Abstract:Based on long-term and often frustrating experiences with the poor quality of tapeworms (Cestoda) collected throughout the world for taxonomic and phylogenetic studies, and considering the increasing obstacles to obtaining new material, a simple, easy-to-use and illustrated methodological guide (manual) is provided. It focusses mainly on key steps in examining hosts, collecting cestodes from poikilothermous vertebrates except elasmobranchs, i.e., from ray-finned fish (Actinopterygii), amphibians and 'reptiles' (a paraphyletic group comprising all sauropsids except birds), and fixing them for subsequent morphological and molecular study. It is proposed that the following methodological points should be followed: (i) ideally only freshly euthanised hosts (not previously frozen) should be used for parasitological examination; (ii) hosts examined should be documented by photographs; host tissue should also be preserved for future genotyping if necessary; (iii) tapeworms should be detached carefully to keep the scolex intact and properly cleaned before fixation; (iv) a small piece of cestode tissue should be always preserved in molecular grade ethanol for DNA sequencing; (v) tapeworms should be fixed as quickly as possible after collecting them and while they are still alive, always using hot (heated) fixatives; this prevents unnatural contraction or deformation and ensures uniform fixation; (vi) each sample (vial) should be properly labelled (a unique code should be given to every cestode sample); (vii) vouchers of sequenced specimens (hologenophores or paragenophores) should always be preserved for identification, and deposited in internationally recognised collections. It is hoped that this guide helps researchers and students to properly process valuable material of cestodes to make it suitable for reliable identification including genotyping and comparative anatomy, which is a prerequisite for any subsequent ecological, biogeographical, phylogenetic life cycle or molecular study.

Abstract:In March, 1929, there was referred to the writer for diagnosis a complete strobila of an unarmed tapeworm, passed by a native white woman from St. John Parish, Louisiana, after administration of oleoresin of male fern. The worm grossly resembled Taenia saginata, but differed in several specific characters, including a more distinctly oval scolex, a pair of lateral elytra along the unsegmented region behind the scolex, longer and narrower terminal proglottids, and more clumped, less discrete branches of the uterus in these proglottids. In view of the fact that only 3 cases of this infection were on record (Nebraska, 2, Ward, 1 Guyer, 2 Texas, 1, Chandler 3 ) and that this specimen was the first one to be diagnosed in the unpreserved condition, it seemed desirable to attempt to determine by experimental feeding what animal served as larval host. Information elicited from the patient indicated that she occasionally ate pork, rabbits and deer meat, and that she was fond of raw beef. A 2-weeks old suckling calf and a rabbit were therefore secured and were each fed macerated terminal proglottids (e. g., gravid segments) of the tapeworm. After a period of 3 months, during which time the animals were kept under rigid supervision, they were autopsied. The rabbit was found to contain 3 specimens of an armed cysticercus, C. pisiformis, a natural infection of rabbits, while the calf was infested with more than a thousand cysticerci, which had unarmed scolices, and which were considerably smaller than those of Taenia saginata. The extremely heavy infestation of the calf under circumstances that practically precluded a natural infection with larvae of T. snginata, together with the smaller size of the cysticerci recovered, has practically established the fact that these larvae were the resultant growth from the eggs which had been fed 3 months previously.

Abstract:The ectoparasitic copepod Lernaea cyprinacea (anchor worm) has more than 100 host species among teleost fishes and affects cyprinids both in fish farms and natural waters. In addition, while L. cyprinacea infection in amphibians has been recorded in Asia, North and South America, there is no data available in the literature on their presence in Europe. In this study, we first reported L. cf. cyprinacea parasitising an anuran tadpole in Europe. Specimens of L. cf. cyprinacea were observed attached to a tadpole of the agile frog (Rana dalmatina), which was caught during fishing of crucian carp fingerlings (Carassius carassius) from a small fish pond in Hungary during the summer of 2012. The infected tadpole was collected from a rearing pond, where juvenile crucian carps were kept. The tadpole was inspected in the laboratory, and digital photos were taken. The parasites were found attached to the body-tail junction and to the leg of the tadpole (at Gosner stage 41). The parasite species was identified as L. cf. cyprinacea based on morphological traits.
Diversity and biology of Spirometra tapeworms (Cestoda: Diphyllobothriidea), zoonotic parasites of wildlife: A review

