Abstract:Electrically sensitive polyvinyl alcohol (PVA) hydrogels loaded with methotrexate (MTX) were prepared via a solution casting process, and characterized by attenuated total reflectance (ATR) spectrometry. In order to determine if the hydrogels were electro-sensitive or not, bending tests were conducted on the sulfoacetic acid modified hydrogels. It was observed that the prepared samples into strip forms started bending towards the cathode, and this bending was reversible when the polarity of the applied voltage was changed. The drug release study was performed on the MTX-loaded hydrogel strips placed in a sodium chloride (NaCl) solution under three different voltages (e.g., 0V, 5V, 10V, and 20V). Subsequently, the solutions were collected every five minutes in order to determine the drug release behaviors of the hydrogels using an ultraviolet-visible (UV-Vis) spectrophotometer. The test results showed that sulfoacetic acid (SA)-modified PVA hydrogels possess electrical sensitive behavior and kept their electric sensitivity for a long period of time. Also, the results confirmed that the control drug release could be achieved under different electrical voltages.

Abstract:A pH responsive nanoparticle-hydrogel hybrid drug delivery system was investigated for in-depth anticancer drug delivery to solid tumours. It consists of acid susceptible polymer nanoparticles loaded in a chitosan hydrogel. The hybrid formulation was characterized by UV-visible spectroscopy, FTIR, SEM, TEM, particle size analysis, zeta potential measurement and viscosity measurement. Drug encapsulation and nanoparticle loading efficiencies were found to be 48% and 72% respectively which describes the efficient interaction of the chemical entities in this hybrid drug delivery system. The hydrogel exhibited pH responsive behaviour: minimal drug and nanoparticle release at physiological pH but an increase in viscosity under acidic conditions and fast nanoparticle and drug release. The cytotoxicity of the drug loaded hydrogel was investigated against the MCF-7 breast cancer cell line along with the drug and nanoparticles without hydrogel. The drug loaded hydrogel showed a better cytotoxic effect on MCF-7 cancer cells. Thus, drug loaded nanoparticles containing hydrogel could be a better option for maximum drug distribution in tumours.
Synthesis and characterization of Gellan gum-based hydrogels for the delivery of anticancer drug etoposide

Abstract:Present research work reports the synthesis of Gellan gum (Gg) and methacrylic acid (MA) based grafted hydrogels (Gg-cl-poly(MA)) crosslinked using N, N'- methylene-bis-acrylamide (MBA) and the evaluation of their efficiency to be used as a sustained drug delivery carrier for anticancer drug i.e., etoposide. Various characterization techniques like Fourier transform infrared spectroscopy (FTIR), X-ray diffraction (XRD), and scanning electron microscopy (SEM) confirmed the grafting of Gg with MA and the formation of crosslinked Gg-cl-poly(MA) hydrogel polymer. The synthesized hydrogel showed pH-dependent swelling properties and exhibited a maximum swelling capacity of 867 % under optimized environmental conditions. The Gg-cl-poly(MA) was biocompatible and non-cytotoxic, which was confirmed by the hemolytic and cytotoxic tests. The release dynamics of etoposide from the Gg-cl-poly(MA) polymer matrix was checked under specific physiological conditions. Drug release was found to be significantly higher in the acidic medium, followed by the neutral and alkaline medium. This clearly indicated that etoposide drug release through synthesized hydrogel was stomach-specific and it is effective for the treatment of stomach cancer. The release mechanism of the etoposide drug was a Fickian-type diffusion mechanism in the acidic medium and a non-Fickian-type diffusion mechanism in the neutral and alkaline medium. The release profile of the etoposide was best fitted to the first-order rate model. The results showed that the synthesized hydrogel (i.e., Gg-cl-poly(MA)) was biocompatible, non-toxic, and could be used for the treatment of stomach cancer.

