Abstract:Fipronil, a wildly used pesticide, exhibits high toxicity to pollinators, aquatic organisms, farmland birds, as well as mammals that have GABA receptors, including humans. Thus, developing an effective method for fipronil determination is of great importance. Although the fluorescence assays have the features of easy-to-use, rapid response, high sensitivity and visualized readout, such assay for fipronil was rather difficult to design because fipronil was both chemically and photophysically stable. Herein, we utilized a chalcone dye bound into albumin inner-cavity to construct a novel supramolecular recognition system, for the first time, successfully realizing ratiometric fluorescence detection of fipronil. The sensing mechanism has been carefully verified by the molecular docking, fluorescent titration, DLS tests and fluorescence lifetime. This reported probe could detect fipronil with a rapid response (< 3 min), high sensitivity (LOD similar to 0.01 ppm) and selectivity, good anti-interference property, and significantly, ratiometric color change from green to yellow, which could be discriminated by naked eyes. Moreover, we demonstrated the application of this method in the ratiometric tracking of fipronil in plants (Arabidopsis thaliana). We anticipate this albumin-based supramolecular recognition system would inspire more probe design for environmental monitoring.
Label-Free Aptasensor for Detection of Fipronil Based on Black Phosphorus Nanosheets

Abstract:A label-free fipronil aptasensor was built based on Polylysine-black phosphorus nanosheets composition (PLL-BPNSs) and Au nanoparticles (AuNPs). A PLL-BP modified glassy carbon electrode (GCE) was fabricated by combining BP NSs and PLL, which included a considerable quantity of -NH2. Au nanoparticles (AuNPs) were placed onto the GCE, and PLL-BPNSs bonded to Au NPs firmly by assembling. The thiolated primers were then added and fixed using an S-Au bond, and competitive binding of the fipronil aptamer was utilized for fipronil quantitative assessment. The sensor's performance was evaluated using differential pulse voltammetry (DPV) method. The linear equation is ΔI (μA) = 13.04 logC + 22.35, while linear correlation coefficient R2 is 0.998, and detection limit is 74 pg/mL (0.17 nM) when the concentration of fipronil is 0.1 ng/mL-10 μg/mL. This aptasensor can apply to quantitative detection of fipronil.

Abstract:A fluorometric method is described for the determination of fipronil, a frequently-used insecticide. It exploits the blue fluorescence of carbon quantum dots (CQDs) and the selectivity of molecularly imprinted silica (MIS). The MIS was prepared via the sol-gel method by using fipronil as the template, 3-aminopropyltriethoxysilane as functional monomer, and tetraethoxysilane as cross-linker in the presence of CQDs. The blue fluorescence of the CQD@MIS, with excitation/emission peaks at 340/422 nm, is quenched by fipronil. The assay works in the 0. 70 pM to 47 μM fipronil concentration range, and the limit of detection is 19 pM. The method was successfully applied to the quantitation of fipronil in spiked eggs, milk, and tap water. Recoveries between 83.8 and 114.0% were achieved. The corresponding relative standard deviations (RSD) are less than 6.67%.Schematic representation of a high sensitivite and selectivite fluorescence nanoprobe constructed by combining the excellent fluorescence property of carbon quantum dots and the predicted selectivity of molecularly imprinted silica. It was applied to analyze fipronil in egg, milk and tap water, respectively.

Abstract:Fipronil, as an excellent insecticide, is widely used around the world, but its residues in agricultural products and the environment cause a series of health and ecological risks. In this study, a novel aptamer sensor for the sensitive, selective, and quantitative detection of fipronil in eggs was prepared using nitrogen-doped carbon quantum dots (NCQDs) modified by fipronil aptamer. It is based on the adsorption of apt-NCQDs on the surface of graphene oxide (GO) through π-stacking and hydrophobic interactions, and quenching the fluorescence of apt-NCQDs through the fluorescence resonance energy transfer between NCQDs and GO. In the presence of fipronil, apt-NCQDs were desorbed from GO and the fluorescence resonance energy transfer (FRET) process was inhibited to recover the fluorescence. Under the optimized conditions, the linear range of fipronil was 10–60 nM, R 2 = 0.9907, and the LOD was 3.58 nM, which showed good selectivity and sensitivity to fipronil. In addition, the recovery of this method for detection in egg samples ranged 89–104% with an RSD < 8%, which indicates that this method has good prospects for practical application.

