Abstract:Biofuel is a renewable and sustainable energy as an substitute of fossil fuel. In China, the key challenge to develop biofuels is how to balance the competition of food and bioenergy and supply a sustainable feedstock. Sweet sorghum is highlighted as a multiple platform plant which can supply grain, sugar, cellulose and feed. In addition, sweet sorghum has so good tolerance of drought and sterilize that it can grow on marginal land. However, the loose stalk pith of sweet sorghum results in the infeasibility of liquid fermentation technology due to the considerably high energy consumption.In present research, an integrated process combined advanced solid-state fermentation technology (ASSF) and alkaline pretreatment was presented in this work. Soluble sugars in sweet sorghum stalks were firstly converted into ethanol by ASSF using crushed stalks directly. Then, the operation combining ethanol distillation and alkaline pretreatment was performed in one distillation-reactor simultaneously. The process was optimized according to the yield of sugar-ethanol and amounts of monosaccharide released from SSB. In our novel process, energy consumption of mechanically squeeze and pretreatment of SSB was avoided. Meanwhile, the recalcitrance of lignocellulose was destructed and the biodegradation of lignocellulose into chemical products is feasible to achieve. Cellulosic ethanol as a model product was studied by using this novel integrated process. The mass balance of the overall process was calculated, and 91.9 kg was achieved from one tonne of fresh sweet sorghum stalk. Energy consumption for raw materials preparation and pretreatment were reduced or avoided in our process. To produce 1 tonne of ethanol, the energy input in this process was 12,481.2 MJ/tonne, while the energy input in other cellulosic ethanol processes is from 17,430 to 33,330 MJ/tonne.Based on this technology, the recalcitrance of lignocellulose was destructed via a cost-efficient process and structural carbohydrates in sweet sorghum stalks were hydrolysed into fermentable sugars. Bioconversion of fermentable sugars released from sweet sorghum bagasse into different products except ethanol, such as butanol, biogas, and chemicals was feasible to operate under low energy-consumption conditions. So it is considered as a promising process to produce biofuels at commercial scale. (C) 2014 Published by Elsevier Ltd.

Abstract:Background: Sweet sorghum is regarded as a very promising energy crop for ethanol production because it not only supplies grain and sugar, but also offers lignocellulosic resource. Cost-competitive ethanol production requires bioconversion of all carbohydrates in stalks including of both sucrose and lignocellulose hydrolyzed into fermentable sugars. However, it is still a main challenge to reduce ethanol production cost and improve feasibility of industrial application. An integration of the different operations within the whole process is a potential solution.Results: An integrated process combined advanced solid-state fermentation technology (ASSF) and alkaline pretreatment was presented in this work. Soluble sugars in sweet sorghum stalks were firstly converted into ethanol by ASSF using crushed stalks directly. Then, the operation combining ethanol distillation and alkaline pretreatment was performed in one distillation-reactor simultaneously. The corresponding investigation indicated that the addition of alkali did not affect the ethanol recovery. The effect of three alkalis, NaOH, KOH and Ca(OH)(2) on pretreatment were investigated. The results indicated the delignification of lignocellulose by NaOH and KOH was more significant than that by Ca(OH)(2), and the highest removal of xylan was caused by NaOH. Moreover, an optimized alkali loading of 10% (w/w DM) NaOH was determined. Under this favorable pretreatment condition, enzymatic hydrolysis of sweet sorghum bagasse following pretreatment was investigated. 92.0% of glucan and 53.3% of xylan conversion were obtained at enzyme loading of 10 FPU/g glucan. The fermentation of hydrolyzed slurry was performed using an engineered stain, Zymomonas mobilis TSH-01. A mass balance of the overall process was calculated, and 91.9 kg was achieved from one tonne of fresh sweet sorghum stalk.Conclusions: A low energy-consumption integrated technology for ethanol production from sweet sorghum stalks was presented in this work. Strain engineering of Bacillus coagulans with high osmotic pressure tolerance for effective L-lactic acid production from sweet sorghum juice under unsterile conditions

Abstract:Open unsterile fermentation of the low-cost non-food crop, sweet sorghum, is an economically feasible lactic acid biosynthesis process. However, hyperosmotic stress inhibits microbial metabolism and lactic acid biosynthesis, and engineering strains with high osmotic tolerance is challenging. Herein, heavy ion mutagenesis combined with osmotic pressure enrichment was used to engineer a hyperosmotic-tolerant Bacillus coagulans for L-lactic acid production. The engineered strain had higher osmotic pressure tolerance, when compared with the parental strain, primarily owing to its improved properties such as cell viability, cellular antioxidant capacity, and NADH supply. In a pilot-scale open unsterile fermentation using sweet sorghum juice as a feedstock, the engineered strain produced 94 g/L L-lactic acid with a yield of 91 % and productivity of 6.7 g/L/h, and optical purity of L-lactic acid at the end of fermentation was 99.8 %. In short, this study provided effective and low-cost approach to produce polymer-grade L-lactic acid.

