Abstract:Publisher This chapter provides a practical guide to the yeast two-hybrid system, emphasizing recent improvements of this method. There are several different designs for the actual hybrid proteins and selection utilized in two-hybrid screens. The chapter focuses on derivatives of the GAL4-based system. The plasmid encoding the fusion protein with the GAL4 DNA-binding domain is often referred to as the bait plasmid. The basis of the two-hybrid system relies on the structure of particular transcription factors that have two physically separable domains: a DNA binding domain and a transcription activation domain. The DNA-binding domain serves to target the transcription factor to specific promoter sequences, whereas the activation domain serves to facilitate assembly of the transcription complex allowing the initiation of transcription. A schematic representation of the two-hybrid genetic selection by activation of transcription of HIS3 and lacZ genes is shown in the chapter.
