Abstract:Mitochondria, as "cell energy stations", are involved in the regulation of various cell functions. Recent investigations revealed that mitochondrial dysfunction that can cause an intracellular viscosity mutation, a process that is associated with an increasing number of diseases that are not curable or manageable. However, conventional viscometers cannot be used to monitor the viscosity changes in living cells and in vivo. In order to cater to the complex biological environment, we present a chemical toolbox, MI-BP-CC, that employs N,N-diethyl and double bonds as sensitive sites for viscosity based on the TICT mechanism (twisted intramolecular charge transfer) to monitor the viscosity of living cells and fatter liver mice. MI-BP-CC features good mitochondrial targeting and a near-infrared emission. Surprisingly, in the presence of viscosity, the MI-BP-CC probe exhibited an ultrasensitive model for viscosity detection showing a red fluorescence signal from a silent "off" state to "on". More importantly, utilizing the satisfactory detection performance of MI-BP-CC, we have successfully visualized increased viscosity under the pathological models of Parkinson's (PD) and fatty liver mice. We anticipate that these findings will provide a convenient and efficient tool to understand physiological functions of viscosity in more biosystems.

Abstract:Studies have shown that the changes of mitochondrial viscosity which is close related to its state can be affected by the destruction of ion balance, excessive production of reactive oxygen species, apoptosis and other factors. However, these processes are usually connected to mitochondrial depolarization, causing probes with cationic mitochondrial targeting groups to escape from the mitochondria. As a result, it is impossible to perform high-fidelity imaging of mitochondrial viscosity changes. To solve this problem, we report a novel viscosity probe (MF-V1) which is able to investigate the changes of mitochondrial viscosity under mitochondrial depolarization. Interestingly, MF-V1 exhibited excellent performance, including large ''off-on'' fluorescence response (130-fold), large Stokes shift (119 nm), and high fluorescence quantum yield in glycerol (0.64). More importantly, MF-V1 has been successfully used to monitor viscosity changes of mitochondria under normal and depolarized conditions. Additionally, the new probe was also used to detect viscosity changes in zebrafish and mice, affording a powerful tool for investigating the relationship between mitochondrial viscosity and related diseases. Abstract:Intracellular viscosity plays an important role in regulating cellular morphology and physiology and is closely related to a host of diseases. Especially, the changes in mitochondrial viscosity will cause some common diseases such as hyperlipidemia, Alzheimer's disease and cancer. In this work, we report the design of a red-emissive molecular rotor for the detection of mitochondrial viscosity in live cells. The probe showed fascinating performance, such as specific targeting to mitochondria, high sensitivity to viscosity, and rapid fluorescence response, especially the dual response mode of fluorescence intensity and fluorescence lifetime. By using this probe, we realized monitoring of the mitochondrial viscosity variations in live cells under different physiological processes. Our study offers an opportunity to discover potential tools for mitochondria-related physiology and pathology investigation. A D-pi-A typed fluorescence lifetime probe for sensitively detecting viscosity has been designed and synthesized. BSOH has been successfully applied to real-time monitoring mitochondrial viscosity in live cells by fluorescence lifetime imaging.

Abstract:It is meaningful to accurately quantify the changes in local viscosity within the mitochondria of living cells, because viscosity influences mitochondrial network organization and metabolite diffusion. Although many molecular probes targeting mitochondria have been reported, almost all of them are not fixed to the mitochondria. Thus, they may not be suitable for sensing in abnormal mitochondria with lowered potential. In order to monitor viscosity in all mitochondria, no matter their working or health status, we develop the first fixable BODIPY (boron-dipyrromethene) rotor, named Vis-A. Vis-A contains an aldehyde group as an anchor to react with amino groups of proteins, which make it immobilizable in mitochondria. Vis-B, the reference compound without such anchor unit, is also synthesized. Both Vis-A and Vis-Bshow excellent mitochondrial targetability, as good as the commercially available mitochondrial dye (Mito Tracker Deep Red). However, when cells are chemically treated to decrease the mitochondrial potential, only Vis-A continues emitting strong fluorescence in mitochondria, but the signals of Vis-B and Mito Tracker Deep Red completely disappear. This comparison confirms that Vis-A not only specifically localizes in mitochondria, but also can be stably retained there. Then, Vis-A is applied to detect the mitochondrial viscosity of living cells by Fluorescence Lifetime Imaging (FLIM). Especially, with the aid of Vis-A, the changes in viscosity under typical pathological conditions (i.e., treatment with rotenone and carbonylcyanide-m-chlorophenylhydrazone (CCCP)) for mitochondria are monitored by FLIM.