Abstract:Fipronil (FP) is a very effective phenylpyrazole insecticide and is now widely used in agriculture. At the same time, the water and soil in the environment are polluted by FP. For the rapid detection of FP toxicants in food and the environment, we have designed an entirely novel electrochemical immunosensor that employs the combined functionalities of a cMWCNTs-AgNPs-CS-FAb-BSA layer to modify an SPCE by the freeze-drying technique. The high porosity of chitosan (CS) coupled with an excellent electron transfer enabled by the cMWCNTs and AgNPs increased the surface area for anti-fipronil (FAb) antibody immobilization and enhanced the current signal of the immunosensor. Cyclic voltammetry (CV) was applied for the quantitative determination of FP under optimized conditions (0.1 M PBS, pH 7.5, 35 °C incubation temperature, and 40 min incubation duration). The modified electrochemical immunosensor displayed excellent analytical performance, including a wide linear concentration range from 0.1 to 1000 ng mL −1 with a very low limit of detection of 0.021 ng mL −1 and good reproducibility (RSD = 2.58%, n = 6), stability (80.4% sensitivity after 5 days), and selectivity. Not only could the modified electrochemical immunosensor be applied in the FP residue analysis of agricultural products, but the present immobilization strategy can also potentially be applied to different biomolecules.

Abstract:Accurate monitoring of trace pesticides in complex matrix remains a challenge in food safety supervision. Herein, we designed a facile zeolitic imidazolate framework (ZIF)-8/aptamer-based assay for the sensitive detection of acetamiprid. ZIF-8 efficiently adsorbs 6-carboxyfluorescein-labeled complementary DNA (cDNA-FAM) via electrostatic interaction, hydrogen bonding and Zn 2+ coordination, which contributed to resistance to cDNA-FAM displacement by biological ligands. ZIF-8 serves as an "ion pump" that contains lots of Zn 2+ who boosts cDNA-FAM adsorption and triggers the photoinduced electron transfer (PET) effect from FAM to ZIF-8, improving the sensing sensitivity. Acetamiprid could trigger the change in the adsorption state of cDNA-FAM, further tuning the PET effect and causing fluorescence conversion. The fluorescence assay showed a high sensitivity for monitoring acetamiprid with a detection limit of 0.05 ng mL −1 in the apple sample. This ZIF/DNA-based analytical platform provides a powerful tool for facile and low-cost screening of pesticide residues, with promising applications in food safety monitoring.

Abstract:The N and Fe doped carbon dot (CD NFe ) was prepared by microwave procedure. Using CD NFe as the nano-substrate, fipronil (FL) as the template molecule and α-methacrylic acid as the functional monomer, the molecular imprinted polymethacrylic acid nanoprobe (CD NFe @MIP) with difunction was synthesized by microwave procedure. The CD NFe @MIP was characterized by transmission electron microscopy, X-ray photoelectron spectroscopy, Fourier infrared spectroscopy, and other techniques. The results show that the nanoprobe not only distinguish FL but also has a strong catalytic effect on the HAuCl 4 –Na 2 C 2 O 4 nanogold indicator reaction. When the nanoprobes specifically recognize FL, their catalytic effect is significantly reduced. Since the AuNPs generated by HAuCl 4 reduction have strong surface-enhanced Raman scattering (SERS) and resonance Rayleigh scattering (RRS) effects, a SERS/RRS dual-mode sensing platform for detecting 5–500 ng/L FL was constructed. The new analytical method was applied to detect FL in food samples with a relative standard deviation (RSD) of 3.3–8.1 % and a recovery rate of 94.6–104.5 %.

