Abstract:As the fastest mode of transport, the aircraft is a major driver for globalization and economic growth. The development of alternative advanced liquid fuels is critical to sustainable development within the sector. Such fuels should be compatible with existing infrastructure and derived from second generation feedstocks to avoid competition with food markets. With properties similar to petroleum based fuels, isoprenoid derived compounds such as limonene, bisabolane, farnesane, and pinene dimers are of increasing interest as "drop-in" replacement jet fuels. In this review potential isoprenoid derived jet fuels and progress toward their microbial production was discussed in detail. Although substantial advancements have been achieved, the use of first generation feedstocks remains ubiquitous. Lignocellulosic biomass is the most abundant raw material available for biofuel production, however, technological constraints associated with its pretreatment and saccharification hinder its economic feasibility for low-value commodity production. Non-conventional microbes with novel characteristics including cellulolytic bacteria and fungi capable of highly efficient lignocellulose degradation and xylose fermenting oleaginous yeast with enhanced lignin-associated inhibitor tolerance were investigated as alternatives to traditional model hosts. Finally, innovative bioprocessing methods including consolidated bioprocessing and sequential bioreactor approaches, with potential to capitalize on such unique natural capabilities were considered.
Microbial alkane production for jet fuel industry: motivation, state of the art and perspectives

Abstract:Bio-jet fuel has attracted a lot of interest in recent years and has become a focus for aircraft and engine manufacturers, oil companies, governments and researchers. Given the global concern about environmental issues and the instability of oil market, bio-jet fuel has been identified as a promising way to reduce the greenhouse gas emissions from the aviation industry, while also promoting energy security. Although a number of bio-jet fuel sources have been approved for manufacture, their commercialization and entry into the market is still a far way away. In this review, we provide an overview of the drivers for intensified research into bio-jet fuel technologies, the type of chemical compounds found in bio-jet fuel preparations and the current state of related pre-commercial technologies. The biosynthesis of hydrocarbons is one of the most promising approaches for bio-jet fuel production, and thus we provide a detailed analysis of recent advances in the microbial biosynthesis of hydrocarbons (with a focus on alkanes). Finally, we explore the latest developments and their implications for the future of research into bio-jet fuel technologies.

Abstract:There is an immediate need to drastically reduce the emissions associated with global fossil fuel consumption in order to limit climate change. However, carbon-based materials, chemicals, and transportation fuels are predominantly made from fossil sources and currently there is no alternative source available to adequately displace them. Gas-fermenting microorganisms that fix carbon dioxide (CO2) and carbon monoxide (CO) can break this dependence as they are capable of converting gaseous carbon to fuels and chemicals. As such, the technology can utilize a wide range of feedstocks including gasified organic matter of any sort (e.g., municipal solid waste, industrial waste, biomass, and agricultural waste residues) or industrial off-gases (e.g., from steel mills or processing plants). Gas fermentation has matured to the point that large-scale production of ethanol from gas has been demonstrated by two companies. This review gives an overview of the gas fermentation process, focusing specifically on anaerobic acetogens. Applications of synthetic biology and coupling gas fermentation to additional processes are discussed in detail. Both of these strategies, demonstrated at bench-scale, have abundant potential to rapidly expand the commercial product spectrum of gas fermentation and further improve efficiencies and yields.

Abstract:Concerns over climate change have necessitated a rethinking of our transportation infrastructure. One possible alternative to carbon-polluting fossil fuels is biofuels produced by engineered microorganisms that use a renewable carbon source. Two biofuels, ethanol and biodiesel, have made inroads in displacing petroleum-based fuels, but their uptake has been limited by the amounts that can be used in conventional engines and by their cost. Advanced biofuels that mimic petroleum-based fuels are not limited by the amounts that can be used in existing transportation infrastructure but have had limited uptake due to costs. In this Review, we discuss engineering metabolic pathways to produce advanced biofuels, challenges with substrate and product toxicity with regard to host microorganisms and methods to engineer tolerance, and the use of functional genomics and machine learning approaches to produce advanced biofuels and prospects for reducing their costs.

