Abstract:Residual injury of atrazine to the succeeding crops has been frequently reported. It is necessary to find a solution for the detoxification of atrazine contaminated soil. A high-efficient bacterial strain Arthrobacter sp. C2 for atrazine degradation was isolated in this study. The genomic information of the isolate C2, and its degradation characteristics and potential application in detoxification of atrazine contaminated soil were investigated. The results indicated that the isolate C2 genome contained 4,305,216 bp nucleotides, three plasmids, and 4705 coding genes. The degradation rates of atrazine at levels of 1, 10, 100 mg/L by the isolate C2 were 0.34, 1.94, 18.64 mg/L/d, respectively. The optimum temperature and pH for the isolate C2 to degrade atrazine were 30 degrees C and 7.0-9.0. Based on the metabolites detected by UPLC-TOF-MS/MS and genome annotation of the isolate C2, a common metabolic pathway of atrazine was proposed as that atrazine is firstly dechlorinated into hydroxyatrazine, and subsequently to N-isopropylammelide via dealkylation, and ultimately deaminated to cyanuric acid. Introduction of the isolate C2 into soil can enhance degradation of atrazine and thus eliminate the toxic effect of this herbicide on wheat growth. Our results indicate that the strain C2 could be a potential bioresource for bioremediation of atrazine contaminated soil. (C) 2020 Elsevier Ltd. All rights reserved.
Characterization of atrazine biotransformation by human and murine glutathione S-transferases

Abstract:Atrazine is one of the most widely used herbicides in the United States and has been detected, occasionally, at low levels in drinking water sources. The biotransformation of atrazine in humans has not been fully characterized. Rodent studies suggest Phase I-dominated biotransformation with minor Phase II-mediated biotransformation by glutathione S-transferase(s) (GST). In human urine, mercapturates of atrazine are significant metabolites, yet the specific GST form(s) responsible for glutathione (GSH) conjugation have not been identified. Using recombinant alpha, mu, pi and theta class human GSTs, we demonstrated that only hGSTP1-1 displays significant activity toward atrazine (7.1 nmol/min/mg protein). We also confirmed that mouse GST Pi (pi) protein is responsible for the GSH-dependent biotransformation of atrazine in mouse liver; recombinant mGSTP1-1 had a specific activity of 7.3-nmol/min/mg protein. Furthermore, cytosolic fractions from mouse and human liver conjugated atrazine with glutathione at rates of 282.3 and 3.0 pmol/min/mg, respectively. Docking studies of the atrazine-GST conjugate in the hGSTP1-1 substrate-binding site were used to elucidate a basis for the dramatic difference in activity between mouse GSTP1-1 and GSTP2-2 (7.14 versus 0.02 nmol/min/mg protein, respectively). The inactivity of mGSTP2-2 appears to be attributable to an indirect structural disruption of the G-site by Pro12. Possible effects of the hGSTP1 polymorphisms were investigated. No significant differences in catalytic-specific activity were noted among purified proteins corresponding to the four hGSTP1 variants: hGSTP1(*)A (most common form), hGSTP1(*)B (Ile105Val), hGSTP1(*)C (Ile105Val, Ala114Val), and hGSTP1(*)D (Ala114Val). Overall, this work supports a physiological role for GSTs in atrazine biotransformation and indicates a novel diagnostic substrate for human and mouse GSTP1-1 proteins.
The toxic effects of chronic atrazine exposure on the intestinal microbiota, metabolism and transcriptome of Pelophylax nigromaculatus larvae

