Abstract:Combustion power generation is still the main way of lignite utilization, but lignite combustion will produce a lot of toxic gases, so how to make lignite clean utilization has become an urgent problem to be solved. Hydrogen is an environmentally friendly, zero-carbon emission clean energy because microorganisms can degrade brown coal to produce hydrogen. Therefore, in this experiment, the anaerobic hydrogen production experiment of lignite was carried out, and the influence of different concentrations of benzyl alcohol on hydrogen production of lignite was studied. The results showed that the addition of 500 mg/L benzyl alcohol had the most significant effect on the hydrogen production of lignite, and the total hydrogen production reached 1.70 mL/g, which was 47.83% higher than that of the blank group. The addition of benzyl alcohol extended the peak time of hydrogen production in lignite fermentation. The peak hydrogen production time of 500 mg/L benzyl alcohol in the middle and late stage was 8 days, 5 days longer than that in the blank group. The peak hydrogen production of 500 mg/L benzyl alcohol in the middle and late stage was 0.11 mL/g, which was 2.75 times that of 0.04 mL/g in the blank group. Hydrogen production of lignite is mainly produced by acetic acid and propionic acid fermentation, which is different from butyric acid metabolic pathway of biomass microbial transformation such as crop straw and kitchen waste. This also provides a new way and theoretical basis for the fermentation of lignite to produce hydrogen.

Assessment of Antioxidant and Antimicrobial Properties of Lignin from Corn Stover Residue Pretreated with Low-Moisture Anhydrous Ammonia and Enzymatic Hydrolysis Process.

Abstract:Lignin accounts for 15–35% of dry biomass materials. Therefore, developing value-added co-products from lignin residues is increasingly important to improve the economic viability of biofuel production from biomass resources. The main objective of this work was to study the lignin extracts from corn stover residue obtained from a new and improved process for bioethanol production. Extraction conditions that favored high lignin yield were optimized, and antioxidant and antimicrobial activities of the resulting lignin were investigated. Potential estrogenic toxicity of lignin extracts was also evaluated. The corn stover was pretreated by low-moisture anhydrous ammonia (LMAA) and then subjected to enzymatic hydrolysis using cellulase and hemicellulase. The residues were then added with sodium hydroxide and extracted for different temperatures and times for enhancing lignin yield and the bioactivities. The optimal extraction conditions using 4% (w/v) sodium hydroxide were determined to be 50 °C, 120 min, and 1:8 (w:v), the ratio between corn stover solids and extracting liquid. Under the optimal condition, 33.92 g of lignin yield per 100 g of corn stover residue was obtained. Furthermore, the extracts produced using these conditions showed the highest antioxidant activity by the hydrophilic oxygen radical absorbance capacity (ORAC) assay. The extracts also displayed significant antimicrobial activities against Listeria innocua. Minimal estrogenic impacts were observed for all lignin extracts when tested using the MCF-7 cell proliferation assay. Thus, the lignin extracts could be used for antioxidant and antimicrobial applications, and improve the value of the co-products from the biomass-based biorefinery.

Abstract:Lignocellulosic biomass is abundant in the environment, which can be used as the substrate for methane production via anaerobic digestion (AD). However, the methane production efficiency from lignocellulosic biomass is relatively low, mainly due to its refractory components, of which lignin is the major barrier to bioconversion and also resists the hydrolysis of lignocellulose, thus limiting the AD process. This study compared the effects of two different gases (N2 and CO2) nanobubble water (NBW) on methane production from lignin using mono-digestion and co-digestion with acetic acid (HAc) at four different total organic carbon (TOC) ratios of HAc to lignin (97.5%:2.5%, 95%:5%, 90%:10%, and 80%:20%). Results show that N2-NBW is the promising NBW which can enhance methane production by 22% compared to the control. In addition, co-digestion of lignin with acetic acid reflects a higher methane production potential in comparison with the mono-digestion of lignin, achieving a much higher methane production (824–1061 mL CH4/g-TOCremoved) when lignin content was less than 20% (TOC basis) and the highest lignin reduction (43%) at the TOC ratio of HAc to lignin of 95%:5%.

