Mitochondrial Toxicity of PM2.5 Leads to Cardiac Failure: A Comparative Evaluation of PM2.5 from Ambient Air, Diesel Exhaust and SRM 2975

Abstract:Different epidemiological and toxicological studies have suggested that the chemical constituents of particulate matter (PM 2.5 ) vary with different sources and can adversely affect human health. Thus, the coherence in different experimental evidences from various locations/ regions makes it difficult to have a comprehensive conclusion on PM 2.5 mediated cardiotoxicity. In the present study, we evaluated the mitochondrial toxicity of PM 2.5 from real ambient air sample (RA_PM) and diesel particulate matter (DPM) from the same location and compared the effect with the standard, SRM 2975. Twenty-one days of daily exposure to 250 μg/m 3 of PM 2.5 resulted in significant alterations in mitochondrial function and prominent decline in mitochondrial copy number and expression of bioenergetics genes. Mitochondrial adaptative genes (Fission, fusion, mitophagy) were modulated differentially in all samples with respect to the magnitude, where the pattern of changes was similar in different sources of PM 2.5 . In addition, the mitotoxic impact of PM 2.5 was also reflected in the downregulation of the mitochondrial regulatory genes like PGC1-α, TFAM and POLG. Eventually, the deteriorated mitochondrial quality and quantity at the basal cardiac tissue of PM 2.5 exposed animals compromised the cardiac ability to withstand ischemia reperfusion injury (IR). Comparative evaluation of cardiotoxicity of PM 2.5 from different sources demonstrated higher mito toxicity with RA_PM sample than DPM and SRM 2975, suggesting the critical role played by the heterogenous composition of PM 2.5 towards cardiotoxicity. Thus, our findings emphasize the need and urgency on the need to revisit the existing guidelines for air quality index.
