Abstract:In light of environmental apprehension, supercritical fluid technology (SFT) exhibits excellent opportunities to accomplish key objectives in the drug delivery sector. Supercritical fluid extraction using carbon dioxide (CO(2)) has been recognized as a green technology. It is a clean and versatile solvent with gas-like diffusivity and liquid-like density in the supercritical phase, which has provided an excellent alternative to the use of chemical solvents. The present commentary provides an overview of different techniques using supercritical fluids and their future opportunity for the drug delivery industry. Some of the emerging applications of SFT in pharmaceuticals, such as particle design, drug solubilization, inclusion complex, polymer impregnation, polymorphism, drug extraction process, and analysis, are also covered in this review. The data collection methods are based on the recent literature related to drug delivery systems using SFT platforms. SFT has become a much more versatile and environmentally attractive technology that can handle a variety of complicated problems in pharmaceuticals. This cutting-edge technology is growing predominantly to surrogate conventional unit operations in relevance to the pharmaceutical production process. Lay abstract: Supercritical fluid technology has recently drawn attention in the field of pharmaceuticals. It is a distinct conception that utilizes the solvent properties of supercritical fluids above their critical temperature and pressure, where they exhibit both liquid-like and gas-like properties, which can enable many pharmaceutical applications. For example, the liquid-like properties provide benefits in extraction processes of organic solvents or impurities, drug solubilization, and polymer plasticization, and the gas-like features facilitate mass transfer processes. It has become a much more versatile and environmentally attractive technology that can handle a variety of complicated problems in pharmaceuticals. This review is focused on different techniques that use supercritical fluids and their opportunities for the pharmaceutical sector.

Abstract:During the past few decades, supercritical fluid (SCF) has emerged as an effective alternative for many traditional pharmaceutical manufacturing processes. Operating active pharmaceutical ingredients (APIs) alone or in combination with various biodegradable polymeric carriers in high-pressure conditions provides enhanced features with respect to their physical properties such as bioavailability enhancement, is of relevance to the application of SCF in the pharmaceutical industry. Herein, recent advances in drug delivery systems manufactured using the SCF technology are reviewed. We provide a brief description of the history, principle, and various preparation methods involved in the SCF technology. Next, we aim to give a brief overview, which provides an emphasis and discussion of recent reports using supercritical carbon dioxide (SC-CO2 ) for fabrication of polymeric carriers, for applications in areas related to drug delivery, tissue engineering, bio-imaging, and other biomedical applications. We finally summarize with perspectives.

Abstract:This article reviews the use of fluids under supercritical conditions in processes related to the modern and innovative polymer industry. The most important processes using supercritical fluids are: extraction, particle formation, micronization, encapsulation, impregnation, polymerization and foaming. This review article briefly describes and characterizes the individual processes, with a focus on extraction, micronization, particle formation and encapsulation. The methods mentioned focus on modifications in the scope of conducting processes in a more ecological manner and showing higher quality efficiency. Nowadays, due to the growing trend of ecological solutions in the chemical industry, we see more and more advanced technological solutions. Less toxic fluids under supercritical conditions can be used as an ecological alternative to organic solvents widely used in the polymer industry. The use of supercritical conditions to conduct these processes creates new opportunities for obtaining materials and products with specialized applications, in particular in the medical, pharmacological, cosmetic and food industries, based on substances of natural sources. The considerations contained in this article are intended to increase the awareness of the need to change the existing techniques. In particular, the importance of using supercritical fluids in more industrial methods and for the development of already known processes, as well as creating new solutions with their use, should be emphasized.

Abstract:In the pharmaceutical industry, supercritical fluid (SCF), particularly supercritical carbon dioxide (SC-CO 2 ), can be utilized as a solvent, extractant, liquid expander, plasticizer, and aerosol. SCF-based particle formation technology, an environmentally friendly technology, has been used in the field of drug development to overcome solubility/dissolution issues of poorly water-soluble drugs and/or improve bioavailability without changing pharmacological activity. However, these technologies have limitations, such as operation in batch or semi-continuous mode and particle collection, and expert suggestions are required to resolve these issues.

Abstract:The rapid advances in the development of formulation and delivery systems based on micron-sized and nanoscale drug particles will create significant benefits to the pharmaceutical industry. Complementary to traditional methods, supercritical fluid techniques have found many useful, and sometimes unique, applications in the production and processing of drug particles. In this article background information is provided on a variety of supercritical fluid techniques relevant to drug formulation and delivery, recent advances and novel applications are highlighted, and the successful development of a new supercritical fluid rapid expansion technique for producing exclusively nanoscale drug particles will be discussed. Challenges and opportunities for further development and future applications are also reviewed.

