Cellulose Nanowhiskers from Moso Bamboo Residues: Extraction and Characterization

Abstract:To take full advantage of moso bamboo processing waste, bamboo cellulose nanowhiskers were isolated from bamboo residues using sulfuric acid hydrolysis. Changes in bamboo cellulose at different stages of processing, as well as the roles of acid concentrations (55 wt% and 65 wt%) and hydrolysis times (1 h to 5 h) on the characteristics of nanowhiskers were investigated by scanning electron microscopy (SEM), transmission electron microscopy (TEM), thermogravimetry-Fourier transform infrared spectroscopy analysis (TGA-FTIR), synchrotron radiation wide angle X-ray scattering (WAXS), composition analysis, and Brunauer-Emmett Teller (BET) analysis. Both rod-like and network-like nanowhiskers were observed. Alkaline pretreatment removed impurities and part of the hemicellulose. Cellulose content increased to nearly 85%, and specific surface area improved as well after bleaching. Nanowhiskers had an average length of 455 nm, diameter of 12 nm, and an aspect ratio of about 37. Cellulose I was the dominant composition in bamboo cellulose; the transformation of cellulose Iα to cellulose Iβ was observed. Nanowhiskers presented greater crystallinity and crystallite size than those of cellulose without hydrolysis, but lower thermal stability. These bionanowhiskers might be used as reinforcements in nanocomposites.
Extraction and Characterization of Phenolic Compounds from Bamboo Shoot Shell Under Optimized Ultrasonic-Assisted Conditions: a Potential Source of Nutraceutical Compounds

Abstract:The extraction and characterization of bioactive compounds from bamboo shoot shell was performed under ultrasonic-assisted extraction (UAE) along with UPLC-Triple-TOF/MS analysis. Under optimized UAE conditions (ethanol concentration (58%), liquid to solid ratio (24 mL/g), extraction temperature (59 °C), and sonication time (29 min)), total phenolic content (TPC) was recorded highest as 85.3 mg GAE/g DW, while DPPH and FRAP antioxidant activities were 77 μmol AAE/g DW and 33 μmol AAE/g DW, respectively. Using UPLC-Triple-TOF/MS analysis, fifteen phenolic acids, seven flavonoids, nineteen organic acids, two iridoid glucosides, and one neoglinan were identified. Among them, p -coumaric acid (119 μg/g DW), chlorogenic acid (87 μg/g DW), rutin (39 μg/g DW), and ferulic acid (17 μg/g DW) were found to be the most abundant phenolic compounds. Furthermore, compounds including phenolic acids (3 -p- coumaroylquinic acid, cryptochlorogenic acid, 3 -O- feruloylquinic acid, 5 -O- feruloylquinic acid, 3 -O- caffeoylshikimic acid, 5 -p- coumaroylquinic acid, 1,3-dicaffeoyl quinic acid, 3,5-dicaffeoyl quinic acid), flavonoids (schaftoside/c-hexosyl-c-pentosylapigenin, apigenin 6,8-di-C-α- l -arabinopyranoside, kaempferide 3 -O- α- l -rhamnopyranosyl(1->6)-β- d -glucopyranoside, 6-C-β- d -glucopyranosyl-8-C-α- l -arabinopyranosylchrysin, narcissin), some organic acids, iridoid glucosides, and neolignan were also detected for the first time from bamboo shoot shell extract. The present study revealed that bamboo shoot shell extract contains a larger number of nutraceutically active compounds which can be further utilized by food and nutraceutical industries.
Isolation, Identification and Determination of Six Nucleosides and Two Amino Acids from Bamboo Shoots of Gramineae Phyllostachys Prominens (W Y Xiong)

