feed additive (NutraGen; NG) in calves following exposure to bovine viral diarrhea virus (BVDV) on d -3 and Mannheimia haemolytica (MH) infection on d 0. For each experiment, steers [body weight (BW = 251 ± 38.2 kg were randomly allocated to 1 of 3 treatments. Treatments included a placebo (CON; Exp 1 n = 5; Exp 2 n = 5), a placebo fed from d -18 to d -3 followed by NG fed from d -3 to d 28 (CHLG; Exp 1 n = 5; Exp 2 n = 6), and NG fed from d -18 to d 28 (PREC; Exp 1 n = 6; Exp 2 n = 5). Data were analyzed using the GLIMMIX procedure of SAS Version 9.4 (SAS Institute, Cary, N.C.). Animal served as the experimental unit and time served as a repeated measure. Treatment, time, and the treatment × time interaction served as fixed effects. Block was included as a random effect. Cytokine data were tested for normality using the Shapiro-Wilk test, determined to be non-normal, and logarithm (base 10) transformed as a result prior to statistical analysis. Multiple time effects and treatment × time interactions occurred for serum MH, BVDV 1, and BVDV 2 antibody concentrations in both experiments (P ≤ 0.05). In Exp 1, there was a tendency for an overall treatment effect for calves assigned to the CON treatment to exhibit the greatest MH serum antibody concentrations (P = 0.06). While no differences were detected between treatment groups, BVDV 1 and BVDV 2 antibody concentrations peaked on the same day in both experiments, d 28 post-challenge. There were no treatment × time interactions, treatment effects, or time effects for serum TNF-α, IFN-γ, or IL-17A concentrations (P ≥ 0.13) in either experiment. In Exp 1, there was a treatment effect for serum IL-10 (P ≤ 0.0001), where calves assigned to the PREC treatment had increased serum IL-10 concentrations when compared with calves in the other treatment groups. In Exp 2, there was a tendency for a treatment effect for IL-10 concentrations, where calves assigned to the PREC treatment displayed decreased IL-10 concentrations when compared with calves in the other treatment groups (P = 0.07). These data suggest that feeding NG to calves prior to a stress experience may alter cellular responses. Overall, these experiments suggest that NG impacted antibody concentrations and cytokine expression during a bovine respiratory disease challenge.

Abstract:This study aimed to evaluate the impacts of a maternal bovine appeasing substance (MBAS) administered to preweaned Holstein calves on their health and performance. In total, 410 calves individually allocated were enrolled (Treatment, n = 205; Control, n = 205). Treated calves received a topical application of MBAS to the nuchal skin (2.5 ml) and the skin above the muzzle (2.5 ml) at one day of age and then every 14 days until weaning at 60 days of life. An extra dose was applied when calves were moved from hutches to collective pens at approximately 100 days of age. Calves allocated to the control group did not receive any kind of treatment. MBAS reduced the incidence of diarrhea (treatment = 58.6% vs control = 70.8%; P = 0.01) as well as mortality risk (treatment = 2.4% vs control = 7.8%; P = 0.02). Furthermore, calves treated with MBAS had greater (P = 0.002) body weight 7 days after calves were moved from the hutches to the collective pens. Therefore, the use of MBAS during the preweaning period appears to be an effective strategy to improve health and growth of dairy calves.

