Abstract:MYB transcription factors are involved in the regulation of plant development and response to biotic and abiotic stress. In this study, TaMyb1D, a novel subgroup 4 gene of the R2R3-MYB subfamily, was cloned from wheat (Triticum aestivum L.). TaMyb1D was localized in the nucleus and functioned as a transcriptional repressor. The overexpression of TaMyb1D in tobacco (Nicotiana tabacum) plants repressed the expression of genes related to phenylpropanoid metabolism and down-regulated the accumulation of lignin in stems and flavonoids in leaves. These changes affected plant development under normal conditions. The expression of TaMyb1D was ubiquitous and up-regulated by PEG6000 and H2O2 treatments in wheat. TaMyb1D-overexpressing transgenic tobacco plants exhibited higher relative water content and lower water loss rate during drought stress, as well as higher chlorophyll content in leaves during oxidative stress. The transgenic plants showed a lower leakage of ions as well as reduced malondialdehyde and H2O2 levels during conditions of drought and oxidative stresses. In addition, TaMyb1D up-regulated the expression levels of ROS- and stress-related genes in response to drought stress. Therefore, the overexpression of TaMyb1D enhanced tolerance to drought and oxidative stresses in tobacco plants. Our study demonstrates that TaMyb1D functions as a negative regulator of phenylpropanoid metabolism and a positive regulator of plant tolerance to drought and oxidative stresses.

Abstract:Abscisic acid (ABA)-, stress-, and ripening-induced (ASR) proteins are reported to be involved in abiotic stresses. However, it is not known whether ASR genes confer drought stress tolerance by utilizing the antioxidant system. In this study, a wheat ASR gene, TaASR1, was cloned and characterized. TaASR1 transcripts increased after treatments with PEG6000, ABA and H(2)O(2). Overexpression of TaASR1 in tobacco resulted in increased drought/osmotic tolerance, which was demonstrated that transgenic lines had lesser malondialdehyde (MDA), ion leakage (IL) and reactive oxygen species (ROS), but higher relative water content (RWC) and superoxide dismutase (SOD) and catalase (CAT) activities than wild type (WT) under drought stress. Overexpression of TaASR1 in tobacco also enhanced the expression of ROS-related and stress-responsive genes under osmotic stress. In addition, transgenic lines exhibited improved tolerance to oxidative stress by retaining more effective antioxidant system. Finally, TaASR1 was localized in the cell nucleus and functioned as a transcriptional activator. Taken together, our results showed that TaASR1 functions as a positive factor under drought/osmotic stress, involved in the regulation of ROS homeostasis by activating antioxidant system and transcription of stress-associated genes.
TaFDL2-1A interacts with TabZIP8-7A protein to cope with drought stress via the abscisic acid signaling pathway

Abstract:Drought has negative effects on cereal production. Studies have shown that many basic leucine zipper transcription factors (bZIP TFs) help to cope with drought stress. In this study, bZIP TF wheat (Triticum aestivum L.) FD-Like2 (TaFDL2) was isolated and functionally analyzed. Three homologs of TaFDL2 were identified and their expression was induced by drought and abscisic acid (ABA) treatment. TaFDL2-1A has transactivation activity and two activation domains, and the domain D region has different effects on the transcriptional activity of the two domains. Analysis of TaFDL2-1A overexpression plants indicated their enhanced drought tolerance and greater sensitivity to ABA. TabZIP8-7A was identified as a protein that interacts with TaFDL2-1A in the nucleus, and the overexpression of TabZIP8-7A conferred greater drought resistance and ABA sensitivity in Arabidopsis. Surprisingly, TaFDL2-1A × TabZIP8-7A double overexpression lines exhibited the highest drought resistance. Genetic and transcriptional regulation analyses demonstrated that stress-response gene transcription was initiated by TaFDL2-1A or TabZIP8-7A via the ABA signaling pathway. Importantly, TaFDL2-1A and TabZIP8-7A synergistically promoted ABA-inducible gene expression in a more efficient manner to form the transcriptional activation complex. Our findings provide new insights into the molecular mechanisms that allow bZIP TFs to regulate ABA signaling in response to drought stress.
A CBL-interacting Protein Kinase TaCIPK27 Confers Drought Tolerance and Exogenous ABA Sensitivity in Transgenic Arabidopsis.

