Abstract:Diethyl 2-(((4-methyl-2-oxo-2H-chromen-7-yl)oxy)methylene)malonate (2) was synthesized from coumarin 1 and diethyl ethoxymethylene malonate in ethanol, followed by cyclization in diphenyl ether to give chromene-9-carboxylate (3). Sugar hydrazones 5a-c were formed by reacting hydrazide 4 with D-galactose, D-mannose, and D-xylose, then acetylated to per-O-acetyl derivatives 6a-c. Heating 5a-c with acetic anhydride at 100 °C gave oxadiazolines 7a-c. Compound 8, obtained by refluxing 4 with carbon disulfide, was alkylated to 9 or reacted to give 10. Further reactions yielded acetoxy derivative 13 and hydroxy derivative 14. Compounds 17a-e and 18a-e were synthesized using thiomorpholinophenyl ureido/thioureido-s-triazine. These compounds were characterized and evaluated for antibacterial activity against Gram (+ve) bacteria (B. subtilis, S. aureus) and Gram (-ve) bacteria (E. coli, P. aeruginosa) in addition to yeast-like fungi (C. albicans). Compounds 11, 13, 15, 16, 17c-e, and 18a-e showed the highest antibacterial activity. Molecular docking was performed to study their binding with transpeptidases.

Abstract:A series of 2-aryl-5-{2-[3-(trifluoromethyl)phenyl]ethenyl}-1,3,4-oxadiazoles were synthesized by cyclization of 3-[3-(trifluoromethyl)phenyl]acrylic acid with substituted benzohydrazides in the presence of phosphoryl chloride. The synthesized compounds were characterized by FT-IR, 1 H NMR, and mass spectroscopic techniques and screened for their in vitro antimicrobial activity against S. aureus , S. pyogenes , E. coli , P. aeruginosa , C. albicans , and A. niger . Most of the synthesized 1,3,4-oxadiazole derivatives exhibited moderate to good activity against the tested microbial strains.

Abstract:A series of 5-aryl-3-(4-oxo-2-phenylquinazolin-3(4H)-yl)-2-(pyridin-4-yl)thiazolidin-4-ones were synthesized and evaluated for their antibacterial and antifungal activities. Various spectral techniques e.g., IR, 1H NMR, 13C NMR and Mass spectrometry were used to determine structure of novel synthesized compounds. The title compounds showed good to excellent inhibition potency for respective Gram-positive bacterial strains and Gram-negative bacterial strains. These compounds exhibited a broad spectrum of inhibitory activity. Molecular docking studies against microbial DNA gyrase sub unit B could provide valuable insights into the binding affinity of these molecules and their plausible mechanism of antimicrobial action. Compounds 5a, 5d, 5f and 5h, 5i, 5j, 5o exhibited excellent activity against bacterial and fungal strains respectively.Graphical

Abstract:A convenient, straightforward, and effective one-step reaction for the synthesis of a three-component compound of biologically relevant novel 2,4-diamino-5-(8-hydroxyquinolin-7-yl)-5H-chromeno[2,3-b] pyridine-3-carbonitrile derivatives was designed and synthesized. The synthesis was developed by the reaction between salicylaldehyde 1, 8-hydroxyquinoline 2, 2-aminopropene-1,1,3-tricarbonitrile 3, and the catalytic amount of triethylamine in ethanol at 78 °C. This methodology has many beneficial features, including the use of inexpensive and non-hazardous starting materials, single-flask reactions, optimized reaction conditions, the termination of intermediate isolation, easy workup, reducing organic waste products, being chromatography-free, and decreasing the reaction time along with quantitative yields with high functional group tolerance. A proposed mechanism with supporting experimental data is presented, including 1H NMR, 13C NMR, 2D NMR (HMBC, COSY, HSQC), mass, and IR spectroscopy, which are used to characterize the complete derivatives. All synthesized compounds were evaluated in vitro for their antibacterial activities against Bacillus subtilis, Staphylococcus aureus, Escherichia coli and Pseudomonas aeruginosa bacterial strains via the agar-well diffusion method compared with the reference drug gentamicin. The data indicated that compounds 4A, 4F, 4G, 4 J, and 4K consistently demonstrated strong antimicrobial activity against Gram-positive and Gram-negative bacteria. Furthermore, a molecular docking investigation was carried out to gain insight into the binding mode of the most promising compounds via the crystal structure of the S. aureus DNA gyrase complex with ciprofloxacin (PDB ID: 2XCT). Density functional theory (DFT) calculations were performed to determine the various molecular properties of the synthesized novel 2,4-diamino-5-(8-hydroxyquinolin-7-yl)-5H-chromeno [2,3-b] pyridine-3-carbonitrile derivatives (4A-4 M). On the basis of the reactive sites explored by the molecular electrostatic potential maps, the antibacterial activities of the compounds were screened.

