Abstract:The rise of a pest species represents a unique opportunity to address how species evolve new behaviors and adapt to novel ecological niches [1]. We address this question by studying the egg-laying behavior of Drosophila suzukii, an invasive agricultural pest species that has spread from Southeast Asia to Europe and North America in the last decade [2]. While most closely related Drosophila species lay their eggs on decaying plant substrates, D. suzukii oviposits on ripening fruit, thereby causing substantial economic losses to the fruit industry [3-8]. D. suzukii has evolved an enlarged, serrated ovipositor that presumably plays a key role by enabling females to pierce the skin of ripe fruit [9]. Here, we explore how D. suzukii selects oviposition sites, and how this behavior differs from that of closely related species. We have combined behavioral experiments in multiple species with neurogenetics and mutant analysis in D. suzukii to show that this species has evolved a specific preference for oviposition on ripe fruit. Our results also establish that changes in mechanosensation, olfaction, and presumably gustation have contributed to this ecological shift. Our observations support a model in which the emergence of D. suzukii as an agricultural pest is the consequence of the progressive modification of several sensory systems, which collectively underlie a radical change in oviposition behavior.

Abstract:Summary: In insects, odorant receptors (ORs) are required for the detection of most olfactory cues. We investigated the function of a clade of four duplicated ORs in the hawkmoth Manduca sexta and found that these paralogs encode broadly tuned receptors with overlapping but distinct response spectra. Two paralogs, which arose after divergence from a related lineage, show high sensitivity to floral esters released by a nectar-rich plant frequently visited by M. sexta. Functional imaging in mutant moths lacking one of the paralogs suggests that olfactory sensory neurons expressing this OR target a previously identified feeding-associated glomerulus in the primary olfactory center of the brain. However, only the response of this glomerulus to the single ligand unique to the now mutated OR disappeared, suggesting neuronal coexpression of the paralogs. Our results suggest a link between the studied OR expansion and enhanced detection of odors emitted by valuable nectar sources in M. sexta.

Abstract:Comparative analyses of multiple genomes are used extensively to examine the gains and losses of chemosensory receptors across the genus Drosophila. However, few studies have delved into functional olfactory characteristics. Here we assess olfactory function across 20 species, and identify and describe several similar elements of evolution. We document (a) minor changes in functional ligands based on amino acid substitutions, (b) major changes in olfactory function or perhaps entire receptor replacements, and (c) that only a few receptors are subject to repeated changes, whereas 32 out of 37 OSNs are largely functionally conserved. In addition, we generate a robust model for identifying olfactory function using genomic data and comprehensive ligand-receptor combinations, which includes the prediction of binding pockets. Moreover, this study highlights that functional olfactory evolution does not affect all chemosensory receptors equally, and that ecological, evolutionary, and developmental forces repeatedly affect only a small subset of available receptor proteins.
The Power of Drosophila Genetics in Studying Insect Toxicology and Chemical Ecology

Abstract:Insect toxicology and chemical ecology are inherently interconnected disciplines, both dedicated to unraveling the intricate relationships between insects and the diverse array of chemical compounds that pervade their surroundings. Drosophila melanogaster , owing to its genetic and physiological similarities to other insects, serves as a robust model system in the study of insect toxicology. Moreover, state-of-the-art techniques in Drosophila neurobiology have extensively probed the chemosensory system of insects, providing significant insights into their adaptation to chemical environments. In this review, we emphasize the advancements achieved through the application of Drosophila genetics in investigations spanning both of these fields, significantly enhancing our understanding of the mode of action and resistance mechanisms of insecticides, as well as unraveling the molecular and cellular mechanisms underlying insect chemosensation and associated behaviors. The profound insights derived through this tiny fly not only enrich our understanding of the broader world of insects but also hold the potential to develop more effective and sustainable strategies for pest management.