Abstract:Tapeworms of the genus Spirometra Faust, Campbell et Kellogg, 1929 have long been known as intestinal parasites of carnivores and their larvae (spargana) have been found in various vertebrates. Nevertheless, their species diversity, host associations and geographical distribution remain poorly understood. Molecular data clearly confirm the validity of the genus, which has been synonymised by several authors with Diphyllobothrium Cobbold, 1858. Despite morphological similarities between the species of the two genera, they are not closely related and also differ in their life cycle. The present review provides a list of the species recognised as valid and additional genotypes that may represent other species, with a basic characterisation of each taxon and comments on their validity, the probable range of definitive and intermediate hosts, and their distribution. The existing taxonomic problems and the insufficient knowledge of the host specificity and distribution of Spirometra tapeworms can only be solved by combining molecular and morphological data, i.e. by comparing genetically characterised specimens with corresponding morphological vouchers (hologenophores). Further targeted sampling and surveys are required to clarify the distribution and host associations.

Abstract:A new species of tapeworm, Taenia lynciscapreoli sp. n. (Cestoda, Cyclophyllidea), is described from the Eurasian lynx (Lynx lynx), the main definitive host, and the roe deer (Capreolus capreolus and Capreolus pygargus), the main intermediate hosts, from Finland and Russia (Siberia and the Russian Far East). The new species was found once also in the wolf (Canis lupus) and the Eurasian elk/moose (Alces alces), representing accidental definitive and intermediate hosts, respectively. The conspecificity of adult specimens and metacestodes of Taenia lynciscapreoli sp. n. in various host species and regions, and their distinction from related species of Taenia, was confirmed by partial nucleotide sequences of the mitochondrial cytochrome c oxidase subunit 1 gene. Morphologically, Taenia lynciscapreoli sp. n. can be separated unambiguously from all other species of Taenia by the shape of its large rostellar hooks, particularly the characteristically short, wide and strongly curved blade. If the large rostellar hooks are missing, Taenia lynciscapreoli may be separated from related species by a combination of morphological features of mature proglottids. It is suggested that Taenia lynciscapreoli has been present in published materials concerning the tapeworms of Lynx lynx and Lynx pardinus in Europe, but has been misidentified as Taenia pisiformis (Bloch, 1780). Taenia lynciscapreoli sp. n. has not been found in lynx outside the range of roe deer, suggesting a transmission pathway based on a specific predator-prey relationship. The present study applies a novel, simple approach to compare qualitative interspecific differences in the shape of rostellar hooks.

Abstract:The life cycles of species of Proteocephalus Weinland, 1858 (Cestoda: Proteocephalidea) parasitizing fishes in the Palearctic Region are reviewed on the basis of literary data and personal experimental observations, with special attention being paid to the development within the intermediate and definitive hosts. Planktonic crustaceans, diaptomid or cyclopid copepods (Copepoda), serve as the only intermediate hosts of all Proteocephalus species considered. A metacestode, or procercoid, develops in the body cavity of these planktonic crustaceans and the definitive host, a fish, becomes infected directly after consuming them. No previous reports of the parenteral location of metacestodes within the second intermediate host as it is in the Nearctic species P. ambloplitis have been recorded. Thus, the life cycles of Proteocephalus tapeworms resemble in their general patterns those of some pseudophyllidean cestodes such as Eubothrium or Bothriocephalus, differing from the latter in the presence of a floating eggs instead of possessing an operculate egg from which a ciliated, freely swimming larva, a coracidium, is liberated. The scolex of Proteocephalus is already formed at the stage of the procercoid within the copepod intermediate host; in this feature, proteocephalideans resemble caryophyllidean rather than pseudophyllidean cestodes. The morphology of procercoids of individual species is described with respect to the possibility of their differentiation and data on the spectrum of intermediate hosts are summarized. Procercoids of most taxa have a cercomer, which does not contain embryonic hooks in contrast to most pseudophyllidean cestodes. The role of invertebrates (alder-fly larvae — Megaloptera) and small prey fishes feeding upon plankton in the transmission of Proteocephalus tapeworms still remains unclear but these hosts are likely to occur in the life cycle. Data on the establishment of procercoids in definitive hosts, morphogenesis of tapeworms within fish hosts, and the length of the prepatent period are still scarce and new observations are needed. Whereas extensive information exists on the development of P. longicollis (syns. P. exiguus and P. neglectus), almost no data are available on the ontogeny of other taxa, in particular those occurring in brackish waters (P. gobiorum, P. tetrastomus). The morphology of P. cernuae and P. osculatus procercoids from experimentally infected intermediate hosts is described for the first time.