Abstract:Drought is one of the major environmental stresses to plants. The calcium sensor, calcineurin B-like (CBL) proteins, and their interacting protein kinases (CIPK) play important roles in responding to abiotic stresses. In this study, we functionally characterized a CIPK gene from Triticum aestivum designated TaCIPK27. The transcriptional levels of TaCIPK27 were increased both in roots and leaves after treatment with polyethylene glycol 8000, abscisic acid and H2O2. Besides, TaCIPK27 interacted with AtCBL1, AtCBL3, AtCBL4, AtCBL5 and AtCBL9 in yeast two-hybrid assays. Ectopic overexpression of TaCIPK27 positively regulates drought tolerance in transgenic Arabidopsis compared with controls, which was demonstrated by seed germination and survival rates experiments, as well as the detection of physiological indices including ion leakage, malonic dialdehyde and H2O2 contents and antioxidant enzyme activities under normal and drought conditions. Moreover, higher concentration of endogenous abscisic acid was detected under drought in TaCIPK27 transgenic plants. In addition, TaCIPK27 transgenic plants were more sensitive to exogenous abscisic acid treatment at seed germination and seedling stage. The expression levels of somedrought stress and abscisic acid related genes were up-regulated in TaCIPK27 transgenic plants. The results suggest that TaCIPK27 functions as a positive regulator under drought partly in an ABA-dependent pathway.

Abstract:WRKYs are one of the largest transcription factor (TF) families and play an important role in plant resistance to various stresses. TaWRKY133, a group I WRKY protein, responds to a variety of abiotic stresses, including PEG treatment. The TaWRKY133 protein is located in the nucleus of tobacco epidermal cells, and both its N-terminal and C-terminal domains exhibit transcriptional activation activity. Overexpression of TaWRKY133 reduced drought tolerance in Arabidopsis thaliana, as reflected by a lower germination rate, shorter roots, higher stomatal aperture, poorer growth and lower antioxidant enzyme activities under drought treatment. Moreover, expression levels of stress-related genes (DREB2A, RD29A, RD29B, ABF1, ABA2, ABI1, SOD (Cu/Zn), POD1 and CAT1) were downregulated in transgenic Arabidopsis under drought stress. Gene silencing of TaWRKY133 enhanced the drought tolerance of wheat, as reflected in better growth, higher antioxidant enzyme activities, and higher expression levels of stress-related genes including DREB1, DREB3, ABF, ERF3, SOD (Fe), POD, CAT and P5CS. In conclusion, these results suggest that TaWRKY133 might reduce drought tolerance in plants by regulating the expression of stress-related genes.

Abstract:Osmotic stresses such as drought, salinity, and cold are major environmental factors that limit agricultural productivity. Transcription factors play essential roles in abiotic stress signaling in plants. Three TaMYB2 members were identified and designated TaMYB2A , TaMYB2B , and TaMYB2D based on their genomic origins. The cis -regulatory elements in the promoter regions were compared, and their diverse expression patterns under different abiotic stress conditions were identified. TaMYB2A was further characterized because of its earlier response to stresses. Subcellular localization revealed that TaMYB2A localized in the nucleus. To examine the role of TaMYB2A under various environmental stresses, transgenic Arabidopsis plants carrying TaMYB2A controlled by the CaMV 35S promoter were generated and subjected to severe abiotic stress. TaMYB2A transgenics had enhanced tolerance to drought, salt, and freezing stresses, which were confirmed by the enhanced expressions of abiotic stress-responsive genes and several physiological indices, including decreased rate of water loss, enhanced cell membrane stability, improved photosynthetic potential, and reduced osmotic potential. TaMYB2A is a multifunctional regulatory factor. Its overexpression confers enhanced tolerance to multiple abiotic stresses while having no obvious negative effects on phenotype under well-watered and stressed conditions; thus, TaMYB2A has the potential for utilization in transgenic breeding to improve abiotic stress tolerances in crops.
Overexpression of a Maize MYB48 Gene Confers Drought Tolerance in Transgenic Arabidopsis Plants