Abstract:Atrazine, a widely used herbicide, is a potential threat to the intestines of animals due to its long-term persistence in water and accumulation in animal tissues, but the underlying mechanism is unclear. In this study, we evaluated the toxicity of atrazine (50, 100, and 500 μg/L) on the intestine of Pelophylax nigromaculatus larvae from Gosner stages 18–42 (about 90 days). We conducted a 16S rRNA gene amplification study, which showed that chronic exposure to atrazine changed the diversity and composition of intestinal microbiota of P. nigromaculatus larvae. Chronic exposure to atrazine (50, 100, and 500 μg/L) significantly decreased the relative abundance of Fusobacteria and U114 , while 50 μg/L and 100 μg/L atrazine resulted in a significant increase of Clavibacter. The LC-MS/MS metabolomics indicated that there were 145 significantly changed metabolites. Furthermore, atrazine led to metabolic abnormalities related to purine metabolism and biosynthesis of amino acids in the intestine. The transcriptomic results showed that atrazine exposure caused 21,120 differentially expressed genes between the control and the atrazine exposure groups. Atrazine exposure primarily disturbed the pathways related to the cytokine-cytokine receptor interaction and cell adhesion molecules. Moreover, 500 μg/L atrazine caused a significant up-regulation in the gene expression of apolipoprotein ( ApoA1, ApoB, and ApoA4 ), and fatty acid binding protein ( FABP1 ), as well as a significant down-regulation in the gene expression of superoxide dismutase ( SOD ). Our work provides insight into the ecotoxicity mechanisms of atrazine in amphibians at the level of intestinal microbiota response, metabolome and transcriptome, and serves as a basis for future research on pesticide pollutants.

Mapping and path planning in complex environments: An obstacle avoidance approach for an unmanned helicopter

Abstract:This paper presents an obstacle avoidance method that is performed with an unmanned helicopter. The approach begins with a mapping step where information from sensor data about previously unknown dangers is extracted into an occupancy grid and eventually converted into a polygonal 3D world model. This continuously updating map is used by a path planner that generates and updates a 3D trajectory guiding the vehicle through safe passages around the detected objects. The algorithms are generic but optimized for unmanned aircraft and a stereo camera as the environmental sensor. Computation is fully executed on board so that a ground control station is only needed for supervision. With successful obstacle detection and avoidance flight tests, the paper shows the qualification of the presented method under real operational conditions.
Transcriptomic profiling of atrazine phytotoxicity and comparative study of atrazine uptake, movement, and metabolism in Potamogeton crispus and Myriophyllum spicatum

Abstract:The accumulation of atrazine in sediments raises wide concern due to its potential negative effects on aquatic environments. Here we collected sediments and different submerged macrophytes to simulate natural shallow lakes and to measure atrazine levels and submerged macrophyte biomass. We determined gene expressions in submerged macrophytes treated with or without atrazine. We also examined atrazine concentrations and its metabolite structures in submerged macrophytes. When the initial concentration of atrazine in sediments ranged from 0.1 to 2.0 mg kg−1 dry weight (DW), atrazine levels in the pore water of the sediments ranged from 0.003 to 0.05 mg L−1 in 90 days. Atrazine did not show obvious long-term effects on the biomass of Potamogeton crispus and Myriophyllum spicatum (P > 0.05). On day 90, gene expressions related to cell wall in P. crispus were changed by atrazine phytotoxicity. Moreover, the decrease in the number genes controlling light-harvesting chlorophyll a/b-binding proteins verified the toxic effects of atrazine on the photosynthesis of M. spicatum. Compared with unexposed plants on day 90, ribosome pathway was significantly enriched with differentially expressed genes after submerged macrophytes were exposed to 2.0 mg kg−1 DW atrazine (P < 0.05). In addition, shoots and roots of P. crispus and M. spicatum could absorb the equal amount of atrazine (P > 0.05). Once absorbed by submerged macrophytes, atrazine was degraded into 1-hydroxyisopropylatrazine, hydroxyatrazine, deethylatrazine, didealkylatrazine, cyanuric acid, and biuret, and some of its metabolites could conjugate with organic acids, cysteinyl β-alanine, and glucose. This study establishes a foundation for aquatic ecological risk assessments and the phytoremediation of atrazine in sediments.

Abstract:Effects of concentration of atrazine on growth of Scenedesmus quadricauda and Microcystis aeruginosa cultured in MA medium were studied by determining growth rate and content of chlorophyll a in laboratory.With the maximum specific growth rate,expressed by count of algal cells as evaluation index,the two algae were compared in sensitivity to atrazine.When low in concentration,atrazine stimulated growth of Scenedesmus quadricauda and Microcystis aeruginosa;but when high,it demonstrated a reverse effect.And its stimulating effect was greater on Scenedesmus quadricauda than on Microcystis aeruginosa,whereas its inhibiting effect showed the other way.

