Abstract:Linear and miktoarm star-shaped diblock copolymers consisting of single-stranded DNA and poly(N-isopropylacrylamide) (PNIPAAm) with various compositions were synthesized via atom transfer radical polymerization and click chemistry. The temperature-responsive phase transition behavior, micellization, was systematically examined using UV vis spectrometry, high-sensitivity differential scanning calorimetry, dynamic light scattering, and small-angle X-ray scattering. The lower critical solution temperature (LCST) increased, and its enthalpy decreased with decreasing PNIPAAm content The copolymers self-assembled into well-defined nanoparticles having a core composed of PNIPAAm and a coronal layer of DNA above LCST. The particle size and micellar aggregation number of copolymer chains depended on the macromolecular composition and chain architecture. On the other hand, regardless of their factors, the surface area occupied by one DNA strand was found to be almost unchanged. The hybridization of DNA on the nanoparticles with fully complementary one induced the aggregation of the particles in a non-cross-linking configuration. The nanoparticle composed of miktoarm star copolymer showed a quicker DNA-hybridization response in this non-cross-linking aggregation compared with the case of a linear analogue.

Abstract:Diblock and miktoarm star-shaped thermoresponsive copolymers composed of single-stranded DNA (ssDNA) and poly(N-isopropylacrylamide) were successfully synthesized with combination of atom transfer radical polymerization (ATRP) and click chemistry. This approach should be generalizable to other DNA-functionalized copolymers. Such copolymers self-assemble into spherical micelles with ssDNA corona in aqueous solution above the lower critical solution temperature. The micellar size can be tuned from the macromolecular architecture. These DNA-encoded micellar particles are able to encapsulate and release hydrophobic guest molecules upon changing temperature.

Abstract:Developing polymers with switchable glass transition temperature (Tg) can solve scientific challenges such as fracture healing of polymers with high Tg and processing of hard polymers without plasticizer at room temperature. The azobenzene-containing polymers, or the so-called azopolymers, are smart materials that show photoswitchable Tg and photoinduced motions, which can be used in fabrication of photoactuators. Here, healable and reprocessable models were constructed by using linear azopolymers. Molecular dynamics (MD) simulation was used to study the behaviors of photo-induced bending, self-healing, and directional photofluidization of the azopolymers. A temporary modification of the C–N=N–C dihedral potential was applied to achieve the isomerization of azopolymer by using polymer consistent force-field (PCFF). The simulation results show that isomerization of azobenzene groups can lead to photoswitchable Tg of the azopolymers, which resulting in shape changes and different mechanical responses. Furthermore, the photoswitchable Tg makes the azopolymers much easier to heal under UV irradiation, and restore the mechanical properties under visible light irradiation. Finally, the mechanism of photoliquefaction was investigated, our results imply that the photoinduced trans–cis-trans cycles can reduce the mechanical properties and Tg values of azopolymers.

Abstract:Over the last decades, water-based lyotropic liquid crystals of nucleic acids have been extensively investigated because of their important role in biology. Alongside, solvent-free thermotropic liquid crystals (TLCs) from DNA are gaining great interest, owing to their relevance to DNA-inspired optoelectronic applications. Up to now, however, only the smectic phase of DNA TLCs has been reported. The development of new mesophases including nematic, hexagonal, and cubic structures for DNA TLCs remains a significant challenge, which thus limits their technological applications considerably. In this work, a new type of DNA TLC that is formed by electrostatic complexation of anionic oligonucleotides and cationic surfactants containing an azobenzene (AZO) moiety is demonstrated. DNA-AZO complexes form a stable nematic mesophase over a temperature range from -7 to 110 degrees C and retain double-stranded DNA structure at ambient temperature. Photoisomerization of the AZO moieties from the E- to the Z-form alters the stiffness of the DNA-AZO hybrid materials opening a pathway toward the development of DNA TLCs as stimuli-responsive biomaterials.

