Abstract:Mandelic acid and its derivatives are important chiral analogs which are widely used in the pharmaceutical synthetic industry. The present study investigated the enantiomeric separation of six mandelic acids (mandelic acid, 2‐chloromandelic acid, 3‐chloromandelic acid, 4‐chloromandelic acid, 4‐bromomandelic acid, 4‐methoxymandelic acid) on the Chiralpak AD‐3 column by supercritical fluid chromatography. The influences of volume fraction of trifluoroacetic acid, type and percentage of modifier, column temperature, and backpressure on the separation efficiency were investigated. And the enantiomer elution order was determined. The results show that, for a given modifier, the retention factor, the separation factor, and the separation resolution decreased gradually with increasing the volume ratio of the modifier. At the same volume ratio of modifier, the retention factor of the mandelic acid and its derivatives increased in the order of methanol, ethanol, and isopropanol, except 3‐chloromandelic acid. The separation factor and the separation resolution decreased with the increase of column temperature (below the temperature limit). The backpressure affected the enantioseparation process: As the backpressure increased, a corresponding decrease in retention factor was observed. Under the same chiral column conditions, the SFC method exhibited faster and more efficient separation with better enantioselectivity than the HPLC method.
R-(-)-acetyl-o-mandelic acid preparation via enzymatic dynamic kinetic resolution

Abstract:In this study,asymmetric hydrolysis of acetyl chloromandelic acid was catalyzed by Pseudomonas sp. ECU1011,and the S-(-)-chloromandelic acid in the resolution mixture was then racemerized using the mutated mandelate racemase and fed back into the hydrolysis reaction after acylation , realizing enzymatic dynamic kinetic resolution for preparation of R-(-)-acetyl chloromandelic acid. Firstly,the conditions for enzymatic resolution,separation of resolution mixture, and enzymatic racemization were optimized. Subsequently,dynamic kinetic resolution of acetyl chloromandelic acid was conducted under the optimal conditions,obtaining optically pure R-(-)-acetyl chloromandelic acid with final yield of 80%. The conditions for enzymatic resolution,chloromandelic acid recycling and racemization were optimized in this study,leading to preliminary establishment of the dynamic kinetic resolution process for R-(-)-acetyl chloromandelic acid preparation,with guiding significance for future industrial applications.
Investigation on the origin of 5-HMF in Shengmaiyin decoction by RP-HPLC method

Abstract:The origin of 5-HMF (5-hydroxymethyl-2-furaldehyde) in a Shengmaiyin decoction was investigated by the RP-HPLC method below. AC 18 column (250 mm×4.6 mm, i.d. 5 μm) with a column temperature of 25°C was used. The mobile phase was a mixture of ultra-pure water-acetonitrile (95∶3, V/V ) and the flow rate was 1.0 ml/min. The detection wavelength was 280 nm. The injection volume was 1 μl and the running time was about 20 min. The addition of Schisandra was regulated to assess the contribution of an acid envrionment to the production of 5-HMF. In order to confirm the role of saccharides in the production of 5-HMF, different amount of fructose was used. The 5-HMF level in decoctions of processed and unprocessed Schisandra was investigated in order to determine the origin of 5-HMF. The results showed that 5-HMF was derived mainly from the decoction of Schisandra only and not the mixed decocition of Ophionpogon and Schisandra . The appearance of 5-HMF is not simply the result of the decomposition of saccharides under the acid environment created by Schisandra , but the processing procedure plays an important role in the production of 5-HMF.
Preparation Method for Reference Substances of Hyperin and Isorhamnetin-3-O-galactoside

Abstract:Objective To establish a method for preparing the reference substances of hyperin and isorhamnetin- 3- O- galactoside. Methods Methanol- extract of Fructus Evodia was isolated and purified by combining solvent extraction with column chromatography and recrystallization. Hyperin and isorhamnetin- 3- O- galactoside were identified by mass spectrometry (MS), 1H nuclear magnetic resonance (1HNMR ) and 13C nuclear magnetic resonance (13CNMR) and their content was determined by HPLC. Results The purity was 99.6 % and 98.0 % for hyperin and isorhamnetin- 3- O- galactoside respectively. Conclusion This method is simple cancl effective to yield high- purity products. And these high- purity products can be used as reference substances for the research of herbal medicine.

