Abstract:Fluorescent bioprobs are in urgent demand to monitor important biological events in biomedicine. However, the aggregation-caused quenching character, high toxicity, water-insolubility and easy leakage property of conventional small molecular dyes hinder the development in this area. In this work, an aggregation-induced emission (AIE) bioconjugate was synthesised by labeling tetraphenylethylene (TPE) to quaternized chitosan (QCS). The TPE-QCS bioconjugate emits strong fluorescence even in solid state, and is cationic and water-soluble over a wide range of pH values. The TPE-QCS aqueous solution stained HeLa cells by dose- and time-depent manner and imaged living cells with bright fluorescence. Futhermore, the cationic bioconjugate was readily internalized by cells through endocytosis, and further aggragated to large sizes and adhered to negatively charged organelle membranes inside cells achieving fluorescent cell imaging with fluorescence enhancement and leakage-free staining. The AIE-active TPE-QCS with cationic nature, good water-solubility over a wide pH range and unique cell imaging properties could trace HeLa cells for as long as 23 passages, that was obviously superior to existing commercial cellular tracer, so has promising application prospects as ultra long-term tracer in biomedical field.

Abstract:ABSTRACTIn this article, a novel zwitterionic conjugated polyelectrolyte containing tetraphenylethene unit was synthesized via Pd‐catalyzed Sonogashira reaction. The resulting polymer (P2), which exhibited typical aggregation‐induced emission (AIE) properties, was weakly fluorescent in dilute DMSO solution and showed bright fluorescence emissions when aggregated in DMSO/water mixtures or fabricated into conjugated polymer nanoparticles (CPNs). The nanoparticles from P2 could be prepared by reprecipitation method with an average diameter around 23 nm. Notably, the cell‐staining efficiencies of lipid‐P2 nanoparticles could be enhanced with lipid encapsulation and these nanoparticles were endocytosed via caveolae‐mediated and clathrin‐mediated endocytosis pathways. Furthermore, the lipid‐P2 nanoparticles with low cytotoxicity, high photostability and efficient cell staining ability could be employed for in vitro detection of Fe3+ ions in A549 cells. © 2016 Wiley Periodicals, Inc. Abstract:Phenylacetylene and 1-pentyne derivates containing aminated tetraphenylethene (TPE) units 1 and 2 are synthesized. The TPE-functionalized luminogens are nonemissive in dilute solutions but luminescent as solid aggregates, Showing H novel behavior of aggregation-induced emission (AIE). The monomers are polymerized to their corresponding polymers P1 and P2 with an E conformation by organorhodium catalysts, although Rh-catalyzed polymerizations normally yield Z-rich polyacetylenes. Whereas P1 fluoresces faintly in THF, its light emission is enhanced by aggregation inaqueous media, exhibiting an aggregation-induced emission enhancement (AIEE) effect. Oil the other hand, P2 is AIE active: the emission of its nanoaggregates Suspended in 90% aqueous mixture is similar to 57-fold stronger than that Of its Solution in THF. A novel superamplification effect is observed in the emission quenching of P2 nanoaggregates by picric acid (PA): the quenching constant (K-SV) is nonlinearly increased with the quencher concentration and reaches a very high value in the high [PA] region (K-SV similar to 350000 M-1). P1 and P2 are transformed to conjugated polyelectrolytes P1(+) and P2+ via ionization by hydrochloric acid. P1+ readily hybridizes with carbon nanotubes, affording functionalized P1+/nanotube hybrids with good water miscibility. Emission of P2+ in aqueous buffer is boosted when it binds to bovine serum albumin (BSA), allowing the polymer to work as a sensitive bioprobe for BSA quantitation at ultralow protein concentration (0-0.6 ppm).

Abstract:Three conjugated poly(tetraphenylethene)s grafted with dibenzo-24-crown-8 (DB24C8) groups, P-O, P-M and P-P, have been synthesized via Suzuki cross-coupling polymerization, in which the tetraphenylethene (TPE) units are connected at the ortho, meta and para positions, respectively. These conjugated polymers inherit the aggregation-induced emission feature of the TPE unit and thus their emission intensities increased significantly in the THF-water mixture solvents. The enhanced emission intensities highly depend on the connection modes of the TPE units and are in the order of P-O < P-M < P-P. Furthermore, the aggregation-induced emission enhancement (AIEE) feature can be activated by the acid-base-controlled complexation of DB24C8 groups with a dibenzylammonium chloride based molecule, protonated C12-2. The reduplicative complexation induces step-by-step emission enhancements together with morphological transitions from micellar to vesicular in the systems of P-O/C12-2 and P-M/C12-2. In the case of P- P/C12-2, of note is that the complexation not only leads to an AIEE feature but also a significant blue shift of the emission band from 515 to 483 nm. Both of these are really reversible under the acid-base-controlled condition. These polymer materials are of great importance for their potential application in the fields of optoelectronic devices and chemical and biomedical sensors.