Abstract:Purpose: To develop a method to identify and quantify the compounds in the shoots of four Phyllostachys bamboo species (Gramineae Phyllostachys prominens W. Y. Xiong, Gramineae Phyllostachys iridescins C. Y. Yao Gramineae Phyllostachys pubescens (Carr.) Mitford, Gramineae Phyllostachys praecox C. D. Chu et C. S. Chao.).Methods: The compounds in bamboo shoots were isolated and identified by ultraviolet (UV) spectroscopy, mass spectrometry (MS), and nuclear magnetic resonance (NMR). Quantitative analysis was performed by reversed-phase high performance liquid chromatography (RP-HPLC) using a C18 column and a mixture (1: 1ratio) of acetonitrile and 15 mM ammonium acetate (pH 6.0) as mobile phase. This method was validated for its reproducibility, chemical stability, and recovery.Results: Six nucleosides and two amino acids were isolated from bamboo shoots, including guanosine, 2'-deoxyguanosine, adenosine, thymidine, uridine, cytidine, tryptophan, and phenylalanine. The HPLC method was rapid and reproducible. The intraday and interday concentrations of the eight identified compounds showed good linearity in the range of 0.22 - 60.00 mu g/mL. The relative standard deviation (RSD) for intraday and interday precision for reproducibility and stability was < 3 %. The validated method was successfully applied to determine the content of the eight compounds in four different Phyllostachys species.Conclusion: Adenosine was isolated from bamboo shoots previously, but the isolation of the other seven compounds are reported here for the first time. The method proposed is sensitive and reproducible, and would facilitate studies of nutritional/medicinal compounds in bamboo shoot.

Abstract:Interest in the antioxidant activity of bamboo leaves is growing. To discover new sources of natural antioxidants, a TLC bioautography method combined with a new image processing method was developed to evaluate the antioxidant activity of leaf extracts from 15 different species of bamboo. The results showed that the methanolic extract of Bambusa. textilis McClure possessed the highest antioxidant activity among the selected bamboo species. To rapidly identify the antioxidant compounds, the crude extract of B. textilis McClure was analysed by HPLC-UV, and HPLC-micro-fractionation of the extract was carried out. Based on TLC bioautography-guided fractionation, three antioxidant fractions were isolated from B. textilis McClure by preparative chromatography. These three antioxidant compounds were identified as isoorientin 4''-O β-D-xylopyranoside (1), isoorientin 2''-O-α-L-rhamnoside (2) and isoorientin (3) according to their UV, MS, and NMR data. The proposed TLC screening method could therefore be an easy way to evaluate the antioxidant activity of bamboo leaves, and the results achieved should prove very helpful for promoting their utilization, as B. textilis McClure can be considered a promising plant source of natural antioxidants.
Switching Carbon Metabolic Flux for Enhancing the Production of Sesquiterpene-Based High-Density Biofuel Precursor in Saccharomyces Cerevisiae

Abstract:Sesquiterpenes are designated as a large class of plant-derived natural active compounds, which have wide applications in industries of energy, food, cosmetics, medicine and agriculture. Neither plant extraction nor chemical synthesis can meet the massive market demands and sustainable development goals. Biosynthesis in microbial cell factories represents an eco-friendly and high-efficient way. Among several microorganisms, Saccharomyces cerevisiae exhibited the potential as a chassis for bioproduction of various sesquiterpenes due to its native mevalonate pathway. However, its inefficient nature limits biosynthesis of diverse sesquiterpenes at industrial grade. Herein, we exploited an artificial synthetic malonic acid-acetoacetyl-CoA (MAAC) metabolic pathway to switch central carbon metabolic flux for stable and efficient biosynthesis of sesquiterpene-based high-density biofuel precursor in S. cerevisiae. Through investigations at transcription and metabolism levels, we revealed that strains with rewired central metabolism can devote more sugars to β-caryophyllene production. By optimizing the MVA pathway, the yield of β-caryophyllene from YQ-4 was 25.8 mg/L, which was 3 times higher than that of the initial strain YQ-1. Strain YQ-7 was obtained by introducing malonic acid metabolic pathway. Combing the optimized flask fermentation process, the target production boosted by about 13-fold, to 328 mg/L compared to that in the strain YQ-4 without malonic acid metabolic pathway. This designed MAAC pathway for sesquiterpene-based high-density biofuel precursor synthesis can provide an impressive cornerstone for achieving a sustainable production of renewable fuels.