Abstract:Aviation sector discharges approximately 2% of the global anthropogenic CO2, and the proportion is growing. The search for cost-effective and environmental-friendly bio-jet fuels derived from natural resources is gaining momentum. The microalgae cultivation conditions including temperature, pH, light intensity and nutrients have shown significant influence on the microalgae growth rate and chemical composition, which create the opportunities to enhance the yield and quality of microalgae bio-jet fuel. This review is focused on the hydroprocessing method for converting microalgae oil into bio-jet fuel, as well as the novel conceptual approaches for bio-jet fuel production such as gasification with Fischer-Tropsch and sugar-to-jet. Fischer-Tropsch synthesis of biomass is one of the best alternative ways to replace natural aviation fuel due to the high maximum energy efficiency and low emission of greenhouse gas. In addition, hydroprocessing with the aid of Ni and zeolites catalysts has successfully converted the microalgae biodiesel to bio-jet fuel with high yield and alkane selectivity. Among these techniques, hydroprocessing used the lowest production cost with the longest duration, whereas the bio-jet fuel with high selectivity (C8–C16) could be produced by using gasification with the Fischer-Tropsch process. Consequently, gasification and Fischer-Tropsch and sugar-to-jet can become the future alternative process to convert microalgae to bio-jet fuel. The development of microalgae bio-jet fuel will increase the security of energy supply and reduce the fuel expenses in aviation industry.

Abstract:Due to the large amount of greenhouse gas emissions and the high dependence on fossil fuels, the sustainable development of aviation industry has attracted worldwide attention. Bio-jet fuel is considered to be a promising alternative to traditional aviation fuel. This article summarizes the representative technological route, development status, opportunities and challenges faced by the development of bio-jet fuel industry. So far, several bio-jet fuel production technologies have been certified by the American Society for Testing and Materials (ASTM). Hydroprocessed esters and fatty acids is currently the most mature process that can be fully commercialized. Considering economic characteristics and technology maturity, Fischer-Tropsch is promising in near term.

Abstract:Low carbon fuels and chemicals can be sourced from renewable materials such as biomass or from industrial and municipal waste streams. Gasification of these materials allows all of the carbon to become available for product generation, a clear advantage over partial biomass conversion into fermentable sugars. Gasification results into a synthesis stream (syngas) containing carbon monoxide (CO), carbon dioxide (CO2), hydrogen (H2) and nitrogen (N2). Autotrophy-the ability to fix carbon such as CO2 is present in all domains of life but photosynthesis alone is not keeping up with anthropogenic CO2 output. One strategy is to curtail the gaseous atmospheric release by developing waste and syngas conversion technologies. Historically microorganisms have contributed to major, albeit slow, atmospheric composition changes. The current status and future potential of anaerobic gas-fermenting bacteria with special focus on acetogens are the focus of this review.

Abstract:This paper describes commonly used processes to produce aviation fuel and alternative routes with potential production yields for sustainable aviation fuels (SAF) like HEFA and ATJ. It also presents the possible sources (crude oil, refinery processes), causes (filter clogging, engine failure), and forms of contamination in both conventional and alternatively produced aviation fuels. Special attention is focused on the threats of fuel contamination with solid particles/trace elements, water, microorganisms, and fatty acid methyl esters (FAME). This review also presents the standard and novel advanced methods (ICP-MS, MALDI, ViPA) for identifying contaminations in aviation fuel. It also identifies possible ways to control and eliminate the risk of contamination, such as the fallowing coherent JIG system to ensure the quality of aviation fuel. Another approach that is very interesting and worth considering for future development is the idea of predictive maintenance and machine learning in monitoring and detecting contamination.

Abstract:Concerns have been raised about the effects of fossil fuel combustion on global warming and climate change. Fuel consumer behavior is also heavily influenced by factors such as fluctuating fuel prices and the need for a consistent and reliable fuel supply. Microalgae fuel is gaining popularity in the aviation industry as a potential source of energy diversification. Microalgae can grow in saltwater or wastewater, capture CO 2 from the atmosphere and produce lipids without requiring a large amount of land. As a result, the production of oil from microalgae poses no threat to food availability. The low carbon footprint of microalgae-derived fuels has the potential to mitigate the impact of traditional aviation fuels derived from petroleum on climate change and global warming. Therefore, aviation fuels derived from microalgae have the potential to be a more environmentally friendly and sustainable alternative to conventional fuels. Gathering microalgal species with a high lipid content, drying them, and turning them into aviation fuel is an expensive process. The use of biofuels derived from microalgae in the aviation industry is still in its infancy, but there is room for growth. This study analyses the potential routes already researched, their drawbacks in implementation, and the many different conceptual approaches that can be used to produce sustainable aviation fuel from microalgal lipids. Microalgae species with fast-growing rates require less space and generate lipids that can be converted into biofuel without imperiling food security. The key challenges in algal-based aviation biofuel include decreased lipid content, harvesting expenses, and drying procedure that should be enhanced and optimized to increase process viability.