Abstract:A novel fluorescent probe NIPF was synthesized by the Suzuki reaction to recognize Cu2+ and CN-. With the addition of Cu2+, NIPF exhibited strong fluorescence quenching (90 % for NIPF) with a Ksv value of 3.4 × 106 M-1 and a detection limit of 9.04 × 10-10 M. Subsequently, CN- was added to the NIPF-Cu2+ solution, and [Cu(CN)x]n- was formed due to the strong interaction between Cu2+ and CN- leading to fluorescence recovery (89 % for NIPF-Cu2+). In addition, a detection limit of 3.6 × 10-8 M was obtained by fluorescence titration. Meanwhile, it was demonstrated that the sensor achieved 93 %-105 % recovery of Cu2+ in the tested environmental samples, and the practicability of Cu2+ and CN- detection were verified using hydrogels test, with significant color changes observed under 365 nm light. Accordingly, the fluorescent probe NIPF was used to recognize Cu2+ and CN- by the "on-off-on" sensors.

Abstract:Fipronil, an insecticide widely employed, poses risks to both the environment and human health. Existing detection methods for fipronil and its metabolites, such as GC–MS, are known; however, these methods are laborious and time-intensive. In this investigation, our aim was to devise a label-free aptasensor strategy for detecting fipronil in food-related scenarios. To achieve this objective, we developed ellipsometric-based aptasensors specifically tailored for fipronil detection. The advantage of ellipsometric sensors lies in their efficacy in analyzing thin and uniform films. Spectrophotometric ellipsometry (SE), utilized in this realm, is preferred due to its sensitivity to molecular accumulation at the substrate surface, as evidenced by the phase shift (Δ). The limit of detection (LOD) observed in the buffer solution for the two aptamers, Anti-FIP1 and Anti-FIP2, used in this study, with lengths of 80 and 73 bases respectively, showed promising results. Additionally, the specificity of these aptamers against bovine serum albumin and ethiprole in the SE sensor approached the reproducibility limit. This is attributed to the wide-ranging cocktail employed during the aptamer selection process. The LOD for Anti-FIP1 was 0.034 nM (33.7 pM), while for Anti-FIP2 it was 0.025 nM (24.9 pM). The limit of quantification (LOQ) was calculated as 0.1 nM (0.044 ng/mL) for Anti-FIP1 and 0.075 nM (0.033 ng/mL) for Anti-FIP2. Utilizing the spike method for fipronil detection by introducing egg and grain samples, the deviation observed was less than ±10 % for recovery. This study presents a dependable aptasensor platform for the testing of fipronil in food samples, employing aptamer-based SE.

Abstract:A new fluorescent sensor for the detection of CN− was developed based on the conjugation of phenothiazine fluorophore and benzofuran unit. By the nucleophilic attacking of CN− to the fluoroacetylamino group in the sensor, the additional reaction of CN− and carbonyl group induced the ICT (intramolecular charge transfer) effect in the molecule and caused the fluorescence quenching sensor. The titration experiments show that the sensor has good sensitivity, selectivity and quick response for CN−. In addition, the fluorescent detection of CN− in the living cell and zebrafish experiments demonstrated the value of the sensor in tracing the CN− in biological systems.

Abstract:A novel fluorogenic receptor 1 was prepared by the reaction of thiosemicarbazide with 1-naphthaldehyde. Based on in situ-formed 1–Fe(III) complexes having the specific binding affinity for F− anions, this sensory system allows rapid recognition and quantitative detection of fluoride in neutral aqueous media in an ‘off–on’ fashion. The fluoride measurement method not only exhibits a low detection limit but also has strong anti-interference ability to common coexisting ions, as evidenced by competitive experiments.

Abstract:Fenitrothion (FN) residue in food is a serious threat to public health. Consequently, a sensitive, cost-effective, and convenient immunoassay for FN urgently needs to be fabricated to safeguard human health. Herein, a nanobody-alkaline phosphatase fusion protein (Nb-ALP)-based fluorescent ELISA using red emissive carbon dots (r-CDs) anchored cobalt oxyhydroxide nanosheet (CoOOH NS) composite was developed for detecting FN. Briefly, a Nb-ALP was obtained by autoinduction expression and employed as a recognition, signal transduction, and amplification element. As the fluorescence signal source, r-CDs were assembled with CoOOH NS to yield the NS composite, leading to the fluorescence quenching of r-CDs via Förster resonance energy transfer (FRET). After competitive immunoreaction, the Nb-ALP bounded to the immobilized antigen can mediate the production of ascorbic acid, which can reduce the CoOOH NS to Co 2+ , breaking the FRET between r-CDs and CoOOH NS, accompanied by the fluorescence recovery of r-CDs. This fluorescent ELISA is highly sensitive to FN with a detection limit of 0.14 ng mL −1 , which is 25-fold lower than that of conventional colorimetric ELISAs. The recovery test of food samples and the validation by GC−MS/MS further demonstrated the proposed assay was an ideal tool for detecting FN.

