Abstract:Lintless mutant is a super-short fiber mutant in upland cotton only 4–8 mm in fiber length and also named Ligon cotton controlled by one dominant geneLi 1. Fiber ultrastructure of the mutant (Li 1) and its wild type (li 1)in situ andin vitro was observed under an electron microscope to understand its cytological characteristics during the fiber cell elongation. The results showed that the mutant fiberin situ had thinner cytoplasm, more small vacuoles, less mitochondria, Gllgi apparatus and endoplasmic reticula, and there were more starch granules which were free or packed in the amyloplast beside the cell wall than that of wild type. It was indicated that scarcity of functional organelles and disability of transformation from starch to sugar might be associated with the fact that the mutant fiber cell was aborted too early to elongate into normal length. Mutant ovule in some media containing GA3 could produce a kind of huge callus that grew faster than normal ovules. The callus was covered with many white, loose, and semitransparent fiber-like cells that apt to get off from ovule. These fiber-like cells were multicellular fibers generated by cell division and had black dots just like pigment glands in the stem and leaf of cotton. There were lots of micro-tubes beside cytoplasm membrane of the multicellular fiber, which were thought to be primary preparation for second wall deposition of multicellular fiber. It was indicated that GA3 might induce the expression of gene(s) that kept inactive in the field condition and then stimulate the original fiber cellin vitro to undergo division again.
Rhamnogalacturonan I with β-(1,4)-Galactan Side Chains as an Ever-Present Component of Tertiary Cell Wall of Plant Fibers

Abstract:The cellulose-enriched tertiary cell walls present in many plant fibers have specific composition, architecture, machinery of formation, and function. To better understand the mechanisms underlying their mode of action and to reveal the peculiarities of fibers from different plant species, it is necessary to more deeply characterize the major components. Next to overwhelming cellulose, rhamnogalacturonan I (RG-I) is considered to be the key polymer of the tertiary cell wall; however, it has been isolated and biochemically characterized in very few plant species. Here, we add RG-I to the list from the phloem fibers of the Phaseolus vulgaris stem that was isolated and analyzed by nuclear magnetic resonance (NMR), dynamic light scattering, and immunolabeling, both within tissue and as an isolated polymer. Additionally, fibers with tertiary cell walls from nine species of dicotyledonous plants from the orders Malphigiales, Fabales, and Rosales were labeled with RG-I-related antibodies to check the presence of the polymer and compare the in situ presentation of its backbone and side chains. The obtained results confirm that RG-I is an obligatory polymer of the tertiary cell wall. However, there are differences in the structure of this polymer from various plant sources, and these peculiarities may be taxonomically related.

Abstract:Transmission electron microscopy was used to study the development and structure of resting spores and sporosori of Polymyxa graminis. Osmiophilic bodies generally distributed in the cytoplasm moved to the periphery as spores matured. The spore wall consisted of four or five layers, two of which extended into spines linking adjacent spores. An unstained plug in the wall, faced towards the outside of the sporosorus. Some spores germinated in situ by producing primary zoospores. Bundle-like structures in some spores may have been barley yellow mosaic virus particles.
Zygospores of the green alga Spirogyra: new insights from structural and chemical imaging