Abstract:The present study investigated a robust method for the preparation of four flavone C-glycosides, i.e. orientin, homoorientin, vitexin and isovitexin, which were prepared from an ethanol aqueous extract, i.e. antioxidant of bamboo leaves (AOB), by AB-8 resin-based column chromatography and preparative high-performance liquid chromatography (HPLC) using a mobile phase consisting of 10% and 15% (v/v) of acetonitrile and 1% acetic acid. These flavone C-glycosides were further purified by the drowning-out crystallization method using methanol and water as drowning-out anti-solvents and salting-out agents, respectively. The purity was assessed by analytical HPLC and the confirmation of chemical structures was performed by IR, MS, NMR and UV spectroscopy. Orientin (49mg), homoorientin (142mg), vitexin (15mg) and isovitexin (62mg) were prepared from 6.5g of crude column chromatography fraction obtained from 5L of AOB concentrated solution. The present method is robust and suitable for preparing available quantities of pure flavone C-glycosides and the quantification of orientin, homoorientin, vitexin and isovitexin in bamboo leaves.

Abstract:The metabolism of flavone C-glucosides and p-coumaric acid from antioxidant of bamboo leaves (AOB) in rats is discussed systematically in the present study. Following single oral administration of AOB, p-coumaric acid was detected in plasma but not in gastrointestinal tract extracts and faeces, and the corresponding absorption pharmacokinetic curve at different time points showed a prolonged elimination phase with p-coumaric acid being detected in the kidneys and excreted as its original form (1.80(SD 0.24)% and 1.90 (SD 0.26) % at 12 and 24 h, respectively). However, the four flavone C-glucosides orientin, homoorientin, vitexin and isovitexin were poorly absorbed in the gastrointestinal tract. More than 50 % recovery of flavone C-glucosides was determined at 12h and faeces containing these four analytes (2123 (SD 1.92)%) were excreted at 24h. These data suggested that the effective time these compounds were in the colon was long enough so that they could exert their antioxidant activity and scavenge free radicals. Besides the excretion of the original forms, moieties of the flavone C-glucosides were hydrolysed by deglycosylation and the opening of the heterocyclic C ring. Some small molecules such as phloroglucinol (PG), hydrocaffeic acid (HCA) and phloretic acid (PA) were detected and identified as metabolites of the flavone C-glucosides. In the present work, we compared the metabolic fate of flavone C-glucosides to that of flavone O-glucosides in rats, and evaluated the absorption, tissue distribution and excretion of flavone C-glucosides in AOB on their metabolism for the first time.
Efficient Delignification of Sugarcane Bagasse by Fenton Oxidation Coupled with Ultrasound-Assisted NaOH for Biotransformation from Agaricus Sinodeliciosus Var. Chaidam

Abstract:Sugarcane bagasse (SCB), a widely available lignocellulosic by-product, could be potentially used in macrofungal bioconversion for high-value bioactive compounds. For macrofungus with poor lignin degradation capacity, its recalcitrant lignin structure blocked the release of fermentable sugars for cell growth. Herein, the main objective was to construct an effective delignification pretreatment strategy for SCB biotransformation and synthesis of bioactive substances by Agaricus sinodeliciosus var. Chaidam. Different pretreatments including chemical (alkali and Fenton oxidation) and ultrasound methods were used to intensify the delignification process. Physico-chemical characteristics of native and pretreated SCB were examined via XRD, XPS, FT-IR and C-13 ssNMR to characterize crystallinity index, surface lignin and chemical structure. Lignocellulose-degrading enzymes were also used to assess the ability of the macrofungus to degrade SCB. By comparing morphological structure, chemical composition and fermentation characteristics, a suitable multiple sequential pretreatment method was finally identified. Our findings revealed that Fenton-ultrasound-alkali pre-treatment has an optimum delignification effect on SCB with never-uncovered excellent ability to detoxify harmful substances produced by Fenton oxidation. Under this pretreatment condition, the cellulose content was increased by 106.45%, whereas hemicelluloses and lignin were decreased by 48.34% and 74.15%, respectively. The macrofungal growth, intracellular polysaccharides, total phenols and terpenoids formation were 2.4, 1.46, 1.36 and 1.66 times higher than those of the untreated, respectively. Fermentation kinetic results showed outstanding fit and prediction of macrofungal growth and enzymatic activity secretion on above pretreated SCB. This study shed light on developing a novel pretreatment strategy for low-cost SCB lignocellulose transformation into high-value products.
Development of a co-culture system for green production of caffeic acid from sugarcane bagasse hydrolysate