Abstract:Long a thermodynamic curiosity, supercritical fluids (SCFs) have gradually gained ground in today's life, generating an increasing number of new, efficient processes in diverse industrial sectors and fueling active R&D programs. Indeed, the versatility of SCFs allows them to serve a wide variety of applications. The list includes not only food processing, biofuel production, extraction of biomolecules marketable as medicines, cosmetics and nutraceuticals, but also emerging technologies for the production of electrical power, based on supercritical or transcritical thermodynamic cycles. This jointly authored article will provide a review of important applications covered by our laboratories in the agri-food, chemical and energy sectors. We will then try to detect recent trends and outline future prospects.

Abstract:The properties of the supercritical fluid near the critical point can be changed by varying the pressure and temperature, allowing selective and faster extraction. Supercritical fluids exhibit liquid-like density and gas – like viscosity & diffusion coefficients and can penetrate more into the solid matrix inaccessible to liquids due to negligible surface tension and viscosity. They are suitable as a substitute for organic solvents in a range of industrial and laboratory processes. These properties of supercritical fluids are well suited for the extraction of solutes, from various plants, fruits, flowers, seeds, leaves, which cannot be easily extracted using conventional extraction processes. In view of above mentioned advantages, supercritical fluid extraction is becoming increasingly popular in many industries such as petroleum, chemical, food, and perfumery. Supercritical fluids are also used for the extraction of metal ions from aqueous solutions and solid and liquid matrices. This review paper provides a concise review of the applications of supercritical fluids from the perspective of feed materials, type of supercritical fluids, co solvents and modifiers used, nature of work, operating conditions, findings, limitations of the work done and scope for further research. The application of supercritical fluids in extraction and purification of various natural extracts and others, using CO2 accounts for more than 90% of the published research in the field of supercritical fluid technology. Other supercritical fluids being used are propane, hexane and butane. Carbon dioxide is a relatively non-polar solvent but has some limited affinity with polar molecules due to its large molecular quadrupole. Co-solvents and modifiers are often be added to improve the solubility of polar molecules. On addition of various co solvents and modifier such as isopropyl alcohol and ethanol, n-hexane, heptane, pentane, toluene, methanol, acetone, formic acid with ammonium formate in methanol etc enhances the performance of supercritical extraction process. Despite this, the status of supercritical fluid technology commercialization is less than satisfactory. A review studies revealed that most of the published research is at laboratory scale with little or no information on scale up and design strategies.

Abstract:This chapter starts with the current status quo of using supercritical fluids (SCFs) technology in the pharmaceutical industry, followed by the application of SCFs as solvents of precipitation for active pharmaceutical ingredients (APIs), particularly those poor, aqueous soluble API. The fundamentals of various particle formation approaches with SCFs acting as “solvents” is discussed. Operating parameters such as thermodynamic parameters and processing parameters that can affect the precipitation of APIs are also discussed and summarized to give the mass an idea on the importance of chemical and process engineering aspects in optimizing the operating conditions to facilitate a possible scale-up in the future. Limitations of the various particle formation approaches and recent modifications on the apparatus setup are deciphered to highlight the potential of SCFs as solvents in the pharmaceutical industry in the near future.

Abstract:Nanotechnology plays a pivotal role in the biomedical field, especially in the synthesis and regulation of drug particle size. Reducing drug particles to the micron or nanometer scale can enhance bioavailability. Supercritical fluid technology, as a green drug development strategy, is expected to resolve the challenges of thermal degradation, uneven particle size, and organic solvent residue faced by traditional methods such as milling and crystallization. This paper provides an insight into the application of super-stable homogeneous intermix formulating technology (SHIFT) and super-table pure-nanomedicine formulation technology (SPFT) developed based on supercritical fluids for drug dispersion and micronization. These technologies significantly enhance the solubility and permeability of hydrophobic drugs by controlling the particle size and morphology, and the modified drugs show excellent therapeutic efficacy in the treatment of hepatocellular carcinoma, pathological scarring, and corneal neovascularization, and their performance and efficacy are highlighted when administered through multiple routes of administration. Overall, supercritical fluids have opened a green and efficient pathway for clinical drug development, which is expected to reduce side effects and enhance therapeutic efficacy.

Abstract:Various active compounds isolated from natural sources exhibit remarkable benefits, making them attractive for pharmaceutical and biomedical applications, such as antioxidant, antimicrobial, and anti-inflammatory activities, which contribute to the treatment of cardiovascular diseases, neurodegenerative disorders, various types of cancer, diabetes, and obesity. However, their major drawbacks are their reactivity, instability, relatively poor water solubility, and consequently low bioavailability. Synthetic drugs often face similar challenges associated with inadequate solubility or burst release in gastrointestinal media, despite being otherwise a safe and effective option for the treatment of numerous diseases. Therefore, drug-eluting pharmaceutical formulations have been of great importance over the years in efforts to improve the bioavailability of active compounds by increasing their solubility and achieving their controlled release in body media. This review highlights the success of the fabrication of micro- and nanoformulations using environmentally friendly supercritical fluid technologies for the processing and incorporation of active compounds. Several novel approaches, namely micronization to produce micro- and nano-sized particles, supercritical drying to produce aerogels, supercritical foaming, and supercritical solvent impregnation, are described in detail, along with the currently available drug delivery data for these formulations.