Abstract:The manuscript represents synthesis of pH and salt responsive hydrogel based on guar gum as a renewable substrate for drug delivery of natural anticancer drug i.e. curcumin. Initially acrylic acid grafted guar gum (GG-g-PAA) hydrogel was obtained in the presence of ammonium persulphate as a redox initiator. Structural stability of grafted copolymer was improved using N, N-methylenebisacrylamide as a crosslinker with simultaneous loading of curcumin as a natural anticancer drug. Delivery rate of curcumin from GG-g-PAA was studied by UV spectrophotometer. The neat and drug loaded hydrogels were characterized using FTIR, NMR, TGA, and SEM techniques for their structural, morphological, and thermal behaviours. Swelling of the hydrogels and water diffusion properties were studied in salt and aqueous solutions of different pH. Finally, the fabricated hydrogel matrices were tested for curcumin release studies to understand the release kinetic. The results indicated that the prepared renewable source based hydrogel can be used in drug delivery of curcumin as a natural medicine. As curcumin and guar gum are products of agricultural food crops <i>viz</i>. turmeric and guar, the current research can be a good contribution towards preparation of an eco-friendly drug delivery system based on agriculture products.

Abstract:The most popular cancer treatment protocols are surgery, radiation therapy, and chemotherapy. Amongst, chemotherapy has received more interest owing to some superior advantages such as adjuvants to local therapy as well as simplicity, availability, and excellent efficacy at suitable dosage. Despite, some drawbacks ( e.g., side effects, drug resistance, low efficiency due to dose-limiting toxicity, and pour solubility of some anti-cancer drugs) of this therapy approach should circumvent to achieve better outcomes. On the other hand, hyperthermia therapy is a promising cancer treatment method due to its low side effects. Nevertheless, lack of homogeneous heat distribution, the inevitable heat treatment of healthy cells, as well as the need for careful control of the patient body's temperature and physiological condition during treatment are the major challenges in the fields. As the superior physicochemical and biological features of natural and stimuli-responsive hydrogels as well as nanomaterials, these biomaterials can be used for formulation of anti-cancer drugs to reduce numerous side effects of conventional chemotherapy as well as combinational synergistic hyperthermia therapy. In present assay, the focus is on recent progress in the combined chemo/hyperthermia therapy based on natural and stimuli-responsive hydrogels as a synergistic cancer treatment strategy. Abstract:Recent focus is on developing cancer treatments that overcome challenges of high-dose chemotherapy, providing improved safety, effectiveness, and site-specific targeting through responsive mechanisms. In our quest for an advanced drug delivery vehicle, herein we have developed a self-healable and injectable polymeric eutectogels that responds to intracellular biological stimuli i.e., pH and temperature. The eutectogels made through interacting imidazolium based deep eutectic solvents with polyvinyl alcohol serve as a local drug delivery system for cancer therapeutics, releasing the anti-cancer drug 5-fluorouracil (5-FU) to the targeted site through intracellular pH, i.e. pH 5.0. Rheology experiments showed strong mechanical strength (>7.3 % strain) for the studied eutectogels that have hierarchical and porous 3D network morphology formed by thick fibers. It exhibits crucial qualities- swelling, injectability, and self-healing- essential for an effective drug delivery vehicle. Additionally, eutectogels demonstrates outstanding biocompatibility, with hemolysis below 2 %, ensuring blood compatibility and sustained viability in HaCaT cells. Leveraging these properties, 5-FU-loaded eutectogels were studied for in vitro cytotoxicity (MCF-7 and HeLa) and drug release that revealed that the eutectogels can be used for targeted delivery under physiological conditions and can more efficiently kill cancerous cells.

Abstract:This research article introduced a novel one-pot method for synthesizing hydrogels, utilizing iron ions (Fe 3+ ) and guar gum succinate (GGS) as cross-linkers. These hydrogels were characterized as cross-linked networks, with hydrogen bonds forming a sacrificial network and coordination bonds serving as the primary network. The reversible nature of these networks was attributed to the hydrogels' exceptional toughness and remarkable self-healing properties. The hydrogel's chemical structure was confirmed through FTIR spectroscopy. XRD analysis highlighted the disruption of the crystalline nature of GGS upon cross-linking with Fe 3+ . By controlling the Fe 3+ concentration, the hydrogels' mechanical properties were tailored. Rheological measurements demonstrated mechanical and self-healing properties, while swelling studies revealed pH-dependent behavior. In vitro studies showed the hydrogels' significant anti-proliferative effect against U-87MG (human glioblastoma) cancer cells, while remaining biocompatible with normal cell lines (HEK-293). These results indicated the potential application of these hydrogels in advancing cancer treatment strategies.

