Abstract:Rabbit,induced ovulation animals,owns vigorous efficiency of reproduction.The traditional reproductive hormones,such as oestrogen and progesterone(receptor) are closely related to the occurrence of oestrous behavior.Reproductive hormones impel signals of rabbit somatic sensation,which activates the central nervous system.Then hypothalamus produces gonadotropins,and later it makes the pituitary gland to emerge luteinizing hormone peak pre-ovulation,inducing rabbit ovulation.Furthermore,recent researchers have found new biological molecules,such as ovarian renin-angiotensin system,induced ovulation factors,participating in the procedure of induced ovulation.Chinning and lordosis are two characteristic estrous behaviors.These different special behaviors are affected by various central neurons.Nevertheless,very little attention was paid by domestic researches to this field.This review made a comprehensive introduction into oestrous behavior with the relationship of reproductive hormones,induced ovulation,and central nervous system,which helps researchers studying on veterinary medicine and human reproductive medicine model.

Abstract:The complexity of processes in the female reproductive system of mammals is extremely sophisticated. The overall relationship between the processes during the oestrus cycle in animals is quite well understood, but the molecular background of these processes still requires an in-depth analysis. Bitches are distinguished by a specific course of sexual cycle during which the oocyte matures after ovulation in the oviduct. Other species of mammals are characterized by maturation of the oocyte within the ovary. Acquisition of developmental competence by cumulus – oocyte complexes seems to be a process with a complex molecular background, and the key to understanding it may be the analysis of intercellular channels. Aquaporins and connexins are structural proteins that are built into the cell membrane. Their location is widespread in many body tissues. Recent years have shown that they exhibit significant expression in different parts of the mammalian reproductive system, although the number of studies on dogs is still negligible. This review paper presents the current state of knowledge of water channels and gap junction connections in different animal species, with particular focus on dogs, and also explores the role of aquaporins and connexins in the acquisition of reproductive competences.

Endometrial receptivity and embryo implantation in carnivores-commonalities and differences with other mammalian species

Abstract:Endometrial receptivity and embryo implantation processes are a major point of pregnancy failure in many mammalian species, including humans. Although reproductive biology in many carnivore species remains enigmatic, the few that have been studied so far are invaluable comparative models. The goals of this review are to (1) summarize current data on the mechanisms involved in uterine receptivity and embryo implantation in carnivores, including commonalities and differences with other mammalian species and (2) identify research priorities to better understand a key phenomenon in a critical group of mammals. Besides unique reproductive traits in some carnivores (induced vs. spontaneous ovulation in cats, ovulation at the germinal vesicle stage in dogs), preimplantation embryo development is comparable with other orders. However, the timing of implantation varies, especially in species having an embryonic diapause. Mechanisms involved in endometrial receptivity and decidualization still remain to be fully understood, but specific markers have already been identified. Importantly, the use of endogenous hormones to control the ovarian activity may impact endometrial receptivity and subsequent embryo implantation. Next, research efforts should take advantage of advanced technologies to further study embryo implantation in carnivores and to provide more relevant models to reproductive medicine or for the conservation of rare and endangered species.

Abstract:Domestic dogs are monoestrous, typically non-seasonal, polytocous, spontaneous ovulators and have a spontaneous luteal phase slightly longer (by approx 5 day) than the 64±1day luteal phases of a 65±1day pregnancy, a phase followed by an obligate anestrus before the next 2-3 week "heat" (proestrus-estrus). The resulting inter-estrus intervals of 5-12 months are variable among bitches, commonly 6-7 months, and range from highly variable to regular (to perhaps within±5-10 day of sequential 7 month cycle, for instance) within bitches, and across studies and do not vary significantly between pregnant and non-pregnant cycles. Hormone levels reported are those observed in this laboratory using previously reported assays and canine gonadotropin standards unless stated otherwise. Endocrine sequences for dog cycles are not unlike those of many other mammals, including selection of ovulatory follicles by increased LH pulsatility, the occurrence of estrus behavior and LH surge during a decline in the estrogen: progestin ratio, a pronounced preovulatory luteinization as in humans and rodents, and luteotrophic roles for both LH and prolactin. Non-pregnant bitches have a spontaneously prolonged luteal phase, often longer and with a more protracted decline in serum progesterone than in pregnancy as there is no uterine luteolytic mechanism. The obligate anestrus of 8-40 weeks is terminated by poorly understood interactions of environment (e.g. pheromones, possibly photoperiod) and a potential endogenous circannual cycle in sensitivities of hypothalamic dopaminergic, serotonergic and/or opioid pathways.

