Abstract:OBJECTIVE:To improve the methods of isolating single ventricular myocytes for purposes of different experiments.METHODS:The cardiac ventricular myocytes were isolated enzymatically by Langendorff perfusion technique at constant flow rate. The patch clamp whole-cell recording, a spectrofluorometric method and a video tracking system were used to verify the basic electrophysiological properties, intracellular calcium transient and cell contraction of the single myocytes.RESULTS:The resting membrance potential of the myocyte was (-74.06+/-4.54)mV. The whole-cell sodium, potassium and calcium currents, intracellular calcium transients and contractile properties were stable and typical.CONCLUSION:The myocytes obtained by adjusting perfusion parameters are well suited for electrophysiological recording, intracellular calcium and single cell contraction measurements.

Abstract:Unintended inhibition of the cardiac potassium channel human ether-a-go-go-related gene (hERG) is considered the main culprit in drug-induced arrhythmias known as torsades de pointes. Electrophysiology is the most reliable in vitro screening method for identifying potential cardiac hERG liabilities, but only the recent advent of planar electrode-based voltage clamp electrophysiology promises sufficient throughput to support the drug testing needs of most drug discovery programs. We have assessed the reliability of this new format of the voltage clamp technology in measuring the activity of small molecules on the hERG channel. Based on the results herein of a screening against a panel of well-characterized hERG-active and -inactive molecules, we demonstrate that planar electrode electrophysiology, utilizing the Sealchip and PatchXpress technology platform (AVIVA Biosciences Corp., San Diego, CA), is comparable to traditional electrophysiology based on glass micropipettes in its reliability and data content. The new technology will allow significantly higher throughput and more thorough testing of pharmaceutical compounds.
Voltage-clamp-controlled current-clamp recordings from neurons: an electrophysiological technique enabling the detection of fast potential changes at preset holding potentials

Abstract:Investigations of the properties of fast, transient potential changes (e.g. receptor potentials or synaptic potentials) in excitable cells by means of current-clamp recording techniques require the exact adjustment and control of membrane potentials. Usually, the desired membrane potential values are set by current injection via the recording electrode and are controlled manually by regulating the current strength necessary to maintain a constant potential. However, this technique is associated with a number of disadvantages. A single-electrode current- and voltage-clamp amplifier was therefore modified to compensate for slow membrane potential changes without affecting faster voltage responses. Basically, low-pass filters with selectable time constants were incorporated into the voltage-clamp feedback circuit to control the amplifier's response speed. In addition, the amplifier's electronic circuits were altered to enable current pulse injection into the cells. Thus, while recording at preset and controlled membrane potentials, it was possible to monitor the cell's input resistance or current/voltage relationship. This new recording technique has been designated "voltage-clamp-controlled current clamp" (VCcCC) and its performance was tested by intracellular recordings from neocortical and neostriatal neurons in vitro using either conventional microelectrodes or patch-clamp electrodes.
Multiple conductance states of lipid pores during Voltage-Clamp electroporation

Abstract:We used voltage-clamp electroporation to obtain single-channel recordings of lipid pores and analyzed the idealized dwell-time sequences using Maximum-Likelihood Fitting. We observed traces with multiple current levels and determined whether they were a result of the presence of multiple pores or a single pore with multiple conductance states. We found that, within the same recording, the bilayer can have a single pore with multiple conductance states or multiple independent pores. Using high sampling rates (100 kHz) we were able to observe pores with 40 μs lifetimes, and in experiments using high-voltage pulses we observed the existence of long-lived fluctuations minutes after the removal of the electric field. These results come closer to reconciling the nanosecond lifetime pores in molecular dynamics simulations and the long-lived permeability of cells after electroporation.

Abstract:Electrophysiology is the study of neural activity in the form of local field potentials, current flow through ion channels, calcium spikes, back propagating action potentials and somatic action potentials, all measurable on a millisecond timescale. Despite great progress in imaging technologies and sensor proteins, none of the currently available tools allow imaging of neural activity on a millisecond timescale and beyond the first few hundreds of microns inside the brain. The patch clamp technique has been an invaluable tool since its inception several decades ago and has generated a wealth of knowledge about the nature of voltage- and ligand-gated ion channels, sub-threshold and supra-threshold activity, and characteristics of action potentials related to higher order functions. Many techniques that evolve to be standardized tools in the biological sciences go through a period of transformation in which they become, at least to some degree, automated, in order to improve reproducibility, throughput and standardization. The patch clamp technique is currently undergoing this transition, and in this review, we will discuss various aspects of this transition, covering advances in automated patch clamp technology both in vitro and in vivo.

