Delignification of Rosa hybrida cv. Rose Sherbet Seed Pericarp Using Deep Eutectic Solvents for Improved Seed Quality Parameters

Abstract:Rose, a perennial flowering plant belonging to the Rosaceae family, is indigenous to the western Himalayan regions of India. The achenes (seeds) of Rosa hybrid cv. Rose sherbet consist of approximately 30–35% embryo and 65–70% pericarp. Dormancy in rose seeds is the major hurdle in the breeding of roses. The dormancy is primarily due to physical (thick pericarp) and endogenous plant growth regulators (high level of abscisic acid (ABA) and low level of gibberellic acid (GA 3 )). Various seed treatment approaches such as physical, chemical, and physiochemical methods are widely used with limited success (ranging from 15–30% improved germination) and each treatment is associated with disadvantages that necessitates the use of an eco-friendly alternative technique for pericarp thickness reduction. Recently, Deep Eutectic Solvents (DES), a class of green solvents composed of hydrogen bond acceptors and donors have been employed for delignifying lignocellulosic biomass. Rose seeds were treated with two types of DESs namely choline chloride (ChCl): Formic acid (FA) and ChCl:Oxalic acid (OA) to analyze the efficacy. Among the treatments, with ChCl:OA the crystallinity index has increased by 10.77 times, and pericarp thickness reduced significantly by 61 folds as deciphered from SEM analysis. This study implies that the DES treatment is a promising method for delignification and can be further exploited for enhancement of seed quality parameters.