Abstract:This study aimed to use a range of non-invasive monitoring technologies to investigate the behavioural and physiological responses of individually housed dairy calves to age at change in milk replacer (MR) feeding frequency. Forty-eight Holstein Friesian calves were individually penned and fed MR (625 g/d) as solids in one of three feeding regimes: (i) once-a-day feeding commencing at age 14 d (OAD14), (ii) once-a-day feeding commencing at age 28 d (OAD28) and (iii) twice-a-day feeding (TAD). Several behavioural (automatic activity sensors), physiological (infrared [IR] thermography and heart rate variability [HRV]) and haematological indicators were used to examine calf responses. Reduction in milk feeding frequency at 14 or 28 d of age increased daily concentrate intakes and drinking water consumption throughout the pre-wean period. Calf lying behaviour was unaffected by reduction in milk feeding frequency; however, TAD calves recorded a significant decrease in total daily lying time during the post-wean period compared with OAD28s. There was no effect of treatment on IR eye or rectal temperature throughout the experiment; however, there was an effect of age, with IR temperature decreasing as calf age increased. OAD14 calves tended to have decreased HRV at days 14 and 16, which is suggestive of an increased stress load. The findings suggest that under high levels of animal husbandry and whilst maintaining the same amount of milk powder/d (625 g/d), reduction in milk feeding frequency from twice to once daily at 28 d can occur without significant impact to behavioural, performance and physiological parameters assessed here.

The Effects of Respiratory Vaccine Type and Timing on Antibody Titers, Immunoglobulins, and Growth Performance in Pre- and Post-Weaned Beef Calves

Abstract:In order to examine the effects of vaccine type and timing of crossbred beef calves (n = 151) were assigned to one of three BRD vaccination protocols stratified by breed of sire, sex, and date of birth, which included: (1) KM-a pentavalent killed viral (KV) vaccine at 2 to 3 months of age (D 0) and a pentavalent modified-live viral (MLV) vaccine at weaning (D 127); (2) MM-MLV on D 0 and revaccinated on D 127 or (3) WN-MLV at weaning and D 140. Vaccination treatment did not affect performance nor BRSV serum-neutralizing antibody titers. Serum-neutralizing antibody titers to BVDV-1 were greatest for the MM through D 154. However, following booster (KM) or initial vaccination (WN) at D 127, titers increased for the other treatment groups to higher values (KM) by the end of the study. Delay of initial vaccination until weaning may have delayed specific antibody response in the WN group and skewed the immune response towards a Th-1 or cell-mediated response. Overall, the inclusion of an MLV in the vaccine protocol resulted in a more robust antibody response, and the timing of vaccination may affect the onset of efficacious and robust vaccine responses.

Abstract:Abstract This study examined the interactions between early life nutrition strategies and their impact on growth, development, and fecal consistency in surplus bull calves. Holstein-Friesian bull calves [n = 120; body weight (BW) = 43.1±3.56 kg] were enrolled. In the first 12 h after birth, calves were fed either 2 L (100 g of IgG) or 6L (300 g of IgG) of colostrum replacer (CR) to obtain either failure (F) or successful (S) transfer of passive immunity, respectively. Upon arrival at the facility, serum IgG concentrations were measured; therefore, F and S calves were identified accordingly and randomly assigned to one of two milk replacer (MR) feeding schemes: Moderate (Mod; ~8 L) or Low (Low; ~4 L). In total, there were 4 treatments resulting from the 2 × 2 factorial design: F-Mod, F-Low, S-Mod, S-Low (n = 30/treatment). Calves were individually housed and fed twice daily until d 42, gradually weaned until d 56, and had free access to straw and water during the study. On d 22, a calf pelleted starter was offered. To evaluate organ and gastroinstetinal tract development, 10 calves/treatment were euthanized at 84 d of age. Other measurements include weekly BW, daily intakes, and daily fecal score until d 21. Data were analyzed by PROC MIXED in SAS accounting for the fixed effects of block, CR, MR, and time, and their interaction. Time entered the model as a repeated statement, and initial BW was used as a covariable when appropriate. Initial BW was not different across treatments, whereas serum IgG was greater in S calves (S = 1,962.9 vs. F = 941.0 mg IgG/dL). During the study, BW was influenced by both CR (P = 0.001) and MR (P < 0.001), where S and Mod calves consistently exhibited greater BW when compared with F and Low calves, respectively. At d 84, Mod calves (F-Mod and S-Mod) were on average 18 kg heavier than Low calves. Similarly, Mod calves exhibited greater fresh tissue weights in the liver, pancreas, spleen, and both the small and large intestines (P < 0.05). Starter intake was influenced by MR (P < 0.001), with Low calves having the greatest intake before and during weaning. However, post-weaning starter intake was greater (P = 0.010) in Mod calves. Fecal consistency was influenced by CR (P = 0.001) and MR (P < 0.001), with S and Low calves having improved fecal consistency. Although no interactions were observed between CR and MR, these findings underscore the importance of considering both colostrum and MR levels in calf nutrition management to optimize growth and improve animal welfare during the early stages of life.