Abstract:Lignocellulosic pretreatment is an important stage in biomass utilization, which usually requires high input. In this study, a low-cost method using combined ensiling and NaOH was developed for lignocellulosic pretreatment. Sweet sorghum bagasse (SSB) was ensiled for 21 days and then treated with diluted NaOH (0%, 1%, and 2%) for fermentation. The results showed that the application of Lactobacillus plantarum (L) reduced fermentation losses of the silages, mainly low water-soluble carbohydrate (WSC) and ammonia nitrogen loss. Meanwhile, the application of Lactobacillus plantarum and ensiling enzyme (LE) promoted lignocellulosic degradation, as evidenced by low neutral detergent fiber (NDF), acid detergent fiber (ADF), lignin (ADL), and hemicellulosic (HC) contents. The dominant bacterial genera were Lactobacillus, uncultured_bacterium_f_Enterobacteriaceae , and Pantoea after silage, which corresponded to the higher lactic acid and acetic contents and lower pH. The reducing sugar yields of SSB increased after combined pretreatment of silage and NaOH and were further enhanced by the 2% NaOH application, as evidenced by the high reducing sugar yield and microstructure damage, especially in the L-2% NaOH group and the LE-2% NaOH group, in which the reducing sugar yields were 87.99 and 94.45%, respectively, compared with those of the no additive control (CK)-0 NaOH group. Therefore, this study provides an effective method for SSB pretreatment to enhance biomass conservation.

Abstract:Sweet sorghum can be used to produce a substantial quantity of biofuel due to its high biological yield and high carbohydrate content. In this study, we investigated the dynamic changes in fermentation characteristics, carbohydrate components, and the bacterial community during the ensiling of wilted and unwilted sweet sorghum. The results revealed a rapid fermentation pattern and high-quality fermentation quality in wilted and unwilted sweet sorghum, wherein lactic acid, and acetic acid accumulated and stabilized during the initial 9 days of ensiling, with the pH values less than 4.2, until 60 days of ensiling. We found that the ensiling of sweet sorghum involved the degradation (5% ~ 10%) of neutral detergent fiber (NDF) and hemicellulose and that the degradation of NDF fit a first-order exponential decay model. A shift in dominance from Lactococcus to Lactobacillus occurred before the first 9 days of ensiling, and the abundance of Lactobacillus (r = -0.68, p < 0.001) was negatively correlated with the NDF content. The relative abundances of Lactobacillus in wilted and unwilted sweet sorghum after ensiling for 60 days were 76.30 and 93.49%, respectively, and relatively high fermentation quality was obtained. In summary, ensiling is proposed as a biological pretreatment for sweet sorghum for subsequent biofuel production, and unlike other materials, sweet sorghum quickly achieves good fermentation quality and has great potential for bioresource production.

Abstract:The preservation of sugars in sweet sorghum stalks is one of the key blocks on the way to realize the industrialization of sweet sorghum talks as a feedstock of fuel ethanol.The "antiseptic and anaerobic" method for sweet sorghum stalks storage was proposed and validated.Two kinds of antiseptics(potassium sorbate and sodium dehydroacetate) were investigated during the anaerobic storage of sorghum stalks at different conditions.The sugar and water contents in sweet sorghum stalks can be maintained effectively by using "antiseptic-anaerobic" storage.The sugar loss rate is less than 5.95% after 6 months,and the sugar conversion rate is above 93.43%.

Abstract:Sweet sorghum, with its multi-platform of resources of starch, sugar and cellulose, is considered a very promising energy crop to produce bioethanol. In this study, bioethanol production of sodium hydroxide (NaOH) pretreated sweet sorghum bagasse without solid substrate washing was investigated. The results showed that direct enzymatic saccharification and ethanol fermentation of NaOH pretreated solid substrate without washing significantly improved the fermentable sugars conversion and ethanol theoretical yield from 44.06±0.93% and 44.85±1.15% to 65.14±0.19% and 61.81±1.28%, respectively. Experiments where NaOH pretreated liquor (SL) was added into the thoroughly washed NaOH pretreated solid substrate showed the fermentable sugars conversion yield improved significantly. Furthermore, analysis of enzymes activities during enzymatic saccharification showed that SL maintained cellulase activity significantly, especially for that of xylanase activity. These results suggested that the NaOH pretreated lignocellulosic solid can be employed for direct enzymatic saccharification and subsequent ethanol fermentation without washing. This process would improve the sustainability and economic viability of bioethanol production.

