Abstract:In two landmark articles, Hanahan and Weinberg synthesized into one conceptual framework 'the hallmarks of cancer', a massive amount of information describing the characteristics of a cancer cell. Although this is neither the intention nor the belief of the authors, hallmarks are often interpreted as applying to a canonic cancer cell, or equally to all cells within a cancer. In this article, we clarify the separate concepts of causes, oncogenic events, signal transduction programs, and hallmarks to show that there is no unimodal relation between these concepts but a complex network of interrelations that vary in different cells, between cells, and at different times in any given cell. We consider cancer as an evolving, dynamic, and heterogeneous system, explaining, at least in part, the difficulty of treating cancer and supporting the use of simultaneous, multitarget therapies.

Abstract:The hallmarks of cancer conceptualization is a heuristic tool for distilling the vast complexity of cancer phenotypes and genotypes into a provisional set of underlying principles. As knowledge of cancer mechanisms has progressed, other facets of the disease have emerged as potential refinements. Herein, the prospect is raised that phenotypic plasticity and disrupted differentiation is a discrete hallmark capability, and that nonmutational epigenetic reprogramming and polymorphic microbiomes both constitute distinctive enabling characteristics that facilitate the acquisition of hallmark capabilities. Additionally, senescent cells, of varying origins, may be added to the roster of functionally important cell types in the tumor microenvironment. SIGNIFICANCE: Cancer is daunting in the breadth and scope of its diversity, spanning genetics, cell and tissue biology, pathology, and response to therapy. Ever more powerful experimental and computational tools and technologies are providing an avalanche of "big data" about the myriad manifestations of the diseases that cancer encompasses. The integrative concept embodied in the hallmarks of cancer is helping to distill this complexity into an increasingly logical science, and the provisional new dimensions presented in this perspective may add value to that endeavor, to more fully understand mechanisms of cancer development and malignant progression, and apply that knowledge to cancer medicine.

Abstract:The hallmarks of cancer comprise six biological capabilities acquired during the multistep development of human tumors. The hallmarks constitute an organizing principle for rationalizing the complexities of neoplastic disease. They include sustaining proliferative signaling, evading growth suppressors, resisting cell death, enabling replicative immortality, inducing angiogenesis, and activating invasion and metastasis. Underlying these hallmarks are genome instability, which generates the genetic diversity that expedites their acquisition, and inflammation, which fosters multiple hallmark functions. Conceptual progress in the last decade has added two emerging hallmarks of potential generality to this list-reprogramming of energy metabolism and evading immune destruction. In addition to cancer cells, tumors exhibit another dimension of complexity: they contain a repertoire of recruited, ostensibly normal cells that contribute to the acquisition of hallmark traits by creating the "tumor microenvironment." Recognition of the widespread applicability of these concepts will increasingly affect the development of new means to treat human cancer.

Abstract:Diagnosis and treatment of disease demand a sound understanding of the underlying mechanisms, determining any Achilles' heel that can be targeted in effective therapies. Throughout history, this endeavour to decipher the origin and mechanism of transformation of a normal cell into cancer has led to various theories—from cancer as a curse to an understanding at the level of single-cell heterogeneity, meaning even among a single sub-type of cancer there are myriad molecular challenges to overcome. With increasing insight into cancer genetics and biology, the disease has become ever more complex to understand. The complexity of cancer as a disease was distilled into key traits by Hanahan and Weinberg in their seminal ‘Hallmarks of Cancer' reviews. This lucid conceptualization of complex cancer biology is widely accepted and has helped advance cancer therapeutics by targeting the various hallmarks but, with the advancement in technologies, there is greater granularity in how we view cancer as a disease, and the additional understanding over the past decade requires us to revisit the hallmarks of cancer. Based on extensive study of the cancer research literature, we propose four novel hallmarks of cancer, namely, the ability of cells to regress from a specific specialized functional state, epigenetic changes that can affect gene expression, the role of microorganisms and neuronal signalling, to be included in the hallmark conceptualization along with evidence of various means to exploit them therapeutically.