Abstract:The influence of post-treatment of as-synthesized mordenite membranes in the hydrochloric acid solution was investigated on the membrane pervaporation performance for the separation of a water/ethanol mixture. It was shown that the water/ethanol separation factor of as-synthesized mordenite membranes could be greatly improved upon post-treatment with hydrochloric acid under controlled conditions. The change in the pervaporation performance of mordenite membranes upon post-treatment was discussed.

Abstract:2-Methyl-2-hydroxymethyl propaldehyde first formed by reaction of formaldehyde with isobutylaldehyde in the presence of triethyl amine, was reduced with formaldehyde by Cannizzaro reaction in the presence of alkali catalyst, and gained product neopentyl glycol and byproduct sodium formate. The product was confirmed to be neopentyl glycol with GC-MS as well as melting point. The total yield of 94%, purity above 98% and melting point of 122~123°C for neopentyl glycol product were obtained under optimized reaction conditions and with an improved purification method. The production cost of neopentyl glycol was decreased as the by-produced sodium formate was also an available chemical product.
Effect of the mobile phase on the retention behavior of optical isomers of the mandelic acid derivative methyl 2-phenyl-2-(tetrahydropyranyloxy) acetate by chiral HPLC.

Abstract:Conditions for separation of enantiomers of a mandelic acid derivative, methyl 2-phenyl-2-(tetrahydropyranyloxy) acetate (the analyte) were studied. Because of the presence of two chiral carbons, the analyte consists of four stereoisomers; stable at ambient temperature. Chiral HPLC of the analyte resulted in four peaks, using an (S,S)Whelk-O1 column with the mobile phase consisting of hexane and the t-butyl methyl ether (TBME). It was found that TBME dramatically changed the retention of the isomers, though it produced the best enantioseparation on (S,S)-Whelk-O1. The amount of TBME in the mobile phase influenced the degree of retention shift; 5% (v/v) TBME gave a bigger shift than 8% (v/v) and 10% (v/v). 2-Propanol did not produce the same results. The chiral separation was also tried on cellulose tris (3, 5-dimethyl phenylcarbamate) (CDMPC), but only three peaks were seen, indicating some but not full enantiomer resolution.

Abstract:The enantio,separation of D,L-alpha-amine acid on(+)-( 18-crown-6) -2,3,11,12-tertcarboxyl acid chiral stationary phase ( CSP), and the influence of the methanol and acid addictives on the separation were investigated. The chiral recognition mechanism was also discussed. It is possible that the solute R-NH2 was protoned to R-NH3+, and the fitness and interaction in the crown ester cavity between the solutes and CSP were the dominant chiral recognition mechanism for the solutes. The concentration of methanol and composition of acid additive influenc ed the equilibrium of R-NH3+ between mobile phase and stationary phase, which resulted in the chiral recognition. In addition, the influence of column temperature on the enantiosepartion of phenylalanine and tyrosine was also investigated. And all the tested temperatures were lowered than the isotemperature T = 467. 3 K. The result showed that the configuratoin of stationary phase didn't have evidence changed, and reservation mechanism and enantiotropy selection action remained changeless. Delta Delta H-0 and Delta Delta S-0 were negative which showed that the enantioseparation'of two solutes was enthalpy control.