Effects of supplementing colostrum beyond the first day of life on growth and health parameters of preweaning Holstein heifers

Abstract:The preweaning period for a dairy calf is characterized by high morbidity and mortality rates, leading to financial losses for producers. Identifying strategies to improve the health and welfare of calves while reducing antimicrobial use continues to be crucial to the success of the dairy industry. The objective of this study was to determine the effects of feeding colostrum replacer (CR) to dairy heifer calves beyond d 1 of life on growth, serum IgG, the incidence of diarrhea and bovine respiratory disease (BRD), and the risk of mortality in the preweaning period. At birth, Holstein heifer calves (n = 200; 50/treatment) weighing 40.7 ± 0.35 kg (mean ± SE) were fed 3.2 L of CR (205 g IgG/feeding) at 0 h and 12 h of life. Calves were then randomly assigned to 1 of 4 treatments: 450 g of milk replacer (MR) from d 2 to 14 (control, CON), 380 g of CR + 225 g of MR from d 2 to 3, then 450 g of MR from d 4 to 14 (transition, TRAN), 45 g of CR + 450 g of MR from d 2 to 14 (extended, EXT); or 380 g of CR + 225 g of MR from d 2 to 3, then 45 g of CR + 450 g of MR from d 4 to 14 (transition + extended, TRAN+EXT). Each treatment was reconstituted to 3 L and fed twice daily. All CR treatments were fed using bovine-derived CR containing 27% IgG. From d 15 to 41, all calves were fed 600 g of MR reconstituted to 4 L twice daily. Body weight was recorded at birth and every 7 d until study completion on d 49. Blood samples were taken daily until d 7 to evaluate serum IgG and then every 7 d until d 49. A health assessment was performed daily to evaluate calves for BRD and diarrhea. Data were analyzed using mixed linear regression, mixed logistic regression, and survival analysis models in SAS 9.4. Serum IgG concentrations were not affected by treatment for the study period. The EXT and TRAN+EXT groups had greater average daily gain (ADG) from d 7 to 14 (0.14 kg/d) and the TRAN group had greater ADG from d 14 to 21 (0.11 kg/d), compared with CON. There was no association of treatment with the odds or the duration of a diarrhea bout. However, provision of CR to the TRAN and EXT calves was associated with a reduced hazard of diarrhea compared with CON calves. Furthermore, TRAN and EXT calves have a lower hazard of mortality compared with CON calves, with TRAN and EXT calves had a 2.8- and 3.8-times lower hazard of mortality, respectively. Our findings suggest that the supplementation of CR to dairy calves positively affects ADG, and reduces the hazard of diarrhea and mortality during the preweaning period. Future research should look to further refine the supplementation strategy of CR to calves and explore the mechanism of action.
Oral alitretinoin (9-cis-retinoic acid) therapy for chronic hand dermatitis in patients refractory to standard therapy: results of a randomized, double-blind, placebo-controlled, multicenter trial.

