Abstract:The first chlororuthenacyclopentatrienes have been realized by the reactions of RuCl2(PPh3)3 with o-phenyldiyne derivatives in the presence of excess HCl. The structures of the unique chlororuthenacycles were fully characterized by spectroscopic data and X-ray diffraction. In addition, computational studies provided the evidence for their aromatic character, making them as an important supplement to the rare aromatic ruthenacycle family.

Abstract:An efficient synthesis of 2,3,4-trisubstituted pyrroles via intermolecular cyclization of alkylidenecyclopropyl ketones with amines was observed. The addition of anhydrous MgSO4 improved the yields of the products 3. A possible mechanism involving the distal cleavage of the C-C bond of cyclopropane ring was proposed.

Abstract:The cyclization of 5,6-diarylpyrrolo[3,4- d ]pyrimidine-2,4-diones containing a halogen atom in the ortho -position of one of the aryl rings, proceeding as intramolecular C–H arylation catalyzed by Ru(II) complexes, was studied. It was found that complexes Ru(NHC)(cymene)Cl 2 , where NHC = IPr [1,3-bis(2,6-diisopropylphenyl)imidazol-2-ylidene], IMes [1,3-bis(2,4,6-trimethylphenyl)imidazol-2-ylidene], and their derivatives containing substituents at positions 4 and 5 of the imidazole ring, exhibit high catalytic activity. Based on the reaction studied, new representatives of pyrimido[5',4':3,4]pyrrolo[1,2- f ]phenanthridine were obtained.

Abstract:A novel K3PO4-catalyzed synthesis of polysubstituted pyrroles by a Michael addition/alkyne carbocyclization of activated alkynes and N-propargylamines has been developed. This transition-metal-free cascade process represents an environmental friendly and efficient way to construct polysubstituted pyrroles in good yields. Catalyzed by CsF, a Michael addition/aza-Claisen rearrangement/cyclization sequential process has been achieved to selectively synthesize pyrroles in moderate yields.

Abstract:The efficient synthesis of pyranones is presented by a three-component cascade reaction from readily available acrylic acids, ethyl glyoxylate, and p-toluenesulfonamide under ruthenium catalysis. For the first time, the nucleophilic addition of the vinylic C-H bond of acrylic acids across aldehyde is achieved, and the intramolecular cyclization as well as subsequent second insertion to aldehyde form the substituted butenolides. The elimination of sulfonamides occurs at higher temperature to give the pyranones.

Abstract:Free radical cyclization has emerged as one of the most important reaction types, which is widely used in natural product synthesis, pharmaceutical chemistry, and materials science. This report described the combination of radical cyclization and ruthenium-catalyzed meta-selective C–H functionalization for the synthesis of arylpyrrolidone derivatives. This method exhibited the highly meta-site selectivity of the primary, secondary, and tertiary alkyl radicals formed by intramolecular addition. A wide spectrum of directing groups bearing diversified N-heterocycles performed well, including biologically active molecules. Abstract:Novel intermediate oxazoline[3,2-a]pyridiniums were facilely prepared from 2-(2,2-dimethoxyethoxy)-pyridines via acid promoted intramolecular cyclization. Sequentially, the quaternary ammonium salts were treated with different nucleophiles for performing regioselective metal-free C-O and C-N bond-cleaving to afford prevalent heterocyclic structures of N-substituted pyridones and 2-substituted pyridines. The reaction mechanism and regioselectivity were then systematically explored by quantum chemistry calculations at B3LYP/6-31 g(d) level. The calculated free energy barrier of the reactions revealed that aniline and aliphatic amines (e.g., methylamine) prefer to attack C8 of intermediate 4a , affording N-substituted pyridones, while phenylmethanamine, 2-phenylethan-1-amine and 3-phenylpropan-1-amine favor to attack C2 of the intermediate to form 2-substituted pyridines. With the optimized geometries of the transition states, we found that the aromatic ring of the phenyl aliphatic amines may form cation-π interaction with the pyridinium of the intermediates, which could stabilize the transition states and facilitate the formation of 2-substituted pyridines.

Abstract:The first example of meta-selective CAr-H bond functionalization of aryl 2-pyridyl ketones has been developed using [Ru(p-cymene)Cl2]2 as the catalyst and alkyl bromide as the coupling reagent. This development provides an efficient strategy for modifying the meta-position of aryl 2-pyridyl ketone skeletons, which are found in various functional molecules.

Abstract:Reported herein is a ruthenium-catalyzed formal dehydrative [4 + 2] cycloaddition of enamides and alkynes, representing a mild and economic protocol for the construction of highly substituted pyridines. Notably, the features of broad substrate scope, high efficiency, good functional group tolerance; and excellent regioselectivities were observed for this reaction. Density functional theory (DFT) calculations and experiments have been carried out to understand the mechanism and regiochemistry. DFT calculations suggested that this formal dehydrative [4 + 2] reaction starts with a concerted metalation deprotonation of the enamide by the acetate group in the Ru catalyst; which generates a six-membered ruthenacycle intermediate. Then alkyne inserts into the Ru-C bond of the six-membered ruthenacycle, giving rise to an eight-membered ruthenacycle intermediate. The carbonyl group (which comes originally from the enamide substrate and is coordinated to the Ru center in the eight-membered ruthenacycle intermediate) then inserts into the Ru-C bond to give an intermediate, which produces the final pyridine product through further dehydration. Alkyne insertion step is a regio-determining step and prefers to have the aryl groups of the used alkynes stay away from the catalyst in order to avoid repulsion of aryl group with the enamide moiety in the six-membered ruthenacycle and to keep the conjugation between the aryl group and the triple C-C bond of the alkynes. Consequently, the aryl groups of the used alkynes are in the beta-position of the final pyridines, and the present reaction has high regioselectivity.