Chronic effects of benzalkonium chlorides on short chain fatty acids and methane production in semi-continuous anaerobic digestion of waste activated sludge

Abstract:As an emerging pollutant, benzalkonium chlorides (BACs) potentially enriched in waste activated sludge (WAS). However, the microbial response mechanism under chronic effects of BACs on acidogenesis and methanogenesis in anaerobic digestion (AD) has not been clearly disclosed. This study investigated the AD (by-)products and microbial evolution under low to high BACs concentrations from bioreactor startup to steady running. It was found that BACs can lead to an increase of WAS hydrolysis and fermentation, but a disturbance to acidogenic bacteria also occurred at low BACs concentration. A noticeable inhibition to methanogenesis occurred when BAC concentration was up to 15 mg/g TSS. Metagenomic analysis revealed the key genes involved in acetic acid (HAc) biosynthesis (i.e. phosphate acetyltransferase, PTA), β-oxidation pathway (acetyl-CoA C-acetyltransferase) and propionic acid (HPr) conversion was slightly promoted compared with control. Furthermore, BACs inhibited the acetotrophic methanogenesis (i.e. acetyl-CoA synthetase), especially BAC concentration was up to 15 mg/g TSS, thereby enhanced short chain fatty acids (SCFAs) accumulation. Overall, chronic stimulation of functional microorganisms with increasing concentrations of BACs impact WAS fermentation.

Abstract:Huge amounts of wastewater that contain aromatic compounds such as benzene and phenols are discharged worldwide. Benzoate is a typical intermediate in the anaerobic transformation of those aromatic compounds. In this study, electrically conductive carbon-based materials of granulated activated carbon (GAC), multiwalled carbon nanotubes (MwCNTs), and graphite were evaluated for the ability to promote the benzoate degradation. The results showed that 82-93% of the electrons were recovered in CH4 production from benzoate. The carbon materials stimulated benzoate degradation in the sequence of GAC (5 g/L) > MwCNTs (1 g/L) similar to Graphite (0.1 g/L) > Control. Acetate was the only detected intermediate in the process of benzoate degradation. Taxonomic analyses revealed that benzoate was degraded by Syntrophus to acetate and H-2, which were subsequently converted to methane by Methanosarcina (both acetoclastic methanogens and hydrogenotrophic methanogens) and Methanoculleus (hydrogenotrophic methanogens), and direct interspecies electron transfer (DIET) of Desulfovibrio and Methanosarcina. Thus, these results suggest a method to effectively enhance the removal of aromatic compounds and methane recovery. (C) 2020 Elsevier B.V. All rights reserved.

Abstract:Abstract Ligninolytic bacteria degrading lignin were isolates and identified, and their biodegradation mechanism of alkaline-lignin was investigated. Four strains with lignin degradation capability were screened and identified from the soil, straw, and silage based on their decolorizing capacity of aniline blue and colony size on alkaline-lignin medium. The degradation ratio of Bacillus aryabhattai BY5, Acinetobacter johnsonii LN2, Acinetobacter lwoffii LN4, and Micrococcus yunnanensis CL32 have been assayed using alkaline-lignin as the unique carbon source. Further, the Lip (lignin peroxidase) and Mnp (manganese peroxidase) activities of strains were investigated. Lip activity of A. lwoffii LN4 was highest after 72 h of incubation and reached 7151.7 U · l –1 . Mnp activity of M. yunnanensis CL32 was highest after 48 h and reached 12533 U · l –1 . The analysis of alkaline-lignin degradation products by GC-MS revealed that the strains screened could utilize aromatic esters compounds such as dibutyl phthalate (DBP), and decomposite monocyclic aromatic compounds through the DBP aerobic metabolic pathway. The results indicate that B. aryabhattai BY5, A. johnsonii LN2, A. lwoffii LN4, and M. yunnanensis CL32 have high potential to degrade alkaline-lignin, and might utilize aromatic compounds by DBP aerobic metabolic pathway in the process of lignin degradation.