Abstract:Atrazine is currently one of the most commonly used agrochemicals in the United States and elsewhere. Here, we studied the immunoexpression of molecular markers of mammalian testicular functions: androgen receptor (AR), promyelocytic leukemia zinc finger (PLZF), GDNF family receptor alpha-1 (GFRA1), VASA/DDX4 (DEAD-Box Helicase 4) as well as the levels of intratesticular and intra-epididymal estradiol (E2) and dihydrotestosterone (DHT), tumor necrosis factor-alpha (TNF-α), interferon-gamma (IFN-γ), interleukins (IL-1β and IL-6, IL-10) and testicular chemokines (CXCL-1, CCL-2 and CCL3) in BalB/c mice after a sub-acute gavage treatment with a gonado-toxin, atrazine (50 mg/kg body wt.) for three days. We found high numbers of AR immunopositive Sertoli cells and low numbers of GFRA1, PLZF and VASA/DDX4-positive germ cells in the seminiferous tubule regions of the testes. While TNF-α level in the testes fell and remained unchanged in the epididymides, IFN-γ levels in the testes remained constant but increased in the epididymides. E2 and DHT concentrations remained unaltered in the testes but were changed in the epididymides. There were no significant changes in the levels of interleukins in the testis and epididymis. Intratesticular chemokines were also not significantly altered, except for CCL-4, which was increased in the testis. Light microscopy of the epididymis showed detached epithelium and some detached cells in the lumen. It is concluded that atrazine changed the inflammatory status of the gonads and highlighted Sertoli and undifferentiated spermatogonia as important targets for atrazine's toxic effects in the testis of mice. Concerning the epididymis, atrazine altered the epididymal hormonal concentrations and promoted histopathological modifications in its parenchyma.

Abstract:Because of an increasing demand for limited health care resources as well as an increasing impact of legal aspects in ultrasound practice, it has become necessary to ensure that screening procedures are cost-effective and clinically effective and to develop systems that could help sonographists in certifying their practice. Based on the literature, various approaches for quality control are reviewed. Special emphasis is put on the use of fetal biometry and statistical tools to develop quality control systems based on both qualitative and quantitative approach. Besides their potentials for audit and quality control, such tools could also be useful during the process of training.

Abstract:Atrazine is a herbicide commonly used on grain crops. It has been identified as a potential endocrine disruptor, especially in freshwater species. However, there is little research on its endocrine disruptor effect in crustaceans. The current study was designed to estimate the median lethal concentration of atrazine (96-h LC50) and to assess the response of the freshwater crayfish Procambarus clarkii as a bioindicator for endocrine disruptors by measuring levels of steroid hormones, total protein, changes in Cytochrome P450 1B1, and histopathology of the ovary and testis. The results showed that the 96-h LC50 of atrazine for male and female P. clarkii was 10.62 and 12.66 mg L−1, respectively. LC10 (1.06 and 1.27mg/l) and LC25 (2.66 and 3.17 mg/l) values for males and females, were chosen for sublethal studies for 28 days. The results indicated that females are more tolerant to atrazine than males. Testosterone and total protein levels were significantly lower, while estradiol and progesterone levels were significantly higher in atrazine-exposed crayfish compared to the control. Also, the levels of CYP450 1B1 increased significantly in the testis, ovary, and hemolymph. The most noticeable histopathological changes in the testis were distorted architecture, spermatogonia distribution disruption in some testicular acini, Vacuolation, and hyperplasia. In the ovary, the separation between the ovarian epithelium and oogenetic pouch and lysis in the ovarian epithelium was noted. These parameters could be used as biomarkers to assess herbicide toxicity. Furthermore, P. clarkii could be used as an indicator of endocrine-disrupting chemicals in waterways.
Developmental atrazine exposure in zebrafish produces the same major metabolites as mammals along with altered behavioral outcomes

