Abstract:BACKGROUND:In our previous work, we constructed short hairpin RNA (shRNA) expression plasmids that targeted human telomerase reverse transcriptase (hTERT) messenger RNA, and we demonstrated that these vectors could inhibit the growth of cancer cells by 76.5% in vivo. In order to maximize the efficiency and versatility of the vector-based siRNA approach, here we have constructed multiple shRNA expression vectors that simultaneously targeted 3 different genes in cancer cells, and then investigated their effect on human laryngeal squamous carcinoma (Hep-2) in vivo.RESULTS:We found that expression of multiple shRNAs led to a significant reduction in VEGF, hTERT and Bcl-xl mRNA and protein expression. Tumor growth curves showed that those tumors treated with the shRNAs were obviously smaller than control, nontransfected tumors. Analysis at 14 days following the final injection showed a tumor inhibition ratio of 91.2%. However, the control shRNA vectors showed none of these effects.MATERIALS AND METHODS:Short hairpin RNA expression vectors targeting the mRNA of VEGF, hTERT and Bcl-xl were constructed and subsequently transfected by direct injections into the tumors formed by Hep-2 cells implanted in nude mice. The expression of targeted genes as well as apoptosis of tumor cells were evaluated after treatment with multiple shRNA vectors or control vectors.CONCLUSIONS:Our results demonstrate that the application of vector-based RNAi technology that involves blocking multiple targets will be a promising therapeutic modality in the gene therapy of human cancers. The data strongly suggest that simultaneously blocking multiple genes in human cancers using an RNAi approach should be considered in cancer therapy.

Abstract:Objective To investigate the effects of inhibition VEGF,hTERT and Bcl-xl by RNA interference on laryngeal carcinoma in vitro and in vivo. Methods A recombinant plasmid containing 3 different short hairpin RNA (shRNA) segments termed pshRNA-VEGF-hTERT-Bcl-xl was used. Cells were treated with these plasmids. Cell viability was examined by using the MTT assay. The cytotoxic effect of pshRNA-VEGF-hTERT-Bcl-xl was evaluated in the subcutaneous xenograft tumors. Results pshRNA-VEGF-hTERT-Bcl-xl exhibited a potent antitumor effect on Hep-2 cells. MTT assay confirmed cytotoxic effect. The in vivo study showed tumor volume was significantly smaller in pshRNA-VEGF-hTERT-Bcl-xl treated group than that in control group. The inhibitory rate was 90. 2%. Conclusion Our study demonstrated that the application of vector-based RNAi technology involved in blocking multiple targets would be a promising therapeutic modality in the gene therapy of human laryngeal cancers.

Abstract:OBJECTIVE:To investigate the biological impact of pEGFP-shVEGF-shTERT-shBcl-xl expression in human laryngeal squamous carcinomas xenografted in nude mice and the related antitumor mechanism.METHODS:A recombinant plasmid vector containing 3 different short hairpin RNA (shRNA) segments including pEGFP-shVEGF-shTERT-shBcl-xl was constructed and directly injected into the grafted tumors of human laryngeal squamous carcinoma in nude mice. The mRNA and protein expressions were determined by real-time RT-PCR and Western blot respectively. Apoptosis was determined by TUNEL assay using a commercial kit. Intratumoral microvessel density (MVD) was assessed by immunhistochemistry.RESULTS:On the 14th days after the final treatment, mRNA and protein expression of VEGF, TERT, and Bcl-xl were markedly suppressed. The tumor sizes were significantly smaller than those in the other two group, with an overall tumor inhibition ratio of 91.2%. MVD counts in the pEGFP-shVEGF-shTERT-shBcl-xl treated group were significantly lower than those of the other two groups, along with increased apoptotic cells.CONCLUSIONS:The data showed that inhibition of VEGF, TERT, Bcl-xl expression by RNAi technique induces cellular apoptosis and suppresses the growth of laryngeal squamous carcinoma in vivo. VEGF, TERT and Bcl-xl may be involved in the development of laryngeal cancers. The findings suggest a synergistic tumor therapeutic effect through simultaneous inhibition of the three genes. Multi-target RNA interference may provide a powerful strategy against human laryngeal cancers.
Suppressing the growth of rectal cancer xenografts derived from patient tumors by an adenovector expressing small hairpin RNA targeting Bcl-XL.