Abstract:OBJECTIVE To assess the efficacy and safety of oral alitretinoin (9-cis-retinoic acid), 10 mg/d, 20 mg/d, and 40 mg/d, compared with placebo control, in the treatment of chronic hand dermatitis. DESIGN Multicenter, randomized, double-blind, placebo-control, prospective trial. SETTING A total of 43 outpatient clinics in 10 European countries. PATIENTS Of 348 patients screened, 319 with moderate or severe refractory chronic hand dermatitis were randomized, in the ratio of 1:1:1:1, to 4 treatment groups and received allocated intervention. Of 75 patients who withdrew, 24 withdrew owing to adverse events. INTERVENTIONS Placebo or 10 mg, 20 mg, or 40 mg of oral alitretinoin (9-cis-retinoic acid) taken once daily for 12 weeks. Safety was assessed for all patients during a follow-up period of 4 weeks, and responders were observed for a follow-up period of 3 months. MAIN OUTCOME MEASURE Physician's global assessment of overall chronic hand dermatitis severity. RESULTS Alitretinoin led to a significant and dose-dependent improvement in disease status, with responses in up to 53% of patients, and up to a 70% mean reduction in disease signs and symptoms. Treatment was generally well tolerated, with dose-dependent effects comprising headache, flushing, mucocutaneous events, hyperlipidemia, and decreased hemoglobin and decreased free thyroxin levels. Three months after discontinuation of treatment, the rate of relapse was 26%, independent of dose. CONCLUSION Alitretinoin given at well-tolerated doses induced substantial clearing of chronic hand dermatitis in patients refractory to conventional therapy.

Abstract:Abstract The Oklahoma Quality Beef Network utilizes three health protocols for producer choice based on vaccine timing and viral components. The study objective was to examine the effects of vaccine type and timing on animal performance, and immune response pre-and post-weaning. A total of 151 Angus, Angus x Hereford, or Angus x Charolais calves were randomly assigned to one of three health protocols stratified by breed of sire, sex, and date of birth. Vaccination treatments were 1) KV/MLV – multivalent inactivated BRD vaccine (KV) administered on d 0 and revaccination with a pentavalent modified-live virus on d 125; 2) MLV/MLV – pentavalent modified-live virus on d 0 and d 125; or 3) WEAN – pentavalent modified-live virus on d 125 and 139. Virus-specific antibody (Ab) titer data was determined using serum-neutralization from serum collected at six time points. Antibody titers, BW, and ADG variables were evaluated following vaccination. Results indicated there was no treatment effect on BW. Bovine viral diarrhea virus (BVD) also displayed significance for treatment x date interaction. The MLV/MLV provided greatest response from d 0 (branding) to d 125 (weaning) than the other two treatments. There was no difference between the KV/MLV group and the WEAN group from d 0 to d 125. By d 167 the KV/MLV group had a greater immune response than the MLV/MLV and WEAN groups. The MLV/MLV provided the greatest protection from branding to weaning. Providing a KV at branding had minimal effect on BVD titers but responded well to MLV vaccine at weaning by d 167. The WEAN group generated the least number of titers overall. Further investigation is needed to determine the long-term protection of vaccination strategies.

Abstract:Introduction: Neonatal calf diarrhea is a multifactorial condition that occurs in early life when calves are particularly susceptible to enteric infection and dysbiosis of the gut microbiome. Good calf health is dependent on successful passive transfer of immunity from the dam through colostrum. There are limited studies on the developing gut microbiota from birth to weaning in calves. Methodology: Therefore, the objective of this study was to examine the effect of immune status and diarrheal incidence on the development of the fecal microbiota in Jersey ( n = 22) and Holstein ( n = 29) heifer calves throughout the pre-weaning period. Calves were hand-fed a colostrum volume equivalent to 8.5% of their birthweight, from either the calf's dam ( n = 28) or re-heated mixed colostrum (≤2 cows, ≤1d; n = 23) within 2 h of birth. All calves were clinically assessed using a modified Wisconsin–Madison calf health scoring system and rectal temperature at day (d) 0, d7, d21, or disease manifestation (DM) and weaning (d83). Weights were recorded at d0, d21, and d83. Calf blood samples were collected at d7 for the determination of calf serum IgG (sIgG). Fecal samples were obtained at d7, d21/DM [mean d22 (SE 0.70)], and at weaning for 16S rRNA amplicon sequencing of the fecal microbiota. Data were processed in R using DADA2 ; taxonomy was assigned using the SILVA database and further analyzed using Phyloseq and MaAsLin 2. Results and discussion: Significant amplicon sequence variants (ASVs) and calf performance data underwent a Spearman rank-order correlation test. There was no effect ( p > 0.05) of colostrum source or calf breed on serum total protein. An effect of calf breed ( p < 0.05) was observed on sIgG concentrations such that Holstein calves had 6.49 (SE 2.99) mg/ml higher sIgG than Jersey calves. Colostrum source and calf breed had no effect ( p > 0.05) on health status or the alpha diversity of the fecal microbiota. There was a relationship between health status and time interaction ( p < 0.001), whereby alpha diversity increased with time; however, diarrheic calves had reduced microbial diversity at DM. No difference ( p > 0.05) in beta diversity of the microbiota was detected at d7 or d83. At the genus level, 33 ASVs were associated (adj. p < 0.05) with health status over the pre-weaning period.

