Abstract:Sensory and cognitive mechanisms allow stimuli to be perceived with properties relating to sight, sound, touch, etc, and ensure, for example, that visual properties are perceived as visual experiences, rather than sounds, tastes, smells, etc. Theories of normal development can be informed by cases where this modularity breaks down, in a condition known as synaesthesia. Conventional wisdom has held that this occurs extremely rarely (0.05% of births) and affects women more than men. Here we present the first test of synaesthesia prevalence with sampling that does not rely on self-referral, and which uses objective tests to establish genuineness. We show that (a) the prevalence of synaesthesia is 88 times higher than previously assumed, (b) the most common variant is coloured days, (c) the most studied variant (grapheme - colour synaesthesia)—previously believed most common—is prevalent at 1%, and (d) there is no strong asymmetry in the distribution of synaesthesia across the sexes. Hence, we suggest that female biases reported earlier likely arose from (or were exaggerated by) sex differences in self-disclosure.

Abstract:Reading acquisition modifies areas of the brain associated with vision, with language, and their connections. Those changes enable reciprocal translation between orthography, and word sounds and meaning. Individual variability in the pre-existing cerebral substrate contributes to the range of eventual reading abilities, extending to atypical developmental patterns, including dyslexia and reading-related synesthesias. The present study is devoted to the little-studied but highly informative ticker-tape synesthesia (TTS), in which speech perception triggers the vivid and irrepressible perception of words in their written form in the mind's eye. We scanned a group of 17 synesthetes and 17 matched controls with functional MRI, while they listened to spoken sentences, words, numbers, or pseudowords (Experiment 1), viewed images and written words (Experiment 2), and were at rest (Experiment 3). First, we found direct correlates of the TTS phenomenon: during speech perception, as TTS was active, synesthetes showed over-activation of left perisylvian regions supporting phonology, and of the occipitotemporal Visual Word Form Area (VWFA), where orthography is represented. Second, we brought support to the hypothesis that TTS results from atypical relationships between spoken and written language processing: the TTS-related regions overlap closely with cortices activated during reading, and the overlap of speech-related and reading-related areas is larger in synesthetes than in controls. Furthermore the regions over-activated in TTS overlap with regions under-activated in dyslexia. Third, during resting state, that is in the absence of current TTS, synesthetes showed increased functional connectivity between left prefrontal and bilateral occipital regions. This pattern may reflect a lowered threshold for conscious access to visual mental contents, and may implement a non-specific predisposition to all synesthesias with a visual content. Those data provide a rich and coherent account of TTS as a non-detrimental developmental condition created by the interaction of reading acquisition with an atypical cerebral substrate.

Abstract:Abstract In some individuals, a visually presented letter or number automatically evokes the perception of a specific color, an experience known as color-grapheme synesthesia. It has been suggested that parietal binding mechanisms play a role in the phenomenon. We used a noninvasive stimulation technique, transcranial magnetic stimulation (TMS), to determine whether the posterior parietal lobe is critical for the integration of color and shape in color-grapheme synesthesia, as it appears to be for normal color-shape binding. Using a color-naming task with colored letters that were either congruent or incongruent with the synesthetic photism, we demonstrate that inhibition of the right posterior parietal lobe with repetitive TMS transiently attenuates synesthetic binding. These findings suggest that synesthesia (the induction of color from shape) relies on similar mechanisms as found in normal perception (where the perception of color is induced by wavelength).

Top-down signal transmission and global hyperconnectivity in auditory-visual synesthesia: Evidence from a functional EEG resting-state study