Abstract:Background Bcl-XL, a mitochondria membrane protein, is overexpressed in colorectal cancers and promotes cell survival. We have previously shown that the adenovector expressing small hairpin (sh)RNA targeting Bcl-XL could induce significantly apoptosis in colon cancer cells. In the present study, we aimed to further detect the anti-cancer effect of adenovector expressing the shRNA targeting Bcl-XL (Ad/Bcl-XL shRNA) on rectal cancer xenografts that were derived from patient tumors. Methods We first established three rectal cancer xenografts. These xenografts were subsequently treated with Ad/Bcl-XL shRNA alone or in combination with 5-fluouracil (5-Fu). Finally, the inhibition of tumor growth, survival time and induction of apoptosis were analyzed. Results The results obtained demonstrated that Ad/Bcl-XL shRNA could effectively suppress the tumor growth of all three rectal cancer xenografts and prolong their survival time. After being combined with 5-Fu, the suppressing effect of Ad/Bcl-XL shRNA was enhanced further. In addition, the data also showed that Ad/Bcl-XL shRNA combined with 5-Fu could significantly increase the apoptotic ratio in the rectal cancer xenograft. Conclusions These data indicate that Ad/Bcl-XL shRNA with or without 5-Fu has effective anti-tumor effects on the patient tumor-derived rectal cancer xenografts, suggesting that it could be a potential strategy for rectal cancer therapy. MicroRNA-145 Inhibits Growth of Laryngeal Squamous Cell Carcinoma by Targeting the PI3K/Akt Signaling Pathway.

Abstract:Purpose: In this study, we used a nude mouse model of human laryngeal squamous cell carcinoma (LSCC) to investigate inhibition of tumor growth by microRNA-145 (miR-145) and the mechanisms underlying this inhibition. Methods: Tumors were established in nude mice by transplantation of the LSCC AMC-HN-8 cell line. Forty-eight nude mice were randomly divided into groups of eight mice each and treated with high (1.0 optical density [OD]) or low (0.5 OD) doses of miR-145, or relevant control treatments. Tumor growth was observed in each group and used to calculate the inhibition rate. Routine pathological and electron microscopic examinations were used to determine tumor apoptosis and proliferation. Changes in levels of miR-145 and PI3K and Akt protein levels were also analyzed. Results: MiR-145 inhibited LSCC growth in a dose-dependent manner, as tumor growth was significantly inhibited in mice injected intratumorally with high-dose miR-145 compared with both the untreated and low-dose miR-145 groups (p<0.05). Pathological examination showed increased tumor necrotic and apoptotic changes in treated mice, which was confirmed by electron microscopy. PI3K and Akt protein expression were significantly lower in tumors treated with high-dose miR-145 group compared with those in the untreated and low-dose miR-145 groups (p<0.05). Conclusions: MiR-145 was associated with inhibited tumor growth in a nude mouse model of LSCC. The underlying mechanism may be inhibition of the PI3K/Akt signaling pathway, which regulates tumor growth, invasion, and metastasis and also plays an important role in tumor angiogenesis and proliferation of tumor stem cells. MiR-145 may act as a tumor suppressor gene and is a promising candidate for cancer treatment.

Abstract:Objective: To develop an RNA-interference (RNAi) approach involving the hitting of multiple targets by a recombinant plasmid and evaluate its antitumor effect on laryngeal squamous carcinoma in vitro and in vivo. Materials and methods A plasmid containing 3 different short hairpin RNA (shRNA) segments termed pEGFP-shVEGF-shTERT-shBcl-xl was constructed. Plasmids containing single shRNA against each target (VEGF, TERT, BCL-xl alone) individually were also constructed as control. Cells were treated with these plasmids. The expression of targeted genes as well as apoptosis of tumor cells were evaluated after treatment with multiple shRNA vectors or control vectors. The mRNA and protein expression were determined by RT-PCR and western blotting. Cell viability was examined using the MTT assay. Apoptotic morphological alterations were observed by Hoechst staining and electron microscopy. The in vivo antitumor effect was characterized in a nude mice model of laryngeal squamous carcinoma. Results: We demonstrated that a recombinant plasmid containing multiple shRNAs could effectively and simultaneously inhibit VEGF, TERT and Bcl-xl mRNA and protein expression in the HEp-2 cells; the plasmid containing the 3 different shRNAs exhibited a potent antitumor effect on LSCC both in vitro and in vivo, and could much more effectively induced cell apoptosis than each single shRNA. We also demonstrated that the simultaneous blockage of these 3 genes have a better inhibitory effect on human HEp-2 cells than the blockage of each single shRNA. Conclusions Our study demonstrates that the application of vector-based RNAi technology that involves hitting multiple targets will be a promising therapeutic modality in the gene therapy of human laryngeal cancers; furthermore, it provides experimental evidence for the clinical application of this technology in the future. ©2008 Landes Bioscience.
Influences of Over-Expression of VEGF-C and Its Inhibition on Biological Characteristics of NCI-H446 Human Small-Cell Lung Cancer Cells

