Abstract:Hydrogels have emerged as promising candidates for biomedical applications, especially in the field of antibacterial therapeutics, due to their unique structural properties, highly tunable physicochemical properties, and excellent biocompatibility. The integration of stimuli‐responsive functions into antibacterial hydrogels holds the potential to enhance their antibacterial properties and therapeutic efficacy, dynamically responding to different external or internal stimuli, such as pH, temperature, enzymes, and light. Therefore, this review describes the applications of hydrogel dressings responsive to different stimuli in antibacterial therapy. The collaborative interaction between stimuli‐responsive hydrogels and antibacterial materials was discussed. This synergistic approach, in contrast to conventional antibacterial materials, not only amplifies the antibacterial effect but also alleviates adverse side effects and diminishes the incidence of multiple infections and drug resistance. The review provides a comprehensive overview of the current challenges and outlines future research directions for stimuli‐responsive antibacterial hydrogels. It underscores the imperative for ongoing interdisciplinary research aimed at unraveling the mechanisms of wound healing. This understanding is crucial for optimizing the design and implementation of stimuli‐responsive antibacterial hydrogels. Ultimately, this review aims to offer scientific guidance for the development and practical clinical application of stimuli‐responsive antibacterial hydrogel dressings. Abstract:Regenerative medicine seeks to assess how materials fundamentally affect cellular functions to improve retaining, restoring, and revitalizing damaged tissues and cancer therapy. As potential candidates in regenerative medicine, hydrogels have attracted much attention due to mimicking of native cell-extracellular matrix (ECM) in cell biology, tissue engineering, and drug screening over the past two decades. In addition, hydrogels with a high capacity for drug loading and sustained release profile are applicable in drug delivery systems. Recently, self-healing supramolecular hydrogels, as a novel class of biomaterials, are being used in preclinical trials with benefits such as biocompatibility, native tissue mimicry, and injectability via a reversible crosslink. Meanwhile, the localized therapeutics agent delivery is beneficial due to the ability to deliver more doses of therapeutic agents to the targeted site and the ability to overcome post-surgical complications, inflammation, and infections. These highly potential materials can help address the limitations of current drug delivery systems and the high clinical demand for customized drug release systems. To this aim, the current review presents the state-of-the-art progress of multifunctional and self-healable hydrogels for a broad range of applications in cancer therapy, tissue engineering, and regenerative medicine.

Abstract:Hydrogels are a network of crosslinked polymers which can hold a huge amount of water in their matrix. These might be soft, flexible, and porous resembling living tissues. The incorporation of different biocompatible materials and nanostructures into the hydrogels has led to emergence of multifunctional hydrogels with advanced properties. There are broad applications of hydrogels such as tissue culture, drug delivery, tissue engineering, implantation, water purification, and dressings. Besides these, it can be utilized in the field of medical surgery, in biosensors, targeted drug delivery, and drug release. Similarly, hyaluronic acid hydrogels have vast applications in biomedicines such as cell delivery, drug delivery, molecule delivery, micropatterning in cellular biology for tissue engineering, diagnosis and screening of diseases, tissue repair and stem cell microencapsulation in case of inflammation, angiogenesis, and other biological developmental processes. The properties like swellability, de-swellability, biodegradability, biocompatibility, and inert nature of the hydrogels in contact with body fluids, blood, and tissues make its tremendous application in the field of modern biomedicines nowadays. Various modifications in hydrogel formulations have widened their therapeutic applicability. These include 3D printing, conjugation, thiolation, multiple anchoring, and reduction. Various hydrogel formulations are also capable of dual drug delivery, dental surgery, medicinal implants, bone diseases, and gene and stem cells delivery. The presented review summarizes the unique properties of hydrogels along with their methods of preparation and significant biomedical applications as well as different types of commercial products available in the market and the regulatory guidance.