Abstract:Auditory-visual (AV) synesthesia is a rare phenomenon in which an auditory stimulus induces a "concurrent" color sensation. Current neurophysiological models of synesthesia mainly hypothesize "hyperconnected" and "hyperactivated" brains, but differ in the directionality of signal transmission. The two-stage model proposes bottom-up signal transmission from inducer- to concurrent- to higher-order brain areas, whereas the disinhibited feedback model postulates top-down signal transmission from inducer- to higher-order- to concurrent brain areas. To test the different models of synesthesia, we estimated local current density, directed and undirected connectivity patterns in the intracranial space during 2 min of resting-state (RS) EEG in 11 AV synesthetes and 11 nonsynesthetes. AV synesthetes demonstrated increased parietal theta, alpha, and lower beta current density compared to nonsynesthetes. Furthermore, AV synesthetes were characterized by increased top-down signal transmission from the superior parietal lobe to the left color processing area V4 in the upper beta frequency band. Analyses of undirected connectivity revealed a global, synesthesia-specific hyperconnectivity in the alpha frequency band. The involvement of the superior parietal lobe even during rest is a strong indicator for its key role in AV synesthesia. By demonstrating top-down signal transmission in AV synesthetes, we provide direct support for the disinhibited feedback model of synesthesia. Abstract:Electrophysiological recordings from human visual cortex were carried out with electrodes chronically implanted in 13 patients for localization of an epileptogenic focus. Visual evoked potentials (VEPs) elicited by red or blue checkerboard stimuli were recorded using an adaptation stimulus-test stimulus design in which color was the most salient feature. A "significant color effect," defined as a statistically significant effect of the adaptation stimulus on test stimulus VEPs evoked by the same or a different color, was determined for various cortical regions: medial lingual gyrus, 20%; lateral lingual gyrus, 38%; posterior fusiform gyrus, 50%; anterior fusiform gyrus, 0%; inferior temporal gyrus, 5%; occipital pole, 30%; lateral surface of non-visual cortex, 6%; inferior parietal and temporal cortex, 5%. The time course of the significant color effects suggests that wave length-selective neuronal activity occurs initially at the first stage of cortical processing in the medial lingual gyrus, followed by progressively later activation of the lateral lingual gyrus, the posterior fusiform gyrus, and the inferior temporal gyrus. In two patients, stimulation of the lateral lingual and fusiform gyri elicited color sensations in the contralateral half-field, whereas stimulation of the medial lingual and cuneate gyri evoked retinotopically appropriate quadrantic "shimmering" devoid of color. These results suggest that a region of inferior occipital cortex, primarily the posterior portion of the fusiform gyrus, is involved in color perception and may be homologous with area V4 in monkeys. There is also a region of dorsolateral surface cortex which exhibits a fairly high percentage of significant color effects and when stimulated may evoke sensations of color. This region may be the same as the dorsolateral region thought to be involved in selective attention to color.
Multiple dimensions in bi-directional synesthesia

Abstract:Grapheme-color synesthetes report seeing a specific color when a number is perceived. The reverse, the synesthetic experience of a specific grapheme after the percept of a color is extremely rare. However, recent studies have revealed these interactions at both behavioral and neurophysiological levels. We investigated whether similar neuronal processes (i.e. perceptual and/or attentional) may underlie this bi-directional interaction by measuring event-related potentials (ERPs) during both a number-color and color-number priming task. In addition, we investigated the unitarity of synesthesia by comparing two distinct subtypes of synesthetes, projectors and associators, and assessed whether consistencies between measures (i.e. behavioral and electrophysiological) were present across synesthetes. Our results show longer reaction times for incongruent compared with congruent trials in both tasks. This priming effect is also present in the P3b latency (parietal electrode site) and P3a amplitude (frontal electrode site) of the ERP data. Interestingly, projector and associator synesthetes did not reveal distinct behavioral or electrophysiological patterns. Instead, a dissociation was found when synesthetes were divided in two groups on the basis of their behavioral data. Synesthetes with a large behavioral priming effect revealed ERP modulation at the frontal and parietal electrode sites, whereas synesthetes with a small priming effect revealed a frontal effect only. Together, these results show, for the first time, that similar neural mechanisms underlie bi-directional synesthesia in synesthetes that do not report a synesthetic experience of a grapheme when a color is perceived. In addition, they add support for the notion of the existence of both 'lower' and 'higher' synesthetes.

Abstract:Synaesthesia has been known to scientific research for over 100 years but has undergone something of a renaissance recently as new investigations begin to uncover its neurological basis. Rather than being an anomaly, it might offer beneficial insights into the basis of normal perception. A new study by Esterman et al. epitomises this current trend and claims to show that the posterior parietal cortex is a crucial locus of synaesthetic experience. As the posterior parietal cortex is commonly linked to normal sensory integration, Esterman et al.'s finding might lend support to the claim that synaesthesia is an extension of the normal perceptual processes assumed to occur in binding.

