Abstract:The mouse/rat hemokinin-1 (m/rHK-1) was discovered nearly 9 years ago. This molecule is a peptide comprising 11 amino acids. The m/rHK-1 was found to be mainly expressed in central immune organs like bone marrow, and was proven to have lymphopoietic roles in B and T lymphocyte development. m/rHK-1was also reported to have analgesic roles in rat spinal cord, in addition to other functions such as relaxing activity on coronary artery. Unlike its analogues SP, NKA, and NKB, m/rHK-1 does not express in the nervous system. To further study the distribution and function of m/rHK-1, we carried out conventional immunization and cell fusion procedures to acquire the hybridomas secreting specific monoclonal antibodies to m/rHK-1. In the 17 positive clones obtained, three antibodies named 1B12, 2B4, and 4G5 were shown representative in cross-reactivity against m/rHK-1 and its analogues by indirect ELISA, competitive indirect ELISA, and immunofluorescence assays. Among the three clones, the 2B4 monoclonal antibody appeared to be the high-titered and specific clone to m/rHK-1. Monoclonal antibodies to m/rHK-1 will function as good tools in the physiological study of m/rHK-1 in the near future.
Rats and mice immunised with chimeric human/mouse proteinase 3 produce autoantibodies to mouse Pr3 and rat granulocytes

Abstract:AIM: In this study, we employed chimeric human/mouse Proteinase 3 (PR3) proteins as tools to induce an autoantibody response to PR3 in rats and mice.METHOD: Rats and mice were immunised with recombinant human PR3 (HPR3), recombinant murine PR3 (mPR3), single chimeric human/mouse PR3 (HHm, HmH, mHH, mmH, mHm, Hmm) or pools of chimeric proteins. Antibodies to mPR3 and HPR3 were measured by ELISA. Antibodies to rat PR3 were determined by indirect immunofluorescence (IIF) on rat white blood cells. Urinalysis was performed by dipstick analysis. Kidney and lung tissue was obtained for pathological examination.RESULTS: In mice, immunisation with the chimeric human/mouse PR3 Hmm led to an autoantibody response to mPR3. Rats immunised with the chimeric human/mouse PR3 Hmm, HmH and mmH, or a pool of the chimeric human/mouse PR3 proteins, produced antibodies selectively binding to rat granulocytes as detected by IIF. No gross pathological abnormalities could be detected in kidney or lungs of mice or rats immunised with chimeric human/mouse PR3.CONCLUSION: Immunisation with chimeric human/mouse proteins induces autoantibodies to PR3 in rats and mice. Chimeric proteins can be instrumental in developing experimental models for autoimmune diseases.

Abstract:Abstract The A region of the major histocompatibility system of the rat, RT1, codes for ubiquitously expressed class I antigens and the RT1.B region determines class II (Ia) molecules. A third region, RT1.C, adjacent to B, has been described recently by histogenetic methods. Data are now presented on the analysis of RT1.C region products by the use of alloantisera and cytotoxic T lymphocytes, both prepared in two, C region congenic strain combinations. C region antigens have been detected serologically by complement-dependent cytotoxicity assays on the majority of B and T lymphocytes, but not on erythrocytes or liver and brain tissue by absorption tests. With the typing sera available only three "alleles" could be identified among 13 different rat strains, so that the RT1.C polymorphism appears to be restricted. Cytotoxic T lymphocytes could be induced against C region products after in vivo immunization and in vitro restimulation; killing was not restricted by the A and B regions. The concept that RT1.C is homologous to H-2 Qa is discussed.

Analysis of shedding of 3 beta-herpesviruses in saliva from patients with connective tissue diseases.