Abstract:Today's research projects propose a modular manufacturing environment for production sites, which adapt itself autonomously and makes manufacturing decisions without human interaction. Therefore, it is necessary that the next generations of production lines, especially the intralogistics transportation systems, are designed more adaptable and flexible. The object in this paper is a cyber-physical material flow system with flexible, autonomous and collaborative vehicles combined with centralized sensors to digitize the workspace. For this purpose, an interface was developed which allows a discrete event simulation tool to communicate with a Multi-Agent-System. Thereby, the decision-making of the agents is integrated directly into the simulation process of the discrete event simulation software. The architecture of this interface is presented as well as a test of its functionality. The architecture is implemented with the Java Agent Development Framework and Plant Simulation as the discrete event simulation tool. The result is an interface, which allows to transfer data from the simulation, in case of an event, to the agent platform. The Multi-Agent-System solves the event specific problem due to its ontology and responses it to the simulation. Therefore, it is possible to integrate the ontology implemented in the physical system as software-in-the-loop in the simulation environment. Furthermore, the possibility is given to improve the ontology iteratively based on historical production data. Different strategies of agents can be combined and improved through machine-learning algorithms by using real production data from the task specific hardware. This leads into a continuous improvement process.

Abstract:This study takes a step in understanding the physiological implications of the nanosecond pulsed electric field (nsPEF) by integrating molecular dynamics simulations and machine learning techniques. nsPEF, a state-of-the-art technology, uses high-voltage electric field pulses with a nanosecond duration to modulate cellular activity. This investigation reveals a relatively new and underexplored phenomenon: protein-mediated electroporation. Our research focused on the voltage-sensing domain (VSD) of the NaV1.5 sodium cardiac channel in response to nsPEF stimulation. We scrutinized the VSD structures that form pores and thereby contribute to the physical chemistry that governs the defibrillation effect of nsPEF. To do so, we conducted a comprehensive analysis involving the clustering of 142 replicas simulated for 50 ns under nsPEF stimuli. We subsequently pinpointed the representative structures of each cluster and computed the free energy between them. We find that the selected VSD of NaV1.5 forms pores under nsPEF stimulation, but in a way that significant differs from the traditional VSD opening. This study not only extends our understanding of nsPEF and its interaction with protein channels but also adds a new effect to further study.

Abstract:The supramolecular architectures of a fluorophore are controlled through the design of a conjugated polycatenar molecule, the self-assembly of which can be addressed toward a columnar liquid-crystalline phase and organogels. Thus, depending on the environmental conditions for self-assembly, compound CA9 organizes into an unprecedented hexagonal columnar mesophase in the condensed state, in which half a molecule constitutes the slice of the column, or into a rectangular mesomorphic-like organization in the presence of apolar solvents such as cyclohexane and dodecane, at a concentration in which fibers form and gelling conditions are fulfilled. In this Col(r)-type arrangement, the organization within the columns depends on the solvent. All of the materials prepared show luminescence, and moreover, a remarkable 3-fold increase in fluorescence intensity was observed in going from the solution to the gel state.

Abstract:Abstract The patch-clamp technique and more recently the high throughput patch-clamp technique have contributed to major advances in the characterization of ion channels. However, the whole-cell voltage-clamp technique presents certain limits that need to be considered for robust data generation. One major caveat is that increasing current amplitude profoundly impacts the accuracy of the biophysical analyses of macroscopic ion currents under study. Using mathematical kinetic models of a cardiac voltage-gated sodium channel and a cardiac voltage-gated potassium channel, we demonstrated how large current amplitude and series resistance artefacts induce an undetected alteration in the actual membrane potential and affect the characterization of voltage-dependent activation and inactivation processes. We also computed how dose–response curves are hindered by high current amplitudes. This is of high interest since stable cell lines frequently demonstrating high current amplitudes are used for safety pharmacology using the high throughput patch-clamp technique. It is therefore critical to set experimental limits for current amplitude recordings to prevent inaccuracy in the characterization of channel properties or drug activity, such limits being different from one channel type to another. Based on the predictions generated by the kinetic models, we draw simple guidelines for good practice of whole-cell voltage-clamp recordings.