Abstract:Parasitic protozoans, helminths, alter the gut microbiota in mammals, yet little is known about the influence of intestinal cestodes on gut microbiota in fish. In the present study, the composition and diversity of the hindgut microbiota were determined in the intestine of common carp (Cyprinus carpio) infected with two tapeworm species, Khawia japonensis and Atractolytocestus tenuicollis. The intestine contained a core microbiota composed of Proteobacteria, Fusobacteria and Tenericutes. Infection with the two cestode species had no significant effect on the microbial diversity and richness, but it altered the microbial composition at the genus level. PCoA analysis indicated that microbial communities in the infected and uninfected common carp could not be distinguished from each other. However, a Mantel test indicated that the abundance of K. japonensis was significantly correlated with the microbial composition (P = 0.015), while the abundance of A. tenuicollis was not (P = 0.954). According to Pearsonʼs correlation analysis, the abundance of K. japonensis exhibited an extremely significant (P < 0.001) positive correlation with the following gut microbiota taxa: Epulopiscium, U114, Bacteroides, Clostridium and Peptostreptococcaceae (0.8< r < 0.9); and a significant (P < 0.05) correlation with Enterobacteriaceae, Micrococcaceae, Rummeliibacillus, Lysinibacillus boronitolerans, Veillonellaceae, Oxalobacteraceae, Aeromonadaceae (negative), Marinibacillus and Chitinilyticum (0.4< r < 0.7). These results suggest that the composition of gut microbiota was somewhat affected by the K. japonensis infection. Additionally, increased ratios of pathogenic bacteria (Lawsonia and Plesiomonas) were also associated with the K. japonensis infection, which may therefore increase the likelihood of disease.

Abstract:Presenting new molecular and scanning electron microscope (SEM) features, this study gives additional data to the better knowledge of Thaparocleidus vistulensis (Siwak, 1932) (Monopisthocotyla, Ancylodiscoididae), a parasite of the European catfish Silurus glanis Linnaeus, 1758 (Siluriformes, Siluridae) cultured in a commercial fish farm in Hungary. In addition, notes on the early development of sclerotized anchors are also provided. The main morphological difference of T. vistulensis compared to other congeneric species is associated with the male copulatory organ, which exhibits 5–7 loops in the middle of the penis length and a long open V-shaped sclerotized accessory piece, dividing terminally into two parts, securing the terminal part of the penis tube. The present study provides for the first time molecular characterization data based on the 2694 bp long nucleotide sequence of rDNA (ITS1, 5.8S, ITS2, and flanked with partial 18S and partial 28S) submitted in GenBank with the accession number OR916383. A phylogenetic tree based on ITS1 sequences supports a well-defined clade including T. vistulensis , forming a sister group with T. siluri, a species-specific monopisthocotylan parasite to S. glanis . The morphological characterization of T. vistulensis, especially for the male copulatory organ, together with the molecular data in the present study, extends knowledge about this monopisthocotylan species and provides new information for future phylogeny studies.

Abstract:The tapeworm Hymenolepis diminuta is a common parasite of the small intestine in rodents but it can also infect humans. Due to its characteristics and ease of maintenance in the laboratory, H. diminuta is also an important model species in studies of cestodiasis, including the search for new drugs, treatments, diagnostics and biochemical processes, as well as its host–parasite interrelationships. A great deal of attention has been devoted to the immune response caused by H. diminuta in the host, and several studies indicate that infection with H. diminuta can reduce the severity of concomitant disease. Here, we present a critical review of the experimental research conducted with the use of H. diminuta as a model organism for over more than two decades (in the 21st century). The present review evaluates the tapeworm H. diminuta as a model organism for studying the molecular biology, biochemistry and immunology aspects of parasitology, as well as certain clinical applications. It also systematizes the latest research on this species. Its findings may contribute to a better understanding of the biology of tapeworms and their adaptation to parasitism, including complex correlations between H. diminuta and invertebrate and vertebrate hosts. It places particular emphasis on its value for the further development of modern experimental parasitology.