Abstract:An integrated thermochemical-biochemical Biomass-to-Liquid (BtL) pathway for the production of aviation and maritime liquid fuels from biogenic residues is introduced. The presence of a semi-commercially proven technology like Dual Fluidized Bed Gasification (DFBG) ensures extended fuel flexibility, syngas of high quality, complete fuel conversion, and optimal heat integration while avoiding CAPEX (Capital Expenditure) intensive equipment like air separation unit. Then, a two-stage biochemical route is proposed: initially syngas fermentation (anaerobic) into acetate and subsequently acetate fermentation (aerobic) into targeted triglycerides (TAGs) that will be finally purified and hydrotreated to form the desired drop-in biofuels. The tolerance of the bacteria to syngas contaminants minimizes the gas cleaning requirements. Moreover, the low-pressure requirements (1–10 bar) along with the mild operating temperatures (30–60 °C) reduce drastically the capital and operational cost of the process. The biological process of syngas fermentation inherently has limited side products, a fact that reduces the risk of deactivation of hydrotreatment catalysts. Heat and mass balances are calculated for the proposed concept via full-scale process simulations in Aspen PlusTM assuming a thermal input of 200 MW th with crushed bark as feedstock. Three different operational scenarios are examined mainly through overall performance indicators such as carbon utilization (CU) and energetic fuel efficiency (EFE). Competitive performance compared to technologies that exploit similar feedstock (i.e., biogenic residues) was noticed, since values in the range of 22–27% and 31–37% were obtained for the CU and EFE, respectively. The aim of this study is to determine the appropriate key process specifications and assess the potential of the proposed concept compared to other competitive technologies.

Abstract:The growing demand for aviation fuel has driven the pursuit of sustainable and eco-friendly alternatives to traditional petroleum-based fuels. The development of sustainable aviation fuel (SAF) through the conversion of greenhouse gases into triacylglycerol (TAG) by microorganisms has emerged as a promising field of research. In this study, Methylomicrobium buryatense 5GB1S was metabolically engineered for the first time to accumulate TAG by introducing heterologous genes encoding enzymes involved in TAG biosynthesis and manipulating carbon flux for endogenous intermediates. The carbon, nitrogen, and energy metabolisms in TAG-producing M. buryatense 5GB1S were elucidated by transcriptomic analyses to further facilitate TAG production. Ultimately, a self-activated two-stage cultivation was developed in 3 L fermenters to maximize TAG production by harnessing the inherent nitrogen-fixing capability of M. buryatense 5GB1S. The final engineered strain TAG6 achieved the highest TAG titer of 224.42 mg/L, representing a 9-fold enhancement over the initial engineered strain TAG1. The methane-derived TAG exhibited a favorable cetane number suitable for SAF production and a significantly reduced carbon footprint. These findings highlight a promising route for upcycling methane into SAF, making a significant step towards mitigating carbon emissions and promoting sustainable biofuel production.
Engineering Acetogens for Biofuel Production: from Cellular Biology to Process Improvement

Abstract:Increasing environmental concerns regarding fossil fuels and potential future supply constraints have driven the exploration of alternative fuel resources. Using syngas to produce biofuels through microbial fermentation processes provides an excellent option for the synthesis of fuels and chemicals in a clean and sustainable way. The fermentation of syngas by anaerobic acetogens via the Wood-Ljungdahl pathway has attracted considerable interest to for the production of biofuels. The major natural fermentation products of these bacteria are acetate, butyrate, ethanol, butanol, and 2,3-butanediol, which can be used directly or serve as precursors for biofuel and industrial chemical production. However, the widespread use of acetogens as production biocatalysts has been partially limited by their metabolic and energetic constraints for efficient conversion of syngas into target products. A comprehensive understanding of the cellular biology that enables syngas fermentation by these versatile microorganisms is necessary to model the electron and carbon flow in specific production routes, which can contribute substantially to design strategies for acetogen cell engineering and to optimize these technologies to an industrially attractive production level. In this review, we summarize the metabolic and energy conservation mechanisms of most known acetogens during syngas fermentation and discuss parameters that can be modulated to improve their metabolic efficiencies. Finally, the potential to utilize metabolic engineering to improve the spectrum of acetogen products is discussed. This will be helpful in developing acetogens as efficient syngas fermentation biocatalysts for biofuel production in large-scale industrial processes and therefore act as a novel microbial production platform that is both environmentally safe and sustainable.
Biofuel production: an odyssey from metabolic engineering to fermentation scale-up