Abstract:The domestic dog is a non-seasonal breeder. The female is monestrus and has a reproductive cycle that is marked by extended periods of proestrus and estrus. The estrous period is characterized by an estrogen peak that coincides with rising circulating progesterone concentration prior to ovulation. After estrus is diestrus and then anestrus, with the ovarian cycle regulated by the hypothalamic-pituitary-gonadal axis. In the male spermatogenesis is controlled by the hypothalamic-pituitary-gonadal axis with testosterone playing a vital role. The reproductive cycle of most wild canids is similar to that of domestic dogs. Diestrus is followed by an extended period of ovarian inactivity. The causes of breeding failure in dogs are numerous and require thorough investigations for accurate diagnosis. Breeding failures can be infectious or non-infectious. Among the infectious causes in the bitch, bacterial endometritis was found to be responsible for the majority of reported cases. Non-infectious causes include primary and secondary anoestrus, cystic conditions of the uterus, and degenerative diseases of the endometrium. In the male, the causes can also be infectious and non-infectious. Non-infectious causes like bilateral cryptorchidism and acquired anatomical abnormalities can also cause male infertility. Spermatocele or sperm granulomas, obstruction of the genital ducts or inguinal or scrotal hernia, and prostatitis lead to infertility. Infections can lead to orchitis/epididymitis, with alteration of the quality of the semen. Nutrition also has important implications for reproductive performance. Undernutrition can result in loss of body condition, delay the onset of puberty, and ultimately lead to infertility.

Abstract:"The mammary gland is a modified sweat gland specialized in milk secretion, whose development is synchronized with the sexual function, being directly influenced by the stages of the sexual cycle in both canids and felines. Knowledge of morphology and physiology is essential for a correct approach of the mammary diseases, an important pathology affecting two categories: the adult female and the newborns. There are numerous controversies in the literature regarding the terminology and duration of the dog and cat sexual cycle, leading to the existent variability of the information. However, for the bitch it is unanimously accepted that most canine breeds have two estrous periods per year, usually in spring and autumn. This period can be systematically divided into proestrus, estrus, luteal phase and anestrus. In this species, the proestrus and estrus periods are long and necessarily accompanied by a long luteal phase. These are then followed by anestrus, which is not influenced by the season, unlike the cat. The queen is a seasonally polyestrous animal, which in the absence of mating goes into estrus every 2-3 weeks throughout the breeding season. She will have repeated cycles in the breeding season, which will only be interrupted by gestation, pseudo-gestation or various ailments. The aim of this review is to describe the estrus cycle in dogs and cats, and its connection with mammary gland development, a gland with unique growth due to the fact that the final stage of development is reached in the adult female only during gestation."

Abstract:Reproductive biology is a uniquely important topic since it is about germ cells, which are central for transmitting genetic information from generation to generation. In this review, we discuss recent advances in mammalian germ cell development, including preimplantation development, fetal germ cell development and postnatal development of oocytes and sperm. We also discuss the etiologies of female and male infertility and describe the emerging technologies for studying reproductive biology such as gene editing and single-cell technologies.
Ovarian and uterine changes during the oestrous cycle in female dogs

Abstract:Context: An accurate staging of sexual cycle is essential for the optimum timing of medical interventions. Aims: Here, an updated insight into clinical, endocrinological and vagino-cytological parameters, and their correlation with histomorphology of ovarian and uterine tissue samples is presented. Methods: Samples from 39 dogs were collected at various stages of the oestrous cycle: pro-oestrus ( n = 8), oestrus ( n = 12), dioestrus ( n = 9) (luteal phase) and anoestrus ( n = 10), according to clinical observations. Final allocation of samples was done after histomorphological evaluation of all tissues. Peripheral oestradiol-17ß (E2) and progesterone (P4) concentrations were measured, P4 by both chemiluminescent immunoassay (CLIA) and radioimmunoassay (RIA). Key results: Differences were observed between determination of the stage of the oestrous cycle, either by clinical, endocrinological or histomorphological evaluation. Individuals considered to be in clinical and endocrinological oestrus, had entered the luteal phase according to histomorphology. P4 concentrations measured by two different assays differed, underlying the importance to understand that absolute P4 concentrations may deviate depending on the used assay. Comparison of E2 and P4 concentrations is suggested to be useful when defining the transition from early follicular phase to the time of ovulation. Conclusions and implications: Based on parallel histomorphological observations, combined with clinical and endocrinological findings on the same individuals, the present study emphasises that an accurate classification of the stage of the cycle in female dogs based solely on clinical and endocrinological assessments can be difficult. The histomorphological findings presented herein provide new insights into the transitional phases between the different stages of the oestrous cycle in the dog.
Accomplishment of canine cloning through in vitro matured oocytes: a pioneering milestone