Abstract:The hepatic nematode Calodium hepaticum is a zoonotic parasite primarily parasitising small mammals, but it can infect a wide range of mammal species, including humans. Due to its specific life cycle and transmission pattern, it is one of the least studied helminths in the world. The only documented findings of C. hepaticum from Slovakia (former Czechoslovakia) come from the 60s and 70s of the 20th Century, including nine human cases of the infection reported post-mortem. The present study was conducted in the area of these original records in the Tatra National Park (TANAP) and the Košice Zoo. In TANAP, 484 small mammals of six shrew species (Insectivora: Soricidae) and eight rodent species (Rodentia: Muroidea) were collected. In the Košice Zoo, 163 rodents from 10 species were sampled. All specimens were examined for the presence of C. hepaticum eggs using the artificial digestion method. The parasite was recorded in two shrew species (Sorex araneus and Neomys fodiens) and five rodent species (Arvicola amphibius, Microtus agrestis, Clethrionomys glareolus, Apodemus flavicollis, and Rattus norvegicus) from TANAP, while in the Košice Zoo only a single individual of R. norvegicus was found to be infected.

Abstract:Sturgeon species have inhabited the world's seas and rivers for more than 200 million years and hold significant taxonomic significance, representing a strong conservation interest in aquatic biodiversity as well as in the economic sector, as their meat and eggs (caviar) are highly valuable goods. Currently, sturgeon products and byproducts can be legally obtained from aquaculture as a sustainable source. Intensive farming practices are accompanied by parasitic infestations, while several groups of parasites have a significant impact on both wild and farmed sturgeons. The present article is a review of common sturgeon parasites from the genus: Protozoa, Trematoda, Crustacea, Nematodes, Monogenea, Hirudinea, Copepoda, Acanthocephala, Cestoda, Polypodiozoa, and Hyperoartia, while also addressing their pathology and statistical distribution.

Abstract:Parasites are ubiquitous in extant ecosystems but rarely preserved in the geological record, especially parasitic worms (helminths). One such group is Cestoda (tapeworms), a specialized endoparasitic group of platyhelminths (flatworms). They have a complex lifecycle with at least two hosts, infecting all major groups of vertebrates. However, their fossil record is extremely sparse due to their soft tissue and concealed habitats, with the only widely accepted example before the Quaternary being eggs discovered in a shark coprolite from the Permian. The lack of body fossils greatly hampers our understanding of their early evolution. We report a slender, armed fossil from mid-Cretaceous Kachin (Myanmar) amber (ca. 99 Ma). This fossil displays unique external (armature pattern) and internal (partially invaginated tentacle and rootless hooks) features that are most consistent with the tentacles of extant trypanorhynch tapeworms that parasitize marine elasmobranchs (mainly sharks and rays). Our study thus probably provides not only the first partial body fossil of a tapeworm, but also arguably the most convincing body fossil of a flatworm. In addition, the exquisite invaginated tentacle inside the fossil highlights that amber can preserve the internal structure of helminths. Remarkably, nearly all extant trypanorhynchs are endoparasites of marine elasmobranchs, thus our study provides an exceptional example of a marine endoparasite trapped in terrestrial amber.