Abstract:Objective: To investigate the influences of vascular endothelial growth factor C (VEGF-C) on cell proliferation, invasion and apoptosis of NCI-H446 human small-cell lung cancer (SCLC) cells in vitro. Methods: The plasmids for VEGF-C overexpression (pHRi-VEGF-C) and small RNA interference targeting VEGF-C (pHRi-siVEGF-C) were constructed. Then the NCI-H446 cells were infected with the lentiviral packaging plasmids pHRi (an empty vector), pHRi-VEGF-C and pHRi-siVEGF-C, respectively. The expression of VEGF-C was examined by Western blotting; the cell proliferation was detected by MTT method; the invasive ability was detected by Transwell assay; and the apoptosis rate was determined by flow cytometry (FCM). Results: The expression level of VEGF-C protein in the NCI-H446 cells was significantly increased after lentiviral packaging plasmid pHRi-VEGF-C infection while it was significantly decreased after lentiviral packaging plasmid pHRi-siVEGF-C infection. The cell proliferation was significantly increased after the NCI-H446 cells were infected with pHRi-VEGF-C for five days as compared with that of the NCI-H446 cells infected with pHRi-siVEGF-C or pHRi (1.481±0.056, 0.838±0.035 and 1.146±0.07; P < 0.05). The average number of invasive cells in each visual field was significantly reduced in the pHRi-siVEGF-C-infected NCI-H446 cells as compared with that of the pHRi-VEGF-C- or pHRi-infected NCI-H446 cells (39.78±0.77, 84.87±1.27 and 60.82±1.05; P < 0.01). The number of apoptotic cells was also reduced by 90% in the pHRi-VEGF-C-infected NCI-H446 cells as compared with that of the pHRi-infected NCI-H446 cells (P < 0.01). Conclusion: The overexpression of VEGF-C can obviously promote the proliferative and invasive abilities of NCI-H446 cells, and it also can reduce the apoptosis. Meanwhile, these effects can be inhibited by small RNA interference targeting VEGF-C. The results suggest that VEGF-C may become a target of gene treatment for small-cell lung cancer. Inhibition of Human Hepatocellular Carcinoma Tumor Angiogenesis by Sirna Silencing of VEGF Via Hepatic Artery Perfusion.

Abstract:OBJECTIVEHepatocellular carcinoma (HCC) is one of the most common malignant tumors in China. Hepatic arterial chemoembolization transcatheter (TACE) is one of the main treatment methods for liver cancer. However, the long-term therapeutic effect of HCC after TACE is still unsatisfactory, postoperative recurrence and metastasis rate is still very high. Furthermore, TACE operation due to liver cancer tissue ischemia and hypoxia will lead to up-regulation of vascular endothelial growth factor (VEGF) expression. In the current study, we investigated the effects of suppressed VEGF on HCC and its molecular mechanism provided a basis for targeting angiogenesis.MATERIALS AND METHODSWe established rabbits primary HCC model by in situ embedding the VX2 subcutaneous transplantation tumor. Conventional Seldinger femoral artery and hepatic artery catheterization method were used to select the catheter over the tumor-bearing hepatic artery. The different groups were divided into TACE operation, and the experimental group was performed with the VEGF-siRNA molecular preparation in the catheter. 64-slice spiral CT were used to perfusion imaging of liver cancer model before and after TACE operation. We further assessed the efficiency of VEGF silencing and its influence on VX2 cells. The expression of VEGF mRNA and protein were detected by RT-PCR and Western blotting, respectively. Intratumoral microvessel density (MVD), VEGF and CD34 were evaluated by immunohistochemistry. We detected the cell apoptotic by immunofluorescence and flow cytometry.RESULTSOur findings indicated that VEGF-siRNA-2# could effectively suppress the expression of VEGF expression, inhibited the proliferation capability and promoted apoptosis of VX2 cells in vitro. Silencing of VEGF expression also suppress HCC tumor growth and reduce HCC angiogenesis in rabbits primary HCC model in vivo. Furthermore, We found that phosphoinositide 3-kinase (PI3K) and protein kinase B (AKT) activation were considerably reduced while inhibition VEGF expression in VX2 cells.CONCLUSIONSOur data demonstrated that VEGF silencing could suppress cells proliferation, promote cells apoptosis and reduce HCC angiogenesis through inactivation of VEGF/PI3K/AKT signaling pathway.