Abstract:MYBs are a vital family of transcription factors that play critical roles in plant development and stress response. However, knowledge concerning the functions of MYBs in the non-model plants is still limited. In this study, we isolated a R1-type MYB gene from maize ( Zea mays L.), designated as ZmMYB48 . Quantitative RT-PCR analysis demonstrated that ZmMYB48 expression was induced by drought and ABA treatments. Subcellular localization analysis revealed that ZmMYB48 protein was targeted to the nucleus in tobacco leaf epidermal cells. Transactivation assay in yeast demonstrated that ZmMYB48 had transcriptional activation ability. Heterologous overexpression of ZmMYB48 in Arabidopsis remarkably improved plant tolerance to drought stress, as determined through physiological analyses of survival rate, relative water content, malonaldehyde content, relative electrolyte leakage and proline content. Moreover, overexpression of ZmMYB48 enhanced the expression of stress/ABA-responsive genes such as P5CS1 , RD22 , RD29B and ABI1 . In addition, ZmMYB48 -overexpressing plants accumulated higher content of ABA than WT plants under drought stress. These results demonstrate that ZmMYB48 might act as a positive regulator that participates in the drought stress response through ABA signalling.
Wheat F-box Protein TaFBA1 Positively Regulates Plant Drought Tolerance but Negatively Regulates Stomatal Closure

Abstract:The phytohormone abscisic acid (ABA) regulates plant growth and development, as well as responses to various stresses, such as salt and drought. The wheat TaFBA1 gene, which encodes an F-box protein, was previously identified in our laboratory by homologous cloning. We previously found that TaFBA1 expression was induced by ABA and drought stress. In this study, wild-type (WT), TaFBA1 over-expressing (OEs), TaFBA1 homologous gene mutants, and TaFBA1 recovery (Rs) Arabidopsis plants were used. We found that the germination rate, the cotyledon greening rate, the root length, and the photosynthetic performance of TaFBA1 OE plants were better than those of WT under drought and ABA conditions, but mutant plants showed the opposite trend, and overexpression of TaFBA1 in mutants can recover their phenotype. In addition, TaFBA1 was found to be a negative regulator of ABA-induced stoma movement; mRNA transcription of certain ABA signaling-related genes was lower in TaFBA1 OE plants than in WT plants following ABA treatment. Further, we found that TaFBA1 can interact with RCAR1 (an ABA receptor) and ABI5. BiFC assay showed that TaFBA1 may interact with RCAR1 in the plasma membrane. In addition, accumulation of ROS and MDA in TaFBA1 OE plants was lower than that in the WT plants after ABA and drought treatments. Based on these results, we suggest that TaFBA1-regulated ABA insensitivity may be dependent on regulating ABA-mediated gene expression through interacting with RCAR1 and ABI5. Increased antioxidant competence and decreased ROS accumulation may be an important mechanism that underlies improved drought tolerance in TaFBA1 OE plants.

Abstract:Plants have developed various protective mechanisms to survive drought stress. Previously, it was shown that a wheat bZIP transcription factor gene TaFD-Like2-1A (TaFDL2-1A) can confer drought tolerance in Arabidopsis. However, the biological functions related to drought stress tolerance of TaFDL2-1A in wheat (Triticum aestivum L.) remain unclear. In the present study, overexpression of TaFDL2-1A in the wheat cultivar Fielder improved drought resistance and conferred abscisic acid (ABA) hypersensitivity. Further analysis showed that overexpression of TaFDL2-1A increased the hypersensitivity of stomata to drought stress and endogenous ABA content under drought conditions. Genetic analysis and transcriptional regulation analysis indicated that TaFDL2-1A binds directly to the promoter fragments of TaRAB21s and TaNCED2s via ACGT core cis-elements, thereby activating their expression, leading to enhanced ABA responses and endogenous ABA accumulation. In addition, our results demonstrate that overexpression of TaFDL2-1A results in higher SOD and GPX activities in wheat under drought conditions by promoting the expression of TaSOD1 and TaGPx1-D, indicating enhanced reactive oxygen species (ROS) scavenging. These results imply that TaFDL2-1A positively regulates ABA biosynthesis, ABA responses, and ROS scavenging to improve drought stress tolerance in transgenic wheat. Our findings improve our understanding of the mechanisms that allow the wheat bZIP transcription factor to improve drought resistance and provide a useful reference gene for breeding programs to enhance drought resistance.
The zinc-finger transcription factor ZFP8 negatively regulates the drought stress response in Arabidopsis thaliana by inhibiting the transcriptional activity of ABF2

