Abstract:Manganese peroxidases (MnP) from the white-rot fungi Phanerochaete chrysosporium catalyse the oxidation of Mn2+ to Mn3+, a strong oxidizer able to oxidize a wide variety of organic compounds. Different approaches have been used to unravel the enzymatic properties and potential applications of MnP. However, these efforts have been hampered by the limited production of native MnP by fungi. Heterologous expression of MnP has been achieved in both eukaryotic and prokaryotic expression systems, although with limited production and many disadvantages in the process. Here we described a novel molecular approach for the expression and purification of manganese peroxidase isoform 1 (MnP1) from P. chrysosporium using an E. coli-expression system. The proposed strategy involved the codon optimization and chemical synthesis of the MnP1 gene for optimised expression in the E. coli T7 shuffle host. Recombinant MnP1 (rMnP1) was expressed as a fusion protein, which was recovered from solubilised inclusion bodies. rMnP1 was purified from the fusion protein using intein-based protein purification techniques and a one-step affinity chromatography. The designated strategy allowed production of an active enzyme able to oxidize guaiacol or Mn2+.

Abstract:A Mn(II)-dependent peroxidase found in the extracellular medium of ligninolytic cultures of the white rot fungus, Phanerochaete chrysosporium, was purified by DEAE-Sepharose ion-exchange chromatography, Blue Agarose chromatography, and gel filtration on Sephadex G-100. Sodium dodecyl sulfate-gel electrophoresis indicated that the homogeneous protein has an Mr of 46,000. The absorption spectrum of the enzyme indicates the presence of a heme prosthetic group. The pyridine hemochrome absorption spectrum indicates that the enzyme contained one molecule of heme as iron protoporphyrin IX. The absorption maximum of the native enzyme (406 nm) shifted to 433 nm in the reduced enzyme and to 423 nm in the reduced-CO complex. Both CN- and N3- readily bind to the native enzyme, indicating an available coordination site and that the heme iron is high spin. The absorption spectrum of the H2O2 enzyme complex, maximum at 420 nm, is similar to that of horseradish peroxidase compound II. P. chrysosporium peroxidase activity is dependent on Mn(II), with maximal activity attained above 100 μM. The enzyme is also stimulated to varying degrees by α-hydroxy acids (e.g., malic, lactic) and protein (e.g., gelatin, albumin). The peroxidase is capable of oxidizing NADH and a wide variety of dyes, including Poly B-411 and Poly R-481. Several of the substrates (indigo trisulfonate, NADH, Poly B-411, variamine blue RT salt, and Poly R-481) are oxidized by this Mn(II)-dependent peroxidase at considerably faster rates than those catalyzed by horseradish peroxidase. The enzyme rapidly oxidizes Mn(II) to Mn(III); the latter was detected by the characteristic absorption spectrum of its pyrophosphate complex. Inhibition of the oxidation of the substrate diammonium 2,2-azino-bis(3-ethyl- 6-benzothiazolinesulfonate) (ABTS) by Na-pyrophosphate suggests that Mn(III) plays a role in the enzyme mechanism. © 1985 Academic Press, Inc.

Abstract:Trichoderma parestonica is a novel source of manganese peroxidase (MnP), [E.C.1.11.1.13] enzyme. The following was the sequence in which T. parestonica induced the production of MnP in the liquid culture medium: rice husk, corncob, sawdust, coir-dust and dried luffa on days 3, 4, 5, 2 and 3, respectively. The molecular weight of the enzyme was found to be 45kDa, determined from SDS-PAGE analysis which was further confirmed by mass analysis and was found to be 51.9kDa. Enzyme characterization parameters like optimum pH, temperature, Km, K cat and catalytic efficiency are 5, 20oC, 0.014mM (for Mn 2+ ) and 0.028mM (for H 2 O 2 ), 23.9 S -1 and 1.7 μM -1 S -1 respectively. Effect of various metal ions, such as bivalent Copper(Cu), Cobalt(Co), Mercury(Hg), Nickel(Ni), Cadmium(Cd), trivalent Chromium(Cr), Iron(Fe), Tetra-valent cerium(Ce), Penta-valent vanadium(V) and hexavalent molybdenum(Mo) the enzyme-catalyzed steady-state velocity was studied. The inhibition constant and nature of inhibition by these metal ions were determined. In the current investigation, MnP catalytic activities demonstrated that it could effectively catalyze the oxidation of phenols, aromatic amines and wastewater treatment. Abstract:The full-length cDNA sequence of Ginkgo biloba manganese superoxide dismutase gene(designated as GbMnSOD EF633506) were isolated from G.biloba for the first time. The full-length cDNA of GbMnSOD contained a 681 bp open reading frame(ORF) encoding a 226-amino-acid protein. The deduced protein have a predicted molecular weight of 25.5 kD and a calculated pI of 8.97. 3D structure modeling showed that GbMnSOD contain 12 helixes and 3 sheets and have a basketry motif in the enzyme core. Phylogenetic tree analysis revealed that GbMnSOD shared the same ancestor with other MnSOD. The result of southern analysis show that the MnSOD genes is encoded by a small gene family in the G. biloba. Northern hybridization analysis show that GbMnSOD expressed in leaves,stem,root and fruit and have the highest expression in leaves. The expression of MnSOD could be induced by ABA,IAA,sucrose,mannitol,NaCl and low temperature.