Abstract:OBJECTIVE:To investigate the expressions of p53 and PCNA by inhibition of hTERT with siRNA in xenografted laryngeal squamous carcinoma in nude mice.METHOD:pshRNA involved fluorescein gene was synthesized according to the sequence of hTERT cDNA. The xenografted tumor in nude mice was established by using human laryngeal squamous carcinoma cell line (Hep-2). pshRNA packaged by METAFECTENE was transfected into the transplantation tumors. The volumes of the subcutaneous tumor were determined. Fluorescence expression in the tumors was detected by confocal microscopy. The expressions of p53 and PCNA were detected using immunohistochemistry.RESULT:The subcutaneous tumors appeared in all the nude mice after transplantation five days later. The diameter of subcutaneous tumors become 5-7 mm after transplantation. Many green fluorescent cells were observed by confocal microscopy after transfection of pshRNA and blank plasmid. The growth of laryngeal squamous carcinoma in nude mice was significantly decreased as compared with the control group. The proliferation activity was inhibited by 76.50%. The expressions of p53 was upregulated and the expression of PCNA was downregulated.CONCLUSION:The transfection of pshRNA could significantly induce inhibitory effects on growth of laryngeal squamous carcinoma in nude mice and the expression of PCNA could be effectively downregulated. The inhibition of telomerase reverse transcriptase (hTERT) mRNA could upregulate the expression of p53 and promote the apoptosis of carcinoma cells as well as inhibit the proliferation of laryngeal squamous carcinoma cells.

Abstract:Background Treatment that targets to cancer stem cells plays an important role in controlling and eliminating of tumor initiation, development, progression, and chemotherapy/radiotherapy resistance.In our previous study, we cultured and harvested human laryngeal cancer stem cells (CSCs), applied microRNA biochips to screen differentially expressed miRNAs that were related to radiation tolerance in irradiated human laryngeal CSCs.According to the predicted genes and pathways of differential miRNAs target, hsa-miR-138-2-3p that was down-regulated expression under radiation, was thought to play a key role in enhancing the radio-sensitivity in human laryngeal squamous cancer stem cells.Method To investigate the radiational enhancement of hsa-miR-138-2-3p, we transfected hsa-miR-138-2-3p mimics that were synthesized based on the sequences of hsa-miR-138-2-3p in vitro into human laryngeal CSCs (Hep-2, M2e, and TU212 cell lines) to make hsa-miR-138-2-3p overexpressed, and the tumorous specialities of CSCs, like cell proliferation, invasion, apoptosis, cell cycle arrest, and DNA damage were evaluated by CCK-8 assay, clone formation assay, invasion assay, flow cytometry, and comet assay.Furthermore, we explored the signal transduction pathways that regulated the cancer stem cell initiation, development, invasion, apoptosis and cell cycle arrest, which were controlled by hsa-miR-138-2-3p.Result Overexpressed hsa-miR-138-2-3p played an key role in many anti-cancer biological process in human laryngeal CSCs: (1) decreased laryngeal CSCs proliferation and invasion in response to radiotherapy; (2) increased the proportion of early and late apoptosis in laryngeal CSCs after radiation, raised G1 phase arrest in laryngeal CSCs after radiation, and decreased the proportion of S stage cells of cell cycle that were related to radio-resistance in laryngeal CSCs; (3) downregulated the expression of β-catenin in Wnt signal pathway that was related to the