Abstract:The in vitro maturation (IVM) rate of canine oocytes remains low compared to other mammals due to their unique reproductive characteristics. This study aimed to explore the effect of hormone supplementation during the IVM of canine immature oocytes on nuclear maturation and subsequently assess its potential application in canine somatic cell nuclear transfer (SCNT). Immature oocytes were collected and cultured in an IVM medium supplemented with hormones (follicle-stimulating hormone [FSH] and progesterone [P4]) or without hormones (control) for 24 hours. The maturation rates of oocytes in the hormone-treated group (94.92 ± 3.15%) were significantly higher than those in the control group (61.01 ± 4.23%). Both in vitro and in vivo matured oocytes underwent NT to evaluate their utility, and the fusion rates were higher in the in vitro matured group than those in the vivo matured group, not significant between in vivo and in vitro matured group (73.28% and 82.35%, respectively). As a result, 14 fused embryos from the in vitro matured group were transferred into two surrogates, with one surrogate achieving a successful pregnancy and delivering four puppies. Whereas in the in vivo matured group, 85 fused embryos were transferred to 8 surrogate mothers, leading to three surrogates becoming pregnant and delivering one, four, and two puppies. The pregnancy rates were not significant between both groups (50% and 37.50%), but the number of offspring exhibited a significant difference (28.57% and 8.23%). In conclusion, we achieved a remarkable milestone by successfully producing cloned puppies using in vitro matured oocytes, underscoring the feasibility of canine cloning from in vitro recovered oocytes. It is important to note that this study focused only on immature oocytes after ovulation and only during the estrus stage. Further research targeting other stages of the estrous cycle could potentially enhance canine cloning efficiency.
Influence of estrous stages on electrocardiography, clinical pathology and ovarian weight of experimental beagle dogs: a retrospective analysis

Abstract:Abstract Estrous cycle is a repetitive phenomenon occurring during the reproductive life of a female dog. The duration of the canine estrous cycle is considerably longer than one in the most of the other animals and is broadly grouped into follicular phase (proestrus and estrus), luteal phase (diestrus) and non-seasonal anestrus. Dogs in the same stage of cycle can be inadvertently assigned to same group during routine safety and metabolic studies leading to possible erroneous interpretation of test-item related effects. This retrospective analysis was conducted by analyzing data of 86 female beagle dogs from control/placebo treated groups to correlate any possible effect of estrous stages with electrocardiography, clinical pathology and ovarian weight. Different estrous cycle stages of beagles were confirmed histologically by evaluating ovary, uterus, vagina and mammary glands. The incidence of beagles in diestrus was the highest, followed by anestrus, proestrus and estrus. No significant effect was noticed on heart rate, P–A, P–D, RR, QRS and QT intervals across different stages of estrous cycle. However, significantly higher PQ (PR) interval in dogs in proestrus stage was observed compared to dogs in anestrus and estrus. Marginally higher WBCs, neutrophils, lymphocytes, RBCs, hemoglobin, AST and lower hematocrit, lipid profile (total cholesterol, HDL, LDL, triglycerides), ALP level was evident in estrous period. Relative ovary weight was significantly higher in dogs in diestrus stage. Considering these results, one may need to exercise caution while interpreting experimental data from female beagle dogs.

Abstract:Over the last 10 years, new drugs have been applied to canine reproduction, widening the spectrum of therapeutic possibilities for diseases that were previously surgically treated, and facilitating better control of the estrous cycle and fertility. Some are not approved for use in dogs; their use is experimental and further clinical trials are necessary. Dopamine agonists such as cabergoline, bromocriptine or metergoline are ergoderivative alkaloids that exert an anti-prolactinergic effect via stimulation of D2 pituitary receptors or inhibition of central serotoninergic ones. Their main indication is suppression of lactation. Anti-prolactinergic compounds have also been successfully used for pregnancy termination and shortening of interestrous intervals. Anti-progestins, (e.g. mifepristone and aglepristone) are synthetic steroids that bind with high affinity to progesterone (P4) receptors, preventing P4 from exerting its biological effects. Anti-progestins have been indicated in P4-dependent conditions, such as pregnancy termination, induction of parturition and the medical treatment of pyometra. Several groups of drugs have been described to have anti-androgenic properties through different mechanisms of action: progestins, receptor binding anti-androgens (e.g. flutamide), competitive enzyme inhibitors (e.g. finasteride), aromatase inhibitors, and GnRH agonists. Their main application is medical treatment of benign prostatic hyperplasia. Long-term release formulations of GnRH agonists (e.g. leuprolide or deslorelin acetate) postponed puberty and reversibly suppressed reproductive function in male and female dogs for periods exceeding 1 year. Anti-estrogens (e.g. clomiphene and tamoxifen citrate) are synthetic non-steroidal type I anti-estrogenic compounds that competitively block estrogen receptors with a combined antagonist-agonistic effect. In dogs, their action is more agonistic than antagonistic.