Abstract:Zygnematophyceae, a class of streptophyte green algae and sister group to land plants (Embryophytes) live in aquatic to semi-terrestrial habitats. The transition from aquatic to terrestrial environments requires adaptations in the physiology of vegetative cells and in the structural properties of their cell walls. Sexual reproduction occurs in Zygnematophyceae by conjugation and results in the formation of zygospores, possessing unique multi-layered cell walls, which might have been crucial in terrestrialization. We investigated the structure and chemical composition of field sampled Spirogyra sp. zygospore cell walls by multiple microscopical and spectral imaging techniques: light microscopy, confocal laser scanning microscopy, transmission electron microscopy following high pressure freeze fixation/freeze substitution, Raman spectroscopy and atomic force microscopy. This comprehensive analysis allowed the detection of the subcellular organization and showed three main layers of the zygospore wall, termed endo-, meso- and exospore. The endo- and exospore are composed of polysaccharides with different ultrastructural appearance, whereas the electron dense middle layer contains aromatic compounds as further characterized by Raman spectroscopy. The possible chemical composition remains elusive, but algaenan or a sporopollenin-like material is suggested. Similar compounds with a non-hydrolysable character can be found in moss spores and pollen of higher plants, suggesting a protective function against desiccation stress and high irradiation. While the tripartite differentiation of the zygospore wall is well established in Zygnematopyhceae, Spirogyra showed cellulose fibrils arranged in a helicoidal pattern in the endo- and exospore. Initial incorporation of lipid bodies during early zygospore wall formation was also observed, suggesting a key role of lipids in zygospore wall synthesis. Multimodal imaging revealed that the cell wall of the sexually formed zygospores possess a highly complex internal structure as well as aromatics, likely acting as protective compounds and leading to impregnation. Both, the newly discovered special three-dimensional arrangement of microfibrils and the integration of highly resistant components in the cell wall are not found in the vegetative state. The variety of methods gave a comprehensive view on the intricate zygospore cell wall and its potential key role in the terrestrial colonization and plant evolution is discussed.

Abstract:Abstract One hallmark of plant cells is their pecto-cellulosic cell walls. They protect cells against the environment and high turgor and mediate morphogenesis through the dynamics of their mechanical and chemical properties. The walls are a complex polysaccharidic structure. Although their biochemical composition is well known, how the different components organize in the volume of the cell wall and interact with each other is not well understood and yet is key to the wall’s mechanical properties. To investigate the ultrastructure of the plant cell wall, we imaged the walls of onion ( Allium cepa ) bulbs in a near-native state via cryo-Focused Ion Beam milling (cryo-FIB-milling) and cryo-Electron Tomography (cryo-ET). This allowed the high-resolution visualization of cellulose fibers in situ ( in muro ). We reveal the coexistence of dense fiber fields bathed in a reticulated matrix we termed “meshing,” which is more abundant at the inner surface of the cell wall. The fibers adopted a regular bimodal angular distribution at all depths in the cell wall and bundled according to their orientation, creating layers within the cell wall. Concomitantly, employing homogalacturonan (HG)-specific enzymatic digestion, we observed changes in the meshing, suggesting that it is at least in part composed of HG pectins. We propose the following model for the construction of the abaxial epidermal primary cell wall: The cell deposits successive layers of cellulose fibers at −45° and +45° relative to the cell’s long axis and secretes the surrounding HG-rich meshing proximal to the plasma membrane, which then migrates to more distal regions of the cell wall.

Abstract:Several species of freshwater green algae belonging to the order Zygnematales (Spirogyra crassa (Ktz.) Czurda, S. condensata (Vauch.) Czurda, S. longata (Vauch.) Ktz., S. juergensii Ktz., S. olivascens Rabenh. and Mougeotia viridis (Ktz.) Wittr.) were shown to have specific sterol content and characteristic monosaccharide composition of the biomass. Polysaccharide fractions were obtained by stepwise extraction of S. condensata and characterised by a determination of monosaccharides liberated after acid hydrolysis. Action of amyloglucosidase was used to prove the presence of starch in algal biomass. The main polysaccharide seems to be a complex mucilage composed of rhamnose, arabinose, xylose, galactose, and uronic acid, and the structure of this mucilage needs further investigation.
Interactions between Cellulose and (1,3;1,4)-β-glucans and Arabinoxylans in the Regenerating Wall of Suspension Culture Cells of the Ryegrass Lolium multiflorum