Abstract:Introduction: Lichen planus is a human exclusive, chronic inflammatory disease affecting skin and mucosa, particularly oral mucosa affecting the stratified squamous epithelia. Epithelial alterations in oral lichen planus (OLP) may lead to superimposed Candida infection, especially the normal flora Candida albicans. Therefore, this study was designed to assess the prevalence of Candida infection in OLP lesions. Materials and methods: Thirty-four OLP patients were subjected to clinical, mycological and histopathological investigations for the presence of candidal organisms. The data obtained was analyzed statistically using Chi-square test and Student’s t-test. Results: Among the 34 patients in the study group 15 (44.11%) showed positive candidal culture on Sabouraud’s dextrose agar medium and none of the 34 histological sections of OLP showed candidal hyphae on Periodic acid-Schiff staining. No significant association of positive candidal culture with respect to symptoms and patterns of lichen planus were found. Conclusion: Candidal infection in OLP is insignificant.

Abstract:Studies have shown that calves fed milk replacers (MR) with crude protein (CP) concentrations greater than 20%, as typically found in conventional MR, have higher dry matter intakes (DMI) and greater average daily gains (ADG) but consume less starter, which can lead to stress during weaning and reduced rumen development. The greater amount of CP being fed to preweaned calves may alter their nitrogen (N) balance, and excess N may be excreted in the urine. The objective of this study was to determine N utilization in preweaned calves fed diets varying in the amount of CP and MR fed. This study used 24 newborn dairy heifer calves blocked by birth and randomly assigned to 1 of 3 treatments: (1) 446g dry matter (DM) of a conventional MR (CON; 20% CP, 20% fat), (2) 669g DM of a moderately high protein MR (moderate; MOD; 26% CP, 18% fat), or (3) 892g DM of a moderately high protein MR (aggressive; AGG; 26% CP, 18% fat). All calves had ad libitum access to starter and water. Both MR and starter were medicated with decoquinate. During weaning (d 43-49), the morning MR feeding ceased. On d 50, all MR feedings ended; however, starter and water intakes were continuously recorded until d 56. At 5wk of age, urine was collected using urinary catheters for 3d and chromium oxide was administered by bolus at 2g/d for 7d to estimate N efficiency. Calves fed MOD and AGG had similar starter intakes, feed efficiencies, and ADG, with the combined treatments having reduced starter intakes (258 vs. 537g/d), greater ADG (674 vs. 422g/d), and improved feed efficiency (0.57 vs. 0.45 gain:feed) compared with CON calves preweaning. However, DMI and water intake were similar across all treatments. Results from the N utilization phase showed that MOD and AGG treatments had similar but lower N efficiency compared with CON calves (45.5 vs. 52.7%). This could be due to MOD- and AGG-fed calves having greater urine volume and thereby, greater combined urine N output compared with CON calves (17.6 vs. 12.1 g/d). In summary, feeding >0.66kg (DM) from a 26% CP MR increased ADG and improved feed efficiency during the preweaning period but reduced starter intake and lowered N efficiency.