Abstract:Compared to classical block copolymers, the self-assembly of small molecules shows an advantage in addressing small features. As a new type of solvent-free ionic complexes, azobenzene-containing DNA thermotropic liquid crystals (TLCs) form an assembly as block copolymers when using small DNA. However, the self-assembly behavior of such biomaterials has not been fully investigated. In this study, photoresponsive DNA TLCs are fabricated by employing an azobenzene-containing surfactant with double flexible chains. For these DNA TLCs, the self-assembly behavior of DNA and surfactants could be guided by the factors of the molar ratio of azobenzene-containing surfactant, dsDNA/ssDNA, and presence or absence of water, which addresses the bottom-up control on domain spacing of mesophase. Meanwhile, such DNA TLCs also gain top-down control on morphology via photoinduced phase change. This work would provide a strategy for regulating the small features of solvent-free biomaterials, facilitating the development of patterning templates based on photoresponsive biomaterials. Statement of Significance : The relationship between nanostructure and function is attractive in the science of biomaterials. With biocompatibility and degradability, photoresponsive DNA materials in solutions have been widely studied in biological and medical areas, but they are still hard to obtain in a condensed state. The complex created with designed azobenzene-containing surfactants paves the way for obtaining condensed photoresponsive DNA materials. However, fine control of the small features of such biomaterials has not yet been achieved. In this study, we present a bottom-up strategy of controlling the small features of such DNA materials and, simultaneously, the top-down control of morphology via photoinduced phase change. This work provides a bi-directional approach to controlling the small features of condensed biomaterials, facilitating the development of nanopatterning templates based on photoresponsive biomaterials.

Abstract:The azobenzene polymer with photoswitchable glass transition temperatures and reversible solid-to-liquid transitions has attracted much attention in recent years. The different glass transition temperatures of the azopolymer in the trans-state and the cis-state introduce evident dynamical heterogeneity during cis-trans interconversion. The correlation between molecular dynamics and rheology in azopolymers remains elusive. In this work, we studied the molecular dynamics of an unentangled amorphous azopolymer without a liquid crystal phase. We found that the Lodge-Mcleish self-concentration (SC) model can quantitatively describe the composition dependence of the glass transition temperature, revealing the local environment of the cis (or trans) segments. The terminal relaxation behavior can be well predicted by the Rouse model with the homogeneous bead friction coefficient and the heterogeneous bead-spring (HBS) model with varying friction coefficients for cis or trans subchains. The difference between two models lies in the relaxation time at small length scales and its distribution. The HBS model predicts broadening the relaxation time spectrum, whose width varies with temperature and the cis content. This prediction is consistent with the broadening of the glass transition temperature in azopolymers. Then, the models were used to study the kinetics of thermal cis-to-trans isomerization of the azopolymer in the melt, proving to be a first-order kinetic process and further justified by the ultraviolet absorption spectrum.
Investigation of Azobenzene Photoisomerization Effect on Morphologies and Properties of Nanostructured Thermosets Involving Epoxy and a Diblock Copolymer.

Abstract:This work highlights the effect of azobenzene photoisomerization on the morphologies and properties of the nanostructured thermosets involving epoxy and a diblock copolymer. First, a diblock copolymer composed of poly(ethylene oxide) (PEO) and poly(6-(4-(4-cyanophenylazo)phenoxy)hexyl methacrylate) (PCPHM) was synthesized, and this diblock copolymer was composed of an epoxy-philic block (i.e., PEO) and an azobenzene moiety-beating block (viz., PCPHM). This diblock copolymer was introduced into epoxy to obtain the nanostructured thermosets via reaction-induced microphase separation approach. To control the configuration of azobenzene moieties of the PCPHM block, the curing reactions were performed in the absence and/or presence of ultraviolet (UV) irradiation, respectively. It was found that, without UV irradiation, the PCPHM microdomains were generated with the trans isomers of azobenzene. Under UV irradiation, however, the PCPHM microdomains were formed with the cis configuration of azobenzene moieties. The ultraviolet-visible light (UV-vis) spectroscopy showed that the trans and cis configurations of azobenzene moieties were significantly fixed with the occurrence of curing reactions. The photoluminescent measurements showed that the nanostructured thermosets with trans-azobenzene moieties can emit fluorescence, which was in sharp contrast to those with cis-azobenzene moieties. The results of small-angle X-ray and atomic force microscopy showed that the nanostructured thermosets with trans and cis isomers of azobenzene moieties had quite different morphologies. It was found that the sizes of the PCPHM microdomains with cis configuration of azobenzene moieties were significantly larger than those with trans configuration. The difference in configuration of azobenzene moieties also resulted in the difference in glass-transition temperatures and dielectric properties of the materials. The results suggest a new approach to modulate the morphologies and physical properties of the nanostructured thermosets by means of photoisomerization of azobenzene moieties.