Abstract:Plant cell walls (PCWs) form the outer barrier of cells that give the plant strength and directly interact with the environment and other cells in the plant. PCWs are composed of several polysaccharides, of which cellulose forms the main fibrillar network. Enmeshed between these fibrils of cellulose are non-cellulosic polysaccharides (NCPs), pectins, and proteins. This study investigates the sequence, timing, patterning, and architecture of cell wall polysaccharide regeneration in suspension culture cells (SCC) of the grass species Lolium multiflorum (Lolium). Confocal, superresolution, and electron microscopies were used in combination with cytochemical labeling to investigate polysaccharide deposition in SCC after protoplasting. Cellulose was the first polysaccharide observed, followed shortly thereafter by (1,3;1,4)-β-glucan, which is also known as mixed-linkage glucan (MLG), arabinoxylan (AX), and callose. Cellulose formed fibrils with AX and produced a filamentous-like network, whereas MLG formed punctate patches. Using colocalization analysis, cellulose and AX were shown to interact during early stages of wall generation, but this interaction reduced over time as the wall matured. AX and MLG interactions increased slightly over time, but cellulose and MLG were not seen to interact. Callose initially formed patches that were randomly positioned on the protoplast surface. There was no consistency in size or location over time. The architecture observed via superresolution microscopy showed similarities to the biophysical maps produced using atomic force microscopy and can give insight into the role of polysaccharides in PCWs.
Metabolomic and Transcriptomic Insights into How Cotton Fiber Transitions to Secondary Wall Synthesis, Represses Lignification, and Prolongs Elongation

Abstract:Background: The morphogenesis of single-celled cotton fiber includes extreme elongation and staged cell wall differentiation. Designing strategies for improving cotton fiber for textiles and other uses relies on uncovering the related regulatory mechanisms. In this research we compared the transcriptomes and metabolomes of two Gossypium genotypes, Gossypium barbadense cv Phytogen 800 and G. hirsutum cv Deltapine 90. When grown in parallel, the two types of fiber developed similarly except for prolonged fiber elongation in the G. barbadense cultivar. The data were collected from isolated fibers between 10 to 28 days post anthesis (DPA) representing: primary wall synthesis to support elongation; transitional cell wall remodeling; and secondary wall cellulose synthesis, which was accompanied by continuing elongation only in G. barbadense fiber.Results: Of 206 identified fiber metabolites, 205 were held in common between the two genotypes. Approximately 38,000 transcripts were expressed in the fiber of each genotype, and these were mapped to the reference set and interpreted by homology to known genes. The developmental changes in the transcriptomes and the metabolomes were compared within and across genotypes with several novel implications. Transitional cell wall remodeling is a distinct stable developmental stage lasting at least four days (18 to 21 DPA). Expression of selected cell wall related transcripts was similar between genotypes, but cellulose synthase gene expression patterns were more complex than expected. Lignification was transcriptionally repressed in both genotypes. Oxidative stress was lower in the fiber of G. barbadense cv Phytogen 800 as compared to G. hirsutum cv Deltapine 90. Correspondingly, the G. barbadense cultivar had enhanced capacity for management of reactive oxygen species during its prolonged elongation period, as indicated by a 138-fold increase in ascorbate concentration at 28 DPA.Conclusions: The parallel data on deep-sequencing transcriptomics and non-targeted metabolomics for two genotypes of single-celled cotton fiber showed that a discrete developmental stage of transitional cell wall remodeling occurs before secondary wall cellulose synthesis begins. The data showed how lignification can be transcriptionally repressed during secondary cell wall synthesis, and they implicated enhanced capacity to manage reactive oxygen species through the ascorbate-glutathione cycle as a positive contributor to fiber length.

