Continuous Monoclonal Antibody Production by a Composite Gel Perfusion Bioreactor in a Protein-Free Medium

Abstract:An alginate/agarose composite gel system for the immobilisation of hybridoma cells, was developed, integrating the simplicity of gelation but instability of calcium alginate and the stability, rigidity and porous structure of an agarose gel. The structural heterogeneity of the gel matrix was able to provide an ideal location for cell growth. During the operation of a packed-bed perfusion bioreactor using this system, the alginate component gradually leached out of the gel and increased the internal space, which provided more space for the cells to grow and improved the gel porosity. As a result, a high cell density with high viability was achieved in the form of many small cell aggregates.