Abstract:Metabolic engineering has developed microbial cell factories that can convert renewable carbon sources into biofuels. Current molecular biology tools can efficiently alter enzyme levels to redirect carbon fluxes toward biofuel production, but low product yield and titer in large bioreactors prevent the fulfillment of cheap biofuels. There are three major roadblocks preventing economical biofuel production. First, carbon fluxes from the substrate dissipate into a complex metabolic network. Besides the desired product, microbial hosts direct carbon flux to synthesize biomass, overflow metabolites, and heterologous enzymes. Second, microbial hosts need to oxidize a large portion of the substrate to generate both ATP and NAD(P)H to power biofuel synthesis. High cell maintenance, triggered by the metabolic burdens from genetic modifications, can significantly affect the ATP supply. Thereby, fermentation of advanced biofuels (such as biodiesel and hydrocarbons) often requires aerobic respiration to resolve the ATP shortage. Third, mass transfer limitations in large bioreactors create heterogeneous growth conditions and micro-environmental fluctuations (such as suboptimal O2 level and pH) that induce metabolic stresses and genetic instability. To overcome these limitations, fermentation engineering should merge with systems metabolic engineering. Modern fermentation engineers need to adopt new metabolic flux analysis tools that integrate kinetics, hydrodynamics, and (13)C-proteomics, to reveal the dynamic physiologies of the microbial host under large bioreactor conditions. Based on metabolic analyses, fermentation engineers may employ rational pathway modifications, synthetic biology circuits, and bioreactor control algorithms to optimize large-scale biofuel production.

Abstract:Biogas generated from food waste through anaerobic digestion is typically utilized for heat and electricity. However, recent developments in synthetic biology have enabled the conversion of carbon dioxide (CO2) and methane (CH4) in biogas into desired products by microorganisms. This study introduced a novel bioroute for the production of sustainable aviation fuel (SAF) using food waste in a carbon–neutral and cost-effective manner. Both CO2 and CH4 derived from food waste are sequentially bioconverted into lipids by photosynthetic and methanotrophic microorganisms, which are then upgraded into SAF. A comparative life cycle assessment was conducted, revealing that this proposed bioroute has a lower environmental impact than a thermo-route involving hydrothermal liquefaction. Furthermore, a techno-economic analysis of the bioroute was also performed to evaluate both the capital and operating costs, showcasing the feasibility of this concept with current cutting-edge technologies. With potential food waste disposal subsidies taken into consideration, the bioroute-based SAF could yield an internal rate of return of up to 38%, demonstrating its economic viability in the market. By leveraging food waste and innovative biotechnology, this bioroute has the potential to contribute to a more eco-friendly and economically sustainable aviation industry.

Abstract:Combating climate change and ensuring energy supply to a rapidly growing global population has highlighted the need to replace petroleum fuels with clean, and sustainable renewable fuels. Biofuels offer a solution to safeguard energy security with reduced ecological footprint and process economics. Over the past years, lignocellulosic biomass has become the most preferred raw material for the production of biofuels, such as fuel, alcohol, biodiesel, and biohydrogen. However, the cost-effective conversion of lignocellulose into biofuels remains an unsolved challenge at the industrial scale. Recently, intensive efforts have been made in lignocellulose feedstock and microbial engineering to address this problem. By improving the biological pathways leading to the polysaccharide, lignin, and lipid biosynthesis, limited success has been achieved, and still needs to improve sustainable biofuel production. Impressive success is being achieved by the retouring metabolic pathways of different microbial hosts. Several robust phenotypes, mostly from bacteria and yeast domains, have been successfully constructed with improved substrate spectrum, product yield and sturdiness against hydrolysate toxins. Cyanobacteria is also being explored for metabolic advancement in recent years, however, it also remained underdeveloped to generate commercialized biofuels. The bacterium Escherichia coli and yeast Saccharomyces cerevisiae strains are also being engineered to have cell surfaces displaying hydrolytic enzymes, which holds much promise for near-term scale-up and biorefinery use. Looking forward, future advances to achieve economically feasible production of lignocellulosic-based biofuels with special focus on designing more efficient metabolic pathways coupled with screening, and engineering of novel enzymes.