Abstract:Trematodes and their snail hosts have developed intimate parasite-host associations, with snails supporting a diverse and often species-specific trematode fauna. In the faucet snail, Bithynia tentaculata (Caenogastropoda, Littorinimorpha), a unique trematode fauna has been recorded recently. However, knowledge of the exact species identity, phylogenetic relationships, and geographical distribution remains limited as many of the species belong to groups with unclear or controversial taxonomical assignment. To contribute to our knowledge of the trematodes, we investigated the trematode fauna of B. tentaculata by examining a total of 556 snails from lakes in County Galway, Ireland. Using an integrative taxonomic approach including DNA sequence data analyses (28S rRNA gene, ITS1-5.8S-ITS2, ITS2, cox1, nad1) and morphological tools (taxonomical drawings and measurements), we identified nine trematode species of seven families, with seven species occurring as cercariae (Cyathocotyle prussica, Lecithodendrium linstowi, Lecithodendrium sp., Asymphylodora progenetica, Sphaerostoma bramae, Metorchis xanthosomus, and Notocotylus sp.) and three species occurring as metacercariae (A. progenetica, Parasymphylodora parasquamosa, and Sphaeridiotrema sp.). Except for S. bramae, all are new species records for Ireland and provide the most western distribution of these trematodes in Europe. The trematode species recorded are known to use a wide range of definitive hosts and have a wide geographical distribution; among them are species members of genera that are zoonotic (Metorchis) and pathogenic to wildlife (Cyathocotyle, Sphaeridiotrema, and Notocotylus). There remains an ongoing need for precise identification of the trematode species to ensure that wider ecological contexts are correctly understood and biodiversity and disease threats can be accurately evaluated.

Abstract:The article provides data on the spread of parasitic diseases in carp and grass carp in the farming ponds of Aqvarest LLC and Aquacity LLC of the Odesa region. Fish diseases are registered both in natural reservoirs and in fish farms, as a result of which the growth rates, reproductive capacity, marketability of fish decrease, quality indicators and biological value deteriorate; in addition, mass kills of fish may occur. The causes of diseases can be both non-contagious (adverse environmental conditions) and contagious (viruses, bacteria, parasitic diseases). Thus, constant monitoring of fish parasitofauna in pond farms of Ukraine is necessary, since the study of patterns of occurrence and spread of fish diseases, forecasting of these diseases affects the effectiveness of aquaculture breeding and preservation of fish products. The purpose of the work was to monitor parasitic diseases in carp and white amur in the farming ponds of Aqvarest LLC and Aquacity LLC of the Odesa region. During the parasitological examination of this year's and two-year-old carp and white amur in the ponds of LLC "Aqvarest" and LLC "Aquasti" of Odesa region, five diseases were diagnosed, the causative agents of which were: Gyrodastylus birmani, Dactylogyrus vastator, Khavia sinensis, Caryophyllaeus fimbriceps and Philometroides lusiana. Gyrodastylus birmani and Caryophyllaeus fimbriceps were recorded the most in the deep pond of "Aqvarest" LLC with the extent of invasion of 25.7%, in two-year-old carp and two-year-old white amur - 21.4%. In the shallow reservoir of "Aquacity" LLC, Gyrodastylus birmani (31.4%) and Caryophyllaeus fimbriceps (25.7%) were most recorded in two-year-old carp, and in two-year-old white amur Khavia sinensis (28.5%) and Caryophyllaeus fimbriceps (32.1 %). The intensity of infestation with cestodoses (caviasis and caryophyllosis), as well as nematodes (phyllometroidosis) was low, 1-2 or 3-4 specimen for fish, and clinical manifestations of the disease were not always recorded. However, the presence of these pathogens in the fauna represents a significant danger for the development of commercial fish species, namely carp and white carp.

Abstract:The morphology of the different larval stages and life cycle of Hypoderaeum conoideum (Trematoda: Echinostomatidae) are described. The freshwater snail species Lymnaea peregra (Gastropoda: Lymnaeidae) serves as the natural first intermediate host and this and L. corvus serve as experimental first intermediate hosts. These and other freshwater snails, such as Physella acuta and Gyraulus chinensis, in turn serve as second intermediate hosts. Adult worms were obtained from chicks and ducks, but not from rats, mice and golden hamsters. The morphology of the larval stages is compared with previous work on H. conoideum. Several aspects of the biology of the life history stages are described with emphasis on the transmission dynamics of the free-living stages. Differential suitability of the snail species that may act as first and/or second intermediate hosts is studied and discussed.