Abstract:In search for new compounds capable of overcoming antimicrobial resistance, we synthesized a series of previously unknown 1-[(2-aryl-5-(pyridine-4-yl)-1,3,4-oxadiazol-3(2 H )-yl]-3-(pyridine-2-yl)prop-2-en-1-ones and assessed their composition and structures using elemental analysis, FTIR, 1 H NMR, and 13 C NMR spectroscopy, and mass spectrometry. The synthesized compounds were screened for antibacterial activity to show that 1-[2-(3-fluorophenyl)-5-(pyridin-4-yl)-1,3,4-oxadiazol-3(2 H )-yl])-3-(pyridin-2-yl)prop-2-en-1-one and 1-[2-(4-fluorophenyl)-5-(pyridin-4-yl)-1,3,4-oxadiazol-3(2 H )-yl]-3-(pyridin-2-yl)prop-2-en-1-one were effective against C. albicans and A. clavatus fungal strains, while 1-[(2-phenyl-5-(pyridine-4-yl)-1,3,4-oxadiazol-3(2 H )-yl)-3-(pyridine-2-yl)prop-2-en-1-one and 1-[2-(4-fluorophenyl)-5-(pyridin-4-yl)-1,3,4-oxadiazol-3(2 H )-yl]-3-(pyridin-2-yl)prop-2-en-1-one demonstrated activity against E. coli and P. aeruginosa bacterial strains.

Abstract:1,3-Thiazolidine-2,4-dione 2 has been synthesized by the cyclisation reaction of thiourea and chloroacetic acid in the presence of ethanol. The reaction of compound 2 with substituted aromatic aldehyde afforded the corresponding derivatives of substituted 5-benzylidene-1,3-thiazolidinone-2,4-dione 3a–d, which upon reflux with ω-bromoalkoxyphthalimide gave 2-{[-5-(substituted benzylidine)-2,4-dioxo-1,3-thiazolidine-3-yl]alkoxy} -1H-isoindole-1,3(2H)-dione 4a–i. Further, compounds 4a–i were treated with phenyl hydrazine and 2,4 dinitro phenyl hydrazine in the DMF to yield the title compound 2-[5-oxo-2,3-substituted diphenyl-2H-pyrazolo[3,4-d][1,3]thiazol-6(5H)-yl)alkoxy]-1H-isoindole-1,3(2H)-dione 5a–r. Structures of newly synthesized compounds were established based on elemental analysis, IR, 1H NMR and mass spectral data. Synthesized compounds have been assayed for their antibacterial activities against B. subtilis, K. pneumoniae, P. aeruginosa and S. aureus and antifungal activities against A. fumigatus and C. albicans.

Abstract:Introduction: A series of 15 thiazolyl hydrazone derivatives of chromone-3- carbaldehyde have been designed and synthesized by the cyclization of thiosemicarbazone derivatives of chromone-3-carbaldehydes with 4’-substituted-2-bromo acetophenones. Method: All these derivatives were evaluated for antioxidant activity by their direct scavenging activity objects to reactive oxygen species such as DPPH, and nitric oxide, as well as in vitro antiinflammatory activity by a protein denaturation method. Most of these synthesized compounds have shown significant antioxidant activity, among which the compounds 5b, 5c, 5e, 5g, and 5j showed very good antioxidant activities in comparison with the standard ascorbic acid. The in vitro anti-inflammatory activity revealed that the compounds 5b, 5g, and 5h possessed significant activity compared to standard diclofenac sodium. Result: Additionally, molecular docking studies of these molecules using ovalbumin as the protein showed remarkable interactions with its active site residues, and the results indicated that the binding mode of these compounds closely resembled that of the reference compound, diclofenac sodium. Conclusion: Thus, these compounds represent an attractive template for the evaluation of new antiinflammatory and antioxidant agents and might be useful for exploring new therapeutic tools.