Abstract:Quaternary ammonium compounds (QACs) was frequently detected in wastewater treatment plants and leads to potential toxicity to the related biological processes. In this study, the effect of benzalkonium bromide (BK) on anaerobic sludge fermentation process for short chain fatty acid (SCFAs) production was investigated. Batch experiments indicated that BK exposure significantly enhanced the SCFAs production from anaerobic fermentation sludge and the maximum concentration of total SCFAs increased from 474.40 ± 12.35 mg/L to 916.42 ± 20.35 mg/L with BK increasing from 0 to 8.69 mg/g VSS. Mechanism exploration exhibited that the presence of BK enhanced much more bioavailable organic matters release, little affected on hydrolysis, acidification, but seriously inhibited methanogenesis. Microbial community investigation revealed that BK exposure importantly enhanced the relative abundances of hydrolytic-acidifying bacteria and also improved the metabolic pathways and functional genes for sludge lysis. This work further supplement the information for environmental toxicity of emerging pollutants.

Abstract:The biogenic methane production experiment of FOG combined with lignite was performed to enhance the methane output of lignite anaerobic fermentation. Three-dimensional fluorescence, gas chromatography-mass spectrometry, volatile fatty acids and 16S rDNA tests were carried out to analyze the composition of the liquid substances at the biomethane peak production. The results showed that the yield of this compound in the combined fermentation of lignite and FOG could be increased by 380.25 %. Moreover, the degradable organic matter and acetic acid content increased, while the degree of humification decreased. In addition, the microbial activity was enhanced. In the combined substrate of the anaerobic fermentation process, both Acinetobacter and Methanosarcina were activated as the dominant bacteria and archaea, respectively, which improved the degradation efficiency of organic matter and the overall biomethane yield. This research showed that the combined anaerobic fermentation of FOG and lignite could promote the biomethane production efficiency in the context of anaerobic fermentation, which provided a new idea for the clean transformation of lignite and utilization of FOG.

Abstract:Lignin is a complex biopolymer comprising phenolic monomers with different degrees of methoxylation and may potentially enhance the degradation of soil pollutants such as polycyclic aromatic hydrocarbons (PAHs) through co-metabolism. However, the contribution of lignin constituents, including phenolic and methoxy subunits, to PAH biodegradation remains unclear. Here, p -hydroxybenzoate (pHBA), vanillate and methanol were selected to simulate phenolic units and methoxy groups of lignin. Soil microcosms receiving these compounds were established to evaluate their regulation on the bacterial community and PAH co-metabolism. There were different effects of different components on the biodegradation of a four-ring PAH, benzo(a)anthracene (BaA), as characterized using an isotopic tracer. Only vanillate significantly stimulated BaA mineralization to CO 2 , with pHBA and methanol leading to no appreciable change in the allocation of BaA in soil compartments. The lignin constituents had differential impacts on the soil bacterial community, with substantial enrichment of methylotrophs occurring in methanol-supplemented microcosms. Both vanillate and pHBA selected several aromatic degraders. Vanillate caused additional enrichment of methylotrophs, suggesting structure-dependent stimulation of bacterial functional guilds by lignin monomers. Compared with its constituents, lignin produced more extensive responses in terms of bacterial diversity and composition and the fate of BaA. However, it was difficult to link BaA co-metabolism to any specific bacterial taxa in the presence of lignin or its subunits. The results indicate that the co-metabolism effects of lignin may not be directly associated with phenolic or methoxy metabolism but with its regulation of the soil microbiome.

Abstract:This study explores the feasibility of using lignosulfonate, a byproduct of the pulp and paper industry, to facilitate sludge anaerobic digestion. Biochemical methane potential assays revealed that the maximum methane production was achieved at 60 mg/g volatile solids (VS) lignosulfonate, 22.18 % higher than the control. One substrate model demonstrated that 60 mg/g VS lignosulfonate boosted the hydrolysis rate, biochemical methane potential, and degradation extent of secondary sludge by 19.12 %, 21.87 %, and 21.11 %, respectively, compared to the control. Mechanisms unveiled that lignosulfonate destroyed sludge stability, promoted organic matter release, and enhanced subsequent hydrolysis, acidification, and methanogenesis by up to 31.30 %, 74.42 % and 28.16 %, respectively. Phytotoxicity assays confirmed that lignosulfonate promoted seed germination and root development of lettuce and Chinese cabbage, with seed germination index reaching 170 ± 10 % and 220 ± 22 %, respectively. The findings suggest that lignosulfonate addition offers a sustainable approach to sludge treatment, guiding effective management practices.