Abstract:There is no doubt that anyone who has participated in cancer care or research has once read the 'Hallmarks of Cancer' papers published by Hanahan and Weinberg in 2001 and 2011. They initially defined the six qualities of cancer cells as cancer hallmarks in 2001, but expanded that to 11 as a next generation in 2011. In their papers, they discussed the potential treatment strategies against cancer corresponding to each of the 11 hallmarks, and to date, proposed therapies that target genes and signaling pathways associated with each of these hallmarks have guided a trail that cancer treatments should take, some of which are now used as standard in clinical practice and some of which have yet to progress that far. Along with the recent advances in cancer research such as genomic analysis with next generation sequencing, they can be reconverged to an alternative six categories defined as selective proliferative advantages, altered stress response, deregulated cellular metabolism, immune modulation and inflammation, tumor microenvironment, tissue invasion and metastasis. In this paper, we will overview the current state of these alternative hallmarks and their corresponding treatments in the current sarcoma practice, then discuss the future direction of sarcoma treatment.

Abstract:Ten years after the publication of the position paper “The hallmarks of cancer” (Hanahan and Weinberg Cell 100:57–70, 2000), it has become increasingly clear that mutated cells on their way to giving rise to a tumor have also to learn how to thrive in a chronically inflamed microenvironment, evade immune recognition, and suppress immune reactivity. Genetic and molecular definition of these three immune hallmarks of cancer offers the opportunity to learn how to deploy specific countermeasures to reverse the situation in favor of the immune system and, eventually, the patient. This new information could be channeled to address what seem to be the three major hallmarks for the immune control of cancer progression: effective procedures to activate immune reactivity; characterization of not-disposable oncoantigens; and counteraction of immune suppression.

Abstract:A thought-provoking article by Gems and de Magalhães suggests that canonic hallmarks of aging are superficial imitations of hallmarks of cancer. I took their work a step further and proposed hallmarks of aging based on a hierarchical principle and the hyperfunction theory.To do this, I first reexamine the hallmarks of cancer proposed by Hanahan and Weinberg in 2000. Although six hallmarks of cancer are genuine, they are not hierarchically arranged, i.e., molecular, intra-cellular, cellular, tissue, organismal and extra-organismal. (For example, invasion and angiogenesis are manifestations of molecular alterations on the tissue level; metastasis on the organismal level, whereas cell immortality is observed outside the host).The same hierarchical approach is applicable to aging. Unlike cancer, however, aging is not a molecular disease. The lowest level of its origin is normal intracellular signaling pathways such as mTOR that drive developmental growth and, later in life, become hyperfunctional, causing age-related diseases, whose sum is aging. The key hallmark of organismal aging, from worms to humans, are age-related diseases. In addition, hallmarks of aging can be arranged as a timeline, wherein initial hyperfunction is followed by dysfunction, organ damage and functional decline.

Abstract:Cancer can be viewed as the loss of cooperative cell behaviors that normally facilitate multicellularity, including the formation of tissues and organs. Hanahan and Weinberg describe the phenotypic differences between healthy and cancerous cells in an article titled “The Hallmarks of Cancer” (Cell, 100, 57–70, 2000). Here the authors propose six phenotypic changes at the cellular level as the essential hallmarks of cancer. They investigate the dynamics and interactions of these hallmarks in a model known as CancerSim. They describe how CancerSim implements the hallmarks in an agent-based simulation which can help test the hypotheses put forth by Hanahan and Weinberg. Experiments with CancerSim are described that study the interactions of cell phenotype alterations, and in particular, the likely sequences of precancerous mutations, known as pathways. The experiments show that sequencing is an important factor in tumorigenesis, as some mutations have preconditions—they are selectively advantageous only in combination with other mutations. CancerSim enables a modeler to study the dynamics of a developing tumor and simulate how progression can be altered by tuning model parameters.