Abstract:Drought stress limits plant growth and development. To cope with drought stress, abscisic acid (ABA) accumulates in plants. Although ABA-dependent drought tolerance pathways have been widely investigated, the feedback mechanisms and the negative regulatory roles within these pathways remain largely unknown. Here we characterize the roles of a C2H2 transcription factor, ZFP8, whose expression is repressed by ABA in the tolerance of drought stress. ZFP8-overexpressing plants were hyposensitive to ABA and exhibited less dehydration tolerance while ABA or drought-induced marker genes were more highly expressed in zfp8, suggesting that ZFP8 functions as a negative regulator in the ABA-mediated drought response. A transcriptome assay showed that ZFP8 positively regulates gene expression for cellular function and negatively regulates hormone and stress response gene expression. Moreover, we found that ZFP8 can interact with ABF2, one of the basic leucine zipper (bZIP) family transcription factor members, to inhibit its transcription activity. In conclusion, our results demonstrate a novel negative regulation pathway of ZFP8, which contributes to plants' ability to fine-tune their drought responses.
Upregulation of Wheat Heat Shock Transcription Factor TaHsfC3-4 by ABA Contributes to Drought Tolerance

Abstract:Drought stress can seriously affect the yield and quality of wheat (Triticum aestivum). So far, although few wheat heat shock transcription factors (Hsfs) have been found to be involved in the stress response, the biological functions of them, especially the members of the HsfC (heat shock transcription factor C) subclass, remain largely unknown. Here, we identified a class C encoding gene, TaHsfC3-4, based on our previous omics data and analyzed its biological function in transgenic plants. TaHsfC3-4 encodes a protein containing 274 amino acids and shows the basic characteristics of the HsfC class. Gene expression profiles revealed that TaHsfC3-4 was constitutively expressed in many tissues of wheat and was induced during seed maturation. TaHsfC3-4 could be upregulated by PEG and abscisic acid (ABA), suggesting that this Hsf may be involved in the regulation pathway depending on ABA in drought resistance. Further results represented that TaHsfC3-4 was localized in the nucleus but had no transcriptional activation activity. Notably, overexpression of TaHsfC3-4 in Arabidopsis thaliana pyr1pyl1pyl2pyl4 (pyr1pyl124) quadruple mutant plants complemented the ABA-hyposensitive phenotypes of the quadruple mutant including cotyledon greening, root elongation, seedling growth, and increased tolerance to drought, indicating positive roles of TaHsfC3-4 in the ABA signaling pathway and drought tolerance. Furthermore, we identified TaHsfA2-11 as a TaHsfC3-4-interacting protein by yeast two-hybrid (Y2H) screening. The experimental data show that TaHsfC3-4 can indeed interact with TaHsfA2-11 in vitro and in vivo. Moreover, transgenic Arabidopsis TaHsfA2-11 overexpression lines exhibited enhanced drought tolerance, too. In summary, our study confirmed the role of TaHsfC3-4 in response to drought stress and provided a target locus for marker-assisted selection breeding to improve drought tolerance in wheat.

Transgenic expression of MYB15 confers enhanced sensitivity to abscisic acid and improved drought tolerance in Arabidopsis thaliana

Abstract:Abiotic stresses cause serious crop losses. Knowledge on genes functioning in plant responses to adverse growth conditions is essential for developing stress tolerant crops. Here we report that transgenic expression of MYB15, encoding a R2R3 MYB transcription factor in Arabidopsis thaliana, conferred hypersensitivity to exogenous abscisic acid (ABA) and improved tolerance to drought and salt stresses. The promoter of MYB15 was active in not only vegetative and reproductive organs but also the guard cells of stomata. Its transcript level was substantially upregulated by ABA, drought or salt treatments. Compared with wild type (WT) control, MYB15 overexpression lines were hypersensitive to ABA in germination assays, more susceptible to ABA-elicited inhibition of root elongation, and more sensitive to ABA-induced stomatal closure. In line with the above findings, the transcript levels of ABA biosynthesis (ABA1, ABA2), signaling (ABI3), and responsive genes (AtADH1, RD22, RD29B, AtEM6) were generally higher in MYB15 overexpression seedlings than in WT controls after treatment with ABA. MYB15 overexpression lines displayed improved survival and reduced water loss rates than WT control under water deficiency conditions. These overexpression lines also displayed higher tolerance to NaCl stress. Collectively, our data suggest that overexpression of MYB15 improves drought and salt tolerance in Arabidopsis possibly by enhancing the expression levels of the genes involved in ABA biosynthesis and signaling, and those encoding the stress-protective proteins.
Abscisic acid-mediated autoregulation of the MYB41-BRAHMA module enhances drought tolerance in Arabidopsis