Abstract:The anionic polytriazole PI with a weight-average molecular weight of 190 300 was facilely and directly prepared in an excellent yield of 98.1% in 6 h by the well-established Cu(I)-catalyzed click polymerization of 4,4'-diazido-2,2'-stilbenedisulfonic acid disodium salt hydrate (1) and AIE-active tetraphenylethene (TPE)-containing diyne (2) under mild reaction conditions. PI is soluble in high polar solvents, and thermally stable with 5% loss of its weight at temperatures higher than 290 degrees C. The photo-physical property investigation showed that PI possesses the unique feature of aggregation-enhanced emission (AEE). By taking this feature, PI could be used for the specific detection of aluminum ions with a threshold as low as 31 ppb, which could potentially be used for strict drinking water purity control of the aluminum ion content.

Abstract:A new fluorescence technique for monitoring layer-by-layer self-assembling processes of polycations and polyanions is developed in this work. The fluorescent probe is a fluorogenic dye named 1,1-bis[p-(diethylaminomethyl)phenyl]-2,3,4,5-tetraphenylsilole (A2HPS). Whereas fluorescence of a “normal” fluorophore is often quenched by aggregate formation, the protonated salt of A2HPS, i.e., [H2A2HPS]2+, emits strong light in the suspensions of its nanoaggregates and in the solid films of its blends with poly(diallyldimethylammonium chloride) (PDDAC), thanks to its novel aggregation-induced emission (AIE) characteristics. When ([H2A2HPS]2++PDDAC) cations and poly(styrenesulfonate) (PSS) anions were used to fabricate thin films via layer-by-layer deposition processes on quartz and glass substrates, the emission intensity of [H2A2HPS]2+ showed linear relationship with the number of ([H2A2HPS]2++PDDAC)/PSS bilayers, due to the uniform co-deposition of [H2A2HPS]2+ cations into the PDDAC/PSS bilayers. This proves that the AIE fluorophore is an excellent probe for monitoring the layer-by-layer self-assembling processes of the polyelectrolytes on various substrates.

Abstract:The two-photon absorption properties of anionic poly(phenylene ethynylene)-type conjugated oligo- and polyelectrolytes are studied in molecularly dissolved and aggregated forms in aqueous solution. Several different polyvalent cations are used to induce aggregation. It is found that both materials in the aggregated form exhibit enhanced two-photon excited fluorescence (2PEF) and two-photon cross section (σ2) compared with the molecularly dissolved structures. The 2PEF and σ2 are unaffected by the nature of the polyvalent cation that is used to induce aggregation. The two-photon absorption cross section enhancement arises because of the increase in the difference dipole moment (Δμ) in the aggregates of the conjugated materials, an effect that is attributed to the introduction of charge transfer character into the aggregate excited state.

Abstract:A novel chitosan-based fluorescent bioconjugate (TPE-CS) with aggregation-induced emission (AIE) characteristic is synthesized and used as a fluorescent probe for monitoring layer-by-layer self-assembly process of natural polyelectrolytes. QCM results and contact angle measurement indicate that this AIE active TPE-CS bioconjugate can be assembled with alginate (ALG) through layer-by-layer deposition. Ellipsometry and fluorescence (FL) spectroscopy show an exponential growth of the TPE-CS/ALG multilayer films. Moreover, the exponential relationship between the FL intensity and the number of bilayers, which is in accordance with the thickness variation of multilayer films, provides solid evidence for its capacity to monitor the layer-by-layer self-assembly process.

Abstract:Fluorescence correlation spectroscopy (FCS) is applied to investigate aggregation behavior of an anionic conjugated polyelectrolyte (CPE) featuring branched carboxylate groups (PPE-(d)CO2) upon the addition of different metal ions. FCS results reveal that monovalent metal ions (Na(+), K(+)) bind to the CPE chain but do not induce aggregation, and divalent metal ions (Ca(2+), Cu(2+)) bind and induce the formation of small aggregates. Iron cations (Fe(2+) and Fe(3+)) bind strongly and induce the formation of large CPE aggregates. This is believed to be because of the propensity of these metals to bind to six ligands and thus bridge two of the triacid units on adjacent PPE-(d)CO2 chains. A comparison between the Stern-Volmer (SV) fluorescence quenching and the relative diffusion time change from FCS demonstrates that the most efficient SV quenching is observed for the ions that give rise to large increases in FCS diffusion time, underscoring the importance of ion-induced aggregation in the amplified quenching effect.