Abstract:Atrazine (ATZ) is the second most commonly applied agricultural herbicide in the United States. Due to contamination concerns, the U.S. EPA has set the maximum contaminant level in potable water sources at 3 parts per billion (ppb; μg/l). Depending on the time of year and sampling location, water sources often exceed this limit. ATZ is an endocrine disrupting chemical in multiple species observed to target the neuroendocrine system. In this study the zebrafish vertebrate model was used to test the hypothesis that a developmental ATZ exposure generates metabolites similar to those found in mammals and alters morphology and behavior in developing larvae. Adult AB zebrafish were bred, embryos were collected, and exposed to 0, 0.3, 3, or 30 ppb ATZ from 1 to 120 h post fertilization (hpf). Targeted metabolomic analysis found that zebrafish produce the same major ATZ metabolites as mammals: desethyl atrazine (DEA), deisopropyl atrazine (DIA), and diaminochloroatrazine (DACT). The visual motor response test at 120 hpf detected hyperactivity in larvae in the 0.3 ppb treatment group and hypoactivity in the 30 ppb treatment group (p < 0.05). Further analysis into behavior during the dark and light phases showed zebrafish larvae exposed to 0.3 ppb ATZ had an increase in total distance moved in the first light phase and time spent moving in the first dark and light phase (p < 0.05). Alternatively, a decrease in total distance moved was observed in the second and third dark phase in zebrafish exposed to 30 ppb ATZ (p < 0.05). No significant differences were observed for any of the morphological measurements following ATZ exposure from 1 to 120 hpf (p > 0.05). These findings suggest that a ATZ exposure during early development generates metabolite profiles similar to mammals and leads to behavioral alterations supporting ATZ as a neurodevelopmental toxicant.

Inhibitory Effects of Atrazine on Chlorella Vulgaris As Assessed by Real-Time Polymerase Chain Reaction.

Abstract:Atrazine, a highly toxic herbicide, is frequently detected in surface water because of its heavy application. Algae are among the aquatic organisms most susceptible to atrazine pollution in water. In the present study, the aquatic alga Chlorella vulgaris Beijerinck was chosen to assess the acute toxicity of atrazine (48–96 h) in terms of gene transcription and physiological changes. A real‐time polymerase chain reaction (PCR) assay was used to quantify transcript levels of three photosystem genes in C. vulgaris . The diel patterns for regulation of the psaB (photosystem I reaction center protein subunit B), psbC (an integral membrane protein component of photosystem II), and rbcL (large subunit of ribulose‐1,5‐bisphosphate carboxylase oxygenase) gene transcripts were successfully quantified. Results showed that atrazine reduced the transcript abundances of three target genes and that the abundances decreased with increasing atrazine concentration. The determined smallest transcript levels of psaB, psbC , and rbcL , which occurred at the highest atrazine concentration tested (400 μg/L), were only 34.6, 34.6, and 8.1%, respectively, of the control sample value. Exposure to atrazine increased the level of malondialdehyde by 1.74‐fold (the highest value) in C. vulgaris , suggesting potential oxidative damage to the alga. The activities of antioxidation enzymes (e.g., superoxide dismutase, peroxidase, and catalase) also increased markedly in the presence of atrazine, with maximum increases of 1.82‐, 1.59‐, and 2.31‐fold, respectively. These elevated activities may help to alleviate the oxidative damage. Our results demonstrate that atrazine is highly toxic to this alga and that real‐time PCR is an efficient technique for assessing the toxicity of xenobiotic compounds in algae.
Contribution of differential alteration in oxidative stress and anti-oxidation related molecular signals to toxicity difference between atrazine and its main metabolites in nematodes

Abstract:As a widely used herbicide, atrazine and its two main metabolites of deethylatrazine (DEA) and deisopropylatrazine (DIA) pose an exposure risk for both human beings and animals in the environment. In this study, Caenorhabditis elegans was selected as an in vivo model to compare the toxicity between atrazine and its main metabolites. Upon exposure from the larval stage L1 to adult day 3, both DEA and DIA showed less toxicity on locomotion and reproduction compared with atrazine at concentration of 0.001, 0.01 0.1 and 1 mg/L for parental generation. In addition, exposure to DEA and DIA at concentration of 0.1 mg/L also induced less transgenerational toxicity on locomotion than exposure to atrazine for both parental generation and offspring of F1-F4. Accordingly, exposure to DEA and DIA caused less ROS production and alteration in the expression of some genes (mev-1, gas-1, and clk-1) governing oxidative stress compared to atrazine. Meanwhile, DEA and DIA lead to less increase in expression of superoxide dismutase genes (sod-2 and sod-3) and SOD-3::GFP than atrazine. Moreover, atrazine and its two main metabolites differentially activated the daf-16 encoding FOXO transcriptional factor in insulin signaling pathway during the control of downstream target of SOD-3. Overall, our results highlighted the important role of oxidative stress and anti-oxidation related molecular signals in mediating toxicity of atrazine, DEA and DIA, which provided a novel explanation for the different toxicity between atrazine and its main metabolites.