Abstract:BACKGROUND & OBJECTIVE:RNA interference (RNAi) is triggered by the presence of double-stranded RNA (dsRNA) in the cell and results in rapid destruction and post-transcriptional gene silencing of target mRNA. The roles of RNAi in gene function and antiviral therapy have been reported, but its effect on human telomerase reverse transcriptase (hTERT) gene, which is highly expressed in laryngeal squamous cell carcinoma and other head and neck neoplasms, has seldom been reported. This study was to explore the inhibitory effect of silencing hTERT gene by short hairpin RNA (shRNA) on growth of laryngeal squamous cell carcinoma xenograft in nude mice with RNAi technique.METHODS:Plasmid pshRNA containing fluorescein gene and hTERT cDNA sequences was synthesized. Human laryngeal squamous cell carcinoma Hep-2 cells were transplanted into nude mice to establish xenograft tumors. pshRNA was transfected into the tumors. The tumor volume was observed. Fluorescence expression in the tumors was observed by laser confocal microscopy. Tumor morphology was observed with HE staining. Cell apoptosis was detected by terminal deoxynucleotidyl transferase-mediated dUTP nick end-labeling (TUNEL) assay. The expression of hTERT protein in the tumors was detected by SP immunohistochemistry. The structural change of heart, liver, kidney and spleen was observed, and peripheral blood hematological and biochemical parameters were determined.RESULTS:The tumor volume was significantly smaller in pshRNA group than in normal saline (NS) group and blank plasmid group (P<0.01). The inhibition rate was 76.50% in pshRNA group. After transfection of pshRNA or blank plasmid, many green fluorescent cells were observed under confocal microscope. The necrosis and apoptosis of tumor cells were found under light microscope in shRNA group. The apoptotic index of tumor cells was significantly higher in pshRNA group than in NS group and blank plasmid group [(26.47+/-4.25)% vs. (2.73+/-1.35)% and (3.40+/-1.41)%, P<0.05]. pshRNA directly down-regulated hTERT protein expression in the tumors, but did no damage to the heart, liver, kidney, spleen, and blood system of nude mice.CONCLUSION:shRNA plasmid containing specific sequences of hTERT gene could significantly inhibit the growth of laryngeal carcinoma in nude mice, with no side effect on the heart, liver, kidney, spleen, and blood system.
Hvegf Sirna Inhibits Initial Growth of A549 Tumor Cells in Nude Mice and Angiogenesis on CAM Assay

Abstract:OBJECTIVE:To investigate the effect of small interfering RNA (siRNA) targeting human vascular endothelial growth factor (hVEGF) on A549 cell growth in nude mice and angiogenesis on chorioallantoic membrane (CAM) assay.METHODS:Three pairs of hVEGF siRNA-plasmid and non-silencing-plasmid were constructed, and transfected into A549 cells through lipofectamine 2000, respectively. The most effective pair of hVEGF siRNA-plasmid was selected by ELISA and real-time RT-PCR. A549 cells transfected with selected hVEGF siRNA- plasmid, A549 cells transfected with non-silencing-plasmid and A549 cells without transfection were inoculated into nude mice, respectively. Chick embryos were randomly divided into four groups and CAM was treated by different solutions for 48 h: culture media DMEM as negative control group,un-transfected A549 cell culture supernatants as positive control group, hVEGF siRNA A549 cell culture supernatants as hVEGF siRNA group and nonsilencing siRNA A549 cell culture supernatants as non-silencing siRNA group. The CAMs were harvested on d12 for microscopic assays.RESULT:Compared with control group, hVEGF siRNA-plasmid induced 48% reduction in hVEGF secretion by A549 cells accompanied by 70% reduction in hVEGF mRNA. Compared with non-silencing siRNA group, the mean tumor volume of murine xenograft was reduced by 58% in hVEGF siRNA group; time for xenografts growing to 50 mm(3)was delayed by 5.4 d. hVEGF contents in xenograft were reduced by 54%; but mean doubling time of tumors and the growth rate of tumors were not significantly reduced. In CAM assays, hVEGF content was zero in negative group, and in hVEGF siRNA group that was 40%-44% of non-silencing siRNA group or positive group; vessels branch points of CAM in hVEGF siRNA group or non-silencing siRNA group or positive group were increased by 45%-55% compared with negative group; total vessel length of CAM in hVEGF siRNA group was increased by 53% compared with negative group, while in non-silencing siRNA group or positive group that was increased by 97% or 99%. Compared with negative control group, the proliferation of microvessels was increased when cell culture supernatant with hVEGF was added in hVEGF siRNA group, significant proliferated vessels were observed in non-silencing siRNA group or positive group.CONCLUSION:A plasmid-mediated hVEGF siRNA has been constructed and verified, which can effectively downregulate the expression of hVEGF in human A549 cells, resulting in the inhibition of angiogenesis. hVEGF siRNA can delay initial growth of A549 tumor xenograft but not reduce the growth rate.