Abstract:In modern times, biological materials have received a lot of attention due to their potentials in the treatment of body defects. Among biomaterials, hydrogels have a high advantage due to their multifunctional structures and behavior. However, the mechanical failure of conventional hydrogels under normal conditions remains a significant problem. To overcome this lesion, self-healing hydrogel materials have been created that have the property of self-healing and are minimally invasive for repairing biological defects in the whole body. In this review study, by combining the developments of recent articles in synthesis and discussion of clinical applications for repairing various types of body defects, the development of self-healing hydrogels is presented. By reviewing the characteristics and behaviors of healable hydrogels, the repair of different types of defects as well as their mechanism is presented. Some properties of polymers such as molecular mass, hydrophilicity, functional groups, intramolecular and extramolecular interactions can affect the properties of self-healing hydrogels. The review examines the development of smart self-healing hydrogel processes over the past 10 years, focusing primarily on their recent ability to repair tissue defects. This paper also presents various methods of synthesis and types of polymers and discusses clinical applications that facilitate the improvement of lesions and enhance organs function. The issues presented here may be of interest to researchers interested in improving self-healing hydrogels.

Abstract:<p>Advanced forms of hydrogels have many inherently desirable properties and can be designed with different structures and functions. In particular, bioresponsive multifunctional hydrogels can carry out sophisticated biological functions. These include in situ single-cell approaches, capturing, analysis, and release of living cells, biomimetics of cell, tissue, and tumor-specific niches. They can allow in vivo cell manipulation and act as novel drug delivery systems, allowing diagnostic, therapeutic, vaccination, and immunotherapy methods. In the present review of multitasking hydrogels, new approaches and devices classified into point-of-care testing (POCT), microarrays, single-cell/rare cell approaches, artificial membranes, biomimetic modeling systems, nanodoctors, and microneedle patches are summarized. The potentials and application of each format are critically discussed, and some limitations are highlighted. Finally, how hydrogels can enable an "all-in-one platform" to play a key role in cancer therapy, regenerative medicine, and the treatment of inflammatory, degenerative, genetic, and metabolic diseases is being looked forward to.</p>

Abstract:Injectable hydrogels, as a kind of highly hydrated soft materials, are of interest for biomedicine due to their precise implantation and minimally invasive local drug delivery around the implantation site. The combination of in situ gelation ability and versatile therapeutic agents/cells loading capabilities makes injectable hydrogels ideal materials for drug delivery, tissue engineering, wound dressing and tumor treatment. Especially, the stimuli-responsive injectable hydrogels that can respond to different stimuli in and out of the body (e.g., temperature, pH, redox conditions, light, magnetic fields, etc.) have significant advantages in biomedicine. Here, we summarize the design strategies, advantages, and recent developments of stimuli-responsive injectable hydrogels in different biomedical fields. Challenges and future perspectives of stimuli-responsive injectable hydrogels are also discussed, where we highlight the future steps necessary to fulfill the potential of these promising materials.

Abstract:Introduction: With the development of hydrogels from half a century ago, their application in various medical fields, including drug delivery has been widely expanded. Hydrogels used in this field are produced with synthetic polymers such as polyvinylpyrrolidone, polyvinyl alcohol, or natural polymers like chitosan, agarose, and hyaluronic acid to develop biocompatible, biodegradable, and non-immunogenic drug carriers. However, limitations such as inadequate response to stimulation, low homogeneity, and poor loading capacity for hydrophobic drugs have limited the use of hydrogels for drug delivery due to the hydrophilic nature of the hydrogel. The use of nanoparticles in the structure of hydrogels to produce hydrogel nanocomposites leads to more diverse interactions such as hydrogen and electrostatic bonds in addition to covalent interactions between hydrogel polymers. In addition to enhancing the mechanical properties of the hydrogel and further homogeneity, these interactions lead to the formation of platforms responsive to various stimuli, attaining sustained release, and ameliorating the poor loading of hydrophobic drugs used in cancer treatment and wound dressing. Conclusion: A review of a research conducted in the last 20 years represents that the application of nanocomposite hydrogels in drug delivery includes a wide range of production methods, nanoparticles to create various stimulation mechanisms, and therapeutic applications. Indeed, research has been focused on developing smart systems for controlled release with stimuli to reduce side effects of conventional cancer treatment methods, such as chemotherapy, by targeting drug delivery and reducing drug administration frequency and mitigating chronic wound complications by the release of growth factors.