Abstract:Caffeic acid (CA) is a phenolic acid compound widely used in pharmaceutical and food applications. However, the efficient synthesis of CA is usually limited by the resources of individual microbial platforms. Here, a cross-kingdom microbial consortium was developed to synthesize CA from sugarcane bagasse hydrolysate using Escherichia coli and Candida glycerinogenes as chassis. In the upstream E. coli module, shikimate accumulation was improved by intensifying the shikimate synthesis pathway and blocking shikimate metabolism to provide precursors for the downstream CA synthesis module. In the downstream C. glycerinogenes module, conversion of p -coumaric acid to CA was improved by increasing the supply of the cytoplasmic cofactor FAD(H 2 ). Further, overexpression of ABC transporter-related genes promoted efflux of CA and enhanced strain resistance to CA, significantly increasing CA titer from 103.8 mg/L to 346.5 mg/L. Subsequently, optimization of the inoculation ratio of strains SA-Ec4 and CA-Cg27 in this cross-kingdom microbial consortium resulted in an increase in CA titer to 871.9 mg/L, which was 151.6% higher compared to the monoculture strain CA-Cg27. Ultimately, 2311.6 and 1943.2 mg/L of CA were obtained by optimization of the co-culture system in a 5 L bioreactor using mixed sugar and sugarcane bagasse hydrolysate, respectively, with 17.2-fold and 14.6-fold enhancement compared to the starting strain. The cross-kingdom microbial consortium developed in this study provides a reference for the production of other aromatic compounds from inexpensive raw materials.

Abstract:Orientin and vitexin, important components of bamboo-leaf extracts, are C-glycosylflavones which exhibit a number of interesting biological properties. In this work, we developed an efficient biocatalytic cascade for orientin and vitexin production consisting of Trollius chinensis C-glycosyltransferase (TcCGT) and Glycine max sucrose synthase (GmSUS). In order to relieve the bottleneck of the biocatalytic cascade, the biocatalytic efficiency, reaction condition compatibilities and the ratio of the enzymes were determined. We found that the specific activity of TcCGT was significantly influenced by enzyme dose and Triton X-100 or Tween 20 (0.2%). Coculture of BL21-TcCGT-Co and BL21-GmSUS-Co affected the catalytic efficiency of TcCGT and GmSUS, and the maximum orientin production rate reached 47 mu M/min at the inoculation ratio of 9:1. The optimal pH and temperature for the biocatalytic cascade were pH 7.5 and 30 degrees C, respectively. Moreover, the high dose of the enzymes can improve the tolerance of biocatalytic cascade to substrate inhibition in the one-pot reaction. By using a fed-batch strategy, maximal titers of orientin and vitexin reached 7090 mg/L with a corresponding molar conversion of 98.7% and 5050 mg/L with a corresponding molar conversion of 97.3%, respectively, which is the highest titer reported to date. Therefore, the method described herein for efficient production of orientin and vitexin by modulating catalytic efficiencies of enzymes can be widely used for the C-glycosylation of flavonoids.
Lignocellulosic biomass-derived carbon quantum dots (CQDs): A novel approach utilizing organosolv lignin from Moso bamboo waste