Abstract:Development of a high-performance electrochemical aptasensor that can detect fipronil in fruits and vegetables is important for human health improvement. This paper presents a method for synthesising Rubus Corchorifolius –like gold (RC-Au) nano-crystals using a type of 21 peptide as a shape inducer. The synthesis process involves the concurrent growth of nano-crystals and their asymmetric development, resulting in the formation of RC-Au nano-particles with increased exposure of their high-index facets, which enhances their catalytic activity. The RC-Au nano-crystals were then combined with an aptamer/auxiliary DNA hybrid to fabricate an electrochemical aptasensor that can sense fipronil via the release of the free auxiliary DNA strand, triggering target-induced DNA cycling. One fipronil molecule was found to transfer many RC-Au–thionine probes to the aptasensor surface, with the resulting oxidation and reduction of the probes leading to signal amplification. Increasing the concentration of fipronil from 1 × 10 −17 to 1 × 10 −12 M led to a linear increase in the amperometric current. A limit of detection of 3.7 × 10 −18 M (S/N = 3) was obtained. The developed sensor exhibits higher sensitivity than other electrochemical fipronil sensors and was satisfactorily used to detect fipronil in fruits and vegetables.

A "Turn-On" FRET Aptasensor Based on the Metal-Organic Framework-Derived Porous Carbon and Silver Nanoclusters for Zearalenone Determination

Abstract:Zearalenone (ZEN) is a fungal secondary metabolite with an estrogen-like structure. Its widespread existence and potential hazards to human and animal health call for alternative rapid determination methods to monitor ZEN in food and the environment. Herein, a robust and enzyme-free "turn-on" aptasensor based on fluorescence resonance energy transfer (FRET) was developed for the sensitive identification of ZEN. Aptamer-modified silver nanoclusters (AgNCs) and porous Fe3O4/carbon octahedra derived from the metal-organic framework (MOF) acted as the FRET energy donor-acceptor pair in fluorescence quenching. DNA-templated AgNCs can be adsorbed onto the surface of porous Fe3O4/carbon octahedra through pi-pi stacking, which will lead to the FL quenching of AgNCs. However, in the presence of the ZEN, the formation of the ZEN/aptamer complex promoted the release of AgNCs and thereby brought about the recovery of fluorescence, which can be used to indicate the concentration of ZEN. The prepared aptasensor was characterized by various physicochemical techniques such as scanning electron microscopy (SEM), transmission electron microscopy (TEM), powder X-ray diffraction (PXRD), energy-dispersive X-ray spectrometry (EDX), X-ray photoelectron spectroscopy (XPS) and circular dichroism spectroscopy (CD). By combining the excellent quenching properties and easy separation of magnetic porous Fe3O4/carbon octahedra, as well as the appealing photostability and water solubility of AgNCs, the proposed FRET platform demonstrated superior analytical performance with a linear range of 0.01-250 ng mL(-1) and a LOD as low as 2 x 10(-3) ng mL(-1). Most importantly, the practicability and reliability of the FRET-based strategy were confirmed by conducting the determination of ZEN in real samples.