Abstract:We have developed an efficient divergent strategy to access various functionalized 5‐substituted 1,2,4‐oxadiazole‐3‐yl benzoxazole derivatives in good to excellent yields by one pot reaction involving nucleophilic addition reaction of amidoxime with aryl or alkyl acid chloride, followed by cyclodehydration in presence of base. A wide range of functional groups were tolerated during this process. Discovery of novel antimicrobial drugs is one of the hottest topics in the field of medicinal chemistry. An efficient and catalyst free synthesis of novel fluorinated 5‐substituted 1,2,4‐oxadiazole‐3‐yl benzoxazole derivatives is described. A series of novel compounds have been synthesized and characterized by different spectroscopic techniques such as FTIR, 1H NMR, 13C NMR, and LCMS. When tested for antibacterial activity, the synthesized benzoxazole derivatives exhibited outstanding to good activities in comparison to the conventional medications. Moreover, the theoretical forecasts grounded in molecular docking against microbial DNA gyrase may offer a glimpse into the conceivable mode of operation and establish a connection between the detected antimicrobial efficacy and the binding affinity, illuminating particular thermodynamic (bonded and nonbonded) interactions that regulate the activity. The produced compounds also exhibited promising qualities to develop into viable oral medication candidates when their absorption, distribution, metabolism, and excretion were examined. In summary, the research proposes that these novel compounds have emerged as interesting contender for the development of next generation antimicrobial drugs. Our protocol demonstrates broad substrate scope, dramatic decrease of synthesis time, and energy consumption with excellent yields.

Synthesis and antioxidant activity of 1,3,4-oxadiazole tagged thieno[2,3-d]pyrimidine derivatives

Abstract:This study represents the synthesis of a new series of N-substituted phenyl-5-methyl-6-(5-(4-substituted phenyl)-1,3,4-oxadiazol-2-yl)thieno[2,3-d]pyrimidin-4-amine derivatives (4a-l) and substituted phenylamino-5-methylthieno[2,3-d]pyrimidine-6-carboxylic acid derivatives (3a-d). The newly synthesized compounds were characterized by (1)H NMR, (13)C NMR, LC-MS and IR analyses. All these novel compounds were screened for their in vitro antioxidant activity by employing DPPH, hydrogen peroxide, and nitric oxide radical scavenging assays. Compounds 4k, 4j, 4d, and 4e showed significant radical scavenging due to the presence of electron donating substituent on both sides of the thienopyrimidine ring enhances the activity and electron withdrawing groups like nitro decrease.

Polycyclic Aromatic Compounds

Abstract:In the present work, a series of novel oxazolidinone derivatives containing thieno-pyridine ring system ( 11a–n ) were synthesized in six steps. Synthesis of amino oxazolidinone scaffold ( 10 ) involved nucleophilic substitution of thienopyridine ( 4 ) with P-chloro-nitrobenzene ( 3 ) in dimethyl formamide at 65 °C give nitro compound ( 5 ) which was further reduced in catalytic hydrogenation condition using Raney-Nickel in isopropyl alcohol afforded amine ( 6 ). Reaction of compound ( 6 ) with 2-[(2S)-oxiran-2-ylmethyl]-1H-isoindole-1,3(2H)-dione ( 7 ) at mild reflux condition in isopropyl alcohol gives compound ( 8 ). Hydroxy amine compound ( 8 ) further undergo carbonyl insertion reaction with 1, 1'-carbonylbis(1H-imidazole) afforded oxazolidinone compound ( 9 ). The de-protection of phthalamide group of compound ( 9 ) carried by treating with aqueous solution of hydrazine hydrate in methanol at room temperature give compound ( 10 ). Finally, compound ( 10 ) reacts with acetyl chloride, carboxylic acid, sulfonyl chloride and chloro format by customary method provided amides, sulfonamide and carbamate derivative of (5S)-5-(aminomethyl)-3-[4-(6,7-dihydrothieno[3,2-c]pyridin-5(4H)-yl)phenyl]-1,3-oxazolidin-2-one. The developed synthetic approach was operationally simple and high yielding. The structures of the synthesized compounds were elucidated by IR, MS, 1 H and 13 C-NMR. Synthesized compounds ( 11a–n ) were tested for antibacterial activity against a panel of Gram-positive bacteria comprising Staphylococcus aureus (ATCC5638) , Streptococcus pyogenes (ATCC12344) , Bacillus subtilis (ATCC6051) , Bacillus pumilus (ATCC27142) , and Enterococcus faecalis (NCIM5253) . The investigation of antimicrobial screening data revealed that, most of the compounds tested have demonstrated sensible to good bacterial activity. In summary, preliminary results of activity indicate that, acetyl derivative ( 11a ), methane sulfonamide derivative ( 11c ) and p -toluene sulfonamide derivative ( 11e ) found to be good activity and di-(methane sulfonamide) derivative ( 11d) showed comparable activity to reference drug substances.