Abstract:Introduction: Ligninolytic bacteria can secrete extracellular enzymes to depolymerize lignin into small-molecular aromatics that are subsequently metabolized and funneled into the TCA cycle. Carbohydrates, which are the preferred carbon sources of bacteria, influence the metabolism of lignin-derived aromatics through bacteria. Methods: In this study, untargeted metabolomics and transcriptomics analyses were performed to investigate the effect of carbohydrates on lignin degradation mediated by Bacillus amyloliquefaciens MN-13, a strain with lignin-degrading activity that was isolated in our previous work. Results: The results demonstrated that the cell growth of the MN-13 strain and lignin removal were promoted when carbohydrates such as glucose and sodium carboxymethyl cellulose were added to an alkaline lignin-minimal salt medium (AL-MSM) culture. Metabolomics analysis showed that lignin depolymerization took place outside the cells, and the addition of glucose regulated the uptake and metabolism of lignin-derived monomers and activated the downstream metabolism process in cells. In the transcriptomics analysis, 299 DEGs were screened after 24 h of inoculation in AL-MSM with free glucose and 2 g/L glucose, respectively, accounting for 8.3% of the total amount of annotated genes. These DEGs were primarily assigned to 30 subcategories, including flagellar assembly, the PTS system, RNA degradation, glycolysis/gluconeogenesis, the TCA cycle, pyruvate metabolism, and tryptophan metabolism. These subcategories were closely associated with the cell structure, generation of cellular energy, and precursors for biosynthetic pathways, based on a − log 10 (P adjust) value in the KEGG pathway analysis. Conclusion: In summary, the addition of glucose increased lignin degradation mediated by the MN-13 strain through regulating glycolysis, TCA cycle, and central carbon metabolism.

Abstract:To improve the methanogenic efficiency of lignite anaerobic fermentation and explore innovative approaches to sludge utilization, a co-fermentation technique involving lignite and sludge was employed for converting biomass into biomethane. Volatile suspended solids were introduced as a native enrichment of the sludge and mixed with lignite for fermentation. The synergistic fermentation mechanism between sludge and lignite for biomethane production was analyzed through biochemical methane potential experiments, measurement of various parameters pre- and post-fermentation, observation of bacterial population changes during the peak of reaction, carbon migration assessment, and evaluation of rheological characteristics. The results showed that the addition of sludge in the anaerobic fermentation process improved the microorganisms' ability to degrade lignite and bolstered biomethane production. Notably, the maximum methane production recorded was 215.52 mL/g-volatile suspended solids, achieved at a sludge to coal ratio of 3:1, with a synergistic growth rate of 25.37%. Furthermore, the removal rates of total suspended solids, and total chemical oxygen demand exhibited an upward trend with an increasing percentage of sludge in the mixture. The relative abundance and activity of the methanogens population were found to increase with an appropriate ratio of sludge to lignite. This observation confirmed the migration of carbon between the solid–liquid–gas phases, promoting enhanced system affinity. Additionally, the changes in solid–liquid phase parameters before and after the reaction indicated that the addition of sludge improved the system's degradation capacity. The results of the study hold significant implications in realizing the resource utilization of sludge and lignite while contributing to environmental protection endeavors.

Abstract:Optimization of microbial community is requisite to boost methane production from inferior coal biodegradation. Here is reported an efficient method of converting lignite into methane by microorganisms, which takes lignite as substrate and obtains efficient microbial flora from sludge by means of enrichment and domestication. The Single Factor Experiment and Response Surface Methodology were carried out to regulate the operating conditions and optimize the medium. The results showed that the optimized gas production of biodegradable lignite reached 79.29 mL (54.32% methane)/2 g coal, which is much higher than the other microbial conversions of lignite to methane reported to date. Furthermore, the combined most probable number counting method and high-throughput sequencing technique revealed that the main bacteria in the fermentation process are acid forming bacteria, sulfate-reducing bacteria and methanogenic bacteria. This study provides new insight into methanation and the clean utilization of lignite.