Abstract:Converting lignocellulosic materials to carbon quantum dots (CQDs) has garnered significant interest to transform biomass wastes into highly valuable products. We chose to use lignin as our feedstock because high-value products more readily utilize cellulose and hemicellulose. Lignin, arguably the most underutilized and complex biopolymer within any biomass, presents an appealing yet overlooked opportunity. While some may argue that lignin-based CQDs are no longer groundbreaking, this is because existing studies typically rely on commercially available lignin, ignoring biomass-derived sources. Therefore, in our pioneering research, we utilized organosolv lignin, prized for its structural purity and extracted from Moso bamboo waste, as the carbon source in a facile two-step technique to synthesize luminous CQDs. The process involved the addition of 2,4-diaminobenzenesulfonic acid, a mild-organic acid that acted as a source for both nitrogen (N) and sulphur (S). A 12-hour hydrothermal treatment at 200 °C with 0.1 M of acid addition attained the best result. The resultant CQDs display strong green fluorescence, with the highest quantum yield of 17.7% seen in N and S co-doped CQDs. With their obvious and specific fluorescence quenching impact on Fe 3+ , CQDs demonstrate great potential as a fluorescent nanoprobe for metal ion detection. This CQD nanoprobe responds very sensitively to Fe 3+ between 0 and 500 μM (R 2 = 0.9803), with a detection limit of 0.15 μM. This research not only presents a unique alternative nanoprobe for the sensing field using renewable biomass lignin, but it also paves the way for the long-term, cost-effective, and scalable manufacturing of CQDs.

Caffeoylquinic Acids with Potential Biological Activity from Plant In vitro Cultures as Alternative Sources of Valuable Natural Products

Abstract:Background: For a long time, the researchers have been looking for new efficient methods to enhance production and obtain valuable plant secondary metabolites, which would contribute to the protection of the natural environment through the preservation of various plant species, often rare and endangered. These possibilities offer plant in vitro cultures which can be performed under strictly-controlled conditions, regardless of the season or climate and environmental factors. Biotechnological methods are promising strategies for obtaining the valuable plant secondary metabolites with various classes of chemical compounds including caffeoylquinic acids (CQAs) and their derivatives. CQAs have been found in many plant species which are components in the daily diet and exhibit a wide spectrum of biological activities, including antioxidant, immunomodulatory, antihypertensive, analgesic, anti-inflammatory, hepato- and neuroprotective, anti-hyperglycemic, anticancer, antiviral and antimicrobial activities. They have also been found to offer protection against Alzheimer's disease, and play a role in weight reduction and lipid metabolism control, as well as modulating the activity of glucose-6-phosphatase involved in glucose metabolism. Methods: This work presents the review of the recent advances in use in vitro cultures of various plant species for the alternative system to the production of CQAs and their derivatives. Production of the secondary metabolites in in vitro culture is usually performed with cell suspension or organ cultures, such as shoots and adventitious or transformed roots. To achieve high production of valuable secondary metabolites in in vitro cultures, the optimization of the culture condition is necessary with respect to both biomass accumulation and metabolite content. The optimization of the culture conditions can be achieved by choosing the type of medium, growth regulators or growth conditions, selection of high-productivity lines or culture period, supplementation of the culture medium with precursors or elicitor treatments. Cultivation for large-scale in bioreactors and genetic engineering: Agrobacterium rhizogenes transformation and expression improvement of transcriptional factor or genes involved in the secondary metabolite production pathway are also efficient strategies for enhancement of the valuable secondary metabolites. Results: Many studies have been reported to obtain highly productive plant in vitro cultures with respect to CQAs. Among these valuable secondary metabolites, the most abundant compound accumulated in in vitro cultures was 5-CQA (chlorogenic acid). Highly productive cultures with respect to this phenolic acid were Leonurus sibiricus AtPAP1 transgenic roots, Lonicera macranthoides and Eucomia ulmoides cell suspension cultures which accumulated above 20 mg g-1 DW 5-CQA. It is known that di- and triCQAs are less common in plants than monoCQAs, but it was also possible to obtain them by biotechnological methods. Conclusion: The results indicate that the various in vitro cultures of different plant species can be a profitable approach for the production of CQAs. In particular, an efficient production of these valuable compounds is possible by Lonicera macranthoides and Eucomia ulmoides cell suspension cultures, Leonurus sibiricus transformed roots and AtPAP1 transgenic roots, Echinacea angustifolia adventitious shoots, Rhaponticum carthamoides transformed plants, Lavandula viridis shoots, Sausera involucrata cell suspension and Cichorium intybus transformed roots.