Abstract:A new series of 1,3,5-trisubstituted 2-pyrazoline derivatives (3a-l) are synthesized in good to excellent yields from the corresponding chalcones (1a-h) and acid hydrazides (2a-e) in polyethylene glycol-400 (PEG-400) as a green reaction medium. The newly synthesized 2-pyrazoline derivatives are screened for their antibacterial and antifungal activity. The synthesized trisubstituted pyrazolines displayed moderate to good antibacterial and antifungal properties as compared with the standard reference penicillin and fluconazole drugs. Additionally, the antioxidant potential of the 1,3,5-trisubstituted 2-pyrazolines is evaluated by OH and DPPH assay. The 1,3,5-trisubstituted 2-pyrazolines showed good radical scavenger activity and were found as good antioxidant agents.

Abstract:A series of thirty-three novel triazolyl pyranochromen-2(1H)-one derivatives have been synthesized via Cu (I) catalysed Huisgen 1,3-dipolar cycloaddition reaction. All of the synthesized compounds have been fully characterized from their spectral data and evaluated for antibacterial activity against both gram-positive and gram-negative bacteria. The information obtained from structure–activity relationship can be used to design and develop the next generation of compounds with higher antibacterial efficacy.

Abstract:Background: Antimicrobial Resistance (AMR) and Tuberculosis (TB) are global concern. According to the WHO fact sheet on tuberculosis, in 2017, 10 million people fell ill with TB, and 1.6 million including 230,000 children died from the disease. There is a critical need of design and development of novel chemotherapeutic agents to combat the emergence and increasing prevalence of resistant pathogens. In the present study, a new series of 1,3,4-oxadiazoles incorporating benzimidazole and pyridine scaffolds in a single molecular framework has been reported. Methods: The structures of the synthesized derivatives (4a to 4e) were assigned by IR, NMR and mass spectral techniques. The hybrid compounds were evaluated for their antimicrobial, antitubercular and antioxidant activities. In addition, docking simulations were performed to study ligand-protein interactions and to determine the probable binding conformations. Results: Molecule 4a has shown anti-tubular activities with MIC 1.6 μg/ml. As compared to ascorbic acid activities (IC50 = 62.91 μg/ml), molecule 4e exhibited better antioxidant activities (IC50 = 24.85 μg/ml). Also, molecule 4e has shown significant antimicrobial activities. Conclusion: The synthesized derivatives from 4a to 4e have exhibited various medicinal activities and could be emerged as lead compounds and further explored as potential therapeutic agents.

Abstract:In the present study, synthesis and antimicrobial activity of 2,5-disubstituted 1,3,4-thiadiazole derivatives 5a-f are described. The structures of the newly synthesized compounds were confirmed by FT-IR, 1 H NMR, 13 C NMR, mass and elemental analysis. All compounds were screened for antitubercular and antimicrobial activity. Molecular modeling studies were performed to dock compounds into the ecKAS III binding site, which suggested probable inhibition mechanism. The results revealed that most of the compounds showed high to moderate biological activity against tested microorganisms.

The treatment of 1-[3,5-dimethyl-1-(4-nitrophenyl)-1H-pyrazol-4-yl]ethan-1-one with methyl hydrazinecarbodithioate or hydrazinecarbothioamide afforded 2-[1-[5-methyl-1-(4-nitrophenyl)-1H-pyrazol-4-yl]ethylidene]hydrazine derivatives. The targeted 1,3,4-thiadiazolyl derivatives were prepared by the reaction of 2-[1-[5-methyl-1-(4-nitrophenyl)-1H-pyrazol-4-yl]ethylidene]hydrazine derivatives with hydrazonoyl chloride derivatives. The reaction of N-(4-nitrophenyl)acetohydrazonoyl bromide with 2-[(methylthio)carbonthioyl]hydrazones in absolute ethanol in the presence of triethylamine afforded the corresponding 1,3,4-thiadiazole derivatives. The newly synthesized compounds were fully characterized by 1H NMR, 13C NMR, IR, MS, and elemental analysis. Moreover, the antimicrobial activity of the synthesized 1,3,4-thiadiazole derivatives were tested against E. coli, B. mycoides, and C. albicans. Four compounds outperformed the other produced compounds in terms of antimicrobial activity.

