Abstract:This study investigates the neural representation of speech in complex listening environments. Subjects listened to a narrated story, masked by either another speech stream or by stationary noise. Neural recordings were made using magnetoencephalography (MEG), which can measure cortical activity synchronized to the temporal envelope of speech. When two speech streams are presented simultaneously, cortical activity is predominantly synchronized to the speech stream the listener attends to, even if the unattended, competing-speech stream is more intense (up to 8 dB). When speech is presented together with spectrally matched stationary noise, cortical activity remains precisely synchronized to the temporal envelope of speech until the noise is 9 dB more intense. Critically, the precision of the neural synchronization to speech predicts subjectively rated speech intelligibility in noise. Further analysis reveals that it is longer-latency (similar to 100 ms) neural responses, but not shorter-latency (similar to 50 ms) neural responses, that show selectivity to the attended speech and invariance to background noise. This indicates a processing transition, from encoding the acoustic scene to encoding the behaviorally important auditory object, in auditory cortex. In sum, it is demonstrated that neural synchronization to the speech envelope is robust to acoustic interference, whether speech or noise, and therefore provides a strong candidate for the neural basis of acoustic-background invariant speech recognition.
Temporal coding of speech in human auditory cortex

Abstract:Human listeners can reliably recognize speech in complex listening environments. The underlying neural mechanisms, however, remain unclear and cannot yet be emulated by any artificial system. In this dissertation, we study how speech is represented in the human auditory cortex and how the neural representation contributes to reliable speech recognition. Cortical activity from normal hearing human subjects is noninvasively recorded using magnetoencephalography, during natural speech listening. It is first demonstrated that neural activity from auditory cortex is precisely synchronized to the slow temporal modulations of speech, when the speech signal is presented in a quiet listening environment. How this neural representation is affected by acoustic interference is then investigated. Acoustic interference degrades speech perception via two mechanisms, informational masking and energetic masking, which are addressed respectively by using a competing speech stream and a stationary noise as the interfering sound. When two speech streams are presented simultaneously, cortical activity is predominantly synchronized to the speech stream the listener attends to, even if the unattended, competing speech stream is 8 dB more intense. When speech is presented together with spectrally matched stationary noise, cortical activity remains precisely synchronized to the temporal modulations of speech until the noise is 9 dB more intense. Critically, the accuracy of neural synchronization to speech predicts how well individual listeners can understand speech in noise. Further analysis reveals that two neural sources contribute to speech synchronized cortical activity, one with a shorter response latency of about 50 ms and the other with a longer response latency of about 100 ms. The longer-latency component, but not the shorter-latency component, shows selectivity to the attended speech and invariance to background noise, indicating a transition from encoding the acoustic scene to encoding the behaviorally important auditory object, in auditory cortex. Taken together, we have demonstrated that during natural speech comprehension, neural activity in the human auditory cortex is precisely synchronized to the slow temporal modulations of speech. This neural synchronization is robust to acoustic interference, whether speech or noise, and therefore provides a strong candidate for the neural basis of acoustic background invariant speech recognition.

Abstract:The hierarchical nature of language requires human brain to internally parse connected-speech and incrementally construct abstract linguistic structures. Recent research revealed multiple neural processing timescales underlying grammar-based configuration of linguistic hierarchies. However, little is known about where in the whole cerebral cortex such temporally scaled neural processes occur. This study used novel magnetoencephalography source imaging techniques combined with a unique language stimulation paradigm to segregate cortical maps synchronized to 3 levels of linguistic units (i.e., words, phrases, and sentences). Notably, distinct ensembles of cortical loci were identified to feature structures at different levels. The superior temporal gyrus was found to be involved in processing all 3 linguistic levels while distinct ensembles of other brain regions were recruited to encode each linguistic level. Neural activities in the right motor cortex only followed the rhythm of monosyllabic words which have clear acoustic boundaries, whereas the left anterior temporal lobe and the left inferior frontal gyrus were selectively recruited in processing phrases or sentences. Our results ground a multi-timescale hierarchical neural processing of speech in neuroanatomical reality with specific sets of cortices responsible for different levels of linguistic units.

Abstract:The ability to focus on and understand one talker in a noisy social environment is a critical social-cognitive capacity, whose underlying neuronal mechanisms are unclear. We investigated the manner in which speech streams are represented in brain activity and the way that selective attention governs the brain's representation of speech using a "Cocktail Party" paradigm, coupled with direct recordings from the cortical surface in surgical epilepsy patients. We find that brain activity dynamically tracks speech streams using both low-frequency phase and high-frequency amplitude fluctuations and that optimal encoding likely combines the two. In and near low-level auditory cortices, attention "modulates" the representation by enhancing cortical tracking of attended speech streams, but ignored speech remains represented. In higher-order regions, the representation appears to become more "selective," in that there is no detectable tracking of ignored speech. This selectivity itself seems to sharpen as a sentence unfolds.