Abstract:Aims: Breast cancer is the most common malignancy among women in both high- and low-resource settings. Conventional breast cancer therapies were inefficient and had low patient compliance. Stimuli-responsive hydrogels possessing similar physicochemical features as soft tissue facilitate diagnostic and therapeutic approaches for breast cancer subtypes. Scope: Polysaccharides and polypeptides are major natural polymers with unique biocompatibility, biodegradability, and feasible modification approaches utilized frequently for hydrogel fabrication. Alternating the natural polymer-based hydrogel properties in response to external stimuli such as pH, temperature, light, ultrasonic, enzyme, glucose, magnetic, redox, and electric have provided great potential for the evolution of novel drug delivery systems (DDSs) and various advanced technologies in medical applications. Stimuli-responsive hydrogels are triggered by specific cancer tissue features, promote target delivery techniques, and modify release therapeutic agents at localized sites. This narrative review presented innovation in preparing and characterizing the most common stimuli-responsive natural polymer-based hydrogels for diagnostic and therapeutic applications in the breast cancer area. Conclusion: Stimuli-responsive hydrogels display bioinspiration products as DDSs for breast cancer subtypes, protect the shape of breast tissue, provide modified drug release, enhance therapeutic efficacy, and minimize chemotherapy agents' side effects. The potential benefits of smart natural polymer-based hydrogels make them an exciting area of practice for breast cancer diagnosis and treatment.

Abstract:Cancer is a highly heterogeneous disease and remains a global health challenge affecting millions of human lives worldwide. Despite advancements in conventional treatments like surgery, chemotherapy, and immunotherapy, the rise of multidrug resistance, tumor recurrence, and their severe side effects and the complex nature of the tumor microenvironment (TME) necessitates innovative therapeutic approaches. Recently, stimulus-responsive nanomedicines designed to target TME characteristics (e.g., pH alterations, redox conditions, enzyme secretion) have gained attention for their potential to enhance anticancer efficacy while minimizing the adverse effects of chemotherapeutics/bioactive compounds. Among the various nanocarriers, hydrogels are intriguing due to their high-water content, adjustable mechanical characteristics, and responsiveness to external and internal stimuli, making them promising candidates for cancer therapy. These properties make hydrogels an ideal nanocarrier for controlled drug release within the TME. This review comprehensively surveys the latest advancements in the area of stimulus-responsive hydrogels for cancer therapy, exploring various stimuli-responsive mechanisms, including biological (e.g., pH, redox), chemical (e.g., enzymes, glucose), and physical (e.g., temperature, light), as well as dual- or multi-stimuli responsiveness. Furthermore, this review addresses the current developments and challenges in hydrogels in cancer treatment. Our aim is to provide readers with a comprehensive understanding of stimulus-responsive hydrogels for cancer treatment, offering novel perspectives on their development for cancer therapy and other medical applications.
Preparation of Composite Hydrogels Based on Cysteine-Silver Sol and Methylene Blue as Promising Systems for Anticancer Photodynamic Therapy

Abstract:In this study, a novel supramolecular composite, "photogels", was synthesized by mixing of cysteine-silver sol (CSS) and methylene blue (MB). A complex of modern physico-chemical methods of analysis such as viscosimetry, UV spectroscopy, dynamic and electrophoretic light scattering, scanning electron microscopy and energy-dispersive X-ray spectroscopy showed that MB molecules are uniformly localized mainly in the space between fibers of the gel-network formed by CSS particles. Molecules of the dye also bind with the surface of CSS particles by non-covalent interactions. This fact is reflected in the appearance of a synergistic anticancer effect of gels against human squamous cell carcinoma even in the absence of light irradiation. A mild toxic influence of hydrogels was observed in normal keratinocyte cells. Photodynamic exposure significantly increased gel activity, and there remained a synergistic effect. The study of free-radical oxidation in cells has shown that gels are not only capable of generating reactive oxygen species, but also have other targets of action. Flow cytometric analysis allowed us to find out that obtained hydrogels caused cell cycle arrest both without irradiation and with light exposure. The obtained gels are of considerable interest both from the point of view of academics and applied science, for example, in the photodynamic therapy of superficial neoplasms.
Injectable and body temperature sensitive hydrogels based on chitosan and hyaluronic acid for pH sensitive drug release