Abstract:In the process of lignin extraction by the organic solvent method, the amount of alkali and the content of 1,4-butanediol are important conditions that affect lignin yield. The effects of alkali and alcohol contents on lignin recovery, removal rate, and structure were studied. In this reaction system, the removal rate of lignin increased with the increase of alkali content but decreased with the increase of alcohol content. Fourier transform infrared (FT-IR) analysis showed that the phenol hydroxyl group and the ether bond in lignin had different trends in different alkali and 1,4-butanediol environments, and four different infrared parameters in lignin had an obvious linear relationship. Gel permeation chromatography (GPC) results showed that high alkali content and high 1,4-butanediol content could lead to the fragmentation of lignin. In addition, lignin extracted from alkali-quantity factor series was selected to prepare activated carbon, CaCl<sub>2</sub> was selected as the activator, and its effects were studied. Results showed that in the process of extracting lignin, on the one hand, NaOH content affects the functional groups of activated carbon by affecting the aromatic structure of lignin; on the other hand, the NaOH content affects the graphitization degree and specific surface area of activated carbon by affecting the removal rate and the molecular weight of lignin.

Abstract:BackgroundLignin is the most abundant aromatic biopolymer in the biosphere and it comprises up to 30% of plant biomass. Although lignin is the most recalcitrant component of the plant cell wall, still there are microorganisms able to decompose it or degrade it. Fungi are recognized as the most widely used microbes for lignin degradation. However, bacteria have also been known to be able to utilize lignin as a carbon or energy source. Bacillus ligniniphilus L1 was selected in this study due to its capability to utilize alkaline lignin as a single carbon or energy source and its excellent ability to survive in extreme environments.ResultsTo investigate the aromatic metabolites of strain L1 decomposing alkaline lignin, GC–MS analysis was performed and fifteen single phenol ring aromatic compounds were identified. The dominant absorption peak included phenylacetic acid, 4-hydroxy-benzoicacid, and vanillic acid with the highest proportion of metabolites resulting in 42%. Comparison proteomic analysis was carried out for further study showed that approximately 1447 kinds of proteins were produced, 141 of which were at least twofold up-regulated with alkaline lignin as the single carbon source. The up-regulated proteins contents different categories in the biological functions of protein including lignin degradation, ABC transport system, environmental response factors, protein synthesis, assembly, etc.ConclusionsGC–MS analysis showed that alkaline lignin degradation of strain L1 produced 15 kinds of aromatic compounds. Comparison proteomic data and metabolic analysis showed that to ensure the degradation of lignin and growth of strain L1, multiple aspects of cells metabolism including transporter, environmental response factors, and protein synthesis were enhanced. Based on genome and proteomic analysis, at least four kinds of lignin degradation pathway might be present in strain L1, including a Gentisate pathway, the benzoic acid pathway and the β-ketoadipate pathway. The study provides an important basis for lignin degradation by bacteria.

Abstract:This study emphasized the synergistic production of bioprocessible lignin and carbohydrates during a sequential liquid hot water and alkali pretreatment of lignocellulose, facilitating their subsequent individual fermentation. Increasing the dose of alkaline lignin from 0 to 8 g/L inhibited cell growth in anaerobic digestion, with varying levels of inhibition observed in the following order: hydrolytic bacteria < acidogens < acetogens. Alkali pretreatment was adapted to maximize yields of bioprocessible lignin liquor without compromising utilization of the carbohydrates. Increasing the NaOH dose from 50 to 200 mg/g-feedstock monotonically improved lignin yields, but further increases in alkali loading led to a decline in lignin recovery. Volatile fatty acids production from anaerobic digestion of the carbohydrate moiety consistently increased with higher NaOH doses. The optimal conditions for maximizing lignin yields were determined to be 105 °C for 30 min, with NaOH loading in the range of 150–200 mg/g-feedstock, resulting in approximately 80 % lignin recovery, of which 35 % was biologically utilizable. Liquid hot water treatment prior to alkali pretreatment was confirmed as necessary to preserve carbohydrates of 0.1 g/g-feedstock at a low temperature of 70 °C. These findings are crucial for economically producing bioprocessible lignin without carbohydrate loss, a key step towards achieving full lignocellulose valorization.