Abstract:The Wernicke-Lichtheim-Geschwind model of the neurology of language has served the field well despite its limited scope. More recent work has updated the basic architecture of the classical model and expanded its scope. This chapter briefly reviews the Wernicke-Lichtheim-Geschwind model and points out its shortcomings, then describes and motivates the dual stream model and how it solves several empirical shortcomings of the classical model. The chapter also (i) underscores how the dual stream model relates to the organization of nonlinguistic cortical networks, integrating language systems with the broader functional-anatomical landscape, (ii) describes recent work that further specifies the computational architecture and neural correlates of the dorsal speech production system, and (iii) summarizes recent extensions of the architectural framework to include syntax.

First published October 5, 2011; doi: 10.1152/jn.00297.2011.-The cortical representation of the acoustic features of continuous speech is the foundation of speech perception. In this study, noninvasive magnetoencephalography (MEG) recordings are obtained from human subjects actively listening to spoken narratives, in both simple and cocktail party-like auditory scenes. By modeling how acoustic features of speech are encoded in ongoing MEG activity as a spectrotemporal response function, we demonstrate that the slow temporal modulations of speech in a broad spectral region are represented bilaterally in auditory cortex by a phase-locked temporal code. For speech presented monaurally to either ear, this phase-locked response is always more faithful in the right hemisphere, but with a shorter latency in the hemisphere contralateral to the stimulated ear. When different spoken narratives are presented to each ear simultaneously (dichotic listening), the resulting cortical neural activity precisely encodes the acoustic features of both of the spoken narratives, but slightly weakened and delayed compared with the monaural response. Critically, the early sensory response to the attended speech is considerably stronger than that to the unattended speech, demonstrating top-down attentional gain control. This attentional gain is substantial even during the subjects' very first exposure to the speech mixture and therefore largely independent of knowledge of the speech content. Together, these findings characterize how the spectrotemporal features of speech are encoded in human auditory cortex and establish a single-trial-based paradigm to study the neural basis underlying the cocktail party phenomenon.

Abstract:Analogous to the visual system, auditory information has been proposed to be processed in two largely segregated streams: an anteroventral ("what") pathway mainly subserving sound identification and a posterodorsal ("where") stream mainly subserving sound localization. Despite the popularity of this assumption, the degree of separation of spatial and non-spatial auditory information processing in cortex is still under discussion. In the present study, a statistical approach was implemented to investigate potential behavioral dissociations for spatial and non-spatial auditory processing in stroke patients, and voxel-wise lesion analyses were used to uncover their neural correlates. The results generally provided support for anatomically and functionally segregated auditory networks. However, some degree of anatomo-functional overlap between "what" and "where" aspects of processing was found in the superior pars opercularis of right inferior frontal gyrus (Brodmann area 44), suggesting the potential existence of a shared target area of both auditory streams in this region. Moreover, beyond the typically defined posterodorsal stream (i.e., posterior superior temporal gyrus, inferior parietal lobule, and superior frontal sulcus), occipital lesions were found to be associated with sound localization deficits. These results, indicating anatomically and functionally complex cortical networks for spatial and non-spatial auditory processing, are roughly consistent with the dual-pathway model of auditory processing in its original form, but argue for the need to refine and extend this widely accepted hypothesis.