Abstract:Electrofluorochromic (EFC) materials, which exhibit electrochemically controllable fluorescence, hold great promise in optoelectronic devices and biological analysis. Here we design such donor acceptor (D-A) conjugated polymers P(TPACO) and P(TCEC)-that contain the same electron-rich and oxidizable polytriphenylamine (PTPA) as pi-backbone, yet with different electron-deficient ketone and cyano units as pendant groups, respectively. They both exhibit solvatochromic effects due to intrinsic characteristics of intramolecular charge transfer (ICT). Compared to P(TPACO), P(TCEC) shows stronger ICT, which leads to higher electrochemical oxidation potential and lower ion diffusion coefficient. Moreover, both polymers present simultaneous electrochromic (EC) and EFC behaviors with multistate display and remarkably rapid fluorescence response. The response time of P(TPACO) is as short as 0.19 s, nearly 4-fold faster than that of P(TCEC) (0.92 s). Such rapid response is found to be determined by the ion diffusion coefficient which is associated with the ICT nature. Finally, the EFC display device based on P(TPACO) is successfully demonstrated, which shows green fluorescence ON/OFF switching upon applied potentials. This work has successfully demonstrated that swift EFCs can be achieved by rational modulation of the ICT effect in such D-A conjugated polymers.

Abstract:Cyano-substituted oligo (alpha-phenylenevinylene)-1,4-bis(R-cyano-4-diphenylaminostyryl)-2,5-diphenylbenzene (CNDPASDB) molecules are Studied in solution and aggregate state by time-resolved fluorescence techniques. CNDPASDB exhibits a strong solvent polarity dependent characteristic of aggregation-induced emission (AIE). By time-dependent spectra, the gradual transition from local excited state to intramolecular charge transfer state with the increasing solvent polarity is clearly resolved. The transition time in high polarity solvent DMF is very fast, around 0.5 ps, resulting in a low fluorescence quantum yield. While in aggregate state, the intramolecular torsion is restricted and the local environment becomes less polar. Thus, the intramolecular charge transfer state is eliminated and efficient AIE Occurs.

Abstract:Aggregation-induced emission (AIE) luminogens with highly tunable properties show great potential for many applications. In this study, we synthesized a new family of AIE-type poly(ethylene glycol)-block-poly(9-anthrylmethyl lysine) (PEG-b-PLys-An) diblock copolymers by taking advantage of amphiphilic self-assembly and rigid helical backbones. These copolymers can self-assemble into various assemblies through nanoprecipitation methods. The micelles using N,N-dimethylformamide (DMF) as a cosolvent present brighter fluorescence than the vesicles prepared from tetrahydrofuran (THF). We demonstrate that the decreased solubility of copolymers in DMF results in the formation of more compact micelles with more excimer formation during the self-assembly process, while better solvent THF favors the formation of vesicles with stretched core chains. In addition, the secondary conformation of the polypeptide block shows pronounced effects on the fluorescence property. We further show the internalization of the assemblies using two types of cells by cellular uptake experiments. By the delicate design of the block copolymer, we successfully prepare the morphology- and conformation-dependent AIE materials for potential biomedical applications.
A New Ratiometric Ag+ Fluorescent Sensor Based on Aggregation-Induced Emission

Abstract:The novel tetraphenylethylene(TPE)-based sensor 1 bearing bis(2-pyridin-2-ylmethyl)amine (BPA) units linked with triazole moieties could be obtained by click reaction efficiently. The results show that 1 can demonstrate a Ag+-specific emission shift and highly sensitive fluorescent enhancement with a 1:2 binding ratio based on the aggregation-induced emission mechanism. Compound 1 is shown to behave as a ratiometric sensor.

Abstract:A series of dicyanomethylene-1,4-dihydropyridine (DCMP) derivatives with aggregation-induced emission (ATE) were designed and synthesized. These target compounds emit low fluorescence in THE solutions because of the free rotation of the phenyl and dihydropyridine rings about the axes of the olefinic double bonds and the resultant nonradiative decay process, but they exhibit strong fluorescence in the aggregate state because of the restriction of intramolecular rotation (RIR), as confirmed by solution thickening and cooling experiments. Compared with the almost-planar dicyanomethylene-4H-pyran (DCM) derivative DCM-1 with aggregation-caused quenching (ACQ), DCMP-5 with AM takes a highly twisted conformation by replacing the oxygen atom of the DCM skeleton with an N-ethyl group, as revealed by crystallographic data and theoretical calculations. Depending on the different electron-donating substituents, the solids of these compounds emit yellow or orange fluorescence. Moreover, the cyano group endows DCMP-5 with a strong self-assembly ability under proper conditions, and the obtained regular microparticles emit bright orange fluorescence. These materials will broaden the new family of ATE-exhibiting fluorophores.