Abstract:BACKGROUND Whether an association exists between infection with beta -herpesviruses and connective tissue diseases remains unclear, as are the mechanisms for the regulation of these infections in the salivary glands. METHODS Human herpesvirus (HHV)-7 was isolated and viral DNA was quantified by real-time polymerase chain reaction (PCR) in serially collected saliva samples, to determine whether viral load correlated with infectivity. Then, to examine the role played by beta -herpesviruses in connective tissue diseases, cytomegalovirus, HHV-6, and HHV-7 DNA loads were examined by real-time PCR in serially collected saliva samples from 21 patients with connective tissue diseases. RESULTS Although subjects with frequent HHV-7 shedding were more likely to have a high viral load than were other subjects, high viral loads were detected in saliva samples from a portion of the subjects with low viral shedding rates. No significant difference between the quantity of HHV-7 DNA in saliva samples from which active virus was isolated and that amplified from samples without detectable virus was observed. Patients with adult-onset Still disease consistently had high HHV-7 DNA loads, in contrast to patients with other connective tissue diseases (P=.0003) and healthy adults (P=.0224). The mean HHV-6 (P=.012) and HHV-7 (P<.0001) DNA loads in patients with connective tissue diseases were lower than those in healthy adults. CONCLUSION These data suggest that a number of host factors in patients with adult-onset Still disease may function to accelerate HHV-7 replication in the salivary glands.

Abstract:Respiratory syncytial virus (RSV) is the most common cause of upper and lower respiratory tract infections in infants and young children. Virus-specific monoclonal antibodies (mAbs) can be used for diagnosis, prophylaxis, and research of RSV pathogenesis. A panel of 16 anti-RSV mAbs was obtained from mice immunized by RSV strain Long. Half of them had virus-neutralizing activity. According to Western blot all of these mAbs effectively bound native oligomeric (homodimeric and homotrimeric) forms of the RSV fusion (F) protein. Only five of the mAbs interacted with the monomeric form, and only one of these possessed neutralizing activity. None of these mAbs, nor the commercial humanized neutralizing mAb palivizumab, reacted with the denaturated F protein. Thus, interaction of all these mAbs with F protein had clear conformational dependence. Competitive ELISA and neutralization assays allowed the identification of nine antigenic target sites for the interaction of mAb with the F protein. Five partially overlapping sites may represent a complex spatial structure of one antigenic determinant, including one neutralizing and four non-neutralizing epitopes. Four sites (three neutralizing and one non-neutralizing) were found to be distinct. As a result of virus cultivation RSV–A, strain Long, in the presence of a large amount of one of the neutralizing mAbs, an escape mutant with a substitution, N240S, in the F protein, was obtained. Thus, it was shown for the first time that position 240 is critical for the protective effect of an anti-RSV antibody. To assess the ability of these mAbs to interact with modern RSV strains circulating in St. Petersburg (Russia) between 2014 and 2022, 73 RSV-A and 22 RSV-B isolates were analyzed. Six mAbs were directed to conserved epitopes of the F protein as they interacted most efficiently with both RSV subtypes in a fixed cell-ELISA and could be used for diagnostic assays detecting RSV.
Monoclonal anti-erythrocyte autoantibodies derived from NZB mice cause autoimmune hemolytic anemia by two distinct pathogenic mechanisms

Abstract:In vivo pathological manifestations of eight monoclonal anti-mouse red blood cell (MRBC) autoantibodies obtained from unmanipulated NZB mice were determined in BALB/c mice. Three (two IgG1 and one IgG2a) of four IgG monoclonal antibodies (mAb) and two of four IgM mAb were able to induce anemia following their i.p. injection. All five pathogenic anti-MRBC mAbs reacted only with MRBC, whereas non-pathogenic anti-MRBC mAbs showed binding to different species of RBC. Competition studies suggested the presence of at least two distinct epitopes recognized by our pathogenic anti-MRBC mAb. Histological examinations revealed that anemia resulted from either marked sequestration of agglutinated MRBC in spleens and livers or erythrophagocytosis, most remarkably by Kupffer cells in livers. This difference was correlated with the ability of each mAb to mediate Fc receptor-dependent phagocytosis by macrophages. The absence of complement-mediated hemolysis in vitro and the development of anemia in C5-deficient or C3-depleted mice indicated a minor role, if any, for complement-mediated lysis in the anemia induced by our anti-MRBC mAb. Our results suggest that (i) at least two different pathogenic epitopes are implicated in autoimmune hemolytic anemia; and (ii) sequestration of agglutinated MRBC in spleens and livers and Fc receptor-dependent phagocytosis, but not complement-mediated hemolysis, are the major mechanisms for the development of autoimmune hemolytic anemia.

