Abstract:OBJECTIVE: To investigate whether the change of embryonic growth factors might explain the embryotoxic effect of peritoneal fluid of infertile women with mild endometriosis (PF-E) on preimplantation embryos.DESIGN: Observe the effect of PF-E on the development of preimplantation mouse embryos in vitro and measure the level of embryonic EGF and IGF-I in the culture medium.MATERIALS AND METHODS: Peritoneal fluids (PF) were obtained from fertile women without endometriosis (PF-NE) and infertile women with mild endometriosis (PF-E). Mouse two-cell-stage embryos were cultured in blank (HTF medium), control (HTF medium with 10% or 30%PF-NE) or treatment (HTF media with 10% or 30% PF-E) groups. Embryo development was observed and cleavage and blastulation rates were assessed (Table 1). EGF and IGF-I levels in culture media were analyzed by ELISA (Table 2).RESULTS: [1] Cleavage and blastulation rates of 10% PF-E, 30% PF-E and 30%PF-NE groups were all significantly lower than HTF group.The rates became lower as PF concentration growing higher in both PF-NE and PF-E groups.At same PF concentration,embryo development of PF-E groups was worse than PF-NE groups. [2] EGF levels of 10%PF-E, 30%PF-NE and 30%PF-E group were all significantly higher than HTF group (p<0.01).The levels of EGF between 10% and 30%PF-NE group,between 10% and 30%PF-E group were statistically different (p<0.01).