Abstract:cer’ often describes malignant tumors. The issue of cancerous stem cells is not a novelty but has existed for centuries. Different civilizations have handled the stem cell subject depending on the knowledge at hand. A hallmark is a distinguishing factor that separates cancers from other conditions. Hence, there have been arguments about the exclusion or inclusion criteria for the six original hallmarks. This review will introduce stemness as a new hallmark for cancer and argue why it should get included. Thus, future studies will focus on how the tumor microenvironment can get used to fight cancer. Methodology. A methodology of literature search using "stemness" as the keyword gets used in the review to avoid biasedness. Results. Stem cells can undergo physiological changes to form malignant cells. Cancer cells are in a variety of cells in the body. The microenvironment of the tumor cells promotes their progression by nourishing them. Conclusions. The results imply that stemness is the behavior of stem cells to self-renew and differentiate to form cells in all organisms. Stem cells are present in all types of hematological and solid cancers. It means that a problem in the self-renewal or differentiation process can lead to the formation of tumors or tumorigenic.

Abstract:The basis for the gene mutation theory of cancer that dominates current molecular cancer research consists of: the belief that gene-level aberrations such as mutations are the main cause of cancers, the concept that stepwise gene mutation accumulation drives cancer progression, and the hallmarks of cancer. The research community swiftly embraced the hallmarks of cancer, as such synthesis has supported the notions that common cancer genes are responsible for the majority of cancers and the complexity of cancer can be dissected into simplified molecular principles. The gene/pathway classification based on individual hallmarks provides explanation for the large number of diverse gene mutations, which is in contrast to the original estimation that only a handful of gene mutations would be discovered. Further, these hallmarks have been highly influential as they also provide the rationale and research direction for continued gene-based cancer research. While the molecular knowledge of these hallmarks is drastically increasing, the clinical implication remains limited, as cancer dynamics cannot be summarized by a few isolated/fixed molecular principles. Furthermore, the highly heterogeneous genetic signature of cancers, including massive stochastic genome alterations, challenges the utility of continuously studying each individual gene mutation under the framework of these hallmarks. It is therefore necessary to re-evaluate the concept of cancer hallmarks through the lens of cancer evolution. In this analysis, the evolutionary basis for the hallmarks of cancer will be discussed and the evolutionary mechanism of cancer suggested by the genome theory will be employed to unify the diverse molecular mechanisms of cancer.

Abstract:Cancer is a complex disease in which several molecular and cellular pathways converge to foster the tumoral phenotype. Notably, in the latest iteration of the cancer hallmarks, "nonmutational epigenetic reprogramming" was newly added. However, epigenetics, much like genetics, is a broad scientific area that deserves further attention due to its multiple roles in cancer initiation, progression, and adaptive nature. Herein, we present a detailed examination of the epigenetic hallmarks affected in human cancer, elucidating the pathways and genes involved, and dissecting the disrupted landscapes for DNA methylation, histone modifications, and chromatin architecture that define the disease. Significance: Cancer is a disease characterized by constant evolution, spanning from its initial premalignant stages to the advanced invasive and disseminated stages. It is a pathology that is able to adapt and survive amidst hostile cellular microenvironments and diverse treatments implemented by medical professionals. The more fixed setup of the genetic structure cannot fully provide transformed cells with the tools to survive but the rapid and plastic nature of epigenetic changes is ready for the task. This review summarizes the epigenetic hallmarks that define the ecological success of cancer cells in our bodies.

Abstract:Since the discovery of the first mammalian Wnt proto-oncogene in virus-induced mouse mammary tumors almost four decades ago, Wnt signaling pathway and its involvement in cancers have been extensively investigated. Activation of this evolutionarily conserved pathway promotes cancer development <i>via</i> diverse mechanisms. Cancer is a complex disease and one outstanding conceptual framework for understanding its biology is the "Hallmarks of Cancer". In this review, we focus on the involvement of Wnt signaling in the ten hallmarks of human cancer. These widespread roles of Wnt signaling in human cancers highlight the importance and feasibility of targeting this signaling pathway for cancer treatment.