Abstract:Drought stress poses a substantial challenge to plant growth and agricultural productivity worldwide. Upon water depletion, plants activate an abscisic acid (ABA) signaling pathway, leading to stomatal closure to reduce water loss. The MYB family of transcription factors plays diverse roles in growth, development, stress responses and biosynthesis, yet their involvement in stomatal regulation remains unclear. Here, we demonstrate that ABA significantly upregulates the expression of MYB41, MYB74, and MYB102, with MYB41 serving as a key regulator that induces the expression of both MYB74 and MYB102. Through luciferase assays, chromatin immunoprecipitation (ChIP) assays and electrophoretic mobility shift assays (EMSA), we reveal that MYB41 engages in positive feedback regulation by binding to its own promoter, thus amplifying its transcription in Arabidopsis (Arabidopsis thaliana). Furthermore, our investigation showed that MYB41 recruits BRAHMA (BRM), the core ATPase subunit of the SWI/SNF complex, to the MYB41 promoter, facilitating the binding of HISTONE DEACETYLASE 6 (HDA6). This recruitment triggers epigenetic modifications, resulting in reduced MYB41 expression characterized by elevated H3K27me3 levels and concurrent decreases in H3ac, H3K27ac, and H3K14ac levels in wild-type plants compared to brm knockout mutant plants. Our genetic and molecular analyses show that ABA mediates autoregulation of the MYB41-BRM module, which intricately modulates stomatal movement in A. thaliana. This discovery sheds light on a drought response mechanism with the potential to greatly enhance agricultural productivity.
A NAC Transcription Factor, TaNAC5D-2, Acts As a Positive Regulator of Drought Tolerance Through Regulating Water Loss in Wheat (triticum Aestivum L.)

Abstract:NAC transcription factors play critical roles in regulating drought stress. However, the drought-responsive NAC transcription factors are largely unexplored in wheat. In this study, we isolated a NAC gene, TaNAC5D-2, and characterized the function of TaNAC5D-2 as a positive regulator of drought tolerance. The expression level of TaNAC5D-2 was induced under drought or abscisic acid treatment. Transient expression and transcriptional activity assays showed that TaNAC5D-2 is localized to the nucleus, and the C-terminal region exhibited transcriptional activity. Overexpression of TaNAC5D-2 in Arabidopsis thaliana led to greater drought tolerance, fresh weight and dry weight than wild-type plants, and the water loss was significant inhibited in the leaves of overexpression lines. Conversely, silencing of TaNAC5D-2 in wheat seedlings led to increased water loss and decreased growth compared to control and negative control plants under drought stress. RNA-Seq and RT-qPCR revealed that many Group A PP2C genes were upregulated in the TaNAC5D-2 silenced plants compared to negative control. And silencing TaNAC5D-2 negatively regulate ABA-induced stomatal closure that led to higher water loss. These data suggest that TaNAC5D-2 acts as a drought-responsive regulator, possibly by an abscisic acid-mediated stomatal closure to control water loss under drought condition.
Overexpression of TaWRKY53 enhances drought tolerance in transgenic Arabidopsis plants

Abstract:The WRKY transcription factors are involved in plant response to multiple biotic and abiotic stress. However, the biological functions of WRKY in wheat are rarely studied. In this work, we isolated a WKRK transcription factor gene TaWRKY53 from wheat. The full-length cDNA of TaWRKY53 is 1436 bp, containing an open reading frame of 1323 bp which encodes a protein of 440 amino acids. TaWRKY53 was identified as part of the WRKY IIc group, and a phylogenetic tree analysis suggested that TaWRKY53 had a closer genetic relationship with the OsWRKY53 of Oryza sativa and ZmWRKY53 of Zea mays . Expression pattern analysis revealed that TaWRKY53 was regulated by various stresses, including drought, salt, cold, ABA and H2O2 treatments. The TaWRKY53 gene was overexpressed in Arabidopsis thaliana. Under drought stress, overexpression of TaWRKY53 plants increased germination rate, survival rate, and relative water content. In addition, overexpression of TaWRKY53 plants generally displayed lower malondialdehyde (MDA) and H2O2 contents, but higher peroxidase (POD), superoxide dismutase (SOD), and catalase (CAT) activities as well as higher chlorophyll content under drought stress. Our results indicate that TaWRKY53 is a Cd-inducible expansin gene, further providing valuable information for crop improvement.