Abstract:Viscosity, at the subcellular level, plays a crucial role as a physicochemical factor affecting microenvironment homeostasis. Abnormal changes in mitochondrial viscosity often lead to various diseases in the organism. Based on the twisted intramolecular charge transfer mechanism, four hemicyanine dye fluorescent probes (HT-SA, HT-SA-S, HT-Bzh, and HT-NA) were designed and synthesized for viscosity response. The single bond between the nitrogen-containing heterocycle and the carbon-carbon double in the structure of the probe bond served as the viscosity response site. Finally, the probe HT-Bzh was screened as the optimal mitochondrial viscosity probe according to its responsiveness, targeting, and interference resistance. The fluorescence intensity of the probe HT-Bzh increased 22-fold when the viscosity was increased from 13.75 to 811.2 cP. In summary, all four viscosity probes we have developed can be used in different applications depending on the external environment, providing a valuable reference for the design of potential tools to address viscosity monitoring in biological systems.

Abstract:Aberrant mitochondrial viscosity is closely associated with many diseases and cellular malfunctions. Thus, the development of reliable methods for monitoring mitochondrial viscosity variations has attracted considerable attention. Herein, through stepwise structural modulation of the dihydroxanthene fluorophore (DHX), we developed three NIR fluorescent probes, named DHX-V-1-3, for detecting mitochondrial viscosity. Among them, DHX-V-3 displayed the highest signal-to-noise ratio (67-fold) for viscosity with outstanding selectivity and showed excellent mitochondria targeting and immobilization ability. At the cellular level, the DHX-V-3 probe was successfully applied to image the mitochondrial viscosity in live cells upon treatment with lipopolysaccharide (LPS) or nystatin. Moreover, benefiting from its NIR emission and the increased depth of tissue imaging, DHX-V-3 demonstrated the ability to visualize the increased viscosity in LPS-treated mice.

Abstract:Viscosity is a key factor that determines the diffusion-controlled processes in biological systems. The matrix of mitochondria contains various enzymes and other proteins with high density, and the diffusion of which are severely restricted by the cristae, making it the most crowded place in the cells. Herein, we reported a new near-infrared probe NV-1 with increased Stokes shift for monitoring viscosity changes in mitochondria. A remarkable increase of the fluorescence was observed in glycerol compared with which was observed in methanol at 744 nm. The probe was applied for measuring viscosity changes not only in mitochondria, but also in vivo (in zebra fishes and mice).

Abstract:Abnormal viscosity variations in the mitochondrial matrix may induce changes in the mitochondrial network organization and further influence metabolite diffusion, which, consequently, are related to cellular aspects of several diseases and malfunctions, such as cell malignancy, atherosclerosis, diabetes, and Alzheimer's disease. Studies have shown that mitochondrial viscosity changes are influenced by many factors, including disruption of ionic balance by drugs, over-production of reactive oxygen species, and cell apoptosis. However, the quantization of mitochondrial viscosity changes caused by these factors is uncertain, mainly because of the lack of a suitable fluorescent viscosity probe to accurately analyze changes in mitochondrial viscosity. By combining two fluorophores that have different sensitivities to viscosity, we designed and synthesized a new mitochondrial-targeted two-photon fluorescent viscosity probe (TM-V). The probe showed a direct linear relationship between the fluorescence intensity ratio of the two fluorophores and the media viscosity, which allowed us to determine the average mitochondrial viscosity in living cells. Using the novel TM-V probe, changes in mitochondrial viscosity under various conditions were quantified. In addition, as an excellent two-proton rotor probe, TM-V was able to report on viscosity in living cells and deeper tissues by ratiometric fluorescence imaging. (C) 2018 Elsevier B.V. Abstract:Abnormal changes in the mitochondrial microenvironment can cause a variety of diseases. Consequently, based on the twisted intramolecular charge transfer (TICT) theory, three hemicyanine fluorescent probes 1a-c were designed and developed by linking the phenanthridine group with different electron donor groups for the dynamic detection of mitochondrial viscosity in this paper. They have the advantages of near-infrared emission, excellent pH stability and viscosity sensitivity. Optical experiments showed that the maximum absorption and emission wavelengths of the probes in different solvents located at 411 - 529 nm and 623 - 684 nm with large Stokes shifts of 133 - 230 nm. In addition, when the viscosity increased from 0.89 cP to 856 cP, their fluorescence intensities in glycerol enhanced by 115-fold, 83-fold and 303-fold compared with water, respectively. The fluorescence intensities of probes showed excellent fit to the Forster Hoffmann equation, and exhibited nearly OFF-ON response to viscosity. Moreover, the probe 1a with good photostability and low cytotoxicity could target mitochondria. It was verified that 1a also could detect variation in mitochondrial viscosity during autophagy by modelling starvation and drug-induced (rapamycin and nystatin) in HeLa cells. Therefore, this study provides a useful probe for the analysis of mitophagy process in HeLa cells by the viscosity variation of mitochondria.