Abstract:Sweet sorghum, as a seasonal energy crop, is rich in cellulose and hemicellulose that can be converted into biofuels. This work aims at investigating the effects of synergistic regulation of Pichia anomala and cellulase on ensiling quality and microbial community of sweet sorghum silages as a storage and pretreatment method. Furthermore, the combined pretreatment effects of ensiling and ball milling on sweet sorghum were evaluated by microstructure change and enzymatic hydrolysis. Based on membership function analysis, the combination of P. anomala and cellulase (PA + CE) significantly improved the silage quality by preserving organic components and promoting fermentation characteristics. The bioaugmented ensiling with PA + CE restructured the bacterial community by facilitating Lactobacillus and inhibiting undesired microorganisms by killer activity of P. anomala . The combined bioaugmented ensiling pretreatment with ball milling significantly increased the enzymatic hydrolysis efficiency (EHE) to 71%, accompanied by the increased specific surface area and decreased pore size/crystallinity of sweet sorghum. Moreover, the EHE after combined pretreatment was increased by 1.37 times compared with raw material. Hence, the combined pretreatment was demonstrated as a novel strategy to effectively enhance enzymatic hydrolysis of sweet sorghum.

[Ethanol production from sweet sorghum stalks by advanced solid state fermentation (ASSF) technology]

Abstract:A robust strain of the species Saccharomyces cerevisiae CGMCC1949 was screened and identified, and advanced solid state fermentation (ASSF) technology for fuel ethanol production from sweet sorghum stalks was thus developed. The fermentation time was shortened to less than 30 h, and ethanol yield was 92% of its theoretical maximum. And in the meantime, the cost-effective storage was established for sweet sorghum stalks, with less than 5% sugar loss after 200 days of storage, making the plant operation could extend up to 200 days without feedstock shortage. With the fermentation kinetics and heat-mass transfer models, modeling of the ASSF process was investigated, and the rotating drum bioreactor was designed. Furthermore, the ASSF technology was successfully applied in the pilot plant in which the rotating drum bioreactor was scaled up to 127 m3, and ethanol yield of 91% was achieved. At the end, techno-economic analysis (TEA) conducted by ASPEN indicated that ethanol production from sweet sorghum stalks by the ASSF is economically competitive.

Abstract:Introduction: Sweet sorghum juice is a typical production feedstock for natural, eco-friendly sweeteners and beverages. Clostridium tyrobutyricum is one of the widely used microorganisms in the food industry, and its principal product, bio-butyric acid is an important food additive. There are no published reports of Clostridium tyrobutyricum producing butyric acid using SSJ as the sole substrate without adding exogenous substances, which could reach a food-additive grade. This study focuses on tailoring a cost-effective, safe, and sustainable process and strategy for their production and application. Methods: This study modeled the enzymolysis of non-reducing sugars via the first/second-order kinetics and added food-grade diatomite to the hydrolysate. Qualitative and quantitative analysis were performed using high-performance liquid chromatography, gas chromatography-mass spectrometer, full-scale laser diffraction method, ultra-performance liquid chromatography-tandem mass spectrometry, the cell double-staining assay, transmission electron microscopy, and Oxford nanopore technology sequencing. Quantitative real-time polymerase chain reaction, pathway and process enrichment analysis, and homology modeling were conducted for mutant genes. Results: The treated sweet sorghum juice showed promising results, containing 70.60 g/L glucose and 63.09 g/L fructose, with a sucrose hydrolysis rate of 98.29% and a minimal sucrose loss rate of 0.87%. Furthermore, 99.62% of the colloidal particles and 82.13% of the starch particles were removed, and the concentrations of hazardous substances were effectively reduced. A food microorganism Clostridium tyrobutyricum TGL-A236 with deep utilization value was developed, which showed superior performance by converting 30.65% glucose and 37.22% fructose to 24.1364 g/L bio-butyric acid in a treated sweet sorghum juice (1:1 dilution) fermentation broth. This titer was 2.12 times higher than that of the original strain, with a butyric acid selectivity of 86.36%. Finally, the Genome atlas view, Gene Ontology (GO), Kyoto Encyclopedia of Genes and Genomes (KEGG), and evolutionary genealogy of genes: Non-supervised Orthologous (eggNOG) functional annotations, three-dimensional structure and protein cavity prediction of five non-synonymous variant genes were obtained. Conclusion: This study not only includes a systematic process flow and in-depth elucidation of relevant mechanisms but also provides a new strategy for green processing of food raw materials, improving food microbial performance, and ensuring the safe production of food additives.
Co-fermentation of Stalk Juice and Clarified Grain Mash of Sweet Sorghum for Ethanol Production