Abstract:Flax ( Linum usitatissimum L.) phloem fibers produce a tertiary cell wall as part of regular plant growth. The fiber tertiary cell wall is responsible for supporting the stem's vertical position and is involved in the implementation of the negative gravitropic reaction when it is tilted. Rhamnogalacturonan I (RG-I) is one of the main matrix polysaccharides of the tertiary cell wall, playing an essential role in its functioning. The distribution of RG-I in fibers isolated from two stem sides (pulling and opposite) during graviresponse was studied. The increased content of galactose-containing polymers in the cell walls of fibers from the pulling stem sides was demonstrated. Differences in the distribution of RG-I epitopes on different sides of the stem were shown. RG-I with branched galactan side chains was found in the fiber tertiary cell walls; the labeling of such side chains in the cell wall of fibers from the pulling side of the stem was increased during gravistimulation. Using transcriptome analysis, the upregulation of genes encoding enzymes involved in RG-I metabolism (synthesis and modifications) in the fibers from the pulling stem side, mainly at the early stage of graviresponse, was revealed. A special role for fasciclin-like arabinogalactan proteins in response to mechanical stimuli was proposed. For the purpose of further modifying the plant cell wall properties, it would be worthwhile to have an understanding of the processes taking place in the fiber cell wall, both in normal conditions and during mechanical stress.

Abstract:The control of the cylindrical cell form in plants appears to reside in the orientation of the reinforcing cellulose microfibrils in the side walls. In elongating cells the fibrils are typically transverse. Control of new synthesis of oriented wall texture is shown to be in turn related to the orientation of cytoplasmic elements in the cell periphery. Three properties of these cytoplasmic elements have been deduced from polarization optical properties of treated and normal cell walls. These deduced properties— namely, possession of a long axis and the ability to build microfibrils perpendicular to it, a tendency to cross-bond to make a parallel array, and a sensitivity of this alignment to colchicine—are all well-known properties of mitotic spindle and phragmoplast fibers which form the cross-wall after mitosis. It is proposed that proteins of spindle fiber nature exist in cortical cytoplasm of plant cells and are active in the control of wall texture and cell form.

Abstract:Oat (Avena sativa) is a cereal crop whose grains are rich in (1,3; 1,4)-β-D-glucan (mixed linkage glucan or MLG), a soluble dietary fiber. In our study, we analyzed oat endosperm development in two Canadian varieties with differing MLG content and nutritional value. We confirmed that oat undergoes a nuclear type of endosperm development but with a shorter cellularisation phase than barley (Hordeum vulgare). Callose and cellulose were the first polysaccharides to be detected in the early anticlinal cell walls at 11 days post-emergence (DPE) of the panicle. Other polysaccharides such as heteromannan and homogalacturonan were deposited early in cellularisation around 12 DPE after the first periclinal walls are laid down. In contrast to barley, heteroxylan deposition coincided with completion of cellularisation and was detected from 14 DPE but was only detectable after demasking. Notably, MLG was the last polysaccharide to be laid down at 18 DPE within the differentiation phase, rather than during cellularisation. In addition, differences in the spatiotemporal patterning of MLG were also observed between the two varieties. The lower MLG-containing cultivar AC Morgan (3.5% w/w groats) was marked by the presence of a discontinuous pattern of MLG labelling, while labelling in the same walls in CDC Morrison (5.6% w/w groats) was mostly even and continuous. RNA-Seq analysis revealed higher transcript levels of multiple MLG biosynthetic Cellulose Synthase-like F (CSLF) and CSLH) genes during grain development in CDC Morrison compared to AC Morgan that likely contributes to the increased abundance of MLG at maturity in CDC Morrison. CDC Morrison was also observed to have smaller endosperm cells with thicker walls than AC Morgan from cellularisation onwards, suggesting the processes controlling cell size and shape are established early in development. This study has highlighted that the molecular processes influencing MLG content and deposition are more complex than previously imagined.

