Abstract:Keratin (K) is a major protein component of hair and is involved in hair growth and development. In this study, we analysed the expression, localization, and polymorphism of the K84 gene (KRT84) in Gansu Alpine Fine-wool sheep using immunofluorescence, RT-qPCR, and PARMS (Penta-primer amplification refractory mutation system). Haplotypes of KRT84 were also constructed and their relationship with wool traits analysed. It was revealed that KRT84 was highly expressed in hair follicles, including the inner root sheath, outer root sheath, and hair medulla and at all six lamb ages investigated from 1 to 270 days of age. Three SNPs were detected in KRT84 exon 1, and they formed three haplotypes (named H1, H2, and H3) and six genotypes. Analyses revealed an association between haplotype combinations (diplotypes) and the mean fibre curvature, mean staple length, mean staple strength, mean fibre diameter, the coefficient of variation of fibre diameter, and comfort factor for these sheep. These results suggest that KRT84 is of importance in determining several key traits in Gansu Alpine Fine-wool sheep and that the gene could possibly be used as a genetic marker for wool trait selection in these sheep.

What problem does this paper attempt to address?
The main problem that this paper attempts to solve is to explore the spatio - temporal expression pattern of the KRT84 gene in Gansu alpine fine - wool sheep, single - nucleotide polymorphisms (SNPs), and the association between its haplotypes and wool traits. Specifically, the researchers hope to reveal its role in determining wool fiber characteristics by analyzing the expression and genetic variation of the KRT84 gene, and to evaluate whether these genetic markers can be used for selective breeding of wool traits. ### Research Background Keratin (K) is the main protein component of hair and plays an important role in hair growth and development. The keratin encoded by the KRT84 gene shows different expression patterns and functions in different species. However, the specific role of the KRT84 gene in wool growth and wool traits is still unclear. Therefore, this study aims to fill this knowledge gap. ### Research Objectives 1. **Spatio - temporal expression analysis**: Determine the spatio - temporal expression pattern of the KRT84 gene at different developmental stages of Gansu alpine fine - wool sheep. 2. **Single - nucleotide polymorphism (SNP) detection**: Identify SNPs in exon 1 of the KRT84 gene and construct corresponding haplotypes. 3. **Wool trait association analysis**: Evaluate the association between different haplotype combinations (diplotypes) of the KRT84 gene and wool traits (such as average fiber curvature, average fleece length, average fleece strength, average fiber diameter, coefficient of variation of fiber diameter, and comfort factor). ### Research Significance Through this study, the authors hope to reveal the importance of the KRT84 gene in determining wool fiber characteristics and provide potential genetic markers for selective breeding of wool traits. This will not only help to gain a deeper understanding of the molecular mechanism of wool growth but also provide a scientific basis for the livestock industry to improve the quality and yield of wool. ### Main Conclusions - The KRT84 gene is highly expressed in the hair follicles of Gansu alpine fine - wool sheep, including the inner root sheath, outer root sheath, and hair medulla, and is expressed at six different ages from 1 day to 270 days. - Three SNPs (c.97A/G, c.162C/T, and c.286G/A) were found in exon 1 of the KRT84 gene, and three haplotypes (H1, H2, and H3) and six genotypes were formed. - There is a significant association between haplotype combinations (diplotypes) and wool traits, indicating that the KRT84 gene may play an important role in determining wool fiber characteristics. These results suggest that the KRT84 gene may be one of the key genes affecting wool traits and can be used as a potential genetic marker for selective breeding.