Abstract:Nearly a quarter century ago, Hanahan and Weinberg conceived six unifying principles explaining how normal cells transform into malignant tumors. Their provisional set of biological capabilities acquired during tumor development  cancer hallmarks  would evolve to fourteen tenets as knowledge of cancer genomes, molecular mechanisms, and the tumor microenvironment expanded, most recently adding four emerging enabling characteristics: phenotypic plasticity, epigenetic reprogramming, polymorphic microbiomes, and senescent cells. AKT kinases are critical signaling molecules that regulate cellular physiology upon receptor tyrosine kinases and phosphatidylinositol 3-kinase activation. The complex branching of the AKT signaling network involves several critical downstream nodes that significantly magnify its functional impact, such that nearly every organ system and cell in the body may be affected by AKT activity. Conversely, tumor intrinsic dysregulation of AKT can have numerous adverse cellular and pathological ramifications, particularly in oncogenesis, as multiple tumor suppressors and oncogenic proteins regulate AKT signaling. Herein, we review the mounting evidence implicating the AKT pathway in the aggregate of currently recognized hallmarks of cancer underlying the complexities of human malignant diseases. The challenges, recent successes, and likely areas for exciting future advances in targeting this complex pathway are also discussed.

Abstract:Cancer cells possess a nearly universal set of characteristics termed the hallmarks of cancer, including replicative immortality and resisting cell death. Dysregulated differentiation is present in virtually all cancers yet has not yet been described as a cancer hallmark. Like other hallmarks, dysregulated differentiation involves a breakdown of the cellular cooperation that typically makes multicellularity possible - in this case disrupting the division of labor among the cells of a body. At the time that the original hallmarks of cancer were described, it was not known that dysregulated differentiation was mechanistically distinct from growth inhibition, but now that this is known, it is a further reason to consider dysregulated differentiation a hallmark of cancer. Dysregulated differentiation also has clinical utility, as it forms the basis of pathological grading, predicts clinical outcomes, and is a viable target for therapies aimed at inducing differentiation. Here we argue that hallmarks of cancer should be near universal, mechanistically distinct, and have clinical utility for prognosis and/or therapy. Dysregulated differentiation meets all of these criteria.

Abstract:The complexity of cancer requires a comprehensive approach to understand its diverse manifestations and underlying mechanisms. Initially outlined by Hanahan and Weinberg in 2000 and updated in 2010, the hallmarks of cancer provide a conceptual basis for understanding inherent variability in cancer biology. Recent expansions have further elucidated additional hallmarks, including phenotypic plasticity and senescent cells. The International Agency for Research on Cancer (IARC) has identified the key characteristics of carcinogens (KCCs) to evaluate their carcinogenic potential. We analyzed chemicals of concern for environmental exposure that interact with specific receptors to induce genomic instability, epigenetic alterations, immune suppression, and receptor-mediated effects, thereby contributing to chronic inflammation. Despite their varying degrees of carcinogenicity, these chemicals have similar KCC profiles. Our analysis highlights the pivotal role of receptor binding in activating most other KCCs, underscoring their significance in cancer initiation. Although KCCs are associated with early molecular or cellular events, they do not encompass processes directly linked to full cellular malignancy. Thus, there is a need to integrate clear endpoints that anchor KCCs to the acquisition of a complete malignant phenotype into chemical testing. From the perspective of toxicology and cancer research, an all-encompassing strategy that incorporates both existing and novel KCCs and cancer hallmarks is essential to enable the targeted identification of prevalent carcinogens and facilitate zone-specific prevention strategies. To achieve this goal, collaboration between the KCC and cancer hallmarks communities becomes essential.