Abstract:Objective: The present study envisages a series of oxadiazole fluoroquinolone derivatives that were synthesized (D1–D12) with added derivatives such as phenyl, aminophenyl, amino hydroxyphenyl along with cyclopropyl, ethyl, piperazine, and imidazole. Methods: All of the newly produced molecules were characterized by infrared, 1H nuclear magnetic resonance, mass spectrometry, and elemental analysis technique and screened for docking stimulation to find out binding modes of synthesized derivatives with 3FV5, 5IMW, and 5ESE and evaluated for in vitro antimicrobial activity. Results: From this study, it was found that the compound D8 showed good antibacterial activity against Gram-positive (Staphylococcus aureus), compound D9 showed good antibacterial activity against Gram-negative (Escherichia coli), and compound D3 showed good antifungal activity against fungi (Saccharomyces cerevisiae) in comparison with standard drugs (Ciprofloxacin and fluconazole). The zone of inhibition and minimum inhibitory concentration studies was performed on synthesized compounds. Conclusion: The analogs of oxadiazole flouroquinolone are suggested to be potent inhibitors with sufficient scope for further exploration.

Abstract:Several 1-(1-aryl-1,4-dihydro-3-carboxy-6-methylpyridazine4-one)-4-aryl thio-semicarbazides and their corresponding oxadiazole, thiadiazole and triazole derivatives were prepared and characterized by their spectral data. The preliminary biological tests showed that some new compounds exhibit good anti-fungal activity.

Abstract:Novel (E)‐N‐(4‐(substituted) benzylidene)‐6‐substituted‐1H‐pyrazolo[3,4‐b]quinoline‐1‐carbothioamides/carboxamides were synthesized and characterized by IR, 1H NMR, 13C NMR and Mass spectral analysis. The synthesized series were subjected to antimicrobial activity and DFT studies. The results have shown that some the of the compounds exhibited significant antimicrobial activities comparable with that of the standard drugs. Synthesis of novel (E)‐N‐(4‐(substituted)benzylidene)‐6‐substituted‐1H‐pyrazolo[3,4‐b]quinoline‐1‐carbothioamides/carboxamides was achieved by the condensation of 6‐substituted‐1H‐pyrazolo[3,4‐b]quinoline‐1‐carbothioamides/carboxamides with substituted benzaldehydes in alcoholic medium in the presence of acetic acid. The structures of synthesized compounds are assigned on the basis of FT IR, 1H NMR, 13C NMR and Mass Spectral data. The compounds are subjected for their antibacterial, antifungal and DFT studies. Compounds, (E)‐6‐chloro‐N‐(4‐(dimethylamino)benzylidene)‐1H‐pyrazolo[3,4‐b]quinoline‐1‐carbothioamide (5 b), (E)‐6‐chloro‐N‐(4‐(dimethylamino)benzylidene)‐1H‐pyrazolo[3,4‐b]quinoline‐1‐carboxamide (5 f), and (E)‐6‐chloro‐N‐(4‐hydroxybenzylidene)‐1H‐pyrazolo[3,4‐b]quinoline‐1‐carboxamide (5 j) possessed pronounced antibacterial and antifungal activities due to their chemical structure.

Abstract:In this study, one-pot multicomponent reactions of novel chromeno[2,3- c ]pyrazole derivatives (1–14) were performed using an AlCl 3 catalyst via cyclisation. Various spectral and chromatographic techniques were used to elucidate the structure of the synthesised derivatives (1–14) . The synthesised compounds were then inspected for their antibacterial, antioxidant, and tyrosinase inhibition activities. An in silico screening approach was also employed to identify highly potent derivatives. Besides, we utilised density functional theory (DFT) with the B3LYP/6-31G + (d, p) basis set to optimise the newly modified derivatives. This approach was used to calculate various properties, including electron density, electrostatic potential map, interaction strength, frontier molecular orbital energy, and reactivity characteristics. To examine the binding affinity, modes, and stability of the protein-drug complex, molecular docking with the 2Y9X protein structure were employed. The findings from DFT computations, along with physicochemical information and molecular docking binding affinity, showed promising results than standard and low active compound 1 . The absorption, metabolism, and cytotoxic characteristics of all the novel derivatives were investigated in the ADMET prediction. Our findings could prove valuable in developing novel drugs for medicinal and pharmaceutical fields.

Abstract:A library of new chroman‐4‐one based 1,2,3‐triazole analogues were synthesized involving a series of condensation, cyclization, Suzuki coupling and copper catalysed click chemistry protocols. The newly synthesized compounds 8a‐l were screened for their invitro antioxidant and anti‐inflammatory activities by employing Ascorbic acid and Diclofenac as reference drugs respectively. The compound without any substituent on benzyl ring (8a), compound with ‐Cl substituent in para position of benzyl ring (8i), and compound with ethoxy substituent in para position of benzyl ring (8k) exhibited potent antioxidant and anti‐inflammatory activities with higher percentage of inhibition. To understand their binding affinities, molecular docking study of these three compounds performed against NADPH oxidase with presented outstanding docking scores and promising binding interactions like H‐bond and hydrophobic.