Abstract:Background Quality control of the herbal medicines (HMs) still hits technical bottlenecks due to their complicated multi-step pharmaceutical processes. Nuclear magnetic resonance (NMR) spectroscopy could serve as a powerful technique able to obtain comprehensive information of chemical compounds in the manufacturing process of HMs by a single analytical test. In this study, ethanol precipitation of Salvia miltiorrhiza Radix et Rhizoma (Danshen in China), a critical purification process of a particularly commonly used herbal medicinal material, was employed as the research subject. Methods Processed with different variables, including ethanol-material ratio, apparent ethanol concentration and moisture concentration of the raw material, 40 lab-scale experiments of ethanol precipitation were orchestrated by a full factor design with additional central composite circumscribed experiments. After 1H NMR tests, chemometrics methods, including orthogonal partial least squares-discriminant analysis (OPLS-DA), principle component analysis (PCA) and multiple linear regression (MLR), were applied to analyze the spectra of samples. Results 16 metabolites were recognized as the most critical potential chemical markers in the process of Danshen ethanol precipitation by 1H NMR spectra and OPLS-DA, including γ-aminobutyrate, pyroglutamate, succinate, acetate, malonate, fructose, sucrose, raffinose, stachyose, mannotriose, danshensu, lithospermic acid, protocatechuic aldehyde, rosmarinic acid, salvianolic acid A and B. PCA reasonably distinguished all the process intermediates as a function of different process variables adopted during the ethanol precipitation. MLR made the statistical interpretation by quadratic equations to characterize how do significant variables affect the chemical markers. Conclusions Our study indicates that NMR can track detailed potential chemical markers of ethanol precipitation process, which could promote the further understanding of ethanol precipitation process and open up possibilities to develop a more effective process quality control and consistency evaluation methodology of an HM, through providing a more in-depth and improved theoretical process understanding and characterization.

Abstract:Mandelic acid and its derivatives are important medical intermediates in the pharmaceutical industry. Different stereoisomers exhibited distinct biological properties to human bodies. Given that, enantioselective recognition and separation of mandelic acid are of great importance. In this study, four novel different types of chiral ionic liquids bearing designed functional groups were synthesized and successful enantioselective precipitation with mandelic acid and its derivatives. That is, (R, R)-chiral ionic liquid 1 can coprecipitated with S-mandelic acid and its derivatives was observed. In addition, good correlation coefficient is achieved by using electrospray mass spectrum at negative ion pattern for quick analysis of the enantioselective precipitation, which could be served as a method of enantioselective recognition. The possible intermolecular interactions are established after systematical studies by NMR spectroscopy and DFT calculations.

Abstract:Forty one microorganisms belonging to different taxonomical groups were used to carry out the enantioselective reduction of methyl benzoylformate to afford the corresponding (R)-methyl mandelate, with moderate to high ee. In contrast, the monooxygenase enzyme in Helminthosporium sp. CIOC3.3316 catalyzed the hydroxylation of methyl 2-phenylacetate to (S)-methyl mandelate. This combination of oxidation and reduction biotransformations thus provides a method for preparing the enantiomers of chiral α-hydroxy acid derivatives.

Abstract:Mandelic acid is a valuable chemical that is commonly used in the synthesis of various drugs, in antibacterial products, and as a skin care agent in cosmetics. As it is an important chemical, various methods are used to synthesize and extract this compound. However, the yields of the used processes is not significant. A dilute aqueous solution is obtained when using several production methods, such as a fermentation, etc. In this study, the reactive extraction of mandelic acid from aqueous solutions using tri-n-octylamine extractant at 298.15 K was investigated. Dimethyl phthalate (DMP), methyl isobutyl ketone (MIBK), 2-octanone, 1-octanol, n-pentane, octyl acetate, and toluene were used as diluents. The batch extraction results of the mandelic acid experiments were obtained for the development of a process design. Calculations of the loading factor (Z), distribution coefficient (D), and extraction efficiency (E%) were based on the experimental data. The highest separation yield was obtained as 98.13% for 0.458 mol.L−1 of tri-n-octylamine concentration in DMP. The overall extraction constants were analyzed for the complex of acid-amine by the Bizek approach, including K11, K12, and K23.