Abstract:Hydrogels based on chitosan/hyaluronic acid/β-sodium glycerophosphate demonstrate injectability, body temperature sensitivity, pH sensitive drug release and adhesion to cancer cell. The drug (doxorubicin) loaded hydrogel precursor solutions are injectable and turn to hydrogels when the temperature is increased to body temperature. The acidic condition (pH 4.00) can trigger the release of drug and the cancer cell (Hela) can adhere to the surface of the hydrogels, which will be beneficial for tumor site-specific administration of drug. The mechanical strength, the gelation temperature, and the drug release behavior can be tuned by varying hyaluronic acid content. The mechanisms were characterized using dynamic mechanical analysis, Fourier transform infrared spectroscopy, scanning electron microscopy and fluorescence microscopy. The carboxyl group in hyaluronic acid can form the hydrogen bondings with the protonated amine in chitosan, which promotes the increase of mechanical strength of the hydrogels and depresses the initial burst release of drug from the hydrogel.

Abstract:Designing stimuli-responsive drug delivery vehicles with higher drug loading capacity, sustained and targeted release of anti-cancer drugs and able to mitigate the shortcomings of traditional systems is need of hour. Herein, we designed stimuli-responsive, self-healable, and adhesive hydrogel through synergetic interaction between [Cho][Gly] (Choline-Glycine) and sodium alginate (SA). The hydrogel was formed as a result of non-covalent interaction between the components of the mixture forming the fibre kind morphology; confirmed through FTIR/computational analysis and SEM/AFM images. The hydrogel exhibited excellent mechanical strength, self-healing ability, adhesive character and most importantly; adjustable injectability. In vitro biocompatibility of the hydrogel was tested on HaCaT and MCF-7 cells, showing >92 % cell viability after 48 h. The hemolysis ratio (<4 %) of the hydrogel confirmed the blood compatibility of the hydrogel. When tested for drug-loading capacity, the hydrogel show 1500 times drug loading for the 5-fluorouracil (5-FU) against the SA based hydrogel. In vitro release data indicated that 5-FU have more preference towards the cancerous cell condition, i.e. acidic pH (>85 %), whereas the drug-loaded hydrogel successfully killed the MCF-7 and HeLa cell with a <IC50-1.92 mM value. The studied hydrogel paves way towards controlled and sustained delivery of anti-cancer drug for the treatment of breast cancer.

Abstract:Cancer, as leading cause of death, has a high rate of mortality worldwide. Although there is a wide variety of conventional approaches for the treatment of cancer (such as surgery and chemotherapy), they have considerable drawbacks in terms of practicality, treatment efficiency, and cost-effectiveness. Therefore, there is a fundamental requirement for the development of safe and efficient treatment modalities based on breakthrough technologies to suppress cancer. Chitosan (CS) and hyaluronic acid (HA) polysaccharides, as FDA-approved biomaterials for some biomedical applications, are potential biopolymers for the efficient treatment of cancer. CS and HA have high biocompatibility, bioavailability, biodegradability, and immunomodulatory function which guarantee their safety and non-toxicity. CS-/HA-based hydrogels (HGs)/composites stand out for their potential anticancer function, versatile preparation and modification, ease of administration, controlled/sustained drug release, and active and passive drug internalization into target cells which is crucial for efficient treatment of cancer compared with conventional treatment approaches. These HGs/composites can respond to external (magnetic, ultrasound, light, and thermal) and internal (pH, enzyme, redox, and ROS) stimuli as well which further paves the way to their manipulation, targeted drug delivery, practicality, and efficient treatment. The above-mentioned properties of CS-/HA-based HGs/composites are unique and practical in cancer treatment which can ignore the deficiencies of conventional approaches. The present manuscript comprehensively highlights the advances in the practical application of stimuli-responsive HGs/composites based on CS/HA polysaccharides.