Abstract:Background Most animals, including humans and insects, consume foods based on their senses. Feeding is mostly regulated by taste and smell. Recent insect studies shed insight into the cross-talk between taste and smell, sweetness and temperature, sweetness and texture, and other sensory modality pairings. Five canonical tastes include sweet, umami, bitter, salty, and sour. Furthermore, other receptors that mediate the detection of noncanonical sensory attributes encoded by taste stimuli, such as Ca 2+ , Zn 2+ , Cu 2+ , lipid, and carbonation, have been characterized. Deorphanizing receptors and interactions among different modalities are expanding the taste field. Methods Our study explores the taste system of Drosophila melanogaster and perception processing in insects to broaden the neuroscience of taste. Attractive and aversive taste cues and their chemoreceptors are categorized as tables. In addition, we summarize the recent progress in animal behavior as affected by the integration of multisensory information in relation to different gustatory receptor neuronal activations, olfaction, texture, and temperature. We mainly focus on peripheral responses and insect decision-making. Conclusion Drosophila is an excellent model animal to study the cellular and molecular mechanism of the taste system. Despite the divergence in the receptors to detect chemicals, taste research in the fruit fly can offer new insights into the many different taste sensors of animals and how to test the interaction among different sensory modalities.

Abstract:Behavioural changes in habitat or mate choice can trigger population divergence, leading to speciation. However, little is known about the neurological bases for such changes. Rhagoletis pomonella (Diptera: Tephritidae) is a model for ecological speciation via host plant shifts. Within the past 180 years, Rhagoletis flies infesting hawthorn (Crataegus spp.) shifted to attack domesticated apple (Malus pumila). The two populations differ in their olfactory preferences for apple versus hawthorn fruit. Here, we looked for patterns of sensory organization that may have contributed to this shift by characterizing the morphology, specificity and distribution of olfactory sensory neurons (OSNs) on the antennae of Rhagoletis responding to host fruit and non-host volatiles. Of 28 OSN classes identified, two colocalized OSN pairs were found that specifically responded to the major behavioural attractant and antagonist volatiles for each fly population. A reversal in the response of these OSNs to fruit volatiles, either through a switch in receptor expression between these paired neurons or changes in neuronal projections in the brain, could therefore account for the behavioural difference between apple and hawthorn flies. The finding supports the hypothesis that relatively minor changes in olfactory sensory pathways may contribute to rapid host shifting and divergence in Rhagoletis.

Abstract:Comparative analysis of multiple genomes has been used extensively to examine the evolution of chemosensory receptors across the genus Drosophila . However, few studies have delved into functional characteristics, as most have relied exclusively on genomic data alone, especially for non-model species. In order to increase our understanding of olfactory evolution, we have generated a comprehensive assessment of the olfactory functions associated with the antenna and palps for Drosophila suzukii as well as several other members of the subgenus Sophophora , thus creating a functional olfactory landscape across a total of 20 species. Here we identify and describe several common elements of evolution, including consistent changes in ligand spectra as well as relative receptor abundance, which appear heavily correlated with the known phylogeny. We also combine our functional ligand data with protein orthologue alignments to provide a high-throughput evolutionary assessment and predictive model, where we begin to examine the underlying mechanisms of evolutionary changes utilizing both genetics and odorant binding affinities. In addition, we document that only a few receptors frequently vary between species, and we evaluate the justifications for evolution to reoccur repeatedly within only this small subset of available olfactory sensory neurons.
Navigating the landscape of fear: Fruit flies exhibit distinct antipredator and antiparasite defensive behaviors

Abstract:Most organisms are at risk of being consumed by a predator or getting infected by a parasite at some point in their life. Theoretical constructs such as the landscape of fear (perception of risk) and nonconsumptive effects (NCEs, costly responses sans predation or infection) have been proposed to describe and quantify antipredator and antiparasite responses. How prey/host species identify and respond to these risks determines their survival, reproductive success and, ultimately, fitness. Most studies to date have focused on either predator–prey or parasite–host interactions, yet habitats and ecosystems contain both parasitic and/or predatory species that represent a complex and heterogenous mosaic of risk factors. Here, we experimentally investigated the behavioral responses of a cactophilic fruit fly, Drosophila nigrospiracula, exposed to a range of species that include parasites (ectoparasitic mite), predators (jumping spiders), as well as harmless heterospecifics (nonparasitic mites, ants, and weevils). We demonstrate that D. nigrospiracula can differentiate between threat and non‐threat species, increase erratic movements and decrease velocity in the presence of parasites, but decrease erratic movements and time spent grooming in the presence of predators. Of particular importance, flies could distinguish between parasitic female mites and nonparasitic male mites of the same species, and respond accordingly. We also show that the direction of these NCEs differs when exposed to parasitic mites (i.e., risk of infection) versus spiders (i.e., risk of predation). Given the opposing effects of predation versus infection risk on fly behavior, we discuss potential trade‐offs between parasite and predator avoidance behaviors. Our findings illustrate the complexity of risk assessment in a landscape of fear and the fine‐tuned NCEs that arise in response. Moreover, this study is the first to examine these behavioral NCEs in a terrestrial system.