Transient tension responses of voltage-clamped frog atrial muscle related to sudden changes in external Ca or Na

Abstract:Frog atrial muscle strips were placed in a double sucrose gap chamber and perfusion of the central node was arranged as to allow rapid changes of external Ca or Na concentration during long-lasting (15 s) depolarizing clamps. When the superfusing fluid was suddenly switched, the intercellular space inside the fibre bundle equilibrated with a time constant in the order of 1 s. A fast change of perfusate during clamp was followed by a delayed change of tonic (sustained) contraction. When [Ca]0 was increased from 0.25 to 4 mM, tension rose in a sigmoid manner and the level reached at the end of the clamp was almost identical with the steady control in 4 mM-Ca-Ringer. A similar tension increase occurred upon a reduction of [Na]0 from 100 to 25% of normal. At a given depolarization time course and height of the tension responses followed the ratio of [Ca]0/[Na]20. Transient tension responses are interpreted in terms of a sudden perturbation of a transmembrane Ca-Na exchange system leading to a depression of Ca pumping activity.
Defined extracellular ionic solutions to study and manipulate the cellular resting membrane potential

Abstract:All cells possess an electric potential across their plasma membranes and can generate and receive bioelectric signals. The cellular resting membrane potential (RMP) can regulate cell proliferation, differentiation and apoptosis. Current approaches to measure the RMP rely on patch clamping, which is technically challenging, low-throughput and not widely available. It is therefore critical to develop simple strategies to measure, manipulate and characterize the RMP. Here, we present a simple methodology to study the RMP of non-excitable cells and characterize the contribution of individual ions to the RMP using a voltage-sensitive dye. We define protocols using extracellular solutions in which permeable ions (Na+, Cl- and K+) are substituted with non-permeable ions [N-Methyl-D-glucamine (NMDG), gluconate, choline, SO42-]. The resulting RMP modifications were assessed with both patch clamp and a voltage sensitive dye. Using an epithelial and cancer cell line, we demonstrate that the proposed ionic solutions can selectively modify the RMP and help determine the relative contribution of ionic species in setting the RMP. The proposed method is simple and reproducible and will make the study of bioelectricity more readily available to the cell biology community.This article has an associated First Person interview with the first author of the paper.

Abstract:Human-induced stem cell-derived cardiomyocytes (hiPSC-CMs) are a valuable tool for studying development, pharmacology, and (inherited) arrhythmias. Unfortunately, hiPSC-CMs are depolarized and spontaneously active, even the working cardiomyocyte subtypes such as atrial- and ventricular-like hiPSC-CMs, in contrast to the situation in the atria and ventricles of adult human hearts. Great efforts have been made, using many different strategies, to generate more mature, quiescent hiPSC-CMs with more close-to-physiological resting membrane potentials, but despite promising results, it is still difficult to obtain hiPSC-CMs with such properties. The dynamic clamp technique allows to inject a current with characteristics of the inward rectifier potassium current (I K1 ), computed in real time according to the actual membrane potential, into patch-clamped hiPSC-CMs during action potential measurements. This results in quiescent hiPSC-CMs with a close-to-physiological resting membrane potential. As a result, action potential measurements can be performed with normal ion channel availability, which is particularly important for the physiological functioning of the cardiac SCN5A -encoded fast sodium current (I Na ). We performed in vitro and in silico experiments to assess the beneficial effects of the dynamic clamp technique in dissecting the functional consequences of the SCN5A -1795insD +/− mutation. In two separate sets of patch-clamp experiments on control hiPSC-CMs and on hiPSC-CMs with mutations in ACADVL and GNB5 , we assessed the value of dynamic clamp in detecting delayed afterdepolarizations and in investigating factors that modulate the resting membrane potential. We conclude that the dynamic clamp technique has highly beneficial effects in all of the aforementioned settings and should be widely used in patch-clamp studies on hiPSC-CMs while waiting for the ultimate fully mature hiPSC-CMs.

Abstract:The measurement of cell membrane potential (Vm) is important for understanding ion channel function, and plays a role in several routine cellular functions and disease, particularly in excitable cells such as muscle and nerve. However, measuring Vm is difficult, relying either on labour-intensive direct measurement of single cells (intracellular electrodes, patch clamp) or indirect measurement of fluorescence intensity, using Vm-sensitive labels. Here we demonstrate a direct measurement technique based on determination of the cell's ζ-potential, the electrical potential at the hydrodynamic shear plane, approximately 1 nm beyond the cell surface. We demonstrate this principle using excitable H9c2 cardiomyoblasts, measured in both polarised and depolarised states, before and after extracellular intervention to alter cell ion concentration. Given widespread availability of ζ-potential measurement apparatus (most typically in chemistry and materials science settings), this offers a new method of measuring Vm without the need for fluorescence measurements or calibration curves.