Abstract:Fumonisin (FB) is a pervasive hazardous substance in the environment, presenting significant threats to human health and ecological systems. Thus, the selective and sensitive detection of fumonisin B1 (FB1) is crucial due to its high toxicity and wide distribution in corn, oats, and related products. In this work, we developed a novel and versatile fluorescent aptasensor by combining enzyme-assisted dual recycling amplification with 2D δ-FeOOH-NH 2 nanosheets for the determination of FB1. The established CRISPR/Cas12a system was activated by using activator DNA (aDNA), which was released via a T7 exonuclease-assisted recycling reaction. Additionally, the activated Cas12a protein was utilized for non-specifically cleavage of the FAM-labeled single-stranded DNA (ssDNA-FAM) anchored on δ-FeOOH-NH 2 nanosheets. The pre-quenched fluorescence signal was restored due to the desorption of the cleaved ssDNA-FAM. Due to the utilization of this T7 exonuclease-Cas12a-δ-FeOOH-NH 2 aptasensor for signal amplification, the detection range of FB1 was expanded from 1 pg/mL to 100 ng/mL, with a limit of detection (LOD) as low as 0.45 pg/mL. This study not only provides novel insights into the development of fluorescence biosensors based on 2D nanomaterials combined with CRISPR/Cas12a, but also exhibits remarkable applicability in detecting other significant targets.

Although reported assays showed high sensitivity and selectivity to cyfluthrin, these intensity-based outputs suffered from the interference from a variety of analyte-independent factors, i.e. the instrumental parameters, which often led to the poor accuracy and repeatability of detection results. In this work, we reported a novel host-guest supramolecular probe by using albumin as the host and solvatofluorochromic flavonoid as the guest, for the first time, successfully realizing ratiometric determination of cyfluthrin. This probe was capable of detecting cyfluthrin with the fast response (10 s), high sensitivity (LOD similar to 70 ppb) and selectivity, good anti-interference performance, and remarkably distinct emission color change from orange to green. Moreover, the portable paper-based test strips could be facilely obtained and utilized for ratiometric determination cyfluthrin pesticide in food samples, such as apple, tomato, cucumber. The signal outputs could be captured and analyzed by using a smartphone, demonstrating the application potential of this probe in on-site food analysis. We expected this work may open up a novel avenue for on-site determination of pesticides with the simple testing apparatuses.

Abstract:The monitoring and precise determination of pesticides and pharmaceutical drugs and their residues have become increasingly important in the field of food safety and water contamination issues. Herein, a fluorescent aluminium MOF sensor (1) was developed for the selective recognition of neonicotinoid insecticide dinotefuran and anti‐Parkinson's drug entacapone. Guest‐free MOF 1′ exhibited ultra‐fast response (< 5 s) and ultra‐low detection limits of 2.3 and 7.6 nM for dinotefuran and entacapone, which are lower than the previously reported MOF sensors. In the presence of other competitive analytes, great selectivity was achieved towards both analytes. The probe was recyclable up to five cycles. The sensing ability was explored towards entacapone in human serum, urine and dinotefuran in real soil, rice, honey samples, different fruits, vegetables, real water specimens and a wide range of pH media. A low‐cost, handy MOF‐based polymer thin‐film composite (1′@PVDF‐PVP) was developed for the on‐site detection of dinotefuran and entacapone. Mechanistic studies involving analytical techniques and theoretical calculations suggested that FRET and PET are the probable reasons for entacapone sensing whereas IFE is responsible for dinotefuran detection. The entire work presents a low cost, multi‐use photoluminescent sensor of entacapone and dinotefuran to address the environmental pollution.

Abstract:Pesticide residues have seriously threatened human health and affected the sustainable development of the environment. It is pivotal to establish an efficient detection platform for pesticide assays. A novel ratiometric fluorescent aptasensor for sensitive determination of pesticide has been successfully established. Carbon dots (CDs) are used as the reference emitter, while the T-rich single-stranded DNA templated copper nanoparticles (CuNPs) provide the increasing signal. More precisely, CDs/magnetic beads (MBs) combined with split aptamer are used in this detection platform to present a label-free and enzyme-free isocarbophos (ICP) detection with high sensitivity and specificity. In the presence of ICP, a sandwich-typed complex is formed which attaches a T-rich DNA. After magnetic separation and addition of Cu2+/ascorbic acid (AA), the CuNPs are generated and the strong red luminescent appears. Without ICP, just blue emission from CDs exits. The developed ratiometric aptasensor offers an excellent ICP detection in the linear range of 3–200 nM with a detection limit of 0.81 nM. It provides new ways for the assay of pesticides in agricultural products or environmental samples.