Abstract:Plant in vitro cultures can be an effective tool in obtaining desired specialized metabolites. The purpose of this study was to evaluate the effect of light-emitting diodes (LEDs) on phenolic compounds in Rhaponticum carthamoides shoots cultured in vitro. R. carthamoides is an endemic and medicinal plant at risk of extinction due to the massive harvesting of its roots and rhizomes from the natural environment. The shoots were cultured on an agar-solidified and liquid-agitated Murashige and Skoog's medium supplemented with 0.1 mg/L of indole-3-acetic acid (IAA) and 0.5 mg/L of 6-benzyladenine (BA). The effect of the medium and different treatments of LED lights (blue (BL), red (RL), white (WL), and a combination of red and blue (R:BL; 7:3)) on R. carthamoides shoot growth and its biosynthetic potential was observed. Medium type and the duration of LED light exposure did not affect the proliferation rate of shoots, but they altered the shoot morphology and specialized metabolite accumulation. The liquid medium and BL light were the most beneficial for the caffeoylquinic acid derivatives (CQAs) production, shoot growth, and biomass increment. The liquid medium and BL light enhanced the content of the sum of all identified CQAs (6 mg/g DW) about three-fold compared to WL light and control, fluorescent lamps. HPLC-UV analysis confirmed that chlorogenic acid (5-CQA) was the primary compound in shoot extracts regardless of the type of culture and the light conditions (1.19–3.25 mg/g DW), with the highest level under R:BL light. BL and RL lights were equally effective. The abundant component was also 3,5-di-O-caffeoylquinic acid, accompanied by 4,5-di-O-caffeoylquinic acid, a tentatively identified dicaffeoylquinic acid derivative, and a tricaffeoylquinic acid derivative 2, the contents of which depended on the LED light conditions.

Abstract:Abstract C -glycosylated flavones (CGFs) are promising candidates as anti-nociceptive compounds. The leaves of bamboo and related crops in the grass family are a largely unexploited bioresource with a wide array of CGFs. We report here pathway-specific enzymes including C -glycosyltransferases (CGTs) and P450 hydroxylases from cereal crops and bamboo species accumulating abundant CGFs. Mining of CGTs and engineering of P450s that decorate the flavonoid skeleton allowed the production of desired CGFs (with yield of 20–40 mg/L) in an Escherichia coli cell factory. We further explored the antinociceptive activity of major CGFs in mice models and identified isoorientin as the most potent, with both neuroanalgesic and anti-inflammatory effects superior to clinical drugs such as rotundine and aspirin. Our discovery of the pain-alleviating flavonoids elicited from bamboo and crop leaves establishes this previously underutilized source, and sheds light on the pathway and pharmacological mechanisms of the compounds.

Abstract:The substantial increase of the atmospheric CO2 concentration over the last few decades has steered toward research on direct air capture (DAC). DAC is one of the mobile-source carbon capture and storage (CCS) processes that is variant from the conventional point-source CCS in terms of easing the CO2 concentration levels and promoting negative carbon emission. We propose here a sustainable material based on the quaternization of bamboo fiber and the implication in DAC. The quaternized bamboo cellulose could adsorb CO2 at a medium relative humidity (RH) of 60-80%, with the efficiency of quaternary ammonium groups over 0.65. The CO2 capacity at higher RH strikingly decreased, which indicates the moisture swing characteristics, and that was further validated by a desorption ratio of 0.70 under humid gas purge. The unique phenomenon that a dry atmosphere (e.g., RH < 20%) is not favored by adsorption was revealed and could be attributed to the hydrophobic feature of the sorbent inherited from natural lignocellulose. The results suggest that quaternized cellulose with a low cost may open up new possibilities for the utilization of moisture-swing CO2 adsorbents in damp circumstances.