The temporal evolution of cancer hallmarks

Abstract:Cancer hallmarks describe key physiological characteristics that distinguish cancers from normal tissues. The temporal order in which these hallmarks appear during cancer pathogenesis is of interest from both evolutionary and clinical perspectives but has not been investigated before. Here, we order hallmarks based on the allele frequency and selective advantage of mutations in cancer hallmark genes across >10k untreated primary tumors and >8K healthy tissues. Using this novel approach, we identified a common evolutionary trajectory for 27 of 32 cancer types with genomic instability as the first and immune evasion as the last hallmark. We demonstrated widespread positive selection in cancer and strong negative selection in normal tissues for all hallmarks. Notable exceptions to the hallmark ordering in tumours were melanomas (uveal and skin) suggesting that strong environmental factors could disrupt common evolutionary paths. Clustering of hallmark trajectories across patients revealed 2 clusters defined by early or late genomic instability, with differential prognosis. Our study is the first to identify the temporal order of cancer hallmarks during tumorigenesis and demonstrate a prognostic value that could be exploited for early detection and risk stratification across multiple cancer types.

Abstract:Over the past 25 years, there has been an unparalleled increase in understanding of cancer biology. This transformation is exemplified by Hanahan and Weinberg’s decision in 2011 to expand their original Hallmarks of Cancer from six traits to ten! At the same time, mathematical modeling has emerged as a natural tool for unraveling the complex processes that contribute to the initiation and progression of cancer, for testing hypotheses about experimental and clinical observations and assisting with the development of new approaches for improving its treatment. This article starts by reviewing some of the earliest models of tumor growth and tumor responses to radiotherapy. Following Hanahan and Weinberg’s lead, attention then focuses on how closer collaboration with cancer scientists and access to experimental data are stimulating the development of new and increasingly detailed models that account, for example, for tumor–immune interactions and immunotherapy. The article concludes by discussing the ways in which mathematical modeling is being integrated with experimental and clinical studies, and outlining how this could improve disease diagnosis and the delivery of effective personalized treatments to cancer patients. As such, this article serves as an introduction to mathematical modeling of cancer and its treatments, suitable for researchers seeking to enter the field.

Abstract:Abstract The concept of the "Hallmarks of Cancer" provides a valuable framework for understanding fundamental organizing principles common to various types of cancers. However, the absence of a consensus gene set for cancer hallmarks poses data comparison and integration challenges, resulting in diverse biological interpretations across studies. Here, we first established a consensus cancer hallmark-gene set by merging data from available mapping resources. We searched the scientific literature for relevant publications from which a list of hallmark genes could be reconstructed. By consolidating data from seven publications, we identified 6,763 candidate cancer hallmark genes associated with ten cancer hallmarks. Of these, 1574 genes were part of at least two resources, forming a "core" hallmark gene set. Then, we compared the enrichment of cancer hallmarks by analyzing the prognostic genes associated with overall survival across twelve solid tumor types. Notably, the hallmark "tissue invasion and metastasis" was most prominent in the stomach (p=1.2E-11), pancreatic (p=1.5E-09), bladder (p=1.8E-08), and ovarian cancers (p=0.0002), aligning with their heightened potential to spread. "Sustained angiogenesis" predominated in lung squamous carcinomas (p=4.5E-08), while "genome instability" showed strong enrichment in lung adenocarcinomas (p=1.3E-09), liver (p=1.4E-10), pancreatic (p=1E-5) and kidney cancers (p=0.0025). Compelling evidence links lifestyle choices like alcohol consumption and smoking to genomic instability across tumors, suggesting a potential common underlying mechanism of tumorigenesis. Pancreatic cancers displayed the highest enrichment of hallmarks (8 out of 10), emphasizing the disease's complexity, while in melanomas, liver, and kidney cancer, a single hallmark (,,genomic instability”) was enriched among the prognostic markers of survival. These findings underscore the utility of the hallmark concept as an effective organizational tool, particularly when establishing a clear connection between genes and biological functions. Finally, we integrated the data into a publicly accessible and user-friendly online tool (available at www.cancerhallmarks.com) that allows users to identify the most relevant cancer-associated "candidate hallmark genes" in their datasets. In summary, we have established a consensus list of genes associated with cancer hallmarks.