Abstract:Hydrogels are known for their high water retention capacity and biocompatibility and have become essential materials in tissue engineering and drug delivery systems. This review explores recent advancements in hydrogel technology, focusing on innovative types such as self-healing, tough, smart, and hybrid hydrogels, each engineered to overcome the limitations of conventional hydrogels. Self-healing hydrogels can autonomously repair structural damage, making them well-suited for applications in dynamic biomedical environments. Tough hydrogels are designed with enhanced mechanical properties, enabling their use in load-bearing applications such as cartilage regeneration. Smart hydrogels respond to external stimuli, including changes in pH, temperature, and electromagnetic fields, making them ideal for controlled drug release tailored to specific medical needs. Hybrid hydrogels, made from both natural and synthetic polymers, combine bioactivity and mechanical resilience, which is particularly valuable in engineering complex tissues. Despite these innovations, challenges such as optimizing biocompatibility, adjusting degradation rates, and scaling up production remain. This review provides an in-depth analysis of these emerging hydrogel technologies, highlighting their transformative potential in both tissue engineering and drug delivery while outlining future directions for their development in biomedical applications.

Abstract:In recent years, significant advancements in the field of advanced materials and hydrogel engineering have enabled the design and fabrication of smart hydrogels and nanogels that exhibit sensitivity to specific signals or pathological conditions, leading to a wide range of applications in drug delivery and disease treatment. This comprehensive review aims to provide an in-depth analysis of the stimuli-responsive principles exhibited by smart hydrogels in response to various triggers, such as pH levels, temperature fluctuations, light exposure, redox conditions, or the presence of specific biomolecules. The functionality and performance characteristics of these hydrogels are highly influenced by both their constituent components and fabrication processes. Key design principles, their applications in disease treatments, challenges, and future prospects were also discussed. Overall, this review aims to contribute to the current understanding of gel-based drug delivery systems and stimulate further research in this rapidly evolving field.

Abstract:MXene‐based hydrogels represent a significant advancement in biomedical material science, leveraging the unique properties of 2D MXenes and the versatile functionality of hydrogels. This review discusses recent developments in the integration of MXenes into hydrogel matrices, focusing on their biomedical applications such as wound healing, drug delivery, antimicrobial activity, tissue engineering, and biosensing. MXenes, due to their remarkable electrical conductivity, mechanical robustness, and tunable surface chemistry, enhance the mechanical properties, conductivity, and responsiveness of hydrogels to environmental stimuli. Specifically, MXene‐based hydrogels have shown great promise in accelerating wound healing through photothermal effects, delivering drugs in a controlled manner, and serving as antibacterial agents. Their integration into hydrogels also enables applications in targeted cancer therapies, including photothermal and chemodynamic therapies, facilitated by their high conductivity and tunable properties. Despite the promising progress, challenges such as ensuring biocompatibility and optimizing the synthesis for large‐scale production remain. This review aims to provide a comprehensive overview of the current state of MXene‐based hydrogels in biomedical applications, highlighting the ongoing advancements and potential future directions for these multifunctional materials.

Abstract:Abstract Hydrogel is a type of versatile platform with various biomedical applications after rational structure and functional design that leverages on material engineering to modulate its physicochemical properties (e.g., stiffness, pore size, viscoelasticity, microarchitecture, degradability, ligand presentation, stimulus-responsive properties, etc.) and influence cell signaling cascades and fate. In the past few decades, a plethora of pioneering studies have been implemented to explore the cell–hydrogel matrix interactions and figure out the underlying mechanisms, paving the way to the lab-to-clinic translation of hydrogel-based therapies. In this review, we first introduced the physicochemical properties of hydrogels and their fabrication approaches concisely. Subsequently, the comprehensive description and deep discussion were elucidated, wherein the influences of different hydrogels properties on cell behaviors and cellular signaling events were highlighted. These behaviors or events included integrin clustering, focal adhesion (FA) complex accumulation and activation, cytoskeleton rearrangement, protein cyto-nuclei shuttling and activation (e.g., Yes-associated protein (YAP), catenin, etc.), cellular compartment reorganization, gene expression, and further cell biology modulation (e.g., spreading, migration, proliferation, lineage commitment, etc.). Based on them, current in vitro and in vivo hydrogel applications that mainly covered diseases models, various cell delivery protocols for tissue regeneration and disease therapy, smart drug carrier, bioimaging, biosensor, and conductive wearable/implantable biodevices, etc. were further summarized and discussed. More significantly, the clinical translation potential and trials of hydrogels were presented, accompanied with which the remaining challenges and future perspectives in this field were emphasized. Collectively, the comprehensive and deep insights in this review will shed light on the design principles of new biomedical hydrogels to understand and modulate cellular processes, which are available for providing significant indications for future hydrogel design and serving for a broad range of biomedical applications.

