Abstract:For biomedical applications, mesoporous silica nanoparticles (MSNs)-based theranostic agents have shown to be a promising alternative. Rational design of particulate systems should consider, beside the physicochemical properties of particle size and surface chemistry, shape features as aspect ratio (AR) and morphology. Recent advances of fabrication technologies for manufacturing different shaped MSNs and evaluation means of its in vitro and in vivo biological performance provide new aspects and wisdom in nanomedicine development. In this review, we discussed the recent progress in the preparation of different shaped MSNs and the evaluation of shape-mediated biological effects. Firstly, we provide an overview of preparation strategies for fabricating MSNs with different aspect ratios and different morphologies, including hollow/rattle MSNs, multishell MSNs, and mesoporous silica nanocomposites. We then highlight the aspect ratio- and morphology-mediated biological effects of MSNs respectively. For AR-mediated biological effects of MSNs, we put our focus in the particle ARs effect on cellular uptake, biocompatibility, and drug delivery. For morphology-mediated biological effects of MSNs, we emphasize on how particle shapes could affect tumor therapy. Finally, for application considerations, we conclude with our personal perspectives on the directions in which future studies in this field might be placed.

Abstract:A nanoparticle's shape is a critical determinant of its biological interactions and therapeutic effectiveness. This study investigates the influence of shape on the performance of mesoporous silica nanoparticles (MSNs) in anticancer therapy. MSNs with spherical, rod-like, and hexagonal-plate-like shapes were synthesized, with particle sizes of around 240 nm, and their other surface properties were characterized. The drug loading capacities of the three shapes were controlled to be 47.46%, 49.41%, and 46.65%, respectively. The effects of shape on the release behaviors, cellular uptake mechanisms, and pharmacological behaviors of MSNs were systematically investigated. Through a series of in vitro studies using 4T1 cells and in vivo evaluations in 4T1 tumor-bearing mice, the release kinetics, cellular behaviors, pharmacological effects, circulation profiles, and therapeutic efficacy of MSNs were comprehensively assessed. Notably, hexagonal-plate-shaped MSNs loaded with PTX exhibited a prolonged circulation time (t1/2 = 13.59 ± 0.96 h), which was approximately 1.3 times that of spherical MSNs (t1/2 = 10.16 ± 0.38 h) and 1.5 times that of rod-shaped MSNs (t1/2 = 8.76 ± 1.37 h). This research underscores the significance of nanoparticles' shapes in dictating their biological interactions and therapeutic outcomes, providing valuable insights for the rational design of targeted drug delivery systems in cancer therapy.

Abstract:INTRODUCTION: Micro- and nanoparticles in drug and vaccine delivery have opened up new possibilities in pharmaceutics. In the past, researchers focused mainly on particle size, surface chemistry and the use of various materials to control particle characteristics and functions. Lately, shape has been acknowledged as an important design parameter having an impact on the interaction with biological systems.AREAS COVERED: In this review, we report on the latest developments in fabrication methods to tailor particle geometry, summarize analytical techniques for non-spherical particles and highlight the most important findings regarding their interaction with biological systems and their potential applications in drug delivery.EXPERT OPINION: The impact of shape on particle internalization into different cell types and particle biodistribution has been extensively studied in the past. Current research focuses on shape-dependent uptake mechanisms and applications for tumour therapy and vaccination. Different fabrication methods can be used to produce a variety of different particle types and shapes. Key challenges will be the transfer of new non-spherical particle fabrication methods from lab-scale to industrial large-scale production. Not all techniques may be scalable for the production of high quantities of particles. It will also be challenging to transfer the promising in vitro findings to suitable in vivo models.

Abstract:This review aims to highlight the importance of particle shape in the design of polymeric nanocarriers for drug delivery systems, along with their size, surface chemistry, density, and rigidity. Current manufacturing methods used to obtain non-spherical polymeric nanocarriers such as filomicelles or nanoworms, nanorods and nanodisks, are firstly described. Then, their interactions with biological barriers are presented, including how shape affects nanoparticle clearance, their biodistribution and targeting. Finally, their drug delivery properties and their therapeutic efficacy, both in vitro and in vivo, are discussed and compared with the characteristics of their spherical counterparts.