Abstract:Grapheme-color synesthetes experience specific colors associated with specific number or letter characters. To determine the neural locus of this condition, we compared behavioral and fMRI responses in six grapheme-color synesthetes to control subjects. In our behavioral experiments, we found that a subject's synesthetic experience can aid in texture segregation (experiment 1) and reduce the effects of crowding (experiment 2). For synesthetes, graphemes produced larger fMRI responses in color-selective area human V4 than for control subjects (experiment 3). Importantly, we found a correlation within subjects between the behavioral and fMRI results; subjects with better performance on the behavioral experiments showed larger fMRI responses in early retinotopic visual areas (V1, V2, V3, and hV4). These results suggest that grapheme-color synesthesia is the result of cross-activation between grapheme-selective and color-selective brain areas. The correlation between the behavioral and fMRI results suggests that grapheme-color synesthetes may constitute a heterogeneous group.
Emotions and Consciousness Alterations in Music-color Synesthesia

Abstract:Music-color synesthesia is a neurological condition in which musical notes or melodies trigger automatic and consistent additional perceptions (e.g., colors, shapes, scenes). Music listening is sometimes accompanied by strong experiences related to music (SEMs), including emotions and alterations of consciousness, and these SEMs have been suggested to mediate some music-color associations. We investigated SEMs in a group of music-color synesthetes and controls. Participants underwent a semi-structured interview and completed the OAV questionnaire to assess alterations of consciousness during music listening. All music-color synesthetes reported that their music-color synesthesia were accompanied by intense emotions including euphoria, fear, love, sadness, and disgust. In addition, most music-color synesthetes and no controls reported alterations of consciousness including out-of-body experiences, depersonalization, alterations of time, and alterations in limb ownership. Also, a majority of music-color synesthetes reported other synesthesia involving visceral or emotional perceptions as well as alterations of consciousness in everyday life. Overall, the data suggest that music-color synesthesia can be accompanied by atypical functioning in brain systems linked to emotions and consciousness.

Abstract:Synesthesia entails a special kind of sensory perception, where stimulation in one sensory modality leads to an internally generated perceptual experience of another, not stimulated sensory modality. This phenomenon can be viewed as an abnormal multisensory integration process as here the synesthetic percept is aberrantly fused with the stimulated modality. Indeed, recent synesthesia research has focused on multimodal processing even outside of the specific synesthesia-inducing context and has revealed changed multimodal integration, thus suggesting perceptual alterations at a global level. Here, we focused on audio-visual processing in synesthesia using a semantic classification task in combination with visually or auditory-visually presented animated and in animated objects in an audio-visual congruent and incongruent manner. Fourteen subjects with auditory-visual and/or grapheme-color synesthesia and 14 control subjects participated in the experiment. During presentation of the stimuli, event-related potentials were recorded from 32 electrodes. The analysis of reaction times and error rates revealed no group differences with best performance for audio-visually congruent stimulation indicating the well-known multimodal facilitation effect. We found enhanced amplitude of the N1 component over occipital electrode sites for synesthetes compared to controls. The differences occurred irrespective of the experimental condition and therefore suggest a global influence on early sensory processing in synesthetes.

Abstract:Synesthesia provides an elegant model to investigate neural mechanisms underlying individual differences in subjective experience in humans. In grapheme-color synesthesia, written letters induce color sensations, accompanied by activation of color area V4. Competing hypotheses suggest that enhanced V4 activity during synesthesia is either induced by direct bottom-up cross-activation from grapheme processing areas within the fusiform gyrus, or indirectly via higher-order parietal areas. Synesthetes differ in the way synesthetic color is perceived: "projector" synesthetes experience color externally colocalized with a presented grapheme, whereas "associators" report an internally evoked association. Using dynamic causal modeling for fMRI, we show that V4 cross-activation during synesthesia was induced via a bottom-up pathway (within fusiform gyrus) in projector synesthetes, but via a top-down pathway (via parietal lobe) in associators. These findings show how altered coupling within the same network of active regions leads to differences in subjective experience. Our findings reconcile the two most influential cross-activation accounts of synesthesia.

Abstract:There is considerable interest in understanding the developmental origins and health implications of individual differences in brain structure and function. In this pre-registered study we demonstrate that a hidden subgroup within the general population—people with synesthesia (e.g. who "hear" colors)—show a distinctive behavioral phenotype and wide-ranging differences in brain structure and function. We assess the performance of 13 different brain-based biomarkers (structural and functional MRI) for classifying synesthetes against general population samples, using machine learning models. The features in these models were derived from subject-specific parcellations of the cortex using the Human Connectome Project approach. All biomarkers performed above chance with intracortical myelin being a particularly strong predictor that has not been implicated in synesthesia before. Resting state data show widespread changes in the functional connectome (including less hub-based connectivity). These brain-based individual differences within the neurotypical population can be as large as those that differentiate neurotypical from clinical brain states.

