Creating and Evaluating a Novel Small Diameter PCL/PU Vascular Graft for Finger Reconstruction: A Comprehensive Analysis of Structural, Mechanical, In vitro, and In Vivo Assessments

Abstract:This study addresses vascular repair challenges during digital replantation by developing an electrospun nanofibrous scaffold using PCL and PU for small-diameter vascular grafts (<2 mm). Structural validation employed diverse techniques such as FE-SEM, DSC, and FTIR analysis, while mechanical properties like stress and strain were rigorously evaluated. Functionality was assessed through in vitro assays and in vivo experiments, including subcutaneous implantation in rats and femoral artery insertion in rabbits. The scaffold demonstrated favorable mechanical properties and enhanced cell attachment, with minimal lumen fibrosis and good blood compatibility observed. Post-implantation in rabbits showed well-established blood flow and signs of tissue formation, suggesting potential for regenerative medicine. In conclusion, the PU85/PCL15 scaffold exhibited promising outcomes, highlighting its potential in microvascular tissue engineering Level of Evidence Level V