Abstract:This review is a comprehensive description of the past decade of research into understanding how the geometry and size of nanoparticles affect their interaction with biological systems: from single cells to whole organisms. Recently, there has been a great deal of effort to use both the shape and the size of nanoparticles to target specific cellular uptake mechanisms, biodistribution patterns, and pharmacokinetics. While the successes of spherical lipid-based nanoparticles have heralded marked changes in chemotherapy worldwide, the history of asbestos-induced lung disease casts a long shadow over fibrous materials to date. The impact of particle morphology is known to be intertwined with many physicochemical parameters, namely, size, elasticity, surface chemistry, and biopersistence. In this review, we first highlight some of the morphologies observed in nature as well as shapes available to us through synthetic strategies. Following this we discuss attempts to understand the cellular uptake of nanoparticles through various theoretical models before comparing this with observations from in vitro and in vivo experiments. In addition, we consider the impact of nanoparticle shape at different size regimes on targeting, cytotoxicity, and cellular mechanics.

Abstract:In the past decade, mesoporous silica nanoparticles (MSNs) have attracted more and more attention for their potential biomedical applications. With their tailored mesoporous structure and high surface area, MSNs as drug delivery systems (DDSs) show significant advantages over traditional drug nanocarriers. In this review, we overview the recent progress in the synthesis of MSNs for drug delivery applications. First, we provide an overview of synthesis strategies for fabricating ordered MSNs and hollow/rattle-type MSNs. Then, the in vitro and in vivo biocompatibility and biotranslocation of MSNs are discussed in relation to their chemophysical properties including particle size, surface properties, shape, and structure. The review also highlights the significant achievements in drug delivery using mesoporous silica nanoparticles and their multifunctional counterparts as drug carriers. In particular, the biological barriers for nano-based targeted cancer therapy and MSN-based targeting strategies are discussed. We conclude with our personal perspectives on the directions in which future work in this field might be focused.

Abstract:Morphology control is one of the most interesting issues in the research field of mesoporous materials. It plays a very important role in understanding the basic synthesis mechanism of the kind of materials. Moreover, the future applications of mesoporous materials are strongly depended on their tunable macroscopical morphologies. Mesoporous materials with spherical morphology are quite attractive due to the potential applications in macromolecular separations, drug deliveries, catalysis supports and template agents for photonic crystals. Researches on synthesis and properties of the materials have been carried out for nearly ten years. In this article, several synthetic methods for both siliceous and non-siliceous mesoporous spheres are reviewed, including solution self-assembly, aerosol spraying and hard templating route. The mechanism of self-assembly in solution system is provided and the rational controls for synthetic conditions are specifically discussed. With the viewpoint of applications, some examples are given in the aspects of bioseparations, drug delivery and catalysis. A short summary has listed some overviews for future developments.
The shape effect of PEGylated mesoporous silica nanoparticles on cellular uptake pathway in Hela cells

Abstract:The shape of nanoparticles plays an important role in the interaction between cells and particles. However, the shape effect of mesoporous silica nanoparticles on the cellular uptake pathway and mechanism has not been reported. Herein, three different kinds of fluorescent mesoporous silica nanoparticles (FMSN, aspect ratio=1, 2, and 4, ca. 100nm in diameter) with similar fluorescent intensity at equivalent concentration were fabricated by a co-condensation strategy. In simulated body fluid (SBF), the PEGylated FMSN (FMSN-PEG) have higher dispersity and stability than the naked counterparts. So we chose the FMSN-PEG to research of shape effect on the cellular uptake pathway in Hela cells. We found that the uptake kinetics and pathway of three different shaped FMSN-PEG were obviously shape-dependent. The long-rod FMSN-PEG (NLR-PEG) showed higher intracellular retention amount than the short-rod FMSN-PEG (NSR-PEG) and the sphere FMSN-PEG (NS-PEG) almost over 8h. The NSR-PEG showed the lowest intracellular amount especially with prolonged incubation time. We also found that the cellular entry pathway of NS-PEG, NSR-PEG, and NLR-PEG into Hela cells was regulated by particle shape. Spherical particles preferred to be internalized via the clathrin-mediated pathway, whereas MSN with larger aspect ratios (ARs) favored to be internalized via caveolae-mediated pathway, which could explain their different uptake kinetics. Our findings may provide useful information for optimizing the nano-based drug delivery and bio-imaging systems.