Abstract:Cell viscosity is related to some diseases, such as diabetes, atherosclerosis, and Alzheimer's disease. These diseases can cause abnormal viscosity of the cell mitochondrial matrix. 1,4-Dihydropyridine (DHP) is an important organic compound with biological activity and is widely used in drug research. However, there are few studies on its optical properties, especially in the design of viscous fluorescent probes. In this study, a fluorescent probe for viscosity detection using 1,4-dihydropyridine as the fluorophore and indole iodide salt as the recognition group was designed and synthesized. The probe has the advantages of a deep-red emission, low cytotoxicity, good biocompatibility and excellent anti-interference ability. In addition, the probe also has the ability to target mitochondria and has been successfully applied to the detection of the viscosity response of HeLa cells and living mice, and has good clinical application potential.

Abstract:Mitochondria are the powerhouses in cells, providing the energy needed for cellular activities. However, the abnormalities in the mitochondrial microenvironment (e.g., the increased viscosity) can lead to mitochondrial dysfunctions and diseases. Herein, we develop a series of near-infrared (NIR) fluorescence probes for the detection of viscosity. After screening, probe CQ-4 is selected since it shows a great fluorescence enhancement (89-fold) in the NIR window. Its specific response to viscosity is not influenced by pH, polarity and biological species. Under stimulation with monensin or nystatin, CQ-4 can measure the cellular viscosity changes with good biocompatibility. In addition, we can observe an increase of viscosity during starvation. CQ-4 is applied to distinguish cancer cells from normal cells based on the viscosity differences. Furthermore, the probe has been successfully applied to image viscosity in inflamed and tumor-bearing mice in vivo. Therefore, CQ-4 may contribute to the future study about viscosity in the physiological and pathological processes.

Abstract:As a fundamental physical parameter, viscosity influences the diffusion in biological processes. The changes in intracellular viscosity led to the occurrence of relevant diseases. Monitoring changes in cellular viscosity is important for distinguishing abnormal cells in cell biology and oncologic pathology. Here, we devised and synthesized a viscosity-sensitive fluorescent probe LBX-1. LBX-1 showed high sensitivity, providing a large Stokes shift as well as an enhancement in fluorescent intensity (16.1-fold) from methanol solution to glycerol solution. Furthermore, the probe LBX-1 could localize in mitochondria because of the ability of the probe to penetrate the cell membrane and accumulate in mitochondria. These results suggested that the probe could be utilized in monitoring the changes in mitochondrial viscosity in complex biological systems.

Abstract:Viscosity strongly influences intracellular transportation of mass and signal, and although bulk viscosity can be well measured with macroscopic samples quantities, microscale measurements in live cells still remain a challenge. By introducing a CHO group into the meso-position of the pentamethine chain, a thiazole-Cy5 dye based two-photon fluorescence probe, Mito-V, was developed. It exhibited a selective response to solution viscosity by fluorescence ratio and lifetime methods. More importantly, Mito-V can localize in mitochondria and quantitatively image its viscosity changes by the dual fluorescence mode during the apoptosis.

Abstract:The visual detection of mitochondrial viscosity is of considerable significance in that it is associated with various cellular functions. Herein, a novel red-emitting viscosity probe YD-Mito constructed by linking phenylamine derivatives and benzo[e]indolium through a C–C double bond has been designed and synthesized. It was found that YD-Mito exhibited a special response to viscosity accompanying with a sharp fluorescence enhancement (20-fold) at 620 nm. Importantly, YD-Mito can precisely localized in mitochondria independent from membrane potential and visualized the mitochondrial viscosity. Therefore, probe YD-Mito could provide a powerful tool for the determination of mitochondrial viscosity.