Abstract:Increasing demand for renewable feedstock‐based biofuels is driving the interest and rapid development of processes to produce fuels and chemicals from biomass‐generated syngas. Biomass is gasified to produce syngas that can be converted via thermochemical routes to fuels and chemicals such as alcohols, olefins, and fuel grade hydrocarbons. An alternate route to produce liquid products from syngas is through gas fermentation, a hybrid thermochemical/ biochemical process. Abstract:Acetogenic bacteria employing the Wood-Ljungdahl pathway can be used as biocatalysts in syngas fermentation for the production ofbiofuels such as ethanol or butanol as well as biocommodities such as acetate, lactate, butyrate, 2,3 butanediol, and acetone. The potential of such processes can be projected by the global syngas output, which was 70,817 megawatts thermal in 2010 and is expected to increase up to 72% in 2016. To date, different acetogens are used as commercial production strains for industrial syngas fermentations in pilot or demonstration plants (Coskata, INEOS Bio, LanzaTech) and first commercial units are expected to launch operation in the near future (INEOS Bio, LanzaTech). Considerations on potential yields are quite promising for fermentative production. New methods for metabolic engineering were established to construct novel recombinant acetogenic biocatalysts. Synthetic biology will certainly play a major role in constructing strains for commercial operations. This way, a cheap and abundant carbon source most probably replace, processes based on crude oil or sugar in the near future.

Abstract:Background: Sabinene, one kind of monoterpene, accumulated limitedly in natural organisms, is being explored as a potential component for the next generation of aircraft fuels. And demand for advanced fuels impels us to develop biosynthetic routes for the production of sabinene from renewable sugar.Results: In this study, sabinene was significantly produced by assembling a biosynthetic pathway using the methylerythritol 4-phosphate (MEP) or heterologous mevalonate (MVA) pathway combining the GPP and sabinene synthase genes in an engineered Escherichia coli strain. Subsequently, the culture medium and process conditions were optimized to enhance sabinene production with a maximum titer of 82.18 mg/L. Finally, the fed-batch fermentation of sabinene was evaluated using the optimized culture medium and process conditions, which reached a maximum concentration of 2.65 g/L with an average productivity of 0.018 g h(-1) g(-1) dry cells, and the conversion efficiency of glycerol to sabinene (gram to gram) reached 3.49%.Conclusions: This is the first report of microbial synthesis of sabinene using an engineered E. coli strain with the renewable carbon source as feedstock. Therefore, a green and sustainable production strategy has been established for sabinene.
Carbon-negative production of acetone and isopropanol by gas fermentation at industrial pilot scale

Abstract:Many industrial chemicals that are produced from fossil resources could be manufactured more sustainably through fermentation. Here we describe the development of a carbon-negative fermentation route to producing the industrially important chemicals acetone and isopropanol from abundant, low-cost waste gas feedstocks, such as industrial emissions and syngas. Using a combinatorial pathway library approach, we first mined a historical industrial strain collection for superior enzymes that we used to engineer the autotrophic acetogen Clostridium autoethanogenum. Next, we used omics analysis, kinetic modeling and cell-free prototyping to optimize flux. Finally, we scaled-up our optimized strains for continuous production at rates of up to ~3 g/L/h and ~90% selectivity. Life cycle analysis confirmed a negative carbon footprint for the products. Unlike traditional production processes, which result in release of greenhouse gases, our process fixes carbon. These results show that engineered acetogens enable sustainable, high-efficiency, high-selectivity chemicals production. We expect that our approach can be readily adapted to a wide range of commodity chemicals.

Abstract:Around 65% of the mitigation needed for the targeted net-zero carbon aviation emissions in 2050 is expected to come from Sustainable Aviation Fuels (SAFs). In this study, an alternative gasification-driven Biomass-to-Liquid (BtL) concept for the production of SAFs is introduced and evaluated. In particular, a fuel synthesis scheme based on the double-stage fermentation of the produced syngas (syngas → acetic acid → TAGs) is assessed instead of the conventional Fischer-Tropsch (FT) or Alcohol-to-Jet (AtJ) synthesis. The objective of the present work is the techno-economic evaluation of a large-scale (200 MWth) replication of the mentioned BtL concept, whose performance has been simulated in Aspen PlusTM (V.11) with reasonable upscaling considerations and models validated at a pilot scale. The estimated baseline Total Capital Investment (TCI) of €577 million lies in the typical range of €500–700 million that many recent techno-economic studies adopt for gasification-driven BtL plants of similar capacity, while the estimated annual operating costs of €50 million correspond to a 15–40% OpEx reduction compared to such plants. A discounted cash flow analysis was carried out, and a baseline Minimum Jet Selling Price (MJSP) equal to 1.83 €/L was calculated, while a range of 1.38–2.27 €/L emerged from the sensitivity analysis. This study sets the biological conversion of gasification-derived syngas into triglycerides (TAGs) as a promising alternative route for the production of SAFs. In general, gasification-driven BtL pathways, led by the relatively mature FT and AtJ technologies, are capable of thriving in the coming years based on their capability of advanced feedstock flexibility.