Abstract:Atrazine is an herbicide with several known toxicologically relevant effects, including interactions with other chemicals. Atrazine increases the toxicity of several organophosphates and has been shown to reduce the toxicity of triclosan to D. magna in a concentration dependent manner. Atrazine is a potent activator in vitro of the xenobiotic-sensing nuclear receptor, HR96, related to vertebrate constitutive androstane receptor (CAR) and pregnane X-receptor (PXR). RNA sequencing (RNAseq) was performed to determine if atrazine is inducing phase I-III detoxification enzymes in vivo, and estimate its potential for mixture interactions. RNAseq analysis demonstrates induction of glutathione S-transferases (GSTs), cytochrome P450s (CYPs), glucosyltransferases (UDPGTs), and xenobiotic transporters, of which several are verified by qPCR. Pathway analysis demonstrates changes in drug, glutathione, and sphingolipid metabolism, indicative of HR96 activation. Based on our RNAseq data, we hypothesized as to which environmentally relevant chemicals may show altered toxicity with co-exposure to atrazine. Acute toxicity tests were performed to determine individual LC50 and Hillslope values as were toxicity tests with binary mixtures containing atrazine. The observed mixture toxicity was compared with modeled mixture toxicity using the Computational Approach to the Toxicity Assessment of Mixtures (CATAM) to assess whether atrazine is exerting antagonism, additivity, or synergistic toxicity in accordance with our hypothesis. Atrazine-triclosan mixtures showed decreased toxicity as expected; atrazine-parathion, atrazine-endosulfan, and to a lesser extent atrazine-p-nonylphenol mixtures showed increased toxicity. In summary, exposure to atrazine activates HR96, and induces phase I-III detoxification genes that are likely responsible for mixture interactions.
A web-based, self-controlled mechanism to support students learning SQL

Abstract:Database systems play a very important role in computer science. Most software and mobile applications store and populate data on a server site. Relational databases dominate the professional market which requires students to be familiar with Structured Query Language (SQL). The client-server is the standard architecture in the database industry. Therefore, students are strongly motivated to have more hands-on SQL assignments and projects using a 3-tier architecture in database courses. For the best learning experience, each student should have his own tables and environment so that his data won't interfere with data from other students. However, it is difficult for teachers to manage individual privileges on every student's tables and SQL programs if the students are accessing a common (shared) database. This paper proposes a mechanism that students can use to create their database account and grant proper privileges by themselves. This minimizes the instructor's workload by eliminating the need to manage students' accounts and tables when teaching databases. Our proposal also provides an interface that allows students to perform exercises and allows the instructor to review and share students' realtime answers in class. Our methods will minimize the workload for grading SQL assignments when teaching databases, while providing students with the opportunity to practice with more lab assignments. Students can also verify whether their SQL statements are working or not using the same review module as the instructor. The results show that for assignments on simple SQL statements, grading efficiency is increased by at least 60%. This allows the instructor to increase the amount of assignments by 133%. As a result of this increased practice, the students' grades improved by 12%.