Abstract:Phase transition materials can be utilized in a variety of functionalization scenarios, benefiting from the photoinduced changes in their physicochemical properties. Among them, small molecular crystals or liquid-crystalline polymers containing azobenzene moieties that show phototunable melting points or glass transition temperatures, respectively, have proven to be significant in optically controlled functional materials. In recent years, they have been successfully used in reversible mechanical actuation, adhesion, energy storage, self-healing, athermal nanoimprinting, and other applications, which has attracted the attention of more scientists and engineers. In this review, azobenzene-containing materials with photoinduced phase transition capabilities are systematically discussed in terms of molecular design strategies, phase change mechanisms, performance control, methods of preparation of materials, and potential applications. Existing problems and future challenges are also discussed. We hope to provide some inspiration for the development of photocontrolled phase transition materials.

Solvent‐Free Liquids and Liquid Crystals from Biomacromolecules

Abstract:As DNA exhibits persistent structures with dimensions that exceed the range of their intermolecular forces, solid-state DNA undergoes thermal degradation at elevated temperatures. Therefore, the realization of solvent-free DNA fluids, including liquid crystals and liquids, still remains a significant challenge. To address this intriguing issue, we demonstrate that combining DNA with suitable cationic surfactants, followed by dehydration, can be a simple generic scheme for producing these solvent-free DNA fluid systems. In the anhydrous smectic liquid crystalline phase, DNA sublayers are intercalated between aliphatic hydrocarbon sublayers. The lengths of the DNA and surfactant are found to be extremely important in tuning the physical properties of the fluids. Stable liquid-crystalline and liquid phases are obtained in the -20 °C to 200 °C temperature range without thermal degradation of the DNA. Thus, a new type of DNA-based soft biomaterial has been achieved, which will promote the study and application of DNA in a much broader context.

Abstract:Because of their facile preparation, small size (<100 nm), programmable design, and biocompatibility, lipid-based DNA micelles show enormous potential as a tool to monitor biological events and treat,human diseases. However, their structural stability in biological matrices suffers from spatiotemporal variability, thus limiting their in vivo use. Herein, we have engineered stability-tunable DNA micelle flares using photocontrollable dissociation of intermolecular G-quadruplexes, which confers DNA micelle flares with robust structural stability against disruption by serum albumin. However, once exposed to light, the G-quadruplex formation is blocked by strand hybridization, resulting in the loss of stability in the presence of serum albumin and subsequent cellular uptake. This programmable regulation to stabilize lipid-based micelles in the presence of fatty-acid-binding serum albumin should further the development of biocompatible DNA micelles for in vivo applications.