Abstract:To date, how the shape of nanomaterials influences their biological properties is poorly understood, due to the insufficient controllability of current preparative methods, especially in the shape and size of nanomaterials. In this paper, we achieved the precise syntheses of nanoscale unimolecular cylindrical polymer brushes (CPBs) and spherical polymer nanoparticles (SPNPs) with the same volume and surface chemistry, which ensured that shape was essentially the only variable when their biological performance was compared. Accurate shape effects were obtained. Impressively, the CPBs had remarkable advantage in tissue penetration over the SPNPs. The CPBs also exhibited higher cellular uptake and rapider body clearance than the SPNPs, whereas the SPNPs had longer blood circulation time, rapider tumor vascular extravasation, and higher tumor accumulation than the CPBs. Additionally, this work also provided a controllable synthesis strategy for nanoscale unimolecular SPNPs by integrating 21 CPBs to a β-cyclodextrin core, whose diameter in dry state could be up to 45 nm.

Abstract:<p>Mesoporous silica nanoparticles (MSNs), one of the important porous materials, have garnered interest owing to their highly attractive physicochemical features and advantageous morphological attributes. They are of particular importance for use in diverse fields including, but not limited to, adsorption, catalysis, and medicine. Despite their intrinsic stable siliceous frameworks, excellent mechanical strength, and optimal morphological attributes, pristine MSNs suffer from poor drug loading efficiency, as well as compatibility and degradability issues for therapeutic, diagnostic, and tissue engineering purposes. Collectively, the desirable and beneficial properties of MSNs have been harnessed by modifying the surface of the siliceous frameworks through incorporating supramolecular assemblies and various metal species, and through incorporating supramolecular assemblies and various metal species and their conjugates. Substantial advancements of these innovative colloidal inorganic nanocontainers drive researchers in promoting them toward innovative applications like stimuli (light/ultrasound/magnetic)‐responsive delivery‐associated therapies with exceptional performance in vivo. Here, a brief overview of the fabrication of siliceous frameworks, along with discussions on the significant advances in engineering of MSNs, is provided. The scope of the advancement in terms of structural and physicochemical attributes and their effects on biomedical applications with a particular focus on recent studies is emphasized. Abstract:Nanotechnology has significantly advanced various fields, including therapeutic delivery, through the use of nanomaterials as drug carriers. The biocompatibility of ordered porous silica materials makes them promising candidates for drug delivery systems, particularly in the treatment of cancer and other diseases. This review summarizes the use of microporous zeolites and mesoporous silica materials in drug delivery, focusing on their physicochemical properties and applications as drug carriers. Special emphasis is placed on strategies for encapsulation and functionalization, highlighting their role in enhancing drug loading and enabling targeted delivery. In conclusion, while ordered porous silica materials hold great potential for drug delivery systems, certain challenges remain.

Abstract:Over the last few years, research on silica nanoparticles has rapidly increased. Particularly on mesoporous silica nanoparticles (MSNs), as nanocarriers for the treatment of various diseases because of their physicochemical properties and biocompatibility. The use of MSNs combined with therapeutic agents can provide better encapsulation and effective delivery. MSNs as nanocarriers might also be a promising tool to lower the therapeutic dosage levels and thereby to reduce undesired side effects. Researchers have explored several routes to conjugate both imaging and therapeutic agents onto MSNs, thus expanding their potential as theranostic platforms, in order to allow for the early diagnosis and treatment of diseases. This review introduces a general overview of recent advances in the field of silica nanoparticles. In particular, the review tackles the fundamental aspects of silicate materials, including a historical presentation to new silicates and then focusing on the key parameters that govern the tailored synthesis of functional MSNs. Finally, the biomedical applications of MSNs are briefly revised, along with their biocompatibility, biodistribution and degradation. This review aims to provide the reader with the tools for a rational design of biocompatible MSNs for their application in the biomedical field. Particular attention is paid to the role that the synthesis conditions have on the physicochemical properties of the resulting MSNs, which, in turn, will determine their pharmacological behavior. Several recent examples are highlighted to stress the potential that MSNs hold as drug delivery systems, for biomedical imaging, as vaccine adjuvants and as theragnostic agents.

Abstract:Natural world provides abundant sources of inspiration for the biomimetic engineering. Morphology of organisms, as a result of long-term natural selection, can offer particular functional merits, such as the spiky seeds of Xanthium strumarium that helps them to spread by enhancing affinity to the passing animals. Such strategies have been inspiring in many fields, among which is the construction of novel nanostructures for drug delivery and diagnostic purposes. Given the increasing number of recent reports yet the very limited reviewing work, we give a comprehensive review on the tailored syntheses and applications of biomaterial particles that mimic natural biological entities in morphology. Emphasis has been put on spiky and red blood cell (RBC)-shaped particles due to their unique properties and the wide research interest they have drawn. The importance of computational simulation for rational biomimetic design is also exemplified. Shaping particles by learning from biotic entities hold great potential of benefiting human health by offer performances that cannot be achieved using otherwise shaped products. This review is expected to offer basic rationales for the future biomimetic engineering.