Abstract:The separation of ten epimeric aromatic acid (-)-menthol esters by countercurrent chromatography with hydroxypropyl-beta-cyclodextrin as the mobile phase additive was investigated, and methods for the analysis of all the epimeric esters by reversed-phase high-performance liquid chromatography were established. A biphasic solvent system composed of n-hexane/20-70% methanol containing 50 mmol/L of hydroxypropyl-beta-cyclodextrin (1: 1, v/v) was selected, which provided high separation factors for five of the epimeric esters, and successful separations by countercurrent chromatography were achieved. The complete separation of five pairs of epimeric ester was obtained with the purity being over 98% for each peak fractions, as determined by highperformance liquid chromatography. The recovery of each analyte from the eluted fractions reached around 80-88%.

Abstract:The equilibrium model in enantioseparation of mandelic acid by chiral ligand exchange countercurrent chromatography was investigated using N-n-dodecyl-l-proline as chiral ligand and cupric ion as central metal. Important parameters, including physical partition coefficient and formation constants of binary and ternary coordination complexes in the two-phase solvent system, were determined. This equilibrium model could give an excellent prediction of distribution ratio and enantioseparation factor of the analyte in the biphasic solvent system, which was further verified by experiments. All the average relative deviations were less than 12%, indicating that the established model could provide a simple computational approach for optimization of enantioseparation conditions in chiral ligand exchange countercurrent chromatography.
Separation and Preparation of High-purity Huperzine A by Medium Pressure Preparative Chromatography

Abstract:OBJECTIVE To establish a method for the preparation of high-purity huperzine A from the extract of Huperzia serrata.METHODS By using the polymer reversed phase medium and C1s reversed phase medium and two steps of medium pressure preparation in series,huperzine A monomer was isolated and prepared from the Huperzia serrata extract.RESULTS The purity of 99.7％ huperzine A monomer was obtained,and the overall recovery rate was 87.5％.CONCLUSION This method utilizes the characteristics of polymer medium and C18 medium to provide an efficient,green and economical way to obtain pharmaceutical grade huperzine A through technological linkage.
Separation of the Mixture of Mandelonitrile and Mandelonitrile Acetate by Solvent Extraction

Abstract:In this paper, a solvent extraction system is applied to separate the mixture of mandelonitrile (MN) and mandelonitrile acetates (MA) for the first time. The basic consistence of the system is water and n-hexane with the phase ratio of 1.0. Operation parameters such as temperature, the pH value in the aqueous phase, and the concentration of the mixture and benzaldehyde were investigated to find the optimal conditions. When the temperature was 30 degrees C and the pH value in the aqueous phase was 2.0, a three-stage extraction was performed. The purity of MA in the organic phase was more than 99% (wt) with the yield of 87.7%. And the purity of MN in the aqueous phase was 98.8 % (wt) with the yield of 92.6%. This extraction system can separate the mixture of MN and MA efficiently.
Synthesis and application of novel silver magnetic amino silicone adhesive particles for preparation of high purity α-linolenic acid from tree peony seed oil under applied magnetic field

Abstract:Silver magnetic amino silicone adhesive (Fe3O4@SiO2@NH2@Ag) particles were prepared for the purification of α-linolenic acid from tree peony seed oil under applied magnetic field. First, Fe3O4@SiO2@NH2@Ag particles were prepared and physicochemically characterized, including XRD, TG, FTIR, SEM, magnetic hysteresis curves and elemental analysis. The static process for the purification of α-linolenic acid using Fe3O4@SiO2@NH2@Ag particles was investigated, including adsorption curve, desorption curve, elution solvent composition and adsorption isotherm. The result indicated that 0-1-4% acetone-n-hexane elution solvent was selected for the gradient elution process, 2 h and 60 min were the time required to reach adsorption and desorption equilibrium, 20 °C was selected as the adsorption temperature, Langmuir model was suitable to fit and explain the equilibrium data, and the adsorption process was spontaneous and exothermic. Under applied magnetic field, the dynamic process for the purification of α-linolenic acid using Fe3O4@SiO2@NH2@Ag particles was investigated, and the optimum conditions were 20:1 µL/g loading amount, 0.5 mL/min flow rate and 51.73 Oe magnetic field intensity. After purification, the purity and recovery ratio of α-linolenic acid were calculated to be 94% and 74%, respectively. Furthermore, the recycled Fe3O4@SiO2@NH2@Ag particles still achieved better purification result. Therefore, the developed method shows a good application prospect in the field of separation and purification of α-linolenic acid.