Abstract:Magnetic bio-nanocomposite hydrogels are a promising platform for tissue engineering and drug delivery applications. This research presents the synthesis and characterization of novel magnetic bio-nanocomposite hydrogel beads based on magnetic hydroxyapatite nanoparticles and chitosan for potential pH-sensitive drug delivery. The successful synthesis was confirmed through various analysis methods. The prepared bio-nanocomposites hydrogel beads exhibited pH-sensitive swelling behavior. Furthermore, the bio-nanocomposites demonstrated a high loading capacity for doxorubicin (DOX) anticancer drug and a pH-sensitive drug release profile, with approximately 80% release rate at pH 5 and less than 10% release rate at pH 7.4. The Weibull kinetic model was the best-fitting function for DOX release from bio-nanocomposite hydrogels at pH 5. The cytotoxicity of DOX-loaded bio-nanocomposite hydrogels against HeLa cells was evaluated, which showed high cytotoxicity after incubation for 48 h at a concentration of 2.5 mg/mL. These findings suggest that the prepared magnetic bio-nanocomposite hydrogel beads have good potential for implantable pH-sensitive drug delivery in the treatment of cancerous tissue.

Abstract:Drug delivery techniques based on polymers have been investigated for their potential to improve drug solubility, reduce systemic side effects, and controlled and targeted administration at infection site. In this study, we developed a co-polymeric hydrogel composed of graphene sheets (GNS), polyvinyl alcohol (PVA), and chitosan (CS) that is loaded with methotrexate (MTX) for in vitro liver cancer treatment. Fourier transform infrared spectroscopy (FTIR) and atomic force microscopy (AFM) was employed to check the structural properties and surface morphology. Moreover, tests were conducted on the cytotoxicity, hemolytic activity, release kinetics, swelling behaviour and degradation of hydrogels. A controlled release of drug from hydrogel in PBS at pH 7.4 was examined using release kinetics. Maximal drug release in six hours was 97.34%. The prepared hydrogels did not encourage the HepG2 growth and were non-hemolytic. The current study highlights the potential of GNS-based hydrogel loaded with MTX as an encouraging therapy for hepatocellular carcinoma. HepG2 cell viability of MTX-loaded CS-PVA-GNS hydrogel was (IC50 5.87 μg/200 mL) in comparison to free MTX (IC50 5.03 μg/200 mL). These outcomes recommend that hydrogels with GNS ensure improved drug delivery in cancer microenvironment while lessening adverse consequences on healthy cells.

pH-Sensitive Peptide Hydrogels as a Combination Drug Delivery System for Cancer Treatment

Abstract:Conventional antitumor chemotherapeutics generally have shortcomings in terms of dissolubility, selectivity and drug action time, and it has been difficult to achieve high antitumor efficacy with single-drug therapy. At present, combination therapy with two or more drugs is widely used in the treatment of cancer, but a shortcoming is that the drugs do not reach the target at the same time, resulting in a reduction in efficacy. Therefore, it is necessary to design a carrier that can release two drugs at the same site. We designed an injectable pH-responsive OE peptide hydrogel as a carrier material for the antitumor drugs gemcitabine (GEM) and paclitaxel (PTX) that can release drugs at the tumor site simultaneously to achieve the antitumor effect. After determining the optimal gelation concentration of the OE polypeptide, we conducted an in vitro release study to prove its pH sensitivity. The release of PTX from the OE hydrogel in the medium at pH 5.8 and pH 7.4 was 96.90% and 38.98% in 7 days. The release of GEM from the OE hydrogel in media with pH of 5.8 and 7.4 was 99.99% and 99.63% in 3 days. Transmission electron microscopy (TEM) and circular dichroism (CD) experiments were used to observe the microstructure of the peptides. The circular dichroism of OE showed a single negative peak shape when under neutral conditions, indicating a β-folded structure, while under acidic conditions, it presented characteristics of a random coil. Rheological experiments were used to investigate the mechanical strength of this peptide hydrogel. Furthermore, the treatment effect of the drug-loaded peptide hydrogel was demonstrated through in vitro and in vivo experiments. The results show that the peptide hydrogels have different structures at different pH values and are highly sensitive to pH. They can reach the tumor site by injection and are induced by the tumor microenvironment to release antitumor drugs slowly and continuously. This biologically functional material has a promising future in drug delivery for combination drugs.