Abstract:OBJECTIVE To investigate the effect of RNA interference silencing the human telomerase reverse transcriptase(hTERT), vascular endothelial growth factor(VEGF)and Bcl-xl mRNA simultaneously on the Hep-2 cells in larynx carcinoma cell line, and to compare the effect of silencing the hTERT gene alone. METHODS A recombinant plasmid containing 3 different short hairpin RNA (shTERT, shVEGF, shBcl-xl)segments termed pshRNA2 was constructed. Plasmid containing single shRNA(shTERT)termed pshRNA1 was also constructed. Cells were treated with these plasmids. Cell proliferation was examined using the MTT assay. The apoptosis of Hep-2 cells was detected by flow cytometer(FCM). The protein expression was observed by western blotting. RESULTS After treatment with pshRNA1 and pshRNA2, the proliferation of Hep-2 cells was obviously inhibited . pshRNA2 exhibited a potent antitumor effect on Hep-2 cells, and could much more effectively induce cell apoptosis than pshRNA1. Our results demonstrated that a recombinant plasmid containing multiple shRNAs could effectively and simultaneously inhibit VEGF, hTERT, Bcl-xl mRNA and protein expression in the Hep-2 cells. CONCLUSION The plasmid containing the 3 different shRNAs is much more effective than a single shRNA, supplying a promising therapeutic modality in the gene therapy of human laryngeal carcinomas.

Abstract:The aim of this study was to assess the inhibitory efficacy of short hairpin RNA (ShRNA) targeting vascular endothelial growth factor C (VEGF-C) in an orthotopic pancreatic cancer mouse model. BxPC-3 human pancreatic cancer cells expressing green fluorescent protein (GFP) were orthotopically implanted onto the pancreas of nude mice. All mice were randomly divided into four groups when the average tumor size had reached 100 mm(3) and were treated with either vehicle or gemcitabine at 150 mg/kg; or intravenous VEGF-C ShRNA at 150 mg/kg; or intratumoral VEGF-C ShRNA at 150 mu g/kg. In vivo fluorescence imaging was performed to monitor tumor growth and metastasis during the study. Real-time quantitative polymerase chain reaction (RT-qPCR) and an enzyme-linked immunosorbent assay (ELISA) were performed to determine the mRNA and protein level of VEGF-C in tumor tissues. Lymphatic vessel marker D2-40, blood vessel marker CD31 and proliferation marker Ki67 expression of the tumor tissues were analyzed by immunohistochemistry staining. Intravenous and intratumoral VEGF-C ShRNA treatment significantly inhibited tumor growth, downregulated the expression of VEGF-C mRNA, reduced tumor microlymphatic vessel density (MLVD), and inhibited cancer cell proliferation. Gemcitabine, as the standard treatment for pancreatic cancer, demonstrated a stronger inhibitory effect on tumor growth, with less inhibition of MLVD and more inhibition of microvessel density (MVD) and proliferation than VEGF-C ShRNA. These results indicate that different mechanisms are associated with the efficacy of gemcitabine and VEGF-C ShRNA.

Abstract:Vascular endothelial growth factor (VEGF) plays an important role in the growth and metastasis of non-small-cell lung cancer (NSCLC). The aim of this study was to develop an RNA-interference approach that targets VEGF, using a recombinant plasmid, and to explore its antitumor efficacy in NSCLC in vivo. shRNA-targeting VEGF was cloned into pGenesil-2 plasmid vector and then transfected into A549 human lung cancer cells, using cationic liposome. Reverse-transcriptase polymerase chain reaction (RT-PCR) and enzyme-linked immunosorbent assay (ELISA) analysis were used to evaluate the silencing effects of VEGF-shRNA on A549 cells in vitro. Further, the growth-inhibition capacity of VEGF-shRNA on A549 lung carcinoma xenografts was tested in nude mice. Proliferation, apoptosis, and angiogenesis in tumor tissues were measured by PCNA, TUNEL, and CD31 immunohistochemistry, respectively. shRNA-targeting VEGF significantly silenced VEGF expression in A549 lung cancer cells, as confirmed by RT-PCR and ELISA assay (P < 0.01). In vivo, the VEGF-shRNA delayed tumor growth and reduced tumor weight by similar to 61.96%, compared with control groups (P < 0.05), accompanied with angiogenesis inhibition (P < 0.01) and apoptosis induction (P < 0.01). Our data showed that the knockdown of VEGF by shRNA might be a potential therapeutic approach against human NSCLC.
Human Antisense-Vascular Endothelial Growth Factor Gene Therapy for Laryngeal Tumor by Cationic Liposome-Mediated Transfection