The cortical cytoskeletal network and cell-wall dynamics in the unicellular charophycean green alga Penium margaritaceum

Abstract:Background and aims: Penium margaritaceum is a unicellular charophycean green alga with a unique bi-directional polar expansion mechanism that occurs at the central isthmus zone prior to cell division. This entails the focused deposition of cell-wall polymers coordinated by the activities of components of the endomembrane system and cytoskeletal networks. The goal of this study was to elucidate the structural organization of the cortical cytoskeletal network during the cell cycle and identify its specific functional roles during key cell-wall developmental events: pre-division expansion and cell division. Methods: Microtubules and actin filaments were labelled during various cell cycle phases with an anti-tubulin antibody and rhodamine phalloidin, respectively. Chemically induced disruption of the cytoskeleton was used to elucidate specific functional roles of microtubules and actin during cell expansion and division. Correlation of cytoskeletal dynamics with cell-wall development included live cell labelling with wall polymer-specific antibodies and electron microscopy. Key results: The cortical cytoplasm of Penium is highlighted by a band of microtubules found at the cell isthmus, i.e. the site of pre-division wall expansion. This band, along with an associated, transient band of actin filaments, probably acts to direct the deposition of new wall material and to mark the plane of the future cell division. Two additional bands of microtubules, which we identify as satellite bands, arise from the isthmus microtubular band at the onset of expansion and displace toward the poles during expansion, ultimately marking the isthmus of future daughter cells. Treatment with microtubule and actin perturbation agents reversibly stops cell division. Conclusions: The cortical cytoplasm of Penium contains distinct bands of microtubules and actin filaments that persist through the cell cycle. One of these bands, termed the isthmus microtubule band, or IMB, marks the site of both pre-division wall expansion and the zone where a cross wall will form during cytokinesis. This suggests that prior to the evolution of land plants, a dynamic, cortical cytoskeletal array similar to a pre-prophase band had evolved in the charophytes. However, an interesting variation on the cortical band theme is present in Penium, where two satellite microtubule bands are produced at the onset of cell expansion, each of which is destined to become an IMB in the two daughter cells after cytokinesis. These unique cytoskeletal components demonstrate the close temporal control and highly coordinated cytoskeletal dynamics of cellular development in Penium.
Complex sphingolipids are essential for cell division and plasmodesmal development in the moss Physcomitrium patens

Abstract:Developmental patterning and organ structure are elegantly simple in the moss Physcomitrium patens. In molecular genetic studies, this facilitates both the cultivation of severe mutant alleles and their phenotypic characterization. Essential membrane lipids, such as complex phosphosphingolipids (in plants, glycosyl inositol phosphorylceramides, GIPCs), have been difficult to functionally characterize due to non-viable and pleiotropic phenotypes of mutants affected in their synthesis in Arabidopsis thaliana. Following the isolation and biochemical characterization of mutants affected in GIPC synthesis in P. patens, including sphinganine-C4-hydroxylase (s4h/sbh) and inositol phosphorylceramide synthase (ipcs), we now report some of their morphological, histological, and cytological phenotypes. We observed alteration in cell division, expansion, and differentiation. Specifically, the s4h knock-out mutant had abnormal cell division planes, as well as irregular depositions attached to cell walls. Severe ipcs mutant alleles showed frequent incomplete cell divisions, causing compromised cell autonomy as demonstrated by intercellular motility assays. These phenotypes suggest that sphingolipids impact both the orientation and proper formation of the cell plate during cytokinesis. Transmission electron microscopy revealed dramatic plasmodesmal structural defects in all three mutants, however, qualitative aspects of plasmodesmal transport do not seem to be severely impacted. Our methods can be used as a toolkit for quantifying growth, and specifically cell division and plasmodesmal phenotypes in mosses; our present results elucidate the specific contributions of GIPCs to fundamental cell functions. Finally, the severity of the observed defects in cell functions and ultrastructure highlight the resilience and utility of P. patens for studying basic cellular functions and severe mutant phenotypes.