Monoclonal antibody RG1 against rat Müller cells recognizes optic nerve and cerebral cortex glia in culture

Abstract:A monoclonal antibody designated as RG1 was produced by the fusion of mouse myeloma cells with spleen cells from mice immunized with rat retinas. The monoclonal antibody was shown to bind to Müller cells of the rat retina by the avidin-biotin peroxidase complex method. The screening of antibody-producing hybridomas and the analysis of the monoclonal antibody obtained were performed by enzyme-linked immunosorbent assay. The optic nerve, cerebral cortex and dorsal root ganglia were cultured from 17-day embryonic rats and maintained for 17-20 days. Expressions of the RG1 antigen were examined by indirect immunofluorescence in the cultured glial cells. In both the optic nerve and the cerebral cortex glial cells the RG1 antigen was expressed; however, the RG1 antigen was not expressed in cultured Schwann cells throughout the whole culture period. Our results suggest that the RG1 antigen is a common antigen in the supporting neuroectodermal glial cells in the central nervous system.
Immunochemical studies on the differential binding properties of two monoclonal antibodies reacting with Tn red cells

Abstract:Two monoclonal antibodies (MoAbs), BRIC 66 (IgM) and BRIC 111 (IgG1), were produced by immunizing mice with ovarian cyst blood group A1 glycoprotein and Tn red cells (RBCs), respectively. Their specificities were determined by inhibitions using Tn sialoglycoproteins (SGPs), mucins (armadillo [ASG] and ovine [OSG] submaxillary glycoproteins), and monosaccharides. BRIC 66 agglutinated both Tn and group A RBCs and reacted immunohistochemically with both the vascular endothelium and tumor cells from a group A adenocarcinoma, BRIC 66 was inhibited by N-acetylgalactosamine (GalNAc), Tn SGPs, and mucins on both hemagglutination inhibition tests and radioimmunoassay. BRIC 111 agglutinated Tn RBCs only, and it specifically stained tumor cells from a group O patient's breast carcinoma and a group A patient's adenocarcinoma. In hemagglutination inhibition tests, BRIC 111 was readily inhibited by Tn SGPs, only partially inhibited by GalNAc, and not inhibited by mucins. In a sensitive radioimmunoassay, BRIC 111 was inhibitable by GalNAc. Tn SGP was 2000-fold more effective as an inhibitor than the mucins (ASG and desialized OSG), which contain a high content of terminal alpha-GalNAc-O-serine (threonine) residues. It is postulated that BRIC 66 is specific for terminal alpha-GalNAc units in carbohydrate chains. The exclusive reaction of BRIC 111 with Tn SGP indicates a combining site larger than GalNAc alpha-1, which probably includes amino acid residues in juxtaposition to GalNAc in Tn SGP. In view of its specific agglutination of Tn RBCs, BRIC 111 is a useful reagent for the examination of polyagglutinable RBCs.

Abstract:Inadequate zinc intake may lead to poor growth and developmental outcome in very-low-birth-weight (VLBW; < 1,500 g) infants. Fifty-two infants (mean birth weight, 1,117 +/- 287 g; mean gestational age, 29 +/- 2.9 weeks) were randomly allocated to two groups. SUPP infants received a regular term formula plus zinc supplements (4.4 mg/L; final content, 11 mg/L); PLAC infants received the same formula plus placebo (final content, 6.7 mg/L). Infants started their formula at 1,853 +/- 109 g and consumed the formula for 6 months. All subjects were evaluated at 3, 6, 9, and 12 +/- 0.75 months corrected-for-gestational-age. At each evaluation, weight, length, and head circumference were measured, a Griffiths developmental assessment was performed, and a blood sample was taken. Higher plasma zinc levels (p < 0.05) were found in the SUPP group at 1 and 3 months, and improved linear growth velocity was found in the SUPP group over the study period for the whole group as well as for girls alone. Maximum motor development scores were higher (p = 0.018) in the SUPP (98 +/- 10) than the PLAC (90 +/- 8) group, indicating that increased zinc intake in early infancy may be beneficial to VLBW infants.
A Study on the Glycan Specificity of Natural Antibody Repertoires in Rodents

