Abstract:Key PointsInterleukin-10 (IL-10) is not a cell type-specific cytokine, but instead it is broadly expressed by many immune cells.Several layers of regulation regulate IL-10 production, including changes in the chromatin structure, enhancement or silencing of IL10 transcription and post-transcriptional regulatory mechanisms. Many of the molecular events leading to IL10 expression are similar and common to various IL-10-producing immune cells, but cell type-specific signals also exist.Induction of IL-10 often occurs together with pro-inflammatory cytokines, although pathways that induce IL-10 may actually negatively regulate these pro-inflammatory cytokines.Understanding the specific molecular events that regulate the expression of IL-10 will be important for the design of new strategies of immune intervention.

What problem does this paper attempt to address?
The problem that this paper attempts to solve is to understand the expression regulation mechanism of interleukin - 10 (IL - 10) in different immune cells. IL - 10 is a cytokine with anti - inflammatory properties and plays a central role in immune regulation by limiting the immune response to pathogens during the infection process to prevent host damage. However, there are still many unknowns regarding the specific regulation mechanisms of IL - 10 in different types of immune cells, especially the molecular mechanisms at the levels of signal transduction, epigenetics, transcription factor binding and gene activation. Therefore, the main objective of the paper is to deeply explore these specific molecular events in order to better understand the production regulation of IL - 10 and further provide a theoretical basis for designing new immune intervention strategies. The paper also pays special attention to the following aspects: 1. **IL - 10 expression in innate immune cells**: It discusses how pathogen - derived products induce macrophages and dendritic cells (DCs) to produce IL - 10, as well as the related signal pathways, such as Toll - like receptor (TLRs) signal pathway and its downstream ERK and p38 pathways. 2. **IL - 10 expression in adaptive immune cells**: It explores the production of IL - 10 in T helper cells (TH cells), especially the regulation of IL - 10 expression in TH1, TH2 and TH17 cells, including the required signal molecules and transcription factors. 3. **IL - 10 expression in regulatory T cells (TReg cells)**: It analyzes the expression of IL - 10 in TReg cells and its role in maintaining intestinal immune homeostasis, especially the potential therapeutic value in inflammatory bowel disease (IBD). 4. **Positive and negative feedback mechanisms of IL - 10**: It studies how IL - 10 itself regulates its expression through positive and negative feedback mechanisms and the differences of these mechanisms in different cell types. Through these studies, the paper aims to provide more in - depth insights into understanding and regulating the expression of IL - 10, thus laying the foundation for developing new immunotherapy methods.