Abstract:Introduction: Polysaccharides, key components present in Grifola frondosa , can be divided into those derived from fruiting bodies, mycelium, and fermentation broth based on their source. The structure of G. frondosa fruiting body-derived polysaccharides has been fully characterized. However, the structure of G. frondosa mycelium-derived polysaccharides remains to be elucidated. Methods: In this study, we obtained mycelia from G. frondosa by liquid fermentation and extracted them with water and alkaline solution. Then, the mycelia were isolated and purified to obtain homogeneity and systematically characterized by methylation and FT infrared (FT-IR) and nuclear magnetic resonance (NMR) spectroscopy. Results and discussion: Structural analysis showed that two neutral fractions (WGFP-N-a and AGFP-N-a 1 ) have a common backbone composed of α-1,6-D-Me-Gal p and α-1,6-D-Gal p that were substituted at O-2 by 1,2-Man p , α-1,3-L-Fuc p , and α-T-D-Man p and thus are identified as fucomannogalactans. WGFP-A-a, AGFP-A-b, and AGFP-A-c are β-1,6-glucans with different molecular weights and are branched with β-1,3-D-Glc p and T-D-Glc p at the O-3 of Glc. Our results provide important structural information about G. frondosa mycelium-derived polysaccharides and provide the basis for their further development and application.

Abstract:Gelatinous (G)-fibers are common in the stems of twining vines (twiners), but their role remain unclear given the lack of developmental insights. Here, we characterize the developmental anatomy of G-fiber formation in common bean stems (Phaseolus vulgaris L., Fabaceae). G-fibers in common bean exhibit cell wall organization comparable to other species, consisting of cellulose and Rhamnogalacturonan-I pectins, with possible traces of lignin. We show that G-fibers are absent in the actively circumnutating stems, thus these tensile fibers are not associated with the dynamic searching movements characteristics of twiners. Instead, we found that after a subtle bend or dramatic coil is formed, G-fibers form asymmetrically on the concave side of the stem for posture maintenance. Therefore, G-fibers do not drive movement, but provide support for existing bends, thus stabilizing the helical conformation of twiners around its host to avoid slippage. Finally, we present common bean as an emergent system to study twiners and growth form diversity given its easy cultivation, self-pollination, fast growth, and habit diversity arising from plant breeding, and the ability to induce habit shifts through simple modifications to light conditions.

Abstract:The Neurospora crassa genome has a gene cluster for the synthesis of galactosaminogalactan (GAG). The gene cluster includes the following: (1) UDP-glucose-4-epimerase to convert UDP-glucose and UDP-N-acetylglucosamine to UDP-galactose and UDP-N-acetylgalactosamine (NCU05133), (2) GAG synthase for the synthesis of an acetylated GAG (NCU05132), (3) GAG deacetylase (/NCW-1/NCU05137), (4) GH135-1, a GAG hydrolase with specificity for N-acetylgalactosamine-containing GAG (NCU05135), and (5) GH114-1, a galactosaminidase with specificity for galactosamine-containing GAG (NCU05136). The deacetylase was previously shown to be a major cell wall glycoprotein and given the name of NCW-1 (non-GPI anchored cell wall protein-1). Characterization of the polysaccharides found in the growth medium from the wild type and the GAG synthase mutant demonstrates that there is a major reduction in the levels of polysaccharides containing galactosamine and N-acetylgalactosamine in the mutant growth medium, providing evidence that the synthase is responsible for the production of a GAG. The analysis also indicates that there are other galactose-containing polysaccharides produced by the fungus. Phenotypic characterization of wild-type and mutant isolates showed that deacetylated GAG from the wild type can function as an adhesin to a glass surface and provides the fungal mat with tensile strength, demonstrating that the deacetylated GAG functions as an intercellular adhesive. The acetylated GAG produced by the deacetylase mutant was found to function as an adhesive for chitin, alumina, celite (diatomaceous earth), activated charcoal, and wheat leaf particulates.