Abstract:In insect olfaction, sensitization refers to the amplification of a weak olfactory signal when the stimulus is repeated within a specific time window. In the vinegar fly, Drosophila melanogaster, this occurs already at the periphery, at the level of olfactory sensory neurons (OSNs) located in the antenna. In our study, we investigate whether sensitization is a widespread property in a set of seven types of OSNs, as well as the mechanisms involved. First, we characterize and compare the differences in spontaneous activity, response velocity and response dynamics, among the selected OSN types. These express different receptors with distinct tuning properties and behavioral relevance. Second, we show that sensitization is not a general property. Among our selected OSN types, it occurs in those responding to more general food odors, while OSNs involved in very specific detection of highly specific ecological cues like pheromones and warning signals show no sensitization. Moreover, we show that mitochondria play an active role in sensitization by contributing to the increase in intracellular Ca2+ upon weak receptor activation. Thus, by using a combination of single sensillum recordings (SSRs), calcium imaging and pharmacology, we widen the understanding of how the olfactory signal is processed at the periphery.

Abstract:Significance Detecting and avoiding noxious stimuli is critical for survival. Nociceptive sensory neurons detect these stimuli and activate neural circuits that elicit stereotyped escape responses. However, individual animals may have genetic predispositions to respond differently to the same stimuli. Furthermore, it is not well understood how experiences during development affect these escape responses. Here, we describe a role for the foraging gene in nociception and show that differences in nociceptive sensitivity may arise from genetic variation of this gene. We also demonstrate that altering synaptic activity in a nociceptive circuit during development changes the sensitivity of the escape response elicited. Our findings suggest that both genetic variation and experiences in development can shape escape responses.

Abstract:Abstract Chemosensory tissues exhibit significant between-species variability, yet the evolution of gene expression and cell types underlying this diversity remain poorly understood. To address these questions, we conducted transcriptomic analyses of five chemosensory tissues from six Drosophila species and integrated the findings with single-cell datasets. While stabilizing selection predominantly shapes chemosensory transcriptomes, thousands of genes in each tissue have evolved expression differences. Genes that have changed expression in one tissue have often changed in multiple other tissues but at different past epochs and are more likely to be cell type-specific than unchanged genes. Notably, chemosensory-related genes have undergone widespread expression changes, with numerous species-specific gains/losses including novel chemoreceptors expression patterns. Sex differences are also pervasive, including a D. melanogaster- specific excess of male-biased expression in sensory and muscle cells in its forelegs. Together, our analyses provide new insights for understanding evolutionary changes in chemosensory tissues at both global and individual gene levels.

Abstract:Insects use olfaction to detect ecologically relevant chemicals in their environment. To maintain useful responses over a variety of stimuli, olfactory receptor neurons are desensitized to prolonged or high concentrations of stimuli. Depending on the timescale, the desensitization is classified as short-term, which typically spans a few seconds; or long-term, which spans from minutes to hours. Compared with the well-studied mechanisms of desensitization in vertebrate olfactory neurons, the mechanisms underlying invertebrate olfactory sensitivity regulation remain poorly understood. Recently, using a large-scale functional screen, a conserved critical receptor phosphorylation site has been identified in the model insect Drosophila melanogaster, providing new insight into the molecular basis of desensitization in insects. Here, we summarize the progress in this area and provide perspectives on future directions to determine the molecular mechanisms that orchestrate the desensitization in insect olfaction.
Human-driven evolution of color in a stonefly mimic

Abstract:Rapid adaptation is thought to be critical for the survival of species under global change, but our understanding of human-induced evolution in the wild remains limited. We show that widespread deforestation has underpinned repeated color shifts in wild insect populations. Specifically, loss of forest has led to color changes across lineages that mimic the warning coloration of a toxic forest stonefly. Predation experiments suggest that the relative fitness of color phenotypes varies between forested and deforested habitats. Genomic and coloration analyses of 1200 specimens show repeated selection at the ebony locus controlling color polymorphism across lineages. These findings represent an example of human-driven evolution linked to altered species interactions, highlighting the possibility for populations to adapt rapidly in the wake of sudden environmental change.