Abstract:In most cases, aphasia is caused by strokes involving the left hemisphere, with more extensive damage typically being associated with more severe aphasia. The classical model of aphasia commonly adhered to in the Western world is the Wernicke-Lichtheim model. The model has been in existence for over a century, and classification of aphasic symptomatology continues to rely on it. However, far more detailed models of speech and language localization in the brain have been formulated. In this regard, the dual stream model of cortical brain organization proposed by Hickok and Poeppel is particularly influential. Their model describes two processing routes, a dorsal stream and a ventral stream, that roughly support speech production and speech comprehension, respectively, in normal subjects. Despite the strong influence of the dual stream model in current neuropsychological research, there has been relatively limited focus on explaining aphasic symptoms in the context of this model. Given that the dual stream model represents a more nuanced picture of cortical speech and language organization, cortical damage that causes aphasic impairment should map clearly onto the dual processing streams. Here, we present a follow-up study to our previous work that used lesion data to reveal the anatomical boundaries of the dorsal and ventral streams supporting speech and language processing. Specifically, by emphasizing clinical measures, we examine the effect of cortical damage and disconnection involving the dorsal and ventral streams on aphasic impairment. The results reveal that measures of motor speech impairment mostly involve damage to the dorsal stream, whereas measures of impaired speech comprehension are more strongly associated with ventral stream involvement. Equally important, many clinical tests that target behaviours such as naming, speech repetition, or grammatical processing rely on interactions between the two streams. This latter finding explains why patients with seemingly disparate lesion locations often experience similar impairments on given subtests. Namely, these individuals' cortical damage, although dissimilar, affects a broad cortical network that plays a role in carrying out a given speech or language task. Abstract:<p>Hierarchy is a major organizational principle of the cortex and underscores modern computational theories of cortical function. The local microcircuit amplifies long-distance inter-areal input, which show distance-dependent changes in their laminar profiles. Statistical modeling of these changes in laminar profiles demonstrates that inputs from multiple hierarchical levels to their target areas show remarkable consistency, allowing the construction of a cortical hierarchy based on a principle of hierarchical distance. The statistical modeling that is applied to structure can also be applied to laminar differences in the oscillatory coherence between areas thereby determining a functional hierarchy of the cortex. Close examination of the anatomy of inter-areal connectivity reveals a dual counterstream architecture with well-defined distance-dependent feedback and feedforward pathways in both the supra- and infragranular layers, suggesting a multiplicity of feedback pathways with well-defined functional properties. These findings are consistent with feedback connections providing a generative network involved in a wide range of cognitive functions. A dynamical model constrained by connectivity data shed insights into the experimentally observed signatures of frequency-dependent Granger causality for feedforward versus feedback signaling. Concerted experiments capitalizing on recent technical advances and combining tract-tracing, high-resolution fMRI, optogenetics and mathematical modeling hold the promise of a much improved understanding of lamina-constrained mechanisms of neural computation and cognition. However, because inter-areal interactions involve cortical layers that have been the target of important evolutionary changes in the primate lineage, these investigations will need to include human and non-human primates comparisons.</p>

Abstract:The brains of humans and old-world monkeys show a great deal of anatomical similarity. The auditory cortical system, for instance, is organized into a ventral and a dorsal pathway in both species. A fundamental question with regard to the evolution of speech and language (as well as music) is whether human and monkey brains show principal differences in their organization (e.g., new pathways appearing as a result of a single mutation), or whether species differences are of a more subtle, quantitative nature. There is little doubt about a similar role of the ventral auditory pathway in both humans and monkeys in the decoding of spectrally complex sounds, which some authors have referred to as auditory object recognition. This includes the decoding of speech sounds ("speech perception") and their ultimate linking to meaning in humans. The originally presumed role of the auditory dorsal pathway in spatial processing, by analogy to the visual dorsal pathway, has recently been conceptualized into a more general role in sensorimotor integration and control. Specifically for speech, the dorsal processing stream plays a role in speech production as well as categorization of phonemes during on-line processing of speech.

Abstract:Understanding the organization of language in the brain has been of great interest for neurologists since the early work of Carl Wernicke and Paul Broca in the 19th century. Wernicke himself assumed a network for language consisting of two specialized regions in the frontal (i.e. Broca's area in the inferior frontal gyrus) and temporal lobe (i.e. Wernicke's area in the posterior superior temporal gyrus), which are connected by a direct pathway. He located this direct connection to the 'fibrae propriae', a fibre bundle running in the deep white matter medially to the insula. This ventral pathway was lost to research in the following decades 1 with Norman Geschwind's view of the functional neuroanatomy of language and the dorsal arcuate fascicle connecting both language regions dominating models of language in textbooks. 2

Abstract:Differences in the functional roles of the left and right cortices for speech-related processes have been known since the findings of Broca [ ] and Wernicke [ ]. Nearly 100 years later anatomical asymmetries of speech-related cortex was emphasised as a potential substrate to such functional lateralisations [ ]. Exploration of associations of anatomical asymmetries and functional lateralisations in speech has since continued, with developing technologies and theoretical insights mutually affording increasingly refined understandings. The present study is another such continuance; we outline and report associations of neuroanatomical (morphometrical) and connective (diffusion tractography) measures of speech-related cortex with differences of participant speech lateralisation and rapid temporal acuity (a hypothesised general auditory ability that contributes to superior speech processing). Review and support of developments in methodological approaches to morphometry and tractography to are also provided. Overall, our study affirms complex and selectively overlapping relationships of anatomy and connectivity (especially in the planum temporale) with behavioural language lateralisation and the processing of rapid temporal acoustics. Implications, limitations, and recommendations are discussed.