Abstract:The effects of (Rac)-metolachlor and (S)-metolachlor on the avoidance behavior, bodyweight change and in vivo enzyme activity of earthworms (Eisenia foetida) were determined and compared in this study. The effects of (Rac)-metolachlor on the enzyme activities of E. foetida and bodyweight were more significant than those of (S)-metolachlor at the same concentrations. In the short term (2 d, 7 d), (S)-metolachlor had faster effects on cellulase and catalase activities of E. foetida. However, in the relatively long term (14 d, 28 d), (Rac)-metolachlor had higher toxic effects on cellulase and catalase activities. The inter-group difference between (Rac)-metolachlor and (S)-metolachlor on E. foetida enzyme activities was the most significant for catalase, and the least significant for cellulase. The test of avoidance behavior shows that earthworms are more sensitive to the stimulation of (Rac)-metolachlor than to that of (S)-metolachlor. The results will help to develop an understanding of the biologically mediated environmental processes of these two herbicides.
Biochemical characterization of metabolism-based atrazine resistance in Amaranthus tuberculatus and identification of an expressed GST associated with resistance

Abstract:Rapid detoxification of atrazine in naturally tolerant crops such as maize (Zea mays) and grain sorghum (Sorghum bicolor) results from glutathione S-transferase (GST) activity. In previous research, two atrazine-resistant waterhemp (Amaranthus tuberculatus) populations from Illinois, U.S.A. (designated ACR and MCR), displayed rapid formation of atrazine-glutathione (GSH) conjugates, implicating elevated rates of metabolism as the resistance mechanism. Our main objective was to utilize protein purification combined with qualitative proteomics to investigate the hypothesis that enhanced atrazine detoxification, catalysed by distinct GSTs, confers resistance in ACR and MCR. Additionally, candidate AtuGST expression was analysed in an F2 population segregating for atrazine resistance. ACR and MCR showed higher specific activities towards atrazine in partially purified ammonium sulphate and GSH affinity-purified fractions compared to an atrazine-sensitive population (WCS). One-dimensional electrophoresis of these fractions displayed an approximate 26-kDa band, typical of GST subunits. Several phi- and tau-class GSTs were identified by LC-MS/MS from each population, based on peptide similarity with GSTs from Arabidopsis. Elevated constitutive expression of one phi-class GST, named AtuGSTF2, correlated strongly with atrazine resistance in ACR and MCR and segregating F2 population. These results indicate that AtuGSTF2 may be linked to a metabolic mechanism that confers atrazine resistance in ACR and MCR.
Atrazine and Diuron Effects on Survival, Embryo Development, and Behavior in Larvae and Adult Zebrafish

Abstract:Atrazine and Diuron are widely used herbicides. The use of pesticides contaminates the aquatic environment, threatening biodiversity and non-target organisms such as fish. In this study, we investigated the effects of acute exposure for 96 h hours to atrazine and diuron commercial formulations in zebrafish (Danio rerio, wild-type AB) embryos and larvae and adult stages. We observed a significant concentration-dependent survival decrease and hatching delays in animals exposed to both herbicides and in the frequency of malformations compared to the control groups. Morphological defects included cardiac edema, tail reduction, and head malformation. At 7 days post-fertilization (dpf), atrazine exposure resulted in a reduction in the head length at 2, 2.5, and 5 mg/L and increased the ocular distance at 1, 2, 2.5, and 5 mg/L atrazine when compared to controls. At the same age, diuron increased the ocular distance in animals exposed to diuron (1.0 and 1.5 mg/L) and no effects were observed on the head length. We also evaluated a behavioral repertoire in larvae at 7 dpf, and there were no significant differences in distance traveled, mean speed, time in movement, and thigmotaxis for atrazine and diuron when animals were individually placed in a new environment. The cognitive ability of the larvae was tested at 7 dpf for avoidance and optomotor responses, and neither atrazine nor diuron had significant impacts when treated groups were compared to their corresponding controls. Adults' behavior was evaluated 7 and 8 days after the end of the acute herbicide exposure. Exploration of a new environment and associated anxiety-like parameters, social interaction, and aggressiveness were not altered. Our results highlight the need for further studies on the sublethal effects of both herbicides and the consideration of the effects of commercial formulas vs. isolated active ingredients. It also emphasizes the need to take sublethal effects into consideration when establishing the environmental limits of residues.