Abstract:<p>Development of photoliquefiable solid-state biomaterials at room temperature would address scientific challenges in life science. However, external stimuli-induced phase transitions are difficult for some biomacromolecules based materials, due to the high rigidity of these biomolecules. In this present work, by delicate molecule design on azobenzene-type ammonium surfactants, two new types of DNA-surfactant materials are fabricated. At room temperature, these DNA materials show photoliquefaction of ionic crystals to isotropic liquids under UV light, and fast self-assembly from isotropic liquids back to crystals after ceasing UV light, under the assistance of azobenzene isomerization. To achieve this objective, the designed solid-state DNA materials should have melting points near room temperature and an immediate liquid crystal to isotropic liquid transition process just above the melting points, which highly depends on the stoichiometric charge ratio between DNA and surfactants. As proved by the successful self-healing tests, these DNA ionic crystals are good biomaterials with potential applications in biomedicine and life science. This work would provide a new strategy for designing anhydrous functional biomaterials at room temperature by using rigid biomacromolecules.</p><h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Statement of Significance</h3><p>At room temperature, solid-state biomaterials with photoregulated crystal⇄isotropic liquid phase transition property are attractive functional materials in life science, considering the body temperature and living environment temperature of human beings. Abstract:Artificial DNA tethering azobenzene through a backbone-inserted glycerol via an ether bond is synthesized. The stabilization effect of the hybrid duplex with a native complementary strand of DNA for R-glycerol-inserted DNA is demonstrated even when the molar ratio of azobenzene to nucleotide is 1:2. The half-lives of tens of hours for glycerol-tethered cis-azobenzene and the rapid and highly efficient trans-to-cis photoisomerization in the hybrid DNA duplex at room temperature render the manipulation of DNA nanodevices and functionalities by photoregulation in a green system possible.

Abstract:A macrocyclic azobenzene is synthesized, demonstrating an exceptional half‐life of 120 years at room temperature and above 14 000 years at 0 °C. Switching efficiencies with visible light are high in both directions and the material retains its switchability within a polymer film, underscoring its potential for optical data storage applications. A macrocyclic azobenzene with unique thermal stability, demonstrating potential for use in optical data storage material is presented: The Z‐isomer of this novel photoswitch exhibits unparalleled thermal stability, with a thermal half‐life surpassing 120 years at 25 °C. This stability is attributed to the strategic fluorination at two ortho‐ and both para‐positions. Comparative analyses involving its non‐fluorinated counterpart, ortho‐only‐fluorinated variant, and open‐chain analog are performed. Employing NMR and UV–vis spectroscopy, X‐ray diffraction, alongside Arrhenius, Eyring, and DFT calculations, revealed insights into its extraordinary stability. Furthermore, when incorporated into poly(methylmethacrylate), this material showcase efficient switching with visible light in the solid state, emphasizing its potential for optical data storage applications.

Abstract:The cis to trans isomerization of the azobenzene photochromic units is faster when they are in more polar microenvironment inside the polymers. Photochromic azobenzenes undergo light‐mediated trans–cis isomerization. The cis isomer reverts to the trans isomer thermally. To investigate the effect of different monomer sequences on the thermal stability of cis‐azobenzene, a series of copolymers, namely, block, and random structures containing stearic acid and azobenzene moieties as their side chains have been synthesized through reversible addition–fragmentation chain transfer (RAFT) polymerization. In this study, we investigate the photoisomerization of the trans and cis forms of the polymers and also the thermal reversal of the cis‐azobenzene photochromic systems. The isomerization data revealed significant differences in the isomerization timescales between the polymers and the corresponding monomer. It was observed that the local polarity around the azobenzene units within a polymer was significantly influenced by the chain segment depending on whether it was in the vicinity of the hydrophobic alkyl chains or other azobenzene units. This local environment of the azobenzene units regulates the stabilization of the transition states during the cis–trans thermal isomerization, consequently affecting the half‐life of the cis isomer.

The photochemical trans → cis and thermal cis → trans isomerization pathways of azobenzo-13-crown ether: A computational study on a strained cyclic azobenzene system