Abstract:Protein-based hydrogels are considered ideal biomaterials due to their high biocompatibility, diverse structure, and their improved bioactivity and biodegradability. However, it remains challenging to mimic the native extracellular matrices that can dynamically respond to environmental stimuli. The combination of stimuli-responsive functionalities with engineered protein hydrogels has facilitated the development of new smart hydrogels with tunable biomechanics and biological properties that are triggered by cyto-compatible stimuli. This review summarizes the recent advancements of responsive hydrogels prepared from engineered proteins and integrated with physical, chemical or biological responsive moieties. We underscore the design principles and fabrication approaches of responsive protein hydrogels, and their biomedical applications in disease treatment, drug delivery, and tissue engineering are briefly discussed. Finally, the current challenges and future perspectives in this field are highlighted.

Abstract:In recent years, smart/stimuli-responsive hydrogels have drawn tremendous attention for their varied applications, mainly in the biomedical field. These hydrogels are derived from different natural and synthetic polymers but are also composite with various organic and nano-organic fillers. The basic functions of smart hydrogels rely on their ability to change behavior; functions include mechanical, swelling, shaping, hydrophilicity, and bioactivity in response to external stimuli such as temperature, pH, magnetic field, electromagnetic radiation, and biological molecules. Depending on the final applications, smart hydrogels can be processed in different geometries and modalities to meet the complicated situations in biological media, namely, injectable hydrogels (following the sol-gel transition), colloidal nano and microgels, and three dimensional (3D) printed gel constructs. In recent decades smart hydrogels have opened a new horizon for scientists to fabricate biomimetic customized biomaterials for tissue engineering, cancer therapy, wound dressing, soft robotic actuators, and controlled release of bioactive substances/drugs. Remarkably, 4D bioprinting, a newly emerged technology/concept, aims to rationally design 3D patterned biological matrices from synthesized hydrogel-based inks with the ability to change structure under stimuli. This technology has enlarged the applicability of engineered smart hydrogels and hydrogel composites in biomedical fields. This paper aims to review stimuli-responsive hydrogels according to the kinds of external changes and t recent applications in biomedical and 4D bioprinting.

Abstract:Recent advances and applications of biomolecule-responsive hydrogels, namely, glucose-responsive hydrogels, protein-responsive hydrogels, and nucleic-acid-responsive hydrogels are highlighted. However, achieving the ultimate purpose of using biomolecule-responsive hydrogels in preclinical and clinical areas is still at the very early stage and calls for more novel designing concepts and advance ideas. On the way toward the real/clinical application of biomolecule-responsive hydrogels, plenty of factors should be extensively studied and examined under both in vitro and in vivo conditions. For example, biocompatibility, biointegration, and toxicity of biomolecule-responsive hydrogels should be carefully evaluated. From the living body's point of view, biocompatibility is seriously depended on the interactions at the tissue/polymer interface. These interactions are influenced by physical nature, chemical structure, surface properties, and degradation of the materials. In addition, the developments of advanced hydrogels with tunable biological and mechanical properties which cause no/low side effects are of great importance.

Abstract:Currently, surgical operations, followed by systemic drug delivery, are the prevailing treatment modality for most diseases, including cancers and trauma-based injuries. Although effective to some extent, the side effects of surgery include inflammation, pain, a lower rate of tissue regeneration, disease recurrence, and the non-specific toxicity of chemotherapies, which remain significant clinical challenges. The localized delivery of therapeutics has recently emerged as an alternative to systemic therapy, which not only allows the delivery of higher doses of therapeutic agents to the surgical site, but also enables overcoming post-surgical complications, such as infections, inflammations, and pain. Due to the limitations of the current drug delivery systems, and an increasing clinical need for disease-specific drug release systems, hydrogels have attracted considerable interest, due to their unique properties, including a high capacity for drug loading, as well as a sustained release profile. Hydrogels can be used as local drug performance carriers as a means for diminishing the side effects of current systemic drug delivery methods and are suitable for the majority of surgery-based injuries. This work summarizes recent advances in hydrogel-based drug delivery systems (DDSs), including formulations such as implantable, injectable, and sprayable hydrogels, with a particular emphasis on stimuli-responsive materials. Moreover, clinical applications and future opportunities for this type of post-surgery treatment are also highlighted.