Abstract:This study aimed to investigate the changes in abomasum transcriptome and the associated microbial community structure in young calves with artificially dosed, adult rumen contents. Eight young bull calves were randomly dosed with freshly extracted rumen contents from an adult cow (high efficiency (HE), n = 4), or sterilized rumen content (Con, n = 4). The dosing was administered within 3 days of birth, then at 2, 4, and 6 weeks following the initial dosing. Abomasum tissues were collected immediately after sacrifice at 8 weeks of age. Five genera (Tannerella, Desulfovibrio, Deinococcus, Leptotrichia, and Eubacterium; p < 0.05) showed significant difference in abundance between the treatments. A total of 975 differentially expressed genes were identified (p 1.5, mean read-counts > 5). Pathway analysis indicated that up-regulated genes were involved in immune system process and defense response to virus, while the down-regulated genes involved in ion transport, ATP biosynthetic process, and mitochondrial electron transport. Positive correlation (r > 0.7, p < 0.05) was observed between TRPM4 gene and Desulfovibrio, which was significantly higher in the HE group. TRPM4 had a reported role in the immune system process. In conclusion, the dosing of adult rumen contents to calves can alter not only the composition of active microorganisms in the abomasum but also the molecular mechanisms in the abomasum tissue, including reduced protease secretion and decreased hydrochloric acid secretion.

Abstract:Abstract The objective of this randomized controlled trial was to determine the performance effects of milk replacer supplemented with a feed additive high in beta-glucans for male dairy calves. Holstein male calves (n = 147) were enrolled in a 76-day randomized block trial in two blocks. Upon enrollment, calves were assigned to one of the four following treatments: Control with no beta-glucan supplementation (BG0; n = 36; BW = 48.32 ± 3.73 kg), 1 g· animal-1· d-1 (BG1; n = 38; BW = 48.29 ± 2.89kg), 3 g· animal-1· d-1 (BG3; n = 37; BW = 48.31 ± 3.23 kg), and 5 g· animal-1· d-1 (BG5; n = 36; BW = 48.47 ± 3.00 kg) of beta-glucans. The beta-glucan supplement was blended with milk replacer (24% CP, 20% fat; 130 g/L) and fed to calves during the morning milking. Calves were individually housed, had unlimited access to water, and a pelletized calf starter (20% CP). Calves were fed 5 L/d of milk replacer divided into two equal meals until d 15; milk feeding plan increased until d 35 (8 L/d), on d 36, milk allowance was gradually reduced until weaned on d 56 (2.5 L/d). Calf starter intake and body weight were recorded weekly until d 77. Disease treatment was recorded daily. A mixed linear model determined the effect of treatment on calf starter intake and average daily gain. The model included treatment if the calf was ever treated with antibiotics for respiratory disease and/or diarrhea, study period, and a treatment by period interaction. Block was considered a random factor. The total milk replacer intake was 5.73 ± 0.08, 5.71 ± 0.08, 5.73 ± 0.01, 5.73 ± 0.01 L/d for BG0, BG1, BG3 and BG5, respectively. Calf grain intake by treatment was 58.10 ± 12.02, 54.58 ± 8.11, 56.86 ± 11.8, 56.73 ± 15.52 for BG0, BG1, BG3 and BG5, respectively. Overall, 55% and 71% of all calves were treated at least once for diarrhea, and respiratory disease respectively. The percentage of animals treated for diarrhea by treatment were 45.00, 57.50, 50.00, and 32.50% for BG0, BG1, BG3, and BG5, respectively. For respiratory disease, the percentage of animals treated was 72.5, 72.5, 70.0, and 70.00 for BG0, BG1, BG3, and BG5 treatments, respectively. There was a significant interaction between treatment and study period where the control group had greater starter intake during the postweaning period (BG0 = 0.78 ± 0.01, BG1= 0.82 ± 0.01, BG3 = 0.81 ± 0.02, BG5 = 0.82± 0.01 kg/d; P < 0 .001). No treatment or treatment by study period effects was observed for average daily gain (P = 0.79, P = 0.89, respectively). Future research should investigate the effects of beta-glucans on blood metabolites and palatability throughout the calf-rearing period.