Abstract:Co-fermentation of stalk juice and clarified grain mash of sweet sorghum was carried out with immobilized active dry yeast in a conical flask fermenter at 30 degrees C. The results showed that co-fermentation of stalk juice and clarified grain mash can shorten fermentation time and improve ethanol efficiency compared with the control. Under the experimental conditions, the maximum ethanol concentration of 65.64 +/- 0.57 gL(-1), ethanol productivity of 5.47 +/- 0.04 gL(-1) h(-1), and ethanol yield efficiency of 89.29 +/- 0.77% were obtained from the juice with 80% volume fraction of clarified grain mash, which were 6.8, 33.5, and 13.7% higher than those of using sweet sorghum juice alone, respectively.

Abstract:Coproduction of H2 and volatile fatty acids from sweet sorghum stalks was successfully developed via two-step fermentation process involving alkaline and enzymatic treatment of the residual slurry obtained from the first step fermentation of the raw material. The optimum treatment conditions to obtain the highest yield of the end products included 2% (w/v) alkali at 120 °C for the residual slurry and 32 FPU cellulase per gram of the alkali-treated materials. The two-step fermentation using Clostridium thermosaccharolyticum increased the end product yields of H2 (6.37 mmol/g-substrate) by 95%, acetic acid (2.33 g/L) by 97% and butyric acid (2.36 g/L) by 143% compared with those obtained in the single step fermentation without any treatment. As a result, the H2 energy recovery efficiency of the two-step fermentation process with commercially attractive butyric acid as a coproduct reached 10.54%. These results provide a promising approach for sweet sorghum biorefining.

Abstract:Key technologies and main research contents of ethanol production from sweet sorghum stalk are introduced. More specifically, current researches and existing problems including the breeding and cultivation of sweet sorghum, stalk storage, ethanol fermentation and integrated utilization of fermentation byproducts are investigated. The research indicates that the high yield sweet sorghum variety should be bred by modern breeding technology, thus increasing the biomass yield of sweet sorghum; the storage technology of sweet sorghum stem should be researched according to different fermentation technologies in order to extend the supplying time of raw material; high efficiency fermentation technology and equipment should be developed in order to increase ethanol yield; in terms of byproduct utilization, the production of protein feedstock, paper pulp, plank should be developed and extended in short term in order to increase economic benefit of the project, and ethanol production from cellulose of sweet sorghum stalk should be developed in the long run.
Compacted Silage Fermentation on Whole Sweet Sorghum Plant for Distributed Bioethanol Production

Abstract:The development of the bioethanol industry is facinga bottleneckdue to challenges in the collection and storage of biomass. In thisstudy, we explored the use of compacted silage fermentation to preservewhole pulverized sweet sorghum. To sterilize native and contaminantbacteria, we employed sulfur dioxide and inoculated SO2-tolerant Saccharomyces cerevisiae TSH4 for long-term preservation in an ethanol atmosphere. Baggedsilage experiments present the most effective preservation dosage,which was 2000 ppm SO2 concentration and 2% TSH4 yeastinoculation. Based on these conditions, we found that compacted silagewith different compaction densities enabled all fermentations to becompleted within 60 days. The highest ethanol concentration is achievedat a compaction density of 600, at which ethanol conversion rate reachesto 93.4%. Higher compaction densities produce more leachate, whichresults in ethanol loss up to 9.7%. During the 240 days of long-termstorage, there was no significant change in ethanol content despitean increase in acetic acid concentration. Additionally, distilledvinasse was found to be comparable to whole silage maize in termsof nutrients, thus proving the feasibility of constructing distributedbiofuel plants. Compactedsilage fermentation on whole sweet sorghum plantyielding both green ethanol and high-quality feed is promising fordistributed biofuel industrialization.
Optimization of fermentation conditions for the production of ethanol from stalk juice of sweet sorghum by immobilized yeast using response surface methodology