Abstract:Local delivery of anticancer agents via injectable hydrogels could be a promising method for achieving spatiotemporal control on drug release as well as minimizing the disadvantages related to the systemic mode of drug delivery. Keeping this in mind, we report the development of N,O-carboxymethyl chitosan (N,O-CMCS)-guar gum-based injectable hydrogels for the sustained delivery of anticancer drugs. The hydrogels were synthesized by chemical crosslinking of multialdehyde guar gum (MAGG) and N,O-CMCS through dynamic Schiff base linkages, without requiring any external crosslinker. Fabrication of injectable hydrogels, involving N,O-CMCS and MAGG via Schiff base crosslinking, is being reported for the first time. The hydrogels exhibited pH-responsive swelling behavior and good mechanical properties with a storage modulus of about 1625 Pa. Due to the reversible nature of Schiff base linkages, hydrogels displayed excellent self-healing and thixotropic properties. Doxorubicin (Dox), an anticancer agent, was loaded onto these hydrogels and its release studies were conducted at pH 7.4 (physiological) and pH 5.5 (tumoral). A sustained release of about 67.06% Dox was observed from the hydrogel after 5 days at pH 5.5 and about 32.13% at pH 7.4. The 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide assay on the human embryonic kidney cell line (HEK-293) and the hemolytic assay demonstrated the biocompatible nature of the hydrogels. The Dox-loaded hydrogel exhibited a significant killing effect against breast cancer cells (MCF-7) with a cytotoxicity of about 72.13%. All the data presented support the efficiency of the synthesized N,O-CMCS/MAGG hydrogel as a biomaterial that may find promising applications in anticancer drug delivery.

Abstract:The constraints associated with current cancer therapies have inspired scientists to develop advanced, precise, and safe drug delivery methods. These delivery systems boost treatment effectiveness, minimize harm to healthy cells, and combat cancer recurrence. To design advanced drug delivery vehicle with these character, in the present manuscript, we have designed a self-healing and injectable hybrid hydrogel through synergistically interacting metal organic framework, CuBTC with the poly(vinyl alcohol) (PVA). This hybrid hydrogel acts as a localized drug delivery system and was used to encapsulate and release the anticancer drug 5-Fluorouracil selectively at the targeted site in response to the physiological pH. The hydrogel was formed through transforming the gaussian coil like matrix of PVA-CuBTC into a three-dimensional network of hydrogel upon the addition of crosslinker; borax. The biocompatible character of the hydrogel was confirmed through cell viability test. The biocompatible hybrid hydrogel then was used to encapsulate and studied for the pH responsive release behavior of the anti-cancer drug, 5-FU. The in vitro cytotoxicity of the drug-loaded hydrogel was evaluated against MCF-7 and HeLa cells. The study confirms that the hybrid hydrogel is effective for targeted and sustained release of anticancer drugs at cancer sites.

Abstract:As an emerging drug carrier, hydrogels have been widely used for tumor drug delivery. A hydrogel drug carrier can cause less severe side effects than systemic chemotherapy and can achieve sustained delivery of a drug at tumor sites. In addition, hydrogels have excellent biocompatibility and biodegradability and lower toxicity than nanoparticle carriers. Smart hydrogels can respond to stimuli in the environment (e.g., heat, pH, light, and ultrasound), enabling in situ gelation and controlled drug release, which greatly enhance the convenience and efficiency of drug delivery. Here, we summarize the different sizes of hydrogels used for cancer treatment and their related delivery routes, discuss the design strategies for stimuli-responsive hydrogels, and review the research concerning smart hydrogels reported in the past few years.
