Abstract:Abstract The structural motifs of trifluoromethylated aromatics (Ar‐CF3) have been widely employed in various industrial fields. The high lipophilicity, strong electron‐withdrawing ability, and characteristic size of the trifluoromethyl group is a key influence in the design of pharmaceuticals, agrochemicals, dyes, liquid crystals, and polymers. Therefore, a great deal of attention has been paid to convenient protocols of introducing trifluoromethyl groups into aromatic rings. With respect to the high regiochemical fidelity in aromatic substitution, the cross‐coupling of aryl halides with CF3Cu is one of the most powerful and versatile methods to construct trifluoromethylated aromatics.

Abstract:The trifluoromethylation of aryl iodides catalyzed by copper(I) salt with trifluoromethylzinc reagent prepared in situ from trifluoromethyl iodide and Zn dust was accomplished. The catalytic reactions proceeded under mild reaction conditions, providing the corresponding aromatic trifluoromethylated products in moderate to high yields. The advantage of this method is that additives such as metal fluoride (MF), which are indispensable to activate silyl groups for transmetallation in the corresponding reactions catalyzed by copper salt by using the Ruppert–Prakash reagents (CF 3 SiR 3 ), are not required.

Abstract:DBU-promoted trifluoromethylation of aryl iodides with difluoromethyltriphenylphosphonium bromide (DFPB) in the presence of copper source is described. In this transformation, DBU not only acts as base to deprotonate the difluoromethyl group in DFPB to generate difluoromethylene phosphonium ylide Ph3P+CF2−, but also converts the difluorocarbene generated from ylide Ph3P+CF2− into trifluoromethyl anion, finally resulting in the trifluoromethylation of aryl iodides. The reactions proceeded smoothly to afford expected products in moderate to good yields.

Abstract:A variety of vinylic bromides and iodides undergo smooth trifluoromethylation and pentafluoroethylation with RfH-derived CuRf (Rf = CF3, C2F5) to give the corresponding fluoroalkylated olefins. These reactions employing the low-cost CuRf reagents occur in high yield with excellent chemo- and stereoselectivity under mild conditions (23–80 °C). Crystal structures of one trifluoromethyl and one pentafluoroethyl derivative have been determined.

Abstract:Catalyzed by CuOTf under very mild conditions, aryl triflates which are embedded in the azacalix[1]arene[3]pyridine macrocycle underwent coupling reactions with metal halides and acetates to afford respectively halogenated and acyloxylated arene products in moderate to excellent yields. The unprecedented CuOTf-catalyzed transformations of aryl triflates proceeded through an oxidative addition of intramolecularly chelated Cu(I) into the C–O bond of aryl triflates to form arylcopper(III) intermediates which underwent anion exchange and reductive elimination reactions with nucleophiles to yield functionalized macrocyclic products.

Abstract:Herein, a copper-catalyzed 2,2,2-trifluoroethylthiolation reaction of aryl bromides and iodides with elemental sulfur, and 1,1,1-trifluoro-2-iodoethane is described. The reaction showed excellent functional group tolerance and allowed the synthesis of various substituted aryl 2,2,2-trifluoroethyl thioethers with good to excellent yields. This transformation constitutes a one-pot synthesis of 2,2,2-trifluoroethylthiolated compounds from inexpensive, readily available starting materials. Utility of the protocol was further demonstrated in the late-stage synthesis of the pirfenidone derivative. The copper thiolate species were prepared and proposed as key intermediates in the catalytic cycle.

Abstract:The copper-mediated trifluoromethylation of alkyl radicals is described. The combination of Et3SiH and K2S2O8 initiates the radical reactions of alkyl bromides or iodides with BPyCu(CF3)3 (BPy = 2,2'-bipyridine) in aqueous acetone at room temperature to afford the corresponding trifluoromethylation products in good yield. The protocol is applicable to various primary and secondary alkyl halides and exhibits wide functional group compatibility. A mechanism involving trifluoromethyl group transfer from Cu(II)-CF3 intermediates to alkyl radicals is proposed.

Abstract:A n ew method of constructing "ArSCF2CF2Cu" from ArSCu and TMSCF2Br (TMS = trimethylsilyl) has been developed. The cross-coupling reactions of the obtained "ArSCF2CF2Cu" with diverse aryl iodides provide an efficient access to Ar'CF2CF2SAr. Mechanistic studies demonstrate that the "ArSCF2CF2Cu" species w ere generated through controllable double difluoromethylene insertions into ArS - Cu bond rather than the 1,2-addition to tetrafluoroethylene.

Abstract:Herein, we present a novel strategy for synthesizing polyfluorinated compounds by the fluorotrifluoromethylation of olefins, which was achieved through a new trifluoromethyl-iodine(III) reagent TFNI-1. TFNI-1 was readily synthesized via a three-step process, and its structure was characterized by NMR spectroscopy and X-ray crystallography. It is shown by radical trapping and radical clock experiments that the reaction involves the CF(3) radical intermediate.

Abstract:A Cu(I)‐catalyzed three‐component reaction of 2‐iodo‐2,2‐difluoroacetophenones, alkynes and TMSCN is described. The reaction provided a facile method for the synthesis of difluoroacyl‐substituted nitriles, which might be served as potentially useful fluoroorganic intermediates for further transformation in drug discovery. This method has broad substrate scope, good efficiency and excellent stereoselectivity . Preliminary mechanistic investigation indicated that radical‐mediate process was involved in this cyanodifluoroalkylation reaction.

Abstract:Fluorine-containing compounds have shown unparalleled impacts in the realm of functional molecules, and the ability to prepare novel structures has been crucial in unlocking new properties for pharmaceutical and materials science. Herein, we report a copper-catalyzed, photoinduced defluorinative C‒O coupling between trifluoromethylarenes and alcohols. This method allows for direct access to a wide selection of difluorobenzylether (ArCF2OR) molecules, including a com-pound displaying liquid crystal behavior. Through slight modification of the protocol, we were able to generate ArCF2I products, another class of synthetically useful fluorine-bearing molecules. Comprehensive mechanistic investigations first suggested ArCF2I as a reservoir to steadily supply the key ArCF2• radical species. Furthermore, experimental evi-dence supported a mechanism consisting of two collaborative cycles: C‒F activation operated by a homoleptic Cu(I) co-ordinated by two bisphosphine ligands as the photocatalyst and C‒O coupling promoted by a Cu(I) ligated by a single bisphosphine ligand. The critical roles of the two salt additives, lithium iodide and zinc acetate, in orchestrating the two cycles were also elucidated. This dual-role copper catalyst demonstrates the power of base metal photoredox catalysis in achieving both substrate activation and chemical bond formation via a single catalytic system.