Abstract:Ruthenium(ii)-catalyzed oxidative coupling by C2-alkenylation of N-acyl pyrroles with alkenes has been described. The acyl unit was found to be an effective chelating group for the activation of aryl C-H bonds ortho to the directing group. The alkenylation reaction of benzoyl pyrroles occurred regioselectively at the C2-position of the pyrrole ring, without touching the benzene ring. The reaction provides exclusively monosubstituted pyrroles under the optimized conditions. Disubstituted pyrroles could be obtained using higher loadings of the ruthenium(ii)-catalyst and the additives.

Abstract:A highly efficient protocol for the synthesis of pyrrolidones by the copper‐catalyzed alkynylation/annulation of aliphatic amides with alkynyl carboxylic acids is discussed in this paper. A broad range of easily accessible alkynyl carboxylic acids were introduced at the β‐methyl group of aliphatic amides with the assistance of an 8‐aminoquinolyl auxiliary group via decarboxylation to achieve the subsequent cyclic CN bond formation within one hour. High selectivity of β‐methyl groups over methylene groups was observed, and the extension of this catalytic system to the activation of methylene CH bonds failed. The substrates with two different groups at the α‐position of the aliphatic amides lead to the formation of diastereoisomers which is determined by 1H NMR spectroscopy. The initially produced products with Z‐configurations can be easily transformed to the corresponding products with E‐configurations by the treatment with dilute p‐toluenesulfonic acid after the reaction. This catalytic tandem decarboxylative cyclization provides a new opportunity for the direct functionalization of sp3 CH bonds.

Abstract:Multisubstituted 2-pyridones are prevalent in pharmaceuticals and bioactive molecules. We herein report an efficient and regioselective approach for the synthesis of trisubstituted 2-pyridone derivatives by a rhodium-catalyzed C–H activation/Lossen rearrangement/cyclization cascade reaction between acrylamides and propargyl alcohols. The desirable features of this protocol include a reusable catalytic system, high regioselectivity, uncommon Lossen rearrangement, good functional group tolerance, operation at room temperature, simple purification by filtration in most cases, and scale-up synthesis with as low as 1 mol% catalyst loading. Additionally, deuterium labeling and KIE assays were performed to investigate the reaction mechanism. The vital effect of the hydroxyl group on propargyl alcohols in determining the regioselectivity was demonstrated by control experiments and DFT calculations. In addition, Mulliken atomic charge analysis of the key intermediates was also carried out to probe the origin of the observed preference for the Lossen rearrangement.

Abstract:The first example of the synthesis of m-alkylphenols via a ruthenium-catalyzed CAr-H bond functionalization of phenol derivatives with sec/tert-alkyl bromides is reported. Mechanistic studies indicated that the m-CAr-H bond alkylation may involve a radical process and that a six-membered ruthenacycle complex was the active catalyst. Moreover, this approach can provide an expedited strategy for the atom-/step-economical synthesis of many noteworthy pharmaceuticals and other functional molecules.

Abstract:The activation of various homopropargylic pyridines by cis‐[RuII/OsII(dppm)2Cl2] (dppm = 1,1‐bis(diphenylphosphino)methane) has previously been shown to generate a diverse array of metallacycles and metalated heterocyclic complexes. However, a minor structural modification of introducing a halide onto the pyridyl group of the alkyne substrate resulted in the formation of unprecedented Ru(II)/Os(II)−haloquinolizine complexes. These complexes display (1) κ2(X,C)‐haloquinolizine chelates arising from the cycloisomerization of HC≡CC(OH)(CH2(6‐X‐2‐py))(Ph) on [RuII/OsII(dppm)2]2+ moieties via a vinylidene pathway, (2) five‐membered Ru/Os−X−C−N−C rings (X = F, Cl, Br) ortho‐ and peri‐fused to quinolizinium skeletons, and (3) uncommon M−X−R bonding interactions that are atypical in coordination complexes. Despite being divalent and integrated into a five‐membered Ru−X−C−N−C ring system, the X atoms in the Ru(II) complexes are susceptible to substitution by O in the presence of −OH, resulting in the formation of quinolizinium‐fused ruthenaoxazole complexes. Overall, this work highlights the importance of considering metal−halocarbon bonding interactions in catalytic or coordination designs.

Abstract:The new ruthenium complex [Ru(N(3)P)(OAc)][BPh(4)] (4), in which N(3)P is the N,P mixed tetradentate ligand N,N-bis[(pyridin-2-yl)methyl]-[2-(diphenylphosphino)phenyl]methanamine was synthesized. The complex was found to be catalytically active for the endo cycloisomerization of alkynols. The catalytic reactions can be used to synthesize five-, six-, and seven-membered endo-cyclic enol ethers in good to excellent yields. A catalytic cycle involving a vinylidene intermediate was proposed for the catalytic reactions. Treatment of complex 4 with PhC[triple bond]CH and H(2)O gave the alkyl complex [Ru(CH(2)Ph)(CO)(N(3)P)][BPh(4)] (30), which supports the assumption that the catalytic reactions involve addition of a hydroxyl group to the C=C bond of vinylidene ligands.