Abstract:Visual motion cues are used by tower controllers to support both visual and anticipated separation. Some of these cues are tabulated as part of the overall set of visual features used in towers to separate aircraft. An initial analyses of one motion cue, landing deceleration, is provided as a basis for evaluating how controllers detect and use it for spacing aircraft on or near the surface. Understanding cues like it will help determine if they can be safely used in a remote/virtual tower in which their presentation may be visually degraded.

Abstract:In order to construct an expression system for the particulate methane mono-oxygenase (pMMO) gene (pmo), the structural gene cluster pmoCAB amplified from Methylosinus trichosporium OB3b was inserted into a shuttle vector pBS305 under the control of a dsz promoter and transformed into Rhodococcus erythropolis LSSE8-1. A stable transformant was successfully obtained using ethane as the sole carbon source. Fluorescence in situ hybridization results showed that the dsz promoter allowed the pmo genes to be transcribed in the recombinant strain. The effects of Cu2+ and Zn2+ concentrations on cell growth and pMMO activity in ethane-containing medium were examined. It was discovered that 7.5 microM Cu2+ and 1.8 microM Zn2+ were suitable to achieve high cell concentration and pMMO activity, but the amount of methanol accumulated during methane oxidation by the recombinant strain was still low.

Abstract:A novel thermostable alkaline β-mannanase from alkaliphilic Bacillus sp. N16-5 was expressed successfully in Pichia pastoris GS115. The combined usage of inducible and constitutive promoters (AOX1 and GAP) enhanced the expression of β-mannanase. Among the parameters investigated in shaking flask cultures, the pH value of medium had significant influence on the production of β-mannanase by recombinant P. pastoris. β-Mannanase produced at pH 7.0 was 6.7 times of that at pH value of 6.0. The highest β-mannanase activity of 32.2IU/ml in culture supernatant was achieved at 120h of cultivation in BMGY medium (pH 7.0). The recombinant β-mannanase was purified and characterized. The purified β-mannanase produced by P. pastoris has optimum pH of 10.0 and optimum temperature of 70°C, which are very close to those of the native enzyme from alkaliphilic Bacillus sp. N16-5. However, much higher thermal stability and pH stability were observed in recombinant β-mannanase. These properties make the recombinant β-mannanase more useful in the detergent industries, the pulp and paper processing and other industrial processes.