Abstract:OBJECTIVE:To observe the effect of cationic liposome mediated antisense-vascular endothelial growth factor (VEGF) gene transfection on the growth of laryngeal cancer Hep-2 cells in the nude mice.METHODS:The VEGF-cDNA gene was cloned by reverse transcriptase polymerase chain reaction (RT-PCR) from human laryngeal cancer, and its eukaryotic expression vector pcDNA3-VEGF (-) with antisense-VEGF gene was constructed and identified by PCR and double-enzyme digestion. The pcDNA3-VEGF (-) was transfected into laryngeal cancer Hep-2 cell line by using cationic liposome (LP 2000). Then, the transfected Hep-2 cells were injected into nude mice and the size of tumor from different groups was observed while establishing laryngeal cancer xenografts in nude mice, and then treating the tumor-bearing mice with liposome-plasmid complex, observing the size of tumor from different groups. The expression of VEGF mRNA in different groups was observed by RT-PCR. The transfected cell ultranstructure was observed by transmission electron microscopy.RESULTS:The human VEGF-cDNA was successfully cloned and its eukaryotic expression vector with antisense-VEGF pcDNA3-VEGF (-) was constructed. The antisense-VEGF gene was transfected into Hep-2 cell line by using cationic liposome (LP2000). The size of tumor transfected with pcDNA3-VEGF (-) was significantly smaller than that of control groups. While the size of tumor treated with liposome-pcDNA3-VEGF (-) complex was significantly smaller than that of control groups. Many apoptic tumor cells were observed by transmission electron microscopy and the structure of microvessel was also changed. The expression of VEGF mRNA was evidently weaker than that of the control groups.CONCLUSION:The growth of Hep-2 cells could be inhibited significantly by antisense-VEGF gene transfection.
Targeted Hsp70 Expression Combined with Cik-Activated Immune Reconstruction Synergistically Exerts Antitumor Efficacy in Patient-Derived Hepatocellular Carcinoma Xenograft Mouse Models

Abstract:The patient-derived tumor xenograft (PDTX) models can reproduce a similar natural genetic background and similar biological behaviors to tumor cells in patients, which is conducive to the assessment of personalized cancer treatment. In this study, to verify the targeting and effectiveness of the therapeutic strategy using a Survivin promoter-regulated oncolytic adenovirus expressing Hsp70, the PDTX models of hepatocellular carcinoma (HCC) were established in nude mice and the cytokine-induced killer (CIK) cells were intravenously infused into mice to partially reconstruct the mouse immune function. The results demonstrated that, either the immune anti-tumor effect caused by CIK cell infusion or the oncolytic effect generated by oncolytic adenovirus replication was very limited. However, the synergistic tumor inhibitory effect was significantly enhanced after treatments with oncolytic adenovirus expressing Hsp70 combined with CIK cells. Oncolytic adenovirus mediated the specific expression of Hsp70 in cancer tissues allowed the CIK chemotaxis, and induce the infiltration of CD3+ T cells in tumor stroma, thereby exhibiting anti-tumor activity. The anti-tumor effect was more effective for the highly malignant tumor xenografts with highly Survivin expression. This strategy can synergistically activate multiple antitumor mechanisms and exert effective anti-tumor activities that have a significant inhibitory effect against the growth of HCC xenografts.
Effect of Silencing Key Proteins in Telomerase Mechanism and Alternative Lengthening of Telomeres Mechanism in Laryngeal Cancer Cells