Abstract:Atrazine (ATR) is one of the most commonly used herbicides that could directly impair the growth and health of organisms in mariculture areas and adversely affect human health through the food chain. This study investigated the contaminant occurrence, migration, and transformation of ATR and three of its chlorinated metabolites, namely deethylatrazine (DEA), deisopropylatrazine (DIA), and didealkylatrazine (DDA), in surface seawater, sediment, and aquatic organisms from the Xiangshan Harbor. ATR was detected in all samples, while DIA and DDA were only respectively detected in aquatic and seawater samples. The distribution of ATR and its metabolites presented different patterns depending on the geographic location and showed a higher level in the aquaculture area than that in the non-aquaculture area. The bioaccumulation of ATR in aquaculture organisms showed that benthic organisms, such as Ditrema, and Sinonovacula constricta (Sin), had increased levels. The ecological risks indicated that ATR posed medium or high risks to algae in the water phase of the study area. The microcosm experiment showed that the main fate of ATR in the simulated microenvironment was sedimentation, which followed the first-order kinetic equation. The ATR in the sediment could be enriched 3–5 times in Sin, and its major metabolites were DEA and DIA.

Abstract:The pesticide atrazine poses a potential threat to the health of frogs living in farmland areas. The exposure concentration in traditional pesticide experiments is usually constant, while pesticide pollution in actual water may fluctuate due to periodic or seasonal application. We examined the effects of different concentrations of atrazine (50, 100 and 500 μg/L) over a 14-day exposure and a 7-day recovery on intestinal histology, bacterial composition and intestinal metabolites of male Pelophylax nigromaculatus. HE staining revealed that after a 14-day atrazine exposure, the 100 μg/L and 500 μg/L groups showed obvious cysts and significantly decreased intestinal crypt depth and villus height. After a 7-day recovery, the damaged intestine in the 100 μg/L group was partially recovered, while in the 500 μg/L exposure group there was no improvement. 16S rRNA gene analysis of intestinal bacteria showed that 500 μg/L atrazine exposure significantly caused a persistent decrease in bacterial α diversity. Compared to the control and other atrazine exposure groups, the 500 μg/L group showed significant changes in the relative abundance of predominant bacteria. In addition, most dominant bacteria in the 500 μg/L recovery group showed significant differences with the 50 μg/L and 100 μg/L recovery groups. Nontargeted metabolomics profiling based on UPLC/MS analysis showed that atrazine exposure and recovery induced changes in the intestinal metabolic profile. The changes in metabolites were mainly related to purine/pyrimidine metabolism, glycine, serine and threonine metabolism, and arginine and proline metabolism. In general, these pathways were closely related to energy metabolism and amino acid metabolism. These results suggest that the short-term exposure to 500 μg/L atrazine causes persistent harm to intestinal health. This study is an important step toward a better understanding of the toxic effects of atrazine exposure and recovery in frog intestines.

Abstract:The intestine is the main organ for nutrient absorption in amphibians. It is sensitive to atrazine, which is a herbicide widely used in agricultural areas. At present, there is a lack of systematic research on the effect of atrazine on the amphibian intestine. In this study, we evaluated the effects of atrazine exposure (0, 50 μg/L, 100 μg/L, and 500 μg/L) for 20 days on intestinal histology, microbiota and short chain fatty acids in Pelophylax nigromaculatus tadpoles. Our research showed that 500 μg/L atrazine exposure significantly decreased the height of microvilli and epithelial cells, and altered the composition and diversity of intestinal microbiota in P. nigromaculatus tadpoles compared to the control. At the phylum level, the abundance of Bacteroidetes and Fusobacteria increased significantly, while that of Verrucomicrobia and Firmicutes decreased significantly in the 500 μg/L atrazine treatment group. At the genus level, Akkermansia and Lactococcus had significantly lower abundance in the 100 μg/L and 500 μg/L atrazine exposure group, while Cetobacterium was only detected in the 100 μg/L and 500 μg/L atrazine treated group. Also, function prediction of intestinal microbiota showed that atrazine treatment significantly changed the metabolism pathways of P. nigromaculatus tadpoles. In addition, 500 μg/L atrazine exposure changed the content of short chain fatty acids by significantly increasing the content of total SFCAs, butyric acid, and valeric acid, and decreasing the content of isovaleric acid in the intestine. Taken together, atrazine exposure could affect the intestinal histology and induce intestinal microbiota imbalance and metabolic disorder in amphibian tadpoles.