Abstract:The white-rot basidiomycete Phanerochaete chrysosporium produces lignin peroxidases (LiPs), a family of extracellular glycosylated heme proteins, as major components of its lignin-degrading system. Upto 15 LiP isozymes, ranging in M r values from 38000 to 43000, are produced depending on culture conditions and strains employed. Manganese-dependent peroxidases (MnPs) are a second family of extracellular heme proteins produced by P. chrysosporium that are also believed to be important in lignin degradation by this organism. LiP and MnP production is seen during secondary metabolism and is completely suppressed under conditions of excess nitrogen and carbon. Excess Mn(II) in the medium, on the other hand, suppresses LiP production but enhances MnP production. Nitrogen regulation of LiP and MnP production is independent of carbon and Mn(lI) regulation. LiP activity is also affected by idiophasic extracetlular proteases. Intracellular cAMP levels appear to be important in regulating the production of LiPs and MnPs, although LiP production is affected more than MnP production. Studies on the sequencing and characterization of lip cDNAs and genes of P. chrysosporium have shown that the major LiP isozymes are each encoded by a separate gene. Each lip gene encodes a mature protein that is 343-344 amino acids long, contains 1 putative N-glycosylation site, a number of putative O-glycosylation sites, and is preceded by a 27-28-amino acid leader peptide ending in a Lys-Arg cleavage site. The coding region of each lip gene is interrupted by 8-9 introns (50-63 bp), and the positions of the last two introns appear to be highly conserved. There are substantial differences in the temporal transcription patterns of the major lip genes. The sequence data suggest the presence of three lip gene subfamilies. The genomic DNA of P. chrysosporium strain BKMF-1767 was resolved into 10 chromosomes (genome size of 29 Mb), and that of strain ME-446 into 11 chromosomes (genome size of 32 Mb). The lip genes have been localized to five chromosomes in BKMF-1767 and to four chromosomes in ME-446. DNA transformation studies have reported both integrative and non-integrative transformation in P. cho'sosporium.

Abstract:Aflatoxins are naturally occurring high-toxic secondary metabolites, which cause worldwide environmental contaminations and wastes of food and feed resources and severely threaten human health. Thus, the highly efficient methods and technologies for detoxification of aflatoxins are urgently needed in a long term. In this work, we report the construction of recombinant Kluyveromyces lactis strains GG799(pKLAC1-Phs mnp ), GG799(pKLAC1-Plo mnp ), GG799(pKLAC1-Phc mnp ), and then the food-grade expression of the three manganese peroxidases in these strains, followed by the degradation of aflatoxin B 1 (AFB 1 ) using the fermentation supernatants. The expression of the manganese peroxidases was achieved in a food-grade manner since Kluyveromyces lactis is food-safe and suitable for application in food or feed industries. The inducible expression process of the optimal recombinant strain GG799(pKLAC1-Phc mnp ) and the aflatoxin B 1 degradation process were both optimized in detail. After optimization, the degradation ratio reached 75.71%, which was an increase of 49.86% compared to the unoptimized results. The degradation product was analyzed and determined to be AFB 1 -8,9-dihydrodiol. The recombinant strain GG799(pKLAC1-Phc mnp ) supernatants degraded more than 90% of AFB 1 in the peanut samples after twice treatments. The structural computational analysis for further mutagenesis of the enzyme PhcMnp was also conducted in this work. The food-grade recombinant yeast strain and the enzyme PhcMnp have potential to be applied in food or feed industries.

Abstract:A dwarf transgenic mouse (DTM) line has been established in which mice express relatively high levels of a mutated bovine (b) GH gene. This bGH analog binds to mouse liver membrane preparations with an affinity similar to that of wild-type bGH. The mean growth ratio of these mice is approximately 0.7 relative to that of their nontransgenic littermates. Serum insulin-like growth factor-I (IGF-I) levels of DTM were found to be approximately half those in nontransgenic littermates. Liver GH receptor levels were up-regulated in DTM or wild-type bGH transgenic mice. Pituitary GH levels were negatively correlated with serum IGF-I concentrations. Wild-type bGH transgenic mice contain relatively high serum IGF-I and low pituitary GH levels, whereas DTM possess low serum IGF-I and high pituitary GH levels. The decrease in serum IGF-I resulting from the interaction between the bGH analog, the endogenous mouse GH, and GH receptor(s) apparently leads to a dwarf phenotype. These data suggest that this bGH analog has uncoupled GH ligand-receptor binding from IGF-I production and acts as a functional antagonist to the action of endogenous mGH.
A Novel Extracellular Multicopper Oxidase from Phanerochaete chrysosporium with Ferroxidase Activity