Abstract:The isomerization of azobenzo-13-crown ether can be expected to be hindered due to the polyoxyethylene linkage connecting the 2,2'-positions of azobenzene. The mixed reference spin-flip time-dependent density functional theory results reveal that the planar and rotational minima of the first photo-excited singlet state (S1) of the trans-isomer pass through a barrier (2.5-5.0 kcal/mol) as it goes toward the torsional conical intersection (S0/S1) geometry (<CNNC ≈ 98°), which is responsible for the cis isomer formation. The second excited singlet state (S2) of the trans form has a nearly planar minimum along the N-N stretching mode, which approaches a sloped S2/S1 intersection geometry. This excited state has a rotational minimum (<CNNC ≈ 99°) as well. Both these minima have a characteristic S2-S1 energy gap of 9 kcal/mol and may undergo internal conversion. A comparison of this system with an analogous 2,2'-dimethoxy-substituted azobenzene system reveals less strain in the rotational path of the latter on the S1 surface, indicating the possibility of its better trans → cis yield than the azocrown. The completely planar S2 geometry of the dimethoxy system has easy access to the linear concerted inversion path, which seems to be the reason behind its reported slightly lower π-π*(S2) yield than n-π*(S1). The thermal cis → trans isomerization path of the azobenzo-13-crown passes through a transition state (frequency 453i cm-1), which corresponds to Gibbs free energy of activation value of 26 kcal/mol in the gas-phase and isooctane. Our study also confirms that its trans isomer strongly binds Li+ among the alkali metal ions, and this observation may open up possibilities for practical applications of this azobenzo-crown.
Photo-induced Actuator Using Temperature and Light Dual Responsive Azobenzene Containing Ion Gel in Ionic Liquid

Abstract:A series of well-defined random copolymers comprising butyl acrylate (BA) and 4-phenylazophenylmethacrylate (AzoMA) (P(AzoMA-r-BA)) are prepared successfully by reversible addition fragmentation chain transfer (RAFT) polymerization. P(AzoMA-r-BA)s show LCST-type phase transition in 1-butyl-3-methylimidazolium bis(trifiuoromethanesulfone)amide ([C(4)mim] [NTf2]). LCST depends on the photoisomerization state of azobenzene, as well as on the AzoMA composition in the random copolymers. LCST of (P(cis-AzoMA-r-BA) is significantly higher than that of P(trarts-AzoMA-r-BA), because cis-AzoMA and trans-AzoMA behave as solvato-philic and solvato-phobic comonomer, respectively. At a bistable temperature, photo-induced phase separation is completely reversible. Secondly, based on this phenomenon, a thermo-/photo- responsive ion gel (BA-AzoMA ion gel) can be prepared by free radical polymerization of BA and AzoMA using ethylene glycol dimethacrylate (EGDMA) as crosslinker in [C(4)mim] [NTf2]. BA-AzoMA ion gel shows high temperature contraction and low temperature expansion behavior, due to LCST-type phase behavior of polymer system consisting of AzoMA and BA. Contraction temperature of BA-AzoMA ion gel depends on photoisomerization state of the azobenzene group in polymers. At a bistable temperature, photo-induced expansion-contraction is reversible. Finally, a Photo-induced actuator can be realized using BA-AzoMA ion gel at suitable temperatures, featuring reversible bending by alternate irradiating with visible and UV light. The bending behavior is also demonstrated.

Abstract:Optically-controlled phase change materials, which are prepared by introducing molecular photoswitches into traditional phase change materials (PCMs), can convert and store solar energy into photochemical enthalpy and phase change enthalpy. However, the thermophysical properties of optically controlled PCMs, which are crucial in the practical, are rarely paid attention to. 4-(phenyldiazenyl)phenyl decanoate (Azo-A-10) is experimentally prepared as an optically-controlled PCMs, whose energy storage density is 210.0 kJ·kg-1, and the trans single crystal structure is obtained. The density, phase transition temperature, thermal conductivity, and other parameters in trans state are measured experimentally. Furthermore, a microscopic model of Azo-A-10 is established, and the thermophysical properties are analyzed based on molecular dynamics. The results show that the microstructure parameter (order parameters) and thermophysical properties (density, radial distribution function, self-diffusion coefficient, phase change temperature, and thermal conductivity) of partially or completely isomerized Azo-A-10, which are challenging to observe in experiments, can be predicted by molecular dynamics simulation. The optically-controlled phase change mechanism can be clarified according to the differences in microstructure. The optically-controlled switchability of thermophysical properties of an optically-controlled PCM is analyzed. This study provides ideas for the improvement, development, and application of optically-controlled PCMs in the future.