On body-environment continuities from a laboratory commensalism

Abstract:Social Studies of Science, Ahead of Print. The article attends to everyday practices in a laboratory of neural genetics that studies olfaction, with the fruit fly as its model organism. Practices in neural genetics exhibit one of the 'post' aspects in post-genomic science – a turn to the environment. To get at how laboratory members engage body-environment continuities, I pay attention to an occasion of designing experimental chambers for an optogenetics study. As practitioners deal with the body's continuities with the world by engaging the spatial character of olfaction, their accounts exhibit qualities of feelings of immediate experience, relatable to C.S. Peirce's phenomenological category of Firstness. While these traces of Firstness inevitably manifest themselves in mixtures with the other two of Peirce's categories – namely, Secondness and Thirdness – noticing them allows for an engagement of the environment that goes beyond action and meaning. I reflect on that environment by considering the involvement of scientists' bodies in life with flies, while not forgetting my inhabitation of the laboratory space. Rather than relying on a cross-mapping of attributes known from the human sphere (intentional states or features of the human body) while managing a measurable space observed from the outside, this is an environment lived from within and with others. I conclude the article by proposing its noticing as an orientation toward ecological preoccupations.

Abstract:The spotted wing Drosophila, Drosophila suzukii, has emerged within the past decade as an invasive species on a global scale, and is one of the most economically important pests in fruit and berry production in Europe and North America. Insect ecology, to a strong degree, depends on the chemosensory modalities of smell and taste. Extensive research on the sensory receptors of the olfactory and gustatory systems in Drosophila melanogaster provide an excellent frame of reference to better understand the fundamentals of the chemosensory systems of D. suzukii. This knowledge may enhance the development of semiochemicals for sustainable management of D. suzukii, which is urgently needed. Here, using a transcriptomic approach we report the chemosensory receptor expression profiles in D. suzukii female and male antennae, and for the first time, in larval heads including the dorsal organ that houses larval olfactory sensory neurons. In D. suzukii adults, we generally observed a lack of sexually dimorphic expression levels in male and female antennae. While there was generally conservation of antennal expression of odorant and ionotropic receptor orthologues for D. melanogaster and D. suzukii, gustatory receptors showed more distinct species-specific profiles. In larval head tissues, for all three receptor gene families, there was also a greater degree of species-specific gene expression patterns. Analysis of chemosensory receptor repertoires in the pest species, D. suzukii relative to those of the genetic model D. melanogaster enables comparative studies of the chemosensory, physiology, and ecology of D. suzukii.

Abstract:Navigating toward (or away from) a remote odor source is a challenging problem that requires integrating olfactory information with visual and mechanosensory cues. Drosophila melanogaster is a useful organism for studying the neural mechanisms of these navigation behaviors. There are a wealth of genetic tools in this organism, as well as a history of inventive behavioral experiments. There is also a large and growing literature in Drosophila on the neural coding of olfactory, visual, and mechanosensory stimuli. Here we review recent progress in understanding how these stimulus modalities are encoded in the Drosophila nervous system. We also discuss what strategies a fly might use to navigate in a natural olfactory landscape while making use of all these sources of sensory information. We emphasize that Drosophila are likely to switch between multiple strategies for olfactory navigation, depending on the availability of various sensory cues. Finally, we highlight future research directions that will be important in understanding the neural circuits that underlie these behaviors.

Abstract:Brain evolution has primarily been studied at the macroscopic level by comparing the relative size of homologous brain centers between species. How neuronal circuits change at the cellular level over evolutionary time remains largely unanswered. Here, using a phylogenetically informed framework, we compare the olfactory circuits of three closely related Drosophila species that differ in their chemical ecology: the generalists Drosophila melanogaster and Drosophila simulans and Drosophila sechellia that specializes on ripe noni fruit. We examine a central part of the olfactory circuit that, to our knowledge, has not been investigated in these species—the connections between projection neurons and the Kenyon cells of the mushroom body—and identify species-specific connectivity patterns. We found that neurons encoding food odors connect more frequently with Kenyon cells, giving rise to species-specific biases in connectivity. These species-specific connectivity differences reflect two distinct neuronal phenotypes: in the number of projection neurons or in the number of presynaptic boutons formed by individual projection neurons. Finally, behavioral analyses suggest that such increased connectivity enhances learning performance in an associative task. Our study shows how fine-grained aspects of connectivity architecture in an associative brain center can change during evolution to reflect the chemical ecology of a species.