Abstract:Nanoscale amorphous materials are very important member of the non-crystalline solids family and have emerged as a new category of advanced materials. However, morphological control of amorphous nanomaterials is very difficult because of the atomic isotropy of their internal structures. In this review, we introduce some emerging innovative methods to fabricate well-defined, regular-shaped amorphous nanomaterials. We then highlight some examples to evaluate the use of these amorphous materials in electrodes, and their optical response. There is still plenty of room to explore the amorphous world. As researchers continue to advance the scientific tools that underpin the concepts related to “amorphous”, additional applications of these materials will emerge. Their controlled synthesis will undoubtedly attain new heights in the discipline of nanomaterials, and allow nanoscale amorphous materials to become more sophisticated, diverse, and mainstream.

Abstract:The outstanding journey towards the investigation of mesoporous materials commences with the discovery of high surface area porous silica materials, named MCM-41 (Mobil Composition of Matter-41) according to the inventors’ name Mobile scientists in the United States. Based on a self-assembled supramolecular templating mechanism, the synthesis of mesoporous silica has extended to wide varieties of silica categories along with versatile applications of all these types in many fields. These silica families have some extraordinary structural features, like highly tunable nanoscale sized pore diameter, good Brunauer–Emmett–Teller (BET) surface areas, good flexibility to accommodate different organic and inorganic functional groups, metals etc., onto their surface. As a consequence, thousands of scientists and researchers throughout the world have reported numerous silica materials in the form of published articles, communication, reviews, etc. Beside this, attention is also given to the morphology-oriented synthesis of silica nanoparticles and their significant effects on the emerging fields of study like catalysis, energy applications, sensing, environmental, and biomedical research. This review highlights a consolidated overview of those morphology-based mesoporous silica particles, emphasizing their syntheses and potential role in many promising fields of research.

Abstract:Nano-sized metal particles exhibit different thermal, electronic, magnetic and optical properties from those of bulk ones because of quantum size effect and so on. The properties of nanoparticles depend on not only the size but also the shape. Specially shaped nanoparticles generally have particular surface structures and properties. In recent years, shape-controlled synthesis of nanoparticles has been attracting more and more attention. The progress has been made for the study on subgroup VIII and IB metals. In this review, the syntheses of metal nanoparticles and methods of controlling the size and shape are introduced. Also, the recent investigations on the shape-controlled syntheses of nanoparticles are reported for Pt, Pd, Ni, Au, Ag, Cu and Co, respectively.

In this review, we highlight the recent advances in silica-assisted drug delivery systems, including (1) MSN-based immediate/sustained drug delivery systems and (2) MSN-based controlled/targeted drug delivery systems. Abstract:Drug delivery systems rely heavily on nanoparticles because they provide a targeted and monitored release of pharmaceuticals that maximize therapeutic efficacy and minimize side effects. To maximize drug internalization, this review focuses on comprehending the interactions between biological systems and nanoparticles. The way that nanoparticles behave during cellular uptake, distribution, and retention in the body is determined by their shape. Different forms, such as mesoporous silica nanoparticles, micelles, and nanorods, each have special properties that influence how well drugs are delivered to cells and internalized. To achieve the desired particle morphology, shape-controlled nanoparticle synthesis strategies take into account variables like pH, temperatures, and reaction time. Abstract:Nanocrystals are fundamental to modern science and technology. Mastery over the shape of a nanocrystal enables control of its properties and enhancement of its usefulness for a given application. Our aim is to present a comprehensive review of current research activities that center on the shape-controlled synthesis of metal nanocrystals. We begin with a brief introduction to nucleation and growth within the context of metal nanocrystal synthesis, followed by a discussion of the possible shapes that a metal nanocrystal might take under different conditions. We then focus on a variety of experimental parameters that have been explored to manipulate the nucleation and growth of metal nanocrystals in solution-phase syntheses in an effort to generate specific shapes. We then elaborate on these approaches by selecting examples in which there is already reasonable understanding for the observed shape control or at least the protocols have proven to be reproducible and controllable. Finally, we highlight a number of applications that have been enabled and/or enhanced by the shape-controlled synthesis of metal nanocrystals. We conclude this article with personal perspectives on the directions toward which future research in this field might take.