Abstract:Optimization of three parameters, including initial total sugar concentration, supplement rate of (NH4)(2)SO4, and particles stuffing rate, was attempted using response surface methodology based on a Box-Behnken design for the optimal production of ethanol by immobilized yeast fermentation of stalk juice of Liaotian number 1 sweet sorghum cultivar in shaking flasks. The correlation analysis of the mathematical regression model indicated that the quadratic polynomial model could be employed to optimize ethanol production. The optimum conditions were found to be an initial total sugar concentration of 22.88%, supplement rate of (NH4)(2)SO4 of 0.244%, and particles stuffing rate of 25.15%. At the optimum conditions, the maximum predicted ethanol yield of 93.83% was obtained. The ethanol yield and fermentation time of verification experiments in the shaking flask were 92.37% and 14 h at the corresponding parameters, respectively, while they were 93.23% and 13 h, respectively, in a 5 1, bioreactor, in which the predicted value of ethanol yield was very close to experimental values. In addition, the fermentation time of the stalk juice of sweet sorghum was about 3-4 times shorter with immobilized yeast than that of conventional fermentation technology. Thus, by immobilized yeast fermentation of the stalk juice of sweet sorghum, the Box-Behnken design was found to be the favorable strategy investigated with respect to the optimization of fermentation conditions for ethanol production.

Abstract:This paper aimed to use the major part of carbohydrate fractions in enzymatic hydrolysate of sweet sorghum bagasse, acetone-butanol-ethanol (ABE) fermentation was performed following the ethanol fermentation of hexoses. In batch ethanol fermentation stage, 50.8 ± 3.2 g/L of ethanol was produced from 111.5 g/L of glucose in hydrolysate. After ethanol recovery by batch vacuum distillation, 123.6 ± 15.6 g/L of ethanol was obtained in the distillate, while 47.2 ± 2.5 g/L of xylose was remained in the fermentation broth. The ethanol-free broth was further used as the substrate for ABE fermentation. Compared with yeast remaining scenario, the cell-free ethanol fermentation broth showed a better performance in ABE production. 7.37 ± 0.33 g/L of butanol and 10.93 ± 0.54 g/L of ABE solvent were produced after 120 h of batch fermentation. Based on the biorefinery strategy that cascade two-types of fermentation process, the overall solvents yield boosted significantly, about 144.8 g of ethanol, 17.3 g of butanol and 4.8 g of acetone can be produced from 1 kg sweet sorghum bagasse.

Abstract:This study presents a cost-effective strategy for producing organic acids from glucose and xylose using the acidtolerant yeast, Issatchenkia orientalis. I. orientalis was engineered to produce lactic acid from xylose, and the resulting strain, SD108XL, successfully converted sorghum hydrolysates into lactic acid. In order to enable lowpH fermentation, a self -buffering strategy, where the lactic acid generated by the SD108XL strain during fermentation served as a buffer, was developed. As a result, the SD108 strain produced 67 g/L of lactic acid from 73 g/L of glucose and 40 g/L of xylose, simulating a sugar composition of sorghum biomass hydrolysates. Moreover, techno-economic analysis underscored the efficiency of the self -buffering strategy in streamlining the downstream process, thereby reducing production costs. These results demonstrate the potential of I. orientalis as a platform strain for the cost-effective production of organic acids from cellulosic hydrolysates.

Abstract:Hydrogen, volatile fatty acids (VFAs), and methane coproduction from sweet sorghum stems using bacterial consortium was investigated as an efficient and sustainable pre-treatment strategy to improve energy recovery. Integrated two-stage dark fermentation and methanization approach aimed to reduce fractionation, juice extraction, and pre-treatment steps to improve the efficiency and sustainability of stalks energy bioconversion. Stems biomass loading did not significantly influence hydrogen and VFAs productivities. Energy recovery yields were (7.07) and (10.01) MJ/kg dry matter (DM), respectively, for raw stem single dark fermentation (DF) and methanization processes, corresponding to 41.22% and 58.37% of raw stalk energy potential. Methanogenic potential increase of 31.9% and energy bioconversion yield of 13.21 MJ/kg DM were reached for solid residues from DF (80.75% of their energy content), suggesting that bacterial consortium efficiently pre-treated sorghum stalk fibers. Coupling process led to 88.74% net biomass energy recovery yield, corresponding respectively to 57.38% and 40.23% more than single DF and methanization. Fiber degradation ability of DF bacterial consortium significantly contributed to improve sorghum stalk energy recovery efficiency and cost-competitiveness.