Abstract:The effect of chromatographic conditions including type and proportion of alcohol modifiers and column temperature to resoluting enantiomers of the O,O-dimethyl-1-(phenoxyacetoxy)ethylphosphonate and O,O-dim-ethyl-1-(4-methylphenoxyacetoxy)ethylphosphonate using amylose-tris-(3,5-dimethylphenylcarbamate) chiral sta-tionary phase(chiralpak AD-H columns) on High-performance Liquid Chromatography were studied.Thermodynami-cal mechanism of enantio separation have also been preliminary discussed.In this study,baseline separation of the enantiomers of both analytes were obtained with a mobile phase of n-hexane/isopropanol(90:10,by vol) at a flow rate of 1.0 mL/min under 25 oC on the Chiralpak AD-H column.

Abstract:Background Vanillic acid, an aromatic carboxylic acid and a derivative of vanillin, is used as an additive in fast‐moving consumer goods for its aroma. Previously, the majority of the vanillic acid was produced through a petrochemical route, but due to stringent environmental policies, it is now produced by the biotransformation of ferulic acid. Vanillic acid is also found in the waste‐water streams of the paper‐pulp industries and olive‐oil mills. So, recovering vanillic acid from fermentation broth, either during bioproduction or from a waste‐water stream, while minimising toxicity, is a difficult task. In order to reduce the toxicity substantially, bio‐based solvents such as sunflower, mustard, and rice bran oils were employed as diluents along with tri‐n‐butyl phosphate (TBP) as the extractant in order to recover vanillic acid by reactive extraction. Results The extraction performance was elucidated using terms such as distribution ratio, extraction efficiency, loading ratio, and equilibrium complexation constant for different extractant‐diluent combinations. The highest extraction efficiency and distribution ratio for the different solvents were in the following order: TBP + sunflower‐oil (72.02% ‐ 92.47% ; 2.57 – 12.29) > TBP + rice bran‐oil (65.56% ‐ 90.55% ; 1.90 – 9.58) > TBP + mustard‐oil(64.03% ‐ 89.87% ; 1.78 – 8.87). The theoretical number of stages (NTS) was found to be 4 to achieve the targeted extraction efficiency of 90%, which were found out experimentally. Conclusion In this study, vanillic acid was recovered from the aqueous phase using tri‐n‐butyl phosphate in sustainable, environmentally friendly, bio‐based solvents such as rice bran oil, sunflower oil, and mustard oil. Abstract:A high performance liquid chromatography-diode array detection-electrospray ionization tandem mass spectrometry method was developed for analysis and identification of impurities in lovastatin. Lovastatin and impurities were separated on a Diamonsil C-18 column (5 mu m,4. 6 mm x 250 mm) using acetonitrile-water (containing 0.1% acetic acid) (65:35) as mobile phase. The elutes were detected with diode array detector and ion trap mass spectrometer in positive mode. The ultraviolet (UV), MS and MS' spectra of each chromatographic peak were obtained. The UV spectra of the impurities are similar with those of lovastatin except for dihydrolovastatin. It indicated that they have the same basic structures. Then the [M+H](+) ions and their MS2 spectra were used for determining the molecule mass and side-chain. According to analyses of the UV, MS and MS' as well as the comparison with the published literature, the structures of ten impurities were identified. The results indicated that the described method could be effectively applied to perform identification of impurities in lovastatin.
