The RING-H2 type E3 ubiquitin ligase gene MdATL16 positively regulates salt tolerance in transgenic tomato and apple

Abstract:Salt stress is a major abiotic stress which severely affects crop growth, yield and quality. Arabidopsis Toxicos en Levadura (ATL) proteins comprise a sub-family of the RING-H2 type E3 ubiquitin ligases and play important roles in response to abiotic stress. However, the functions of ATL homologs have not been deeply investigated in apple, which is an economically important fruit tree and is widely planted in the world. In this study, we cloned and characterized the MdATL16 gene from apple. MdATL16 is strongly expressed in roots and is significantly induced by NaCl treatment, and its function was preliminarily investigated by the genetic transformation of tomato and apple callus. Ectopic expression of MdATL16 in transgenic tomato increased salt stress tolerance, and this was associated with lower accumulation of hydrogen peroxide (H 2 O 2 ) and superoxide radicals (O 2 • – ), and less electrolyte leakage and malondialdehyde (MDA). Overexpression of MdATL16 increased apple callus tolerance to salt, along with inducible expression of antioxidant and stress marker genes and significant decrease of H 2 O 2 and O 2 • – content. These findings indicate MdATL16 is an important regulator involved in plant response to salt tolerance, which may provide a favorable target gene for genetic applications to improve drought stress tolerance in apples.