Abstract:Five female beagles were examined periodically throughout their second biannual cycle of proestrus, estrus, metestrus, and anestrus for fluctuations in several physiological and behavioral charateristics. Physiological measures included plasma levels of estradiol and progesterone as well as the vulvar swelling and vaginal bleeding that begin with proestrus and continue in estrus. Behavioral tests revealed changes in female receptivity, attractivity, and proceptivity. Estradiol increased prior to the onset of proestrus and declined during estrus. Progesterone levels were low during most of proestrus, increased from the beginning to the end of estrus, and remained elevated during the first month or more of metestrus. While they were in proestrous females were not receptive but they and their vaginal secretions were highly attractive to males. Attractivity remained high throughout estrus and declined abruptly in the next 24 hr. Receptivity increased over the first 3 days of estrus and continued at a high level until the last 3 days during which it decreased slightly, and then dropped nearly to zero within the next 24 hr. “Sexual reflexes” of the vulva, tail, and hindquarters followed the same course of changes described for attractivity. Proceptive behavior, including seeking proximity to caged males and display of solicitation responses during mating tests was characteristic of females during proestrus and estrus but uncommon or absent in other phases of the cycle. It is hypothesized that in the natural cycle, attractivity and proceptivity develop during proestrus as a consequence of rapidly increasing secretion of estrogen. The onset of receptivity is due to synergistic action of estrogen and progesterone. Estrogen secreted during proestrus primes the system, and progesterone secreted just before and during estrus activates it. Termination of receptivity is thought to be due to the marked decline in estrogen, to inhibition by high concentrations of progesterone, or to a combination of these factors.

A retrospective view of early research on dominance, stress and reproduction in cooperatively breeding carnivores

Abstract:Social carnivores have been central in studies of cooperative breeding, and research using noninvasive methods to examine behavioral and endocrine mechanisms of reproductive suppression started in the 1980s with dwarf mongooses in Serengeti National Park. Here, I synthesize the methods, findings and limitations of a research program that examined relationships between social dominance, age, mass, aggression, mating, gonadal steroids, glucocorticoids and reproduction in female and male dwarf mongooses, African wild dogs and wolves. Infanticide is a reliable backstop for reproductive suppression in females, and reproduction is energetically costly in these species. These conditions favor hypothalamic - pituitary - gonadal (HPG) adaptations that reduce the fertility of subordinate females to avoid the cost of producing doomed offspring. Infanticide also favors close synchronization of reproduction when subordinate females do become pregnant. In males, infanticide is a less reliable backstop and reproduction is less costly, so direct effects of subordination on fertility are less pronounced. Age is a strong predictor of social dominance in these species, but the evolutionary reason for this is not clear. In dwarf mongooses and wild dogs, alpha females were never deposed by younger packmates, but alpha males were: this difference is also not understood. Patterns of reproduction supported models predicting that alphas are less likely to share reproduction when the fitness costs of reproduction are high, when the fitness expected for dispersers is low, and with young subordinates to whom they are more closely related. Correlations between dominance and adrenal glucocorticoid concentrations varied between species and sexes, but did not support the hypothesis that chronic stress causes reproductive suppression.

Abstract:From an economic standpoint, reproductive characteristics are fundamental for sustainable production, particularly for monotocous livestock like cattle. A longer inter-calving interval is indicative of low reproductive capacity. This issue changes the dynamics of current and future lactations since it necessitates more inseminations, veterinary care, and hormone interventions. Various reproductive phenotypes, including ovulation, mating, fertility, pregnancy, embryonic growth, and calving-related traits, are observed in dairy cattle, and these traits have been associated with several QTLs. Calving ease, age at puberty, scrotal circumference, and inseminations per conception have been associated with 4437, 10623, 10498, and 2476 Quantitative Trait Loci (QTLs), respectively. This data offers valuable insights into enhancing and comprehending reproductive traits in livestock breeding. Studying QTLs associated with reproductive traits has far-reaching implications across various fields, from agriculture and animal husbandry to human health, evolutionary biology, and conservation. It provides the foundation for informed breeding practices, advances in biotechnology, and a deeper understanding of the genetic underpinnings of reproduction.