Abstract:ABSTRACT Lignin degradation by the white rot basidiomycete Phanerochaete chrysosporium involves various extracellular oxidative enzymes, including lignin peroxidase, manganese peroxidase, and a peroxide-generating enzyme, glyoxal oxidase. Recent studies have suggested that laccases also may be produced by this fungus, but these conclusions have been controversial. We identified four sequences related to laccases and ferroxidases (Fet3) in a search of the publicly available P. chrysosporium database. One gene, designated mco1 , has a typical eukaryotic secretion signal and is transcribed in defined media and in colonized wood. Structural analysis and multiple alignments identified residues common to laccase and Fet3 sequences. A recombinant MCO1 (rMCO1) protein expressed in Aspergillus nidulans had a molecular mass of 78 kDa, as determined by sodium dodecyl sulfate-polyacrylamide gel electrophoresis, and the copper I-type center was confirmed by the UV-visible spectrum. rMCO1 oxidized various compounds, including 2,2′-azino(bis-3-ethylbenzthiazoline-6-sulfonate) (ABTS) and aromatic amines, although phenolic compounds were poor substrates. The best substrate was Fe 2+ , with a K m close to 2 μM. Collectively, these results suggest that the P. chrysosporium genome does not encode a typical laccase but rather encodes a unique extracellular multicopper oxidase with strong ferroxidase activity.

Comparative characterization of glyoxal oxidase from Phanerochaete chrysosporium expressed at high levels in Pichia pastoris and Trichoderma reesei

Abstract:In the secretome of Phanerochaete chrysosporium, a white-rot fungus serving as a model organism to elucidate lignocellulose deconstruction, the copper containing metalloprotein glyoxal oxidase (GLOX) is potentially involved in the crucial production of hydrogen peroxide to fuel and initiate oxidative biomass degradation by lignin-degrading peroxidases. Its ability to oxidize a variety of aldehydes and α-hydroxy carbonyls with the concomitant reduction of dioxygen to hydrogen peroxide has attracted attention for its application as green biocatalyst in different industrial fields. Here we report and compare two efficient processes for the heterologous production of GLOX from P. chrysosporium using the well-established methanolytic yeast Pichia pastoris and the filamentous fungus Trichoderma reesei as expression hosts with subsequent purification by anion exchange and hydrophobic interaction chromatography. Both processes were shown to be suitable for the production of the target protein at high levels. GLOX produced in T. reesei carries mainly Man5 glycosylation while the enzyme produced in P. pastoris exhibits the typical high-mannose type N-glycosylation. The enzyme expressed in P. pastoris showed slightly higher specific activities which correlates with the higher copper loading of 65.5 % compared to 51.9 % for the protein from T. reesei. The pH optimum for both recombinant proteins was 6.0, however, GLOX activity was found to be highly affected by different buffer species. Both enzymes showed very similar substrate affinities and turnover numbers with the highest catalytic efficiency observed for methylglyoxal. GLOX from both expression hosts is therefore a suitable enzyme for further mechanistic characterization and application studies.
Heterologous Expression of a Synthetic Gene Encoding Lignin Peroxidase H2 in Pichia Pastoris

Abstract:After optimizing the gene sequence,a lignin peroxidase H2(LiPH2) gene was chemically synthesized and then successfully inserted into selected expression vectors. The recombinant vectors containing different secretion signals and selectable markers were then transformed into the Pichia pastoris genome. DOA extracted from the recombinant P. pastoris was amplified using the specific primers and analyzed by electrophoresis to confirm the correct integration of the LiPH2 gene in the DNA sequence. Eight different recombinant strains were constructed. Only 1 of them successfully expressed LiPH2. Factors that affect the expression were explored and the results are summarized. The host strain SMD1168 was not as effective as X-33 in the expression of the recombinant protein,which indicated that disruption of the pep4 gene in P. pastoris had an adverse effect on the in vitro expression of LiPH2. The native secretion signal of LiPH2 was adequate for both the secretion and expression of the protein. Using vectors pPIC9K and pPIC3.5K led to no expression of LiPH2 in GS115 and SMD1168,but vector pPICZA was more successful. The recombinant protein was in an inactive form. The molecular weight of the recombinant protein was larger than that of the native protein,which suggests that over-glycosylation may be responsible for the inactivation of the enzyme. Tag addition may also influence the protein activation.