These include optical transparency, small volume, rapid reaction, and precise manipulation of fluids, making microfluidics a versatile tool for process optimization and fundamental studies of extraction and reaction kinetics in supercritical CO2 applications. Moreover, the small length scale of microfluidics allows for the production of uniform nanoparticles with reduced particle size, beneficial for nanomaterial synthesis. In this perspective, we review microfluidic investigations involving supercritical CO2, with a particular focus on three primary applications, namely, solvent extraction, nanoparticle synthesis, and chemical reactions. We provide a summary of the experimental innovations, key mechanisms, and principle findings from these microfluidic studies, aiming to spark further interest. Finally, we conclude this review with some discussion on the future perspectives in this field.

Abstract:Supercritical fluid technologies offer a propitious method for drug discovery from natural sources. Such methods require relatively short processing times, produce extracts with little or no organic co-solvent, and are able to extract bioactive molecules whilst minimising degradation. Supercritical fluid extraction (SFE) provides a range of benefits, as well as offering routes to overcome some of the limitations that exist with the conventional methods of extraction. Unfortunately, SFE-based methods are not without their own shortcomings; two major ones being: (1) the high establishment cost; and (2) the selective solvent nature of CO₂, i.e., that CO₂ only dissolves small non-polar molecules, although this can be viewed as a positive outcome provided bioactive molecules are extracted during solvent-based SFE. This review provides an update of SFE methods for natural products and outlines the main operating parameters for extract recovery. Selected processing considerations are presented regarding supercritical fluids and the development and application of ultrasonic-assisted SFE methods, as well as providing some of the key aspects of SFE scalability.

Abstract:The unique properties of supercritical fluids, in particular supercritical carbon dioxide (CO2), provide numerous opportunities for the development of processes for pharmaceutical applications. One of the potential applications for pharmaceuticals includes microencapsulation and nanoencapsulation for drug delivery purposes. Supercritical CO2 processes allow the design and control of particle size, as well as drug loading by utilizing the tunable properties of supercritical CO2 at different operating conditions (flow ratio, temperature, pressures, etc.). This review aims to provide a comprehensive overview of the processes and techniques using supercritical fluid processing based on the supercritical properties, the role of supercritical carbon dioxide during the process, and the mechanism of formulation production for each process discussed. The considerations for equipment configurations to achieve the various processes described and the mechanisms behind the representative processes such as RESS (rapid expansion of supercritical solutions), SAS (supercritical antisolvent), SFEE (supercritical fluid extraction of emulsions), PGSS (particles from gas-saturated solutions), drying, and polymer foaming will be explained via schematic representation. More recent developments such as fluidized bed coating using supercritical CO2 as the fluidizing and drying medium, the supercritical CO2 spray drying of aqueous solutions, as well as the production of microporous drug releasing devices via foaming, will be highlighted in this review. Development and strategies to control and optimize the particle morphology, drug loading, and yield from the major processes will also be discussed.

Abstract:The present review considers some physicochemical properties of fluid mixtures that are of importance for fluid extraction and supercitical fluid chromatography (SFC). Firstly, the important types of phase diagrams are treated, the occurrence of solid phases also being considered in some simple cases. Specific examples are given of mixtures of a highly volatile component I (e.g. CO2, C2H6) with a relatively involatile component II (e.g. squalane) of very different molecular size, shape, structure, and/or polarity, and it is shown how the rather complicated types of phase diagrams can be calculated and correlated. The importance of fluid mixtures extends far beyond the fields of science and technology reviewed.

Abstract:Supercritical fluid technology has been used for a variety of applications for decades and received special attention in the fields of analytics, extraction and purification of valuable compounds and preparation of drug carriers. Among supercritical fluids, supercritical CO 2 is the most attractive one as being nontoxic, nonflammable and chemically inert. In addition, the critical temperature and pressure of CO 2 is quite moderate which makes supercritical CO 2 a popular solvent for a wide range of applications from laboratory to industrial scale. In recent years, a promising research field called as supercritical microfluidics was developed to merge the advantages of supercritical fluids and microfluidics, the science and technology of manipulating liquids in extremely small volume. This review discusses the recent progress in supercritical microfluidics with a focus on supercritical CO 2 , the most commonly used solvent in supercritical fluid processes, and some applications of supercritical CO 2 in microscale such as extraction of valuable compounds, determination of dissolution characteristic in various solvents, performing chemical reactions and particle synthesis are presented.