Abstract:Inbred strains of mice and rats are widely used in preclinical investigations evaluating the effectiveness of glycan-based biomecines, however, the glycan specificity repertoires of serum Abs in rodents have not been fully characterized. In the present study, serum antibodies in naïve mice and rats of different inbred strains were analyzed for specificity against 4 representative carbohydrate structures including PGA (1,4-linked α-D-galactopyranosyluronic acids), β-glucan, mannan and α-glucan (dextran). Mannan was not recognized by serum Abs from any of the mouse and rat strains. Serum IgM in naïve F344, BN and Lewis rats recognized PGA and β-glucan and, less strongly, dextran. High titer circulating IgM against PGA were found in mice of BALB/c, C57BL/6, C3H/NeH and BXSB strains. C3H/NeH was the only strain which also produced low titer IgM against β-glucan and dextran. Age-related production of high titer IgM, IgA and IgG Abs against β-glucan was observed in BXSB mice. Intraperitoneal immunization of BALB/c and C57BL/6 mice with β-glucan elicited strong IgM responses, while immunization with PGA also led to an increase of anti-PGA IgM Ab titers. These results provide useful information on the characteristics of glycan-specific natural antibody repertoires in rodents.

Abstract:The hemolytic, agglutinating, and coating antibody activities of the serum of rabbits immunized with rat erythrocytes could be separated at least partially by DEAE and CM cellulose chromatographic fractionation of ΓG globulins. The serologic activities of serums from different animals did not separate identically by these methods, and serums obtained from animals immunized for only two weeks displayed more chromatographic heterogeneity in regard to these properties than did serums obtained after four weeks of immunization. When Cr51‐labeled rat erythrocytes were sensitized with fractionated antibodies and then injected into isologous rats, shortened survival and sequestration of cells in the liver was observed to be a function of the amount of lytic antibodies applied. Cells sensitized with even large amounts of agglutinating or coating (but non‐lytic) antibodies survived normally. Lysis observed with tests in vitro and shortened survival with liver sequestration in vivo were both either abolished or markedly reduced when the complement‐fixing properties of lytic antibodies were altered by pepsin digestion or reduction and alkylation. Heated antibody fractions (56 C for two hours) supported indirect complement antiglobulin tests in vitro and, when tested in vivo, supported a pattern of splenic sequestration with shortened survival time of sensitized cells.

Mouse Monoclonal Antibody Specific To Human Ena Blood Group Antigen

Abstract:Abstract The aim of this study was to produce mouse monoclonal antibodies (mAbs) specific to human blood group antigens. Two percent solution of normal human red blood cells (RBCs) O, RhD+ in normal saline was freshly prepared, and 100 uL was immunized weekly into BALB/mice, intraperitoneal route for 3 weeks. Prior to each immunization, 100 uL of mouse blood was collected and serum was separated. The rising of anti-RBCs antibody titer was performed by standard hemagglutination compared to pre-immunized serum. Mouse was then sacrificed, and the spleen was collected. The fusion between mouse spleen cells and mouse myeloma (X63Ag8.653) cell lines was performed according to the standard hybridoma technique. Hybrid clones were grown in DMEM high glucose supplemented with 20% FCS at 3 °C, 5% CO2 incubator with 5% humidity. The anti-RBCs in hybrid cell culture supernatant were tested by standard hemagglutination and a monoclonal antibody-producing single clone of the hybrid cell was generated by limiting dilution technique. Antibody screening and identification were performed using screening cells, panel cells, and rare blood type RBCs, followed by enzyme treatment technique. The Isotype of mAb was also identified. The results showed that amongst those obtained, mAb clone 1E10-2B2, was confirmed to be anti-Ena characterized as ficin resistant (anti-EnaFR) and isotype IgG2b, kappa. The anti-Ena mAb is very helpful and can be applied as specific mAb for the phenotyping of Ena which is the high-frequency blood group antigens in all populations. Abstract:Monoclonal anti-idiotypic antibody (Mab2-5G2) raised against idiotypic antibodies to membrane glycoprotein GP5 of porcine reproductive and respiratory syndrome virus (PRRSV). The variable regions of the heavy chain (VH) and light chain (VL) of Mab2-5G2 were cloned and connected with a (Gly4Ser)3 linker. The recombinant scFv gene was cloned into the pEasy-E1 vector and expressed in E. coli as inclusion bodies. The expressed scFv-His proteins renatured in a pH and urea gradient buffer retained the same immunological properties as that of Mab2-5G2. Renatured scFv-His protein retained the same characteristics as that of Mab2-5G2 by recognizing and binding to Marc-145 cells. Furthermore, renatured scFv-His along with Mab2-5G2 were used to immunize rabbits to produce anti-anti-idiotypic antibodies (Ab3) that neutralized PRRSV infection of Marc-145 cells. These results demonstrated that the expressed scFv-His protein possessed the same characteristics of Mab2-5G2 and will be suitable for future investigations of Mab2-5G2 antibody structure and its ability to interact with potential PRRSV cellular receptor as well as immunological properties against PRRSV infection.
Detection of T and B cells in many animal species using cross-reactive anti-peptide antibodies.