Cortical Oscillations In Auditory Perception And Speech: Evidence For Two Temporal Windows In Human Auditory Cortex

Abstract:Natural sounds, including vocal communication sounds, contain critical information at multiple time scales. Two essential temporal modulation rates in speech have been argued to be in the low gamma band (similar to 20-80 ms duration information) and the theta band (similar to 150-300 ms), corresponding to segmental and diphonic versus syllabic modulation rates, respectively. It has been hypothesized that auditory cortex implements temporal integration using time constants closely related to these values. The neural correlates of a proposed dual temporal window mechanism in human auditory cortex remain poorly understood. We recorded MEG responses from participants listening to non-speech auditory stimuli with different temporal structures, created by concatenating frequency-modulated segments of varied segment durations. We show that such non-speech stimuli with temporal structure matching speech-relevant scales (similar to 25 and similar to 200 ms) elicit reliable phase tracking in the corresponding associated oscillatory frequencies (low gamma and theta bands). In contrast, stimuli with non-matching temporal structure do not. Furthermore, the topography of theta band phase tracking shows rightward lateralization while gamma band phase tracking occurs bilaterally. The results support the hypothesis that there exists multi-time resolution processing in cortex on discontinuous scales and provide evidence for an asymmetric organization of temporal analysis (asymmetrical sampling in time, AST). The data argue for a mesoscopic-level neural mechanism underlying multi-time resolution processing: the sliding and resetting of intrinsic temporal windows on privileged time scales.

Is the Sensorimotor Cortex Relevant for Speech Perception and Understanding? An Integrative Review

Abstract:In the neuroscience of language, phonemes are frequently described as multimodal units whose neuronal representations are distributed across perisylvian cortical regions, including auditory and sensorimotor areas. A different position views phonemes primarily as acoustic entities with posterior temporal localization, which are functionally independent from frontoparietal articulatory programs. To address this current controversy, we here discuss experimental results from functional magnetic resonance imaging (fMRI) as well as transcranial magnetic stimulation (TMS) studies. On first glance, a mixed picture emerges, with earlier research documenting neurofunctional distinctions between phonemes in both temporal and frontoparietal sensorimotor systems, but some recent work seemingly failing to replicate the latter. Detailed analysis of methodological differences between studies reveals that the way experiments are set up explains whether sensorimotor cortex maps phonological information during speech perception or not. In particular, acoustic noise during the experiment and 'motor noise' caused by button press tasks work against the frontoparietal manifestation of phonemes. We highlight recent studies using sparse imaging and passive speech perception tasks along with multivariate pattern analysis (MVPA) and especially representational similarity analysis (RSA), which succeeded in separating acoustic-phonological from general-acoustic processes and in mapping specific phonological information on temporal and frontoparietal regions. The question about a causal role of sensorimotor cortex on speech perception and understanding is addressed by reviewing recent TMS studies. We conclude that frontoparietal cortices, including ventral motor and somatosensory areas, reflect phonological information during speech perception and exert a causal influence on language understanding.

Abstract:Abstract Human speech can be comprehended using only auditory information from the talker's voice. However, comprehension is improved if the talker's face is visible, especially if the auditory information is degraded as occurs in noisy environments or with hearing loss. We explored the neural substrates of audiovisual speech perception using electrocorticography, direct recording of neural activity using electrodes implanted on the cortical surface. We observed a double dissociation in the responses to audiovisual speech with clear and noisy auditory component within the superior temporal gyrus (STG), a region long known to be important for speech perception. Anterior STG showed greater neural activity to audiovisual speech with clear auditory component, whereas posterior STG showed similar or greater neural activity to audiovisual speech in which the speech was replaced with speech-like noise. A distinct border between the two response patterns was observed, demarcated by a landmark corresponding to the posterior margin of Heschl's gyrus. To further investigate the computational roles of both regions, we considered Bayesian models of multisensory integration, which predict that combining the independent sources of information available from different modalities should reduce variability in the neural responses. We tested this prediction by measuring the variability of the neural responses to single audiovisual words. Posterior STG showed smaller variability than anterior STG during presentation of audiovisual speech with noisy auditory component. Taken together, these results suggest that posterior STG but not anterior STG is important for multisensory integration of noisy auditory and visual speech.