Abstract:Action potentials play a pivotal role in diverse cardiovascular physiological mechanisms. A comprehensive understanding of these intricate mechanisms necessitates a high-fidelity intracellular electrophysiological investigative approach. The amalgamation of micro-/nano-electrode arrays and electroporation confers substantial advantages in terms of high-resolution intracellular recording capabilities. Nonetheless, electroporation systems typically lack precise control, and commonly employed electroporation modes, involving tailored sequences, may escalate cellular damage and perturbation of normal physiological functions due to the multiple or higher-intensity electrical pulses. In this study, we developed an innovative electrophysiological biosensing system customized to facilitate precise single-pulse electroporation. This advancement serves to achieve optimal and uninterrupted intracellular action potential recording within cardiomyocytes. The refinement of the single-pulse electroporation technique is realized through the integration of the electroporation and assessment biosensing system, thereby ensuring a consistent and reliable means of achieving stable intracellular access. Our investigation has unveiled that the optimized single-pulse electroporation technique not only maintains robust biosafety standards but also enables the continuous capture of intracellular electrophysiological signals across an expansive three-day period. The universality of this biosensing system, adaptable to various micro/nano devices, furnishes real-time analysis and feedback concerning electroporation efficacy, guaranteeing the sustained, secure, and high-fidelity acquisition of intracellular data, thereby propelling the field of cardiovascular electrophysiological research.

Abstract:Conventionally, manual patch-clamp electrophysiological approaches are the gold standard for studying ion channel function in neurons. However, these approaches are labor-intensive, yielding low-throughput results, and are therefore not amenable for compound profiling efforts during the early stages of drug discovery. The SyncroPatch 384PE has been successfully implemented for pharmacological experiments in heterologous overexpression systems that may not reproduce the function of voltage-gated ion channels in a native, heterogeneous environment. Here, we describe a protocol allowing the characterization of endogenous voltage-gated potassium (Kv) and sodium (Nav) channel function in developing primary rat cortical cultures, allowing investigations at a significantly improved throughput compared with manual approaches. Key neuronal marker expression and microelectrode array recordings of electrophysiological activity over time correlated well with neuronal maturation. Gene expression data revealed high molecular diversity in Kv and Nav subunit composition throughout development. Voltage-clamp experiments elicited three major current components composed of inward and outward conductances. Further pharmacological experiments confirmed the endogenous expression of functional Kv and Nav channels in primary cortical neurons. The major advantages of this approach compared with conventional manual patch-clamp systems include unprecedented improvements in experimental ease and throughput for ion channel research in primary neurons. These efforts demonstrated feasibility for primary neuronal ion channel investigation with the SyncroPatch, providing the foundation for future studies characterizing biophysical changes in endogenous ion channels in primary systems associated with disease or development.

Automated measurement of intestinal mucosa electrical parameters using a new digital clamp

Abstract:Electrophysiological studies that include measurements of the electrical parameters of the epithelium offer insight into the cell's ability to react to different biological effectors and their functional viability. These parameters are commonly measured using a Ussing permeation chamber; however, most Ussing permeation chambers currently available must follow a strict operational protocol, and the type of electrodes used has to be taken into special consideration. The purpose of this study was to develop a new Ussing permeation chamber device with an automatic digital clamp which uses a microcontroller. Conventional electrodes, such as platinum or Ag/AgCl electrodes, are replaced by stainless steel 316L working electrodes. The electrode-electrolyte interface (inox-Ringer's) study was performed by impedance spectroscopy in the range of 1-10 kHz. The determination of Warburg's model electrical parameters was inferred from the Nyquist diagram. The model validation of the new digital clamp was performed experimentally on isolated segments of mouse jejunum. Two main study results should be mentioned. One is that impedance spectroscopy on stainless steel electrodes has provided Warburg's parameters, allowing the development of a transfer function model. The other is that the new digital clamp can simultaneously measure or calculate conductance, potential difference and short-circuit current. These results have also confirmed the great importance of Warburg's model for determining the electrical parameters of the electrode-electrolyte interface, and have shown that the measurement of intestinal mucosa electrical parameters can be achieved with a digital correction. Finally, the results suggest that stainless steel electrodes can be used successfully in a Ussing permeation chamber as working electrodes.
Resolving artefacts in voltage-clamp experiments with computational modelling: an application to fast sodium current recordings