Abstract:How do category-selective regions arise in human extrastriate cortex? Visually presented words provide an ideal test of the role of experience: Although individuals have extensive experience with visual words, our species has only been reading for a few thousand years, a period not thought to belong enough for natural selection to produce a genetically specified mechanism dedicated to visual word recognition per se. Using relatively high-resolution functional magnetic resonance imaging (1.4 x 1.4 x 2-mm voxels), we identified a small region of extrastriate cortex in most participants that responds selectively to both visually presented words and consonant strings, compared with line drawings, digit strings, and Chinese characters. Critically, we show that this pattern of selectivity is dependent on experience with specific orthographies: The same region responds more strongly to Hebrew words in Hebrew readers than in nonreaders of Hebrew. These results indicate that extensive experience with a given visual category can produce strong selectivity for that category in discrete cortical regions.

Abstract:As first described by Francis Galton, some persons perceive vividly and automatically in their mind's eye the written form of words that they are hearing. This phenomenon, labeled ticker-tape synesthesia (TTS), is thought to reflect an abnormally strong influence of speech processing in language areas on to orthographic representations in the visual cortex. Considering the relevance of TTS for the study of reading acquisition, we looked for objective behavioral advantages or impairments in 22 synesthetes, as compared to 22 matched control participants. In three auditory tasks relying on orthographic working memory (letters counting, backward spelling, and letter shape decision), we predicted and observed better performance in synesthetes than in controls. In two visual tasks (lexical decision and letter decision) with a concurrent auditory stimulation, we predicted that synesthetes should suffer from a larger interference by irrelevant speech than controls, but eventually found no difference between the groups. Those results, which we discuss in relation to orthographic processing, mental imagery, and working memory, promote TTS from pure subjectivity to an experimentally measurable phenomenon.

Abstract:This study was a case investigation of grapheme-texture synestheste TH, a female who subjectively reported experiencing a visual association between grapheme and colour/texture. First, we validated the existence of a synesthetic association in an objective manner. Involuntarily elicited experience is a major hallmark that is common to different types of synesthetes. Our results indicated interference between physical and synesthetic texture, suggesting the involuntary occurrence of synesthetic textural experience. We analysed the behavioural measures using the EZ diffusion model. The result suggested that TH's synesthetic experience was dissociable from that of briefly trained associative processing of non-synesthetes. Second, we investigated how the synesthetic experience of colour and texture dimensions was bound in the visual representation. We found that the interference effects of colour and texture were not independent. This suggested that in the elicited experience, the colour and texture features construct an integrated representation.
Neural Basis of the Sound-Symbolic Crossmodal Correspondence Between Auditory Pseudowords and Visual Shapes

Abstract:Sound symbolism refers to the association between the sounds of words and their meanings, often studied using the crossmodal correspondence between auditory pseudowords, e.g., 'takete' or 'maluma', and pointed or rounded visual shapes, respectively. In a functional magnetic resonance imaging study, participants were presented with pseudoword-shape pairs that were sound-symbolically congruent or incongruent. We found no significant congruency effects in the blood oxygenation level-dependent (BOLD) signal when participants were attending to visual shapes. During attention to auditory pseudowords, however, we observed greater BOLD activity for incongruent compared to congruent audiovisual pairs bilaterally in the intraparietal sulcus and supramarginal gyrus, and in the left middle frontal gyrus. We compared this activity to independent functional contrasts designed to test competing explanations of sound symbolism, but found no evidence for mediation via language, and only limited evidence for accounts based on multisensory integration and a general magnitude system. Instead, we suggest that the observed incongruency effects are likely to reflect phonological processing and/or multisensory attention. These findings advance our understanding of sound-to-meaning mapping in the brain.

Abstract:Functional color-tone synesthesia was induced in 8 subjects by presenting a low tone when a carmine light was shown and a higher tone just before and during the presentation of a blue-green light. After the preliminary conditioning, a wrong tone was sometimes used when desaturated colors of the same hue were presented. The resulting errors are taken to signify the establishment of synesthesia through conditioning.