A Wheat R2R3 MYB Gene TaMpc1-D4 Negatively Regulates Drought Tolerance in Transgenic Arabidopsis and Wheat

Abstract:MYB transcription factors (TFs) are one of the largest TF families, and R2R3-type MYB TFs participate in the multiply abiotic stress responses in wheat. In this study, an R2R3-type MYB gene Myb protein colourless I located on chromosome D (named TaMpc1-D4), was cloned from wheat. TaMpc1-D4-GFP protein was localized in the nucleus. Overexpression of TaMpc1-D4 reduced drought tolerance in transgenic Arabidopsis lines, which was supported by the lower germination rate, the shorter root length, a higher level of O-2(center dot-) and malonaldehyde (MDA), the decreased proline content, and limited activities of peroxidase (POD), superoxide dismutase (SOD), and catalase (CAT). Furthermore, P5CS1, RD29A, RD29B, DREB2A, ABF3, CBFI, CBF2, CBF3, ERFI, PODI, SOD (Cu/Zn), and CATI genes related to the stress and antioxidant system were remarkably down-regulated in TaMpc1-D4 transgenic Arabidopsis lines under drought stress. Silencing TaMpc1-D4 expression in wheat enhanced the relative water content (RWC), the proline content, and the activities of antioxidant enzymes, and activated stress-related and antioxidant-related genes (DREBI, DREB3, ERF3, ERF4b, ABF, P5CS, POD, SOD (Fe), and CAT). Taken together, these results indicated that TaMpc1-D4 negatively modulated drought tolerance by regulating the capacity of the enzyme system and the expression of stress-related and antioxidant-related genes.
AtMYB32 Regulates the ABA Response by Targeting ABI3, ABI4 and ABI5 and the Drought Response by Targeting CBF4 in Arabidopsis.

Abstract:The Arabidopsis thaliana R2R3-MYB transcription factor AtMYB32 and its homologs AtMYB4 and AtMYB7 play crucial roles in the regulation of phenylpropanoid metabolism. In addition, AtMYB4 and AtMYB7 are involved in the response to abiotic stress. However, the function of AtMYB32 remains unclear. In this study, we found that AtMYB32 is induced by abscisic acid (ABA) and repressed by drought stress. AtMYB32 positively regulates ABA-mediated seed germination and early seedling development. The expression of ABSCISIC ACID-INSENSITIVE 3 (ABI3), ABI4 and ABI5, which encode key positive regulators of ABA signaling, was upregulated in response to ABA in AtMYB32-overexpressing plants and downregulated in the atmyb32-1 mutant. In addition, we found that the atmyb32-1 mutant was drought resistant. Consistent with the drought-resistant phenotype, the transcript levels of C-repeat binding factor 4 (CBF4) were higher in the atmyb32-1 mutant in response to drought stress. Electrophoretic mobility shift assays (EMSAs) and chromatin immunoprecipitation (ChIP) assays revealed that AtMYB32 binds directly to the ABI3, ABI4, ABI5 and CBF4 promoters both in vitro and in vivo. Genetically, ABI4 was found to be epistatic to AtMYB32 for ABA-induced inhibition of seed germination and early seedling development. Taken together, our findings revealed that AtMYB32 regulates the ABA response by directly promoting ABI3, ABI4 and ABI5 expression and that the drought stress response likely occurs because of repression of CBF4 expression.
Wheat Transcription Factor TaAREB3 Participates in Drought and Freezing Tolerances in Arabidopsis

Abstract:AREB (ABA response element binding) proteins in plants play direct regulatory roles in response to multiple stresses, but their functions in wheat (Triticum aestivum L.) are not clear. In the present study, TaAREB3, a new member of the AREB transcription factor family, was isolated from wheat. Sequence analysis showed that the TaAREB3 protein is composed of three parts, a conserved N-terminal, a variable M region, and a conserved C-terminal with a bZIP domain. It belongs to the group A subfamily of bZIP transcription factors. TaAREB3 was constitutively expressed in stems, leaves, florets, anthers, pistils, seeds, and most highly, in roots. TaAREB3 gene expression was induced with abscisic acid (ABA) and low temperature stress, and its protein was localized in the nucleus when transiently expressed in tobacco epidermal cells and stably expressed in transgenic Arabidopsis. TaAREB3 protein has transcriptional activation activity, and can bind to the ABRE cis-element in vitro. Overexpression of TaAREB3 in Arabidopsis not only enhanced ABA sensitivity, but also strengthened drought and freezing tolerances. TaAREB3 also activated RD29A, RD29B, COR15A, and COR47 by binding to their promoter regions in transgenic Arabidopsis. These results demonstrated that TaAREB3 plays an important role in drought and freezing tolerances in Arabidopsis.