Abstract:Hydrogels have established their significance as prominent biomaterials within the realm of biomedical research. However, injectable hydrogels have garnered greater attention compared to their conventional counterparts due to their excellent minimally invasive nature and adaptive behavior post-injection. With the rapid advancement of emerging chemistry and deepened understanding of biological processes, contemporary injectable hydrogels have been endowed with an "intelligent" capacity to respond to various endogenous/exogenous stimuli (such as temperature, pH, light and magnetic field). This innovation has spearheaded revolutionary transformations across fields such as tissue engineering repair, controlled drug delivery, disease-responsive therapies, and beyond. In this review, we comprehensively expound upon the raw materials (including natural and synthetic materials) and injectable principles of these advanced hydrogel, concurrently providing a detailed discussion of the prevalent strategies for conferring stimulus responsiveness. Finally, we elucidate the latest applications of these injectable "smart" stimuli-responsive hydrogels in the biomedical domain, offering insights into their prospects.

Abstract:Hydrogels, with self‐healing properties that can self‐repair spontaneously when subjected to mechanical stress, are gaining popularity in the biomedical field. Numerous attempts have been made to create distinctive hydrogels with self‐healing properties, along with stimuli‐responsiveness and biocompatibility. Several techniques exist for fabricating hydrogels, including physical and chemical crosslinking via the creation of covalent bonds, and so on. Here, we prepared self‐healing, stimuli‐responsive, mineralized hydrogel by simply dissolving Kollidon 90‐F, sodium chloride (NaCl), and potassium carbonate (K2CO3) in an aqueous solution. The dissociated CO32− replaces the water molecules from the Kollidon 90‐F polymer backbone and facilitates the cross‐linking of the polymer chain, resulting in hydrogel formation. In addition, the in‐situ produced sodium carbonate (Na2CO3) strengthens the hydrogel network. We optimized the mineralized hydrogels by taking various metal salts and different concentrations of K2CO3. The optimized hydrogel showed good stability over a period of time, was able to maintain viscoelastic properties, possessed good self‐healing ability, and showed a shape retention ability. The shear‐thinning property demonstrated by the optimized hydrogel could open a ray of hope in the bioprinting or 3D printing industry. Further, the stretch‐responsive release of dye from the Self‐healing mineralized hydrogel (SHMH) matrix confirms the mechanoresponsive behavior of the hydrogel. Overall, the findings could be utilized in the future to fabricate a stable drug delivery system that can autonomously release the drug molecules when stretched by daily processes such as joint movements.

Abstract:Self-healing hydrogels have emerged as the most promising alternatives to conventional brittle hydrogels used in the biomedical field due to the features of long-term stability and durability. While, the incompatibility between the fast self-healing property and enough mechanical strength of hydrogels remains a challenge. Therefore, hydrogels that possess not only mechanical toughness but also autonomous self-healing capacity are becoming sought after. This review presents a comprehensive summary of the latest self-healing mechanisms. Specifically, we review various systems based on dynamic bonds, ranging from dynamic covalent bonds to noncovalent bonds. Additionally, this review presents different characterization methods for self-healing hydrogels, along with highlighting their potential applications in the biomedical field, such as tissue engineering, drug delivery carriers, cell therapy, and wound dressing. Furthermore, this review aims to provide valuable guidance for constructing diverse self-healing hydrogels with tailored functions.

Abstract:The capacity of materials to detect and adapt to environmental changes has long fascinated researchers, leading to the development of smart materials. These materials, particularly smart hydrogels with stimulus-responsive and mechanically enhanced properties have been the focus of extensive research and application in various fields. This review delves into the fundamental principles shaping the structure and functionality of smart hydrogels. It examines the complex chemical interactions and critical factors in creating these intelligent networks, providing insights into optimizing smart hydrogel responses through system adjustments. Additionally, the review presents a prospective view, highlighting the challenges and potential for incorporating smart hydrogels into advanced, intelligent systems for biological application. The objective is to uncover the underlying mechanisms of smart hydrogels, paving the way for novel biomaterials and applications in healthcare systems.