The maps problem and the mapping problem: two challenges for a cognitive neuroscience of speech and language

Abstract:Research on the brain basis of speech and language faces theoretical and empirical challenges. Most current research, dominated by imaging, deficit-lesion, and electrophysiological techniques, seeks to identify regions that underpin aspects of language processing such as phonology, syntax, or semantics. The emphasis lies on localization and spatial characterization of function. The first part of the paper deals with a practical challenge that arises in the context of such a research programme. This maps problem concerns the extent to which spatial information and localization can satisfy the explanatory needs for perception and cognition. Several areas of investigation exemplify how the neural basis of speech and language is discussed in those terms (regions, streams, hemispheres, networks). The second part of the paper turns to a more troublesome challenge, namely how to formulate the formal links between neurobiology and cognition. This principled problem thus addresses the relation between the primitives of cognition (here speech, language) and neurobiology. Dealing with this mapping problem invites the development of linking hypotheses between the domains. The cognitive sciences provide granular, theoretically motivated claims about the structure of various domains (the "cognome"); neurobiology, similarly, provides a list of the available neural structures. However, explanatory connections will require crafting of computationally explicit linking hypotheses at the right level of abstraction. For both the practical maps problem and the principled mapping problem, developmental approaches and evidence can play a central role in the resolution.

Abstract:Models of speech perception are centered around a hierarchy in which auditory representations in the thalamus propagate to primary auditory cortex, then to the lateral temporal cortex, and finally through dorsal and ventral pathways to sites in the frontal lobe. However, evidence for short latency speech responses and low-level spectrotemporal representations in frontal cortex raises the question of whether speech-evoked activity in frontal cortex strictly reflects downstream processing from lateral temporal cortex or whether there are direct parallel pathways from the thalamus or primary auditory cortex to the frontal lobe that supplement the traditional hierarchical architecture. Here, we used high-density direct cortical recordings, high-resolution diffusion tractography, and hemodynamic functional connectivity to evaluate for evidence of direct parallel inputs to frontal cortex from low-level areas. We found that neural populations in the frontal lobe show speech-evoked responses that are synchronous or occur earlier than responses in the lateral temporal cortex. These short latency frontal lobe neural populations encode spectrotemporal speech content indistinguishable from spectrotemporal encoding patterns observed in the lateral temporal lobe, suggesting parallel auditory speech representations reaching temporal and frontal cortex simultaneously. This is further supported by white matter tractography and functional connectivity patterns that connect the auditory nucleus of the thalamus (medial geniculate body) and the primary auditory cortex to the frontal lobe. Together, these results support the existence of a robust pathway of parallel inputs from low-level auditory areas to frontal lobe targets and illustrate long-range parallel architecture that works alongside the classical hierarchical speech network model.

Abstract:Built on an analogy between the visual and auditory systems, the following dual stream model for language processing was suggested recently: a dorsal stream is involved in mapping sound to articulation, and a ventral stream in mapping sound to meaning. The goal of the study presented here was to test the neuroanatomical basis of this model. Combining functional magnetic resonance imaging (fMRI) with a novel diffusion tensor imaging (DTI)-based tractography method we were able to identify the most probable anatomical pathways connecting brain regions activated during two prototypical language tasks. Sublexical repetition of speech is subserved by a dorsal pathway, connecting the superior temporal lobe and premotor cortices in the frontal lobe via the arcuate and superior longitudinal fascicle. In contrast, higher-level language comprehension is mediated by a ventral pathway connecting the middle temporal lobe and the ventrolateral prefrontal cortex via the extreme capsule. Thus, according to our findings, the function of the dorsal route, traditionally considered to be the major language pathway, is mainly restricted to sensory-motor mapping of sound to articulation, whereas linguistic processing of sound to meaning requires temporofrontal interaction transmitted via the ventral route.

Abstract:Auditory word-form recognition was originally proposed by Wernicke to occur within left superior temporal gyrus (STG), later further specified to be in posterior STG. To account for clinical observations (specifically paraphasia), Wernicke proposed his sensory speech center was also essential for correcting output from frontal speech-motor regions. Recent work, in contrast, has established a role for anterior STG, part of the auditory ventral stream, in the recognition of species-specific vocalizations in nonhuman primates and word-form recognition in humans. Recent work also suggests monitoring self-produced speech and motor control are associated with posterior STG, part of the auditory dorsal stream. Working without quantitative methods or evidence of sensory cortex' hierarchical organization, Wernicke co-localized functions that today appear dissociable. "Wernicke's area" thus may be better construed as two cortical modules, an auditory word-form area (AWFA) in the auditory ventral stream and an "inner speech area" in the auditory dorsal stream.