Abstract:The current experimental study has been conducted to analyze cladistic taxonomic aspects of the piscian cestode parasites including its morphological characters. Freshwater fishes caught on the spot at selected sampling stations with the help of local fishermen and collected for tapeworm investigations. Freshwater catfish Clarias batrachus (Linnaeus, 1758)[3] collected from Dehradun sampling stations and examined for tapeworm parasites as per experimental design. The collected fishes were brought to the Parasitology laboratory for experimental study and morphological as well as anatomical facts investigation. Parasites obtained from infected host fishes kept intact in 5% formalin for further investigation. Ten cestode (tapeworm) parasites were found from its infested Intestine. The morphological study of tapeworms indicates that parasitic diversity is accessible within the Ganga river at Dehradun (U.K.) region. Morphotaxonomic characterization reported as - H, butterfly flap shaped ovary, testes numerous and well developed, vitelline follicle - cortical and medullary. This study provides an overview of the piscian tapeworm parasites of some freshwater fishes inhabiting the Ganga river.

Abstract:Anisakis spp. (Nematoda, Anisakidae) are parasites known by their economic and health impacts, as their L3 larval stages infect a variety of fish species, many of them commercial species, sometimes causing zoonotic episodes due to consumption of raw or undercooked fish. The aim of this study is to determine the infection process and the potential impact of A. simplex s.l. L3 on gilthead seabream (Sparus aurata L.), one of the most important fish species in Mediterranean aquaculture, by periodic histological monitoring of the infection process. For this, fish were experimentally infected with A. simplex s.l. L3 and periodically analysed for L3 larvae, collecting samples at different time points (hours post ingestion, hpi): 3, 6, 12, 18, 24, 36, 48, 72, 96, 120, 144, 168 and 192, up to 6 months post infection (mpi). All samples were observed under a stereomicroscope and later fixed for histological examination. A. simplex s.l. L3 were only found on the visceral surface and mesenteric tissue, but never free or encapsulated in muscle. Chronological events were found to occur faster than those reported in previous studies. They were first observed 6 hpi in the coelomic cavity, being present up to 48 hpi. While the earliest evidence of fibrocytes surrounding A. simplex s.l. L3 larvae were observed at 18 hpi, complete spiral encapsulation occurred by 72 hpi. Alive parasites were observed up to 6 mpi. Although the infection of gilthead seabream by Anisakis spp. larvae is feasible, it seems unlikely, especially in aquaculture given the hygienically controlled feeding systems. In the event of infection, the transmission would be unlikely due to the poor condition in which specimens of Anisakis spp. are found. Furthermore, since no larvae were detected in the fish’s muscle, human infection seems improbable.

Abstract:Myxozoans are microscopical parasites widely distributed in fish, with over 2,600 described species, but their actual diversity is still underestimated. Among salmonids, more than 70 myxozoan species have been identified. This study focuses on species of Chloromyxum Mingazzini, 1890 that infect salmonid kidneys, particularly C. majori Yasutake et Wood, 1957 and C. schurovi Shulman et Ieshko, 2003. Despite their similar spore morphology, they exhibit distinct host preferences, tissue affinities and geographical distributions. Chloromyxum schurovi predominantly infects the renal tubules of Salmo salar Linnaues and S. trutta Linnaeus in Europe, while C. majori targets the glomeruli of Oncorhynchus mykiss (Walbaum) and O. tshawytscha (Walbaum) in North America. The sequence data for C. majori and C. schurovi have been either missing or questionable. In our study, we examined the kidneys of two salmonid species for chloromyxid infections, using both morphological and molecular data to characterise Chloromyxum species in salmonids. The sequence of C. schurovi obtained in our study did not match the previously published parasite data. Instead, it clustered as an independent lineage sister to the Paramyxidium Freeman et Kristmundsson, 2018 clade gathering the species from various fish organs, including the urinary tract. Our findings clarified the taxonomic origin of the previous C. schurovi sequence as Myxidium giardi Cépède, 1906, highlighting the risks associated with the presence of myxozoan blood stages in the bloodstream of their fish host and the challenges of non-specific PCR amplification. We redescribe C. schurovi, thus contributing to a better understanding of the diversity and phylogeny of kidney-infecting species of Chloromyxum.