Abstract:The microalgae Haematococcus pluvialis is a commercial source of natural astaxanthin. However, mature cells develop rigid three-layer wall structures and a repulsive odor. This study applied a liquid static fermentation system to screen hydrolyzing microorganisms for cell wall hydrolysis. Baijiu jiuqu and Gutian hongqu were found to have promising potential for application. The fermentation using 2% baijiu jiuqu and 2% glucose for pre-activation achieved comparable recovery of carotenoids to homogenizer disruption methods and produced stable fragrance which may be attributed to ethyl octanoate, hexyl formate, and phenethyl butyrate, as revealed by gas chromatography-mass spectrometry analysis. The abundance of astaxanthin molecules was slightly affected by fermentation with fold change 10) were mainly carbohydrates, lipids, and steroids proving the safety of the fermentation. This study provides a new scheme for the biorefining of Haematococcus. pluvialis , potentially contributing to the industrial production of natural astaxanthin.

Abstract:A green strategy that using Bacillus velezensis to degrade cellulose in bamboo forest waste was demonstrated. The cultivation conditions of microorganism, identified as Bacillus velezensis by 16S rDNA, were optimized by response surface methodology (RSM). Results showed that with a temperature of 35 °C, a pH of 6 in a Luria-Bertani (LB) medium, and a rotation speed of 140 r/min, the bamboo forest waste powder was added to the purified bacteria at a dose of 1 % (w/v), the cellulose degradation rate reached the highest value of 33.12 %. Besides, the fourier-transform infrared (FTIR), X-ray diffraction (XRD), scanning electron microscope (SEM) and liquid chromatography-mass spectrometry (LC-MS) indicated that cellulose could be effectively degraded, with possible products including cellobiose, uridine diphosphate (UDP)-glucose, D-glucuronic acid, etc. The metabolic pathways involved in cellulose degradation by this strain include the pentose phosphate, starch, sucrose, and glucuronic acid pathways.

Abstract:Abstract Background Due to the increasing demands of energy and depletion of fossil fuel, bamboo is considered to be one of the most important renewable biological resources on the basis of its advantages of rapid growth ability and rich reserves. Cellulose, hemicellulose, and lignin are the three most important constituents in moso bamboo. Their concentrations and, especially, their microscopic distributions greatly affect their utilization efficiency and other physical properties as a biomass resource. However, no studies have achieved a quantitative visualization of the distribution of lignocellulose concentrations in transverse sections of bamboo. Therefore, this study proposed the use of quantitative multivariate spectral analysis to reveal the micro-chemical distribution of lignocelluloses in bamboo based on an integration of FTIR macro- and micro-spectroscopic imaging techniques. Results Multivariate calibration models for the quantitative determination of lignocelluloses of bamboo were developed based on FTIR macro-spectroscopy, and the quantitative calibration models based on the FTIR characteristic bands showed an excellent performance with determination coefficients of 0.933, 0.878, and 0.912 for cellulose, hemicellulose, and lignin, respectively. These quantitative models were then utilized to the FTIR micro-spectroscopy of bamboo transverse sections which were corrected using a direct standardization algorithm. Subsequently, the micro-chemical distributions of cellulose, hemicellulose, and lignin were obtained based on the integration of the multivariate calibration models and corrected FTIR micro-spectroscopy. The combination of the multivariate calibration models and calibration transfer algorithm resulted in a final quantitative visualization of the chemical distributions of lignocelluloses in moso bamboos. Conclusions Integration of the FTIR macro- and micro-spectroscopic imaging techniques can provide comprehensive information that can be used to exploit the resource of moso bamboo to develop biofuels and biosynthetic materials.