Abstract:The escalating total amount of industrial solid waste is increasingly taxing the ecological environment. Co-combustion technology is seen as a potential solution to this problem. This study investigated the interaction mechanism and carbon dioxide emissions during the co-combustion process of coal slime and biomass (lignin) using a programmed heating method. The experimental results reveal that the two sub-reaction models effectively depict the combustion process of coal slime. In the co-combustion process, the comprehensive combustion and ignition performance of the mixed fuels surpasses that of coal slime, but the stability of the combustion flame is reduced. The combustion performance of the mixed fuel is primarily influenced by two common factors. The interaction between lignin and coal slime significantly changes with the variation of mixing ratio and heating rate. The main product in the combustion process of mixed fuels is carbon dioxide. Due to the competition for oxygen among volatile matter, the emission of carbon dioxide is lower than the theoretical value. When the mass percentage of lignin is 70 %, the impact on carbon dioxide is most pronounced. The experimental results of this study provide a foundation for understanding the interaction mechanism of coal slime and biomass co-combustion. The research findings also provide theoretical guidance for the safe operation of coal slime and biomass co-combustion equipment.

Abstract:Benzethonium chloride (BZC) is viewed as a promising disinfectant and widely applied in daily life. While studies related to its effect on waste activated sludge (WAS) anaerobic fermentation (AF) were seldom mentioned before. To understand how BZC affects AF of WAS, production of short chain fatty acids (SCFAs), characteristics of WAS as well as microbial community were evaluated during AF. Results manifested a dose-specific relationship of dosages between BZC and SCFAs and the optimum yield arrived at 2441.01 mg COD/L with the addition of 0.030 g/g TSS BZC. Spectral results and protein secondary structure variation indicated that BZC denatured proteins in the solid phase into smaller proteins or amino acids with unstable structures. It was also found that BZC could stimulate the extracellular polymeric substances secretion and reduce the surface tension of WAS, leading to the enhancement of solubilization. Beside, BZC promoted the hydrolysis stage (increased by 7.09 % to 0.030 g/g TSS BZC), but inhibited acetogenesis and methanogenesis stages (decreased by 6.85 % and 14.75 % to 0.030 g/g TSS BZC). The microbial community was also regulated by BZC to facilitate the enrichment of hydrolytic and acidizing microorganisms (i.e. Firmicutes ). All these variations caused by BZC were conducive to the accumulation of SCFAs. The findings contributed to investigating the effect of BZC on AF of WAS and provided a new idea for the future study of AF mechanism.

Abstract:Tungstoborate heteropolyacid catalysts have good catalytic degradation performance, especially for selective cleavage of C–C bonds in biomass. In this paper, the product yield and component distribution of tungstoborate heteropolyacid (BW12)-catalyzed lignin liquefaction were investigated at different parameters, including temperatures (120, 140, 160, 180, and 200 °C), catalyst amount (0, 2.5, 5, 10, and 20 wt. %), and reaction time (0, 30, 60, 90, and 120 min). It was found that a higher conversion (72.16 wt. %) and bio-oil yield (68.41 wt. %) could be obtained under suitable reaction conditions (180 °C, 60 min, 5 wt. %). Bio-oil analysis showed that the BW12 catalyst had a significant effect on the distribution of bio-oil fractions, in which mono-aromatic components increased from 32.96% to 47.56% compared to those without the catalyst. In particular, carbonyl substances in the mono-aromatic components increased from 18.66% to 26.97%. Spectroscopic analysis (FT-IR) found that the absorption peaks of C–O and C–C bonds in the liquefied residue catalyzed by BW12 decreased compared to the raw lignin. Moreover, the mechanism of BW12-catalyzed lignin depolymerization was investigated by DFT simulations. The simulation results demonstrated that the shortening of Cα–O bond, the breaking of Cβ–Cγ and Cα–Cβ bonds in lignin promoted the formation of vanillin and benzaldehyde, 3-hydroxy-4-methoxy. Finally, based on the experimental data and simulation results, a possible reaction pathway for the BW12-catalyzed liquefaction of lignin into mono-aromatic substances was proposed.