Abstract:Two types of cell wall glycopolymers of different structure were found in the type strains of two species of phytopathogenic bacteria, Clavibacter insidiosus VKM Ac-1402 T and Clavibacter nebraskensis VKM Ac-1404 T (family Microbacteriaceae , class Actinomycetes ). The first type was represented by new, previously undescribed (1→6)-linked β-D-galactofuranans, which in the studied strains differed in the structure of side oligosaccharide chains (the structures are given in the text). The structure of the second glycopolymer, a pyruvate-containing galactomannan, was identical in both strains. The results obtained in this work, together with those published previously, indicate that the presence of the pyruvate-containing galactomannan and galactofuranans (with an identical core structure and different side oligosaccharide substituents) can be considered as a chemotaxonomic trait of the genus Clavibacter , and galactofuranans with di-, tri-, or tetrasaccharide substituents of different composition and structures can serve as chemotaxonomic markers of the species. The data obtained expand our understanding of the structural diversity of natural glycopolymers and structural features of the bacterial cell walls in various taxa and may be of interest for taxonomic studies and the studies aimed at elucidating the molecular mechanisms of interaction between bacteria and plant cells.

Abstract:A peculiar inward growth, named a "cell wall sac", formed in mulberry (Morus alba) idioblasts, is a subcellular site for production of calcium carbonate crystals. On the basis of ultrastructural observations, a fully expanded cell wall sac could be divided into two parts-an amorphous complex consisting of multi-layered compartments with multiple fibers originating from the innermost cell wall layer, and a peripheral plain matrix with fiber aggregates. Immunofluorescent localization showed that low and highly esterified pectin epitopes were detected at the early stages of development of the cell wall sac, followed by complete disappearance from the both parts of fully enlarged mature sac. In contrast, the xyloglucan epitope remained in the compartment complex; this was supported by the observation that the xyloglucan epitope labeled with immuno-gold particles is found on fibers in the complex part.

Abstract:Plant primary cell walls are composite structures surrounding the protoplast and containing pectins, hemicelluloses, and cellulose polysaccharides, as well as proteins. Their composition changed during the evolution of the green lineage from algae to terrestrial plants, i.e., from an aquatic to a terrestrial environment. The constraints of life in terrestrial environments have generated new requirements for the organisms, necessitating adaptations, such as cell wall modifications. We have studied the cell wall polysaccharide composition of thalli of Marchantia polymorpha, a bryophyte belonging to one of the first land plant genera. Using a collection of specific antibodies raised against different cell wall polysaccharide epitopes, we were able to identify in polysaccharide-enriched fractions: pectins, including low-methylesterified homogalacturonans; rhamnogalacturonan I with arabinan side-chains; and hemicelluloses, such as xyloglucans with XXLG and XXXG modules, mannans, including galactomannans, and xylans. We could also show the even distribution of XXLG xyloglucans and galactomannans in the cell walls of thalli by immunocytochemistry. These results are discussed with regard to the cell wall proteome composition and in the context of the evolution of the green lineage. The cell wall polysaccharides of M. polymorpha illustrate the transition from the charophyte ancestors of terrestrial plants containing xyloglucans, xylans and mannans as hemicelluloses, and embryophytes which do not exhibit mannans as major primary cell wall polysaccharides.

Abstract:The yellow brain mushroom Tremella mesenterica possesses a wide spectrum of medicinal properties, including immunostimulating, protecting against radiation, antidiabetic, anti-inflammatory, hypocholesterolemic, hepatoprotective, and antiallergic effects. A unique feature of T. mesenterica is that most of the above mentioned medicinal properties depend on glucuronoxylomannan (GXM) contained in fruiting bodies or produced in pure culture conditions. We developed a new strain of T. mesenterica CBS 101939, which grows in submerged culture and offers superior yields of one-cell biomass rich in exocellular heteropolysaccharide GXM. The structure of the GXM was analyzed by NMR spectroscopy and chemical methods. The polysaccharide has a defined repeating unit structure, which is O-acetylated at several points: [structure: see text]. These results differ from previously published structure of Tremella extracellular polysaccharides, where mannan backbone was believed to be randomly glycosylated with xylan chains of different length.
