Abstract:the invasion of UPEC to the urothelial cells. Additional bladders were immunostained with anti-E.coli , TLR4, UPIa, NF-k B p65, Ly6G antibodies. Urinary cytokines, IL1 a , IL6, TNF-a , were measured using ELISA kits and normalized with creatinine. Neutrophil in fi ltration into the bladder was assessed using a myeloperoxidase activity assay. RESULTS: Loss of CDKN2A did not affect the expression and distribution of the urothelial receptor for UPEC ee UPIa and TLR4. At 2 hours post-infection, UPEC induced rapid activation of NF-k B in the umbrella cells in WT mice; however, the activation was 50% less in CDKN2A KO mice. Consequently, the urinary secretion of proin fl ammatory cytokines IL1 a , IL6, TNF-a in CDKN2A KO mice was markedly reduced. At 24 hours post-infection, the bacterial colonization was 5.6-fold higher in the CDKN2A KO mice bladders than in the controls. Gentamycin protection assay showed that the invasion of the bacteria was 3-fold higher in the CDKN2A KO mice. Consistently, the number of intracellular bacterial communities at 12 and 24 hours post-infection was signi fi cantly higher in the CDKN2A KO mice. In contrast, neutrophil in fi ltration in the urothelium was signi fi cantly reduced, and neutrophil MPO activity was 2-fold lower in the CDKN2A KO mice than in the controls. CONCLUSIONS: Our study provides the fi rst in vivo evidence that CDKN2A plays a critical role in the host innate immune defense against UTI. De fi ciency in CDKN2A leads to compromised cytokine secretion, neutrophil in fi ltration, increased bacterial burden and may predispose the urinary tract to recurrent infections by UPEC.