Abstract:Understanding seasonal breeding dynamics is essential for maximizing efficiency and welfare in the application of street dog management programs. Humane Society International (HSI) has conducted many animal birth control programmes concerning the street dog populations in urban India. This paper analyses the data on reproductive indicators—oestrus, pregnancy, and pups—collected by HSI sterilization clinics in the cities of Jamshedpur, Dehradun, and Vadodara over a period of 3, 5, and 4 years, respectively. We found a consistent reproductive seasonality dynamic in all three cities, with peak oestrus and pregnancies occurring in the late/post-monsoon season. Pup proportions peaked soon after. Both these findings are consistent with previous studies of free-roaming domestic dog populations both in India and worldwide. Additionally, we identified minor inter-city differences in the temporal breeding dynamic, which we propose are the result of localized seasonal climatic and human factors. Finally, we examine and assert the relevance of breeding seasonality in the implementation of efficient and welfare-sensitive birth control programmes.

Abstract:18 Annual peaks in reproductive activity have been identified in multiple domestic dog populations. 19 However, there is little evidence to describe how these peaks may be associated with environmental 20 factors such as daylength, which plays a well-established role in breeding patterns of seasonally-21 reproductive species. 22 Data were collected from 2016  2020 during 7,743 and 4,681 neuter surgeries on adult female 23 unowned free-roaming dogs in veterinary clinics in Goa and Tamil Nadu respectively. Temperature, 24 precipitation, relative humidity, and daylength data were gathered for time periods preceding the 25 neuter surgery that may have influenced the likelihood of pregnancy (potential influence periods). A 26 multivariable generalised additive model was used to assess the relationship between these factors 27 and pregnancy. 28 The prevalence of pregnancy varied by month in both locations indicating seasonality of 29 reproduction in these groups. The annual pattern was more distinct in Goa with a peak in 30 pregnancies between September and December. In Goa, decreasing daylength was associated with a 31 higher probability of pregnancy ( p = 0.040). Decreasing temperature was associated with decreasing 32 probability of pregnancy in the Nilgiris ( p = 0.034). Bitches had a median of 6 foetuses, with no 33 evidence of seasonal variation.

Integrated Analysis of mRNAs and Long Non-Coding RNAs Expression of Oviduct That Provides Novel Insights into the Prolificacy Mechanism of Goat (Capra hircus)

Abstract:Artificial directional selection has replaced natural selection and resulted in trait differences across breeds in domestic animal breeding. However, the molecular mechanism by which the oviduct regulates litter size remains largely elusive in goats during the follicular phase. Accumulating data have linked lncRNAs to reproductive activities; however, little is known about the modulation mechanism in the oviduct. Herein, RNA-seq was used to measure mRNA and lncRNA expression levels in low- and high-fecundity goats. We observed distinctive differences in mRNA and lncRNA in terms of different kidding numbers and detected the differential expression of 1640 mRNA transcripts and 271 lncRNA transcripts. Enrichment analysis of differentially expressed mRNAs (DEGs) suggested that multiple pathways, such as the AMPK, PI3K–Akt, calcium signaling pathway, oocyte meiosis, ABC transporter, and ECM–receptor interaction pathways, directly or indirectly affected goat reproduction. Additionally, coexpression of differentially expressed lncRNAs (DEL)-genes analysis showed that XLOC_021615, XLOC_119780, and XLOC_076450 were trans-acting as the DEGs ATAD2, DEPDC5, and TRPM6, respectively, and could regulate embryo development. Moreover, XLOC_020079, XLOC_107361, XLOC_169844, XLOC_252348 were the trans-regulated elements of the DEGs ARHGEF2 and RAPGEF6, and the target DEGs CPEB3 of XLOC_089239, XLOC_090063, XLOC_107409, XLOC_153574, XLOC_211271, XLOC_251687 were associated with prolificacy. Collectively, our study has offered a thorough dissection of the oviduct lncRNA and mRNA landscapes in goats. These results could serve as potential targets of the oviduct affecting fertility in goats.