Abstract:Purpose: To explore the influences of telomerase and alternative lengthening of telomeres mechanism on telomere length and laryngeal squamous cell carcinoma in vitro and in vivo.Materials and methods: Short hairpin RNA expression vectors targeting the messenger TERT, TRF2, RAD51 and NB S1 were constructed. The mRNA and protein expression of targeted genes in human laryngeal squamous carcinoma cell line HEp-2 was evaluated by reverse transcription polymerase chain reaction and Western blotting separately. The length of telomere was analyzed by fluorescent in-situ hybridization. Cell viability was examined by cell counting Kit-8. Effects on tumor growth were also investigated in vivo.Results: The transfection of multiple short hairpin RNAs expression plasmid significantly inhibited the mRNA and protein expression of related genes. Silence of alternative lengthening of telomeres mechanism and telomerase mechanism related genes resulted in the shortening of telomere length in HEp-2 cell. However, silence of alternative lengthening of telomeres mechanism related genes could shorten the telomere length but had no significant difference. Both simultaneously and separately blocking telomerase mechanism and alternative lengthening of telomeres mechanism resulted in reduction of tumor cell viability. Silence of alternative lengthening of telomeres mechanism and telomerase mechanism related genes inhibited the tumor growth in vivo.Conclusions: The inhibition of telomere related gene may be a promising strategy for the treatment of laryngeal squamous cell carcinoma. Abstract:Although surgery-based comprehensive therapy is becoming the main approach to treat laryngeal cancer, recurrence, metastasis, radiotherapy resistance and chemotherapy tolerance are still the main causes of death in patients. Targeted inhibition of laryngeal cancer stem cells has been considered as the consensus to cure laryngeal cancer. Our previous study has confirmed proto-oncogene Bmi-1 as a key regulator for self-renewal of laryngeal cancer stem cells. Targeted knockdown of Bmi-1 gene effectively inhibited the self-renewal and differentiation of laryngeal cancer stem cells, leading to the promoted sensitivity to chemotherapy including paclitaxel. However, due to off-target effects and quick degradation of the naked Bmi-1 -RNAi small RNA oligo by nuclease in body fluids, it is urgently needed to develop a tumor-targeted delivery system with a protective shell. In this study, we designed and synthesized cRGD peptide-modified chitosan-polyethylene glycol slow-release nanoparticles (mPEG-CS-cRGD/ Bmi-1RNAi -PTX) containing Bmi-1RNAi siRNA oligo and paclitaxel, which showed spherical in shape, 200 nm diameter in size, low cytotoxicity, strong DNA wrapping, resistance to nuclease degradation and high transfection efficiency to cells. Functional analysis indicated significant suppression of cell proliferation and migration and induction of apoptosis by the nanocomplex in laryngeal cancer cells in vitro . By application to the mouse model with laryngeal cancer, the nanocomplex inhibited tumor growth significantly in vivo . In addition, cRGD peptide, paclitaxel and Bmi-1 siRNA in the nanoparticles showed synergistic effects to suppress laryngeal cancer stem cells. In conclusion, this study not only developed a laryngeal tumor-targeted chemotherapeutic system, but also demonstrated a Bmi-1 RNAi-based chemotherapeutic strategy to inhibit cancer stem cells, having strong potential to treat laryngeal cancer patients suffering therapy resistance and/or tumor recurrence.

Abstract:Objective: Telomerase is an attractive molecular target because it is active in most malignant cells but undetectable in most normal somatic cells. Small, interfering ribonucleic acid segments have been shown to be effective tools for inhibiting the expression of a given gene within human cells. In the present study, we examined the effects of short hairpin ribonucleic acid expression vectors on the growth of laryngeal squamous cell carcinoma in nude mice, and we assessed potential side effects in these animals. Methods: Short hairpin ribonucleic acid expression vectors targeting the messenger ribonucleic acid of the telomerase catalytic unit were constructed and transfected into Hep-2 human laryngeal squamous cells carcinoma in nude mice. Apoptosis and telomerase catalytic unit expression within tumour cells were evaluated after treating with short hairpin ribonucleic acid. Peripheral blood was collected for haematological and biochemical analysis. Results: The findings demonstrated that short hairpin ribonucleic acid plasmids could inhibit tumour cell growth by 76.5 per cent, and that many tumour cells underwent necrotic or apoptotic cell death. There were no significant side effects of short hairpin ribonucleic acid on the heart, liver, kidney, spleen or blood system in this experimental model. Conclusion: These results indicated that the short hairpin ribonucleic acid expression vector targeted at the telomerase catalytic unit of messenger ribonucleic acid significantly inhibited the growth of laryngeal carcinoma in nude mice, with no significant side effects on the experimental animals.