Abstract:Thermophilic fungi are a promising source of thermostable enzymes able to hydrolytically or oxidatively degrade plant cell wall components. Among these enzymes are lytic polysaccharide monooxygenases (LPMOs), enzymes capable of enhancing biomass hydrolysis through an oxidative mechanism. Myceliophthora thermophila (synonym Sporotrichum thermophile), an Ascomycete fungus, expresses and secretes over a dozen different LPMOs. In this study, we report the overexpression and biochemical study of a previously uncharacterized LPMO (MtLPMO9J) from M. thermophila M77 in Aspergillus nidulans. MtLPMO9J is a single-domain LPMO and has 63% sequence similarity with the catalytic domain of NcLPMO9C from Neurospora crassa. Biochemical characterization of MtLPMO9J revealed that it performs C4-oxidation and is active against cellulose, soluble cello-oligosaccharides and xyloglucan. Moreover, biophysical studies showed that MtLPMO9J is structurally stable at pH above 5 and at temperatures up to 50°C. Importantly, LC-MS analysis of the peptides after tryptic digestion of the recombinantly produced protein revealed not only the correct processing of the signal peptide and methylation of the N-terminal histidine, but also partial autoxidation of the catalytic center. This shows that redox conditions need to be controlled, not only during LPMO reactions but also during protein production, to protect LPMOs from oxidative damage.

Expression of Lignin Peroxidase H2 from Phanerochaete Chrysosporium by Multi-Copy Recombinant Pichia Strain

Abstract:The lipH2 gene, encoding the expression of lignin peroxidase, was cloned from Phanerochaete chrysosporium BKM-F-1767 and expressed in Pichia pastoris X-33, a yeast. The cDNA of LiPH2 was generated from total RNA extracted from P. chrysosporium by PCR with primers that do not contain a P. chrysosporium lignin peroxidase secretion signal. The gene was then successfully inserted into the expression vector pPICZalpha, and resulted in the recombinant vector pPICZalpha-lipH2. The transformation was conducted in two ways. One was using the wild Pichia pastoris as the recipients, which results in the recombinant P. pastoris with single or low lipH2 gene copy. The second was using P. pastoris and single or low lipH2 gene copy as the recipients, which results in the recombinant P. pastoris with multi-copies of lipH2 genes. This study firstly expressed the gene lipH2 in P. pastoris and achieved the successful expression of the lipH2 depending upon the generation of a recombinant strain that contained multiple copies. The lignin peroxidase activity reached a maximum of 15 U/L after 12 h induction.
Degradation of Purified Terephthalic Acid and Expression of Mnp Gene for GEM Fhhh

Abstract:The levels of the specific degradation rate (SDR) for the genetically engineered microorganism (GEM) Fhhh from inter\|kingdom protoplast fusion and the parental strain \%Phanerochaete chrysosporium\% (PC) in degradation of purified terephthalic acid (PTA) were affected by (NH\-4)\-2C\-4H\-4O\-6,H\-2O\-2,Mn\+\{2+\},and pH value.Except pH value,the other 3 factors affecting the SDR levels had a same order for both strains.The pH value in the order was at the first site for Fhhh,and that for PC at the third site.At the optimal levels,the SDR value of Fhhh was 11\^11% higher than that of PC;the value of manganese peroxidase specific activity (MnP\|SA) of Fhhh was 15\^22% higher than that of PC.There was a positive relationship between SDR and MnP\|SA significantly.The optimal levels of 4 factors for SDR also were the optimal levels for mnp gene expression.The technique of the mnp gene expression control would be useful to treat PTA wastewater.

Abstract:To better understand the light regulation of ligninolytic systems in Trametes polyzona KU-RNW027, ligninolytic enzymes-encoding genes were identified and analyzed to determine their transcriptional regulatory elements. Elements of light regulation were investigated in submerged culture. Three ligninolytic enzyme-encoding genes, mnp1, mnp2, and lac1, were found. Cloning of the genes encoding MnP1 and MnP2 revealed distinct deduced amino acid sequences with 90% and 86% similarity to MnPs in Lenzites gibbosa, respectively. These were classified as new members of short-type hybrid MnPs in subfamily A.2 class II fungal secretion heme peroxidase. A light responsive element (LRE), composed of a 5′-CCRCCC-3′ motif in both mnp promoters, is reported. Light enhanced MnP activity 1.5 times but not laccase activity. The mnp gene expressions under light condition increased 6.5- and 3.8-fold, respectively. Regulation of laccase gene expression by light was inconsistent with the absence of LREs in their promoter. Blue light did not affect gene expressions but impacted their stability. Reductions of MnP and laccase production under blue light were observed. The details of the molecular mechanisms underlying enzyme production in this white-rot fungus provide useful knowledge for wood degradation relative to illumination condition. These novel observations demonstrate the potential of enhancing ligninolytic enzyme production by this fungus for applications with an eco-friendly approach to bioremediation.