Abstract:Abstract A wide range of lineage-specific Ag are detectable in the human lymphoid system using mAb, but only a few such markers are detectable in animal species. In this paper, we have investigated the interspecies reactivity of antibodies raised against intracytoplasmic peptide sequences from two T cell Ag (CD3 and CD5) and two B cell markers (the Ig-associated polypeptides encoded by the mb-1 and B29 genes). Immunocytochemical labeling of tissue sections showed that these antibodies cross-react widely between different species (including ungulates, rodents, and marsupials), staining B or T cell areas selectively in lymphoid tissue. The specificity of these antibodies for the animal homologues of the human T and B cell markers was confirmed for the rat by Western blotting analysis. The broad cross-reactivity of these antibodies appears to be due to the fact that they were raised against intracytoplasmic peptide sequences that are highly conserved between humans and rodents, i.e., 80% for mb-1, 85% for CD5, and 100% for CD3 and B29. This strategy should, in the future, widen the range of lineage-associated markers detectable in experimental animals.

Abstract:Objective:To prepare and identify the monoclonal antibody against RAET1G2.Methods:Prokaryotic expressed human RAETlG2 was used as an antigen to immunize BALB/c mice, and prepare monoclonal antibodies by means of the B lymphocyte hybridoma technique.Ig subclass and specificity of monoclonal antibodies was analysed by double immunodiffusion and western blot respectively.Results:4 hybridoma cell lines(7A11,9C6,10F9 and 12F8) secreting anti-RAET1G2 mAbs were obtained. Ig subclass of mAbs belonged to IgG1.Conclusion:We obtained four strains of hybridoma cells secreting anti-RAET1G2 antibodies.All of the antibodies could recognize RAET1G expressed on the CHO cell surface.It could be used to study the relation of RAET1G2 with tumor progression and to observe the expression of RAET1G2 on the surface of tumor cells.

Human red blood cells (RBC) sensitized by each of these McAbs have been assayed for their ability to form rosettes with the human histiocytic lymphoma U937 cell line, human B cell line Daudi and erythroblastoid K562 cell line. IgG1 and IgG3 sensitized RBC formed significant rosettes with the FcR- and FcRII+ Daudi and K562 cell lines, the percentage of cells forming rosettes being directly proportional to the degree of sensitization of the RBC. Bromelin treating Daudi cells did not alter this pattern of reactivity, whereas bromelin treated FcRI+ and FcRII+ U937 cells formed significant resettes with IgG1, IgG3 and IgG4 sensitized RBC, demonstrating a difference in the IgG subclass specificity between human FcRI and FcRII. Murine IgG2b anti-NIP sensitized RBC did not form rosettes with any cell line tested; however, RBC sensitized by some members of a panel of murine IgG1 McAb, specific for the glycophorin A molecule, were able to form rosettes with Daudi, U937 and K562 cells. This interaction was enhanced by bromelin treating the Daudi or U937 cells and can be correlated to the disposition of the epitopes recognized, relative to the target cell membrane, those McAbs recognizing epitopes furthest from the RBC surface being most effective in interacting with FcRII. The data are interpreted in terms of a simple model for antibody-mediated cell--cell interaction.