Abstract:Cellular electrophysiology is the foundation of many fields, from basic science in neurology, cardiology, oncology to safety critical applications for drug safety testing, clinical phenotyping, etc. Patch-clamp voltage clamp is the gold standard technique for studying cellular electrophysiology. Yet, the quality of these experiments is not always transparent, which may lead to erroneous conclusions for studies and applications. Here, we have developed a new computational approach that allows us to explain and predict the experimental artefacts in voltage-clamp experiments. The computational model captures the experimental procedure and its inadequacies, including: voltage offset, series resistance, membrane capacitance and (imperfect) amplifier compensations, such as series resistance compensation and supercharging. The computational model was validated through a series of electrical model cell experiments. Using this computational approach, the artefacts in voltage-clamp experiments of cardiac fast sodium current, one of the most challenging currents to voltage clamp, were able to be resolved and explained through coupling the observed current and the simulated membrane voltage, including some typically observed shifts and delays in the recorded currents. We further demonstrated that the typical way of averaging data for current-voltage relationships would lead to biases in the peak current and shifts in the peak voltage, and such biases can be in the same order of magnitude as those differences reported for disease-causing mutations. Therefore, the presented new computational pipeline will provide a new standard of assessing the voltage-clamp experiments and interpreting the experimental data, which may be able to rectify and provide a better understanding of ion channel mutations and other related applications.

Abstract:We provide here detailed electrophysiological protocols to study voltage-gated sodium channels and to investigate how wild-type and mutant channels influence firing properties of transfected mammalian dorsal root ganglion (DRG) neurons. Whole-cell voltage-clamp recordings permit us to analyze kinetic and voltage-dependence properties of ion channels and to determine the effect and mode of action of pharmaceuticals on specific channel isoforms. They also permit us to analyze the role of individual sodium channels and their mutant derivatives in regulating firing of DRG neurons. Five to ten cells can be recorded daily, depending on the extent of analysis that is required. Because of different internal solutions that are used in voltage-clamp and current-clamp recordings, only limited information can be obtained from recording the same neuron in both modes. These electrophysiological studies help to elucidate the role of specific channels in setting threshold and suprathreshold responses of neurons, under normal and pathological conditions.

Abstract:Conventional electrophysiological methods, i.e. nerve conduction studies and electromyography are suitable methods for the diagnosis of neuromuscular disorders, however, they provide limited information about muscle fibre membrane properties and underlying disease mechanisms. Muscle excitability testing is a technique that provides in vivo information about muscle fibre membrane properties such as membrane potential and ion channel function. Since the 1960s, various methodologies have been suggested to examine muscle membrane properties but technical difficulties have limited its use. In 2009, an automated, fast and simple application, the so-called multi-fibre muscle velocity recovery cycles (MVRC) has accelerated the use of muscle excitability testing. Later, frequency ramp and repetitive stimulation protocols have been developed. Though this method has been used mainly in research for revealing disease mechanisms across a broad range of neuromuscular disorders, it may have additional diagnostic uses; value has been shown particularly in muscle channelopathies. This review will provide a description of the state-of-the art of methodological and clinical studies for muscle excitability testing.

Abstract:In this study, electrical impedance spectroscopy measurements are performed during electroporation of monolayers of differentiated myotubes. The time resolution of the system (1 spectrum/ms) enable 860 full spectra (21 frequencies from 5 kHz to 1.3 MHz) to be acquired during the time gap between consecutive pulses (interpulse) of a classical electroporation treatment (8 pulses, 100 μs, 1 Hz). Additionally, the characteristics of the custom microelectrode assembly used allow the experiments to be performed directly in situ in standard 24 multi-well plates. The impedance response dynamics are studied for three different electric field intensities (400, 800 and 1200 V/cm). The multifrequency information, analysed with the Cole model, reveals a short-term impedance recovery after each pulse in accordance with the fast resealing of the cell membrane, and a long-term impedance decay over the complete treatment in accordance with an accumulated effect pulse after pulse. The analysis shows differences between the lowest electric field condition and the other two, suggesting that different mechanisms that may be related with the reversibility of the process are activated. As a result of the multifrequency information, the system is able to measure simultaneously the conductivity variations due to ion diffusion during electroporation. Finally, in order to reinforce the physical interpretation of the results, a complementary electrical equivalent model is used.
